Abstract:Copper (Cu) nanodrugs can be facilely prepared through atom transfer radical polymerization (ATRP) in an aqueous medium. However, it is difficult to control the morphology of Cu nanodrugs and thereby optimize their anticancer activity. In this work, aqueous ATRP was combined with polymerization-induced self-assembly (PISA) to prepare Cu nanodrugs with various morphologies. We mapped the relationship between polymerization condition and product morphology in which each morphology shows a wide preparation window. Decreasing the reaction temperature and feeding more Cu catalysts can improve the mobility of chains, facilitating the morphology evolution from sphere to other high-order morphologies. The resultant Cu nanodrugs with high monomer conversion and high Cu loading efficiency could be easily taken by cancer cells, showing excellent anticancer efficacy in vitro. This work proposed a potential strategy to prepare Cu nanodrugs with a specific morphology in batches, providing the method to optimize the anticancer efficacy through morphology control.

Abstract:In this study, some copper catalysts used for atom transfer radical polymerization (ATRP) were explored as efficient anti-tumor agents. The aqueous solution of copper-containing nanoparticles with uniform spheric morphology was in situ prepared through a copper-catalyzed activator generated by electron transfer (AGET) ATRP in water. Nanoparticles were then directly injected into tumor-bearing mice for antitumor chemotherapy. The copper nanodrugs had prolonged blood circulation time and enhanced accumulation at tumor sites, thus showing potent antitumor activity. This work provides a novel strategy for precise and large-scale preparation of copper nanodrugs with high antitumor activity.

Abstract:Cancer has become a major public health problem, and chemotherapy is one of the main methods for treating cancer. However, traditional anticancer drugs have shortcomings such as poor selectivity, short half-life, and large toxic side effects. Recently, polymeric prodrugs with therapeutic moieties covalently bonding to polymers have attracted considerable interests in anticancer field, which can self-assemble into nanodrugs with improved the durability, targeting and bioavailability comparing to traditional anticancer drugs. Nevertheless, the chemical composition of polymeric prodrugs is complex due to the polydispersity of a polymer, which greatly limits their clinical translation. In order to obtain polymeric prodrugs with accurate chemical composition, we designed and synthesized an amphiphilic supramolecular polymeric prodrug based on host-guest interaction, which can self-assemble into nano-sized micelles in aqueous medium for anticancer chemotherapy. The novel supramolecular polymeric prodrug we report in this work can combine the general advantages of polymeric prodrug and small molecular prodrug, which should have a bright future in clinical chemotherapy.

Abstract:Intravenously administered nanodrugs undergo a number of processes to generate therapeutic effects. The rational design of nanodrugs with high efficiency at every step of the drug deliver process is critical to their therapeutic efficiency. However, many reported nanodrugs that meet these requirements are too complex to prepare for practical use. Herein, we report a simplified and reproducible one step nanodrug fabrication strategy, with only two components, using supramolecular engineering. Di-adamantane-modified camptothecin was precisely synthesized as a prodrug with high purity. An amphiphilic biodegradable block copolymer with pendant ?-cyclodextrin moieties on the hydrophobic block was then synthesized by controlled ring-opening polymerization and ?click? chemistry. The co-self-assembly of the camptothecin prodrug and block copolymer in an aqueous medium, driven by host?guest interactions between the adamantine and ?-cyclodextrin moieties, gave a camptothecin nanodrug with quantitative drug loading content and defined chemical composition. This nanodrug, which consists of a suitable size and stable core-crosslinked structure, has prolonged blood circulation and accumulates efficiently at tumor sites thus showing potent anti-tumor activity. This work provides a strategy to precisely fabricate nanodrugs that is reproducible, and which has great potential for bench-to-bedside translation.
Nanodelivery of a Self-Assembling Prodrug with Exceptionally High Drug Loading Potentiates Chemotherapy Efficacy.

Abstract:Nanomedicines have achieved several successful clinical applications for cancer therapy over the past decades. To date, numerous nanomedicine formats and design rationales have been proposed to improve pharmaceutical delivery and treatment efficacy. Despite these advances, the achievement of high drug loading and loading efficiencies of drug payloads in nanocarriers remains a technical challenge. In addition, study of the correlation between therapeutic potential and drug loading has been ignored. Here, using a self-assembling dimeric cabazitaxel prodrug, we show that the prodrug can be quantitatively entrapped within clinically approved polymer matrices for intravenous injection and that the drug loading in the nanoparticles (NPs) is tunable. The engineered NPs (NPs1-4) with different drug loading values exhibit dissimilar morphologies, release kinetics, in vitro cytotoxic activity, pharmacokinetic properties, tissue distribution, and in vivo anticancer efficacy and safety profiles. Furthermore, the effect of drug loading on the treatment outcomes was explored through detailed in vitro and in vivo studies. Intriguingly, among the constructed NPs, those comprising poly(ethylene glycol)-block-poly(D,L-lactic acid) (PEG-PLA) copolymers showed substantially prolonged pharmacokinetic properties in the blood circulation, which further promoted their intratumoral delivery and accumulation. Furthermore, the PEG-PLA-composed NPs with high drug loading (~50%) demonstrated favorable efficacy and safety profile in animal models. These data provide convincing evidence that the in vivo performance of a given self-assembling drug is not compromised by high drug loading in nanoplatforms, which may potentially reduce concerns over excipient-associated side effects and immunotoxicities. Overall, our study provides new insight into the rationale for designing more effective and less toxic delivery systems.
Size-Controlled Synthesis of Drug-Loaded Zeolitic Imidazolate Framework in Aqueous Solution and Size Effect on Their Cancer Theranostics in Vivo.

Abstract:Recently, metal-organic frameworks (MOFs) or coordination polymers have shown great potential for drug delivery, yet little has been done to study how particle size affects their tumor targeting and other in vivo features. This plight is probably due to two challenges: (1) the lack of a biocompatible method to precisely control the size of drug-loaded MOFs and (2) the lack of a robust and facile radiolabeling technique to trace particles in vivo. Here, we report a one-pot, rapid, and completely aqueous approach that can precisely tune the size of drug-loaded MOF at room temperature. A chelator-free 64Cu-labeled method was developed by taking the advantage of this rapid and aqueous synthesis. Cancer cells were found to take drug-loaded MOFs in a size-dependent manner. The in vivo biodistribution of drug-loaded MOF was analyzed with positron emission tomography imaging, which, as far as we know, was used for the first time to quantitatively evaluate MOF in living animals, unveiling that 60 nm MOF showed longer blood circulation and over 50% higher tumor accumulation than 130 nm MOF. Altogether, this size-controlled method helps to find the optimal size of MOF as a drug carrier and opens new possibilities to construct multifunctional delivery systems for cancer theranostics.

Abstract:Polymeric nanoparticles (NPs) play a key role in nanoscale formulations for bioimaging, cancer treatment and theranostics. In this work, we designed and synthesized a series of hydrophobic polymers (P1-6) with different pendent groups via one-step multicomponent Passerini reaction. These polymers possessed similar molecular structures and various biomedical functions. Interestingly, they could self-assemble into stable NPs in aqueous media. All formed NPs were redox-sensitive because of the existence of disulfide bonds in the backbone. The stability of NPs in aqueous media with or without glutathione was systematically evaluated and compared. The optical performance, including fluorescence resonance energy transfer (FRET), was characterized in different conditions for those polymers with fluorescent components. Importantly, all formed NPs showed good cytocompatibility towards HeLa cells, and different biological functions, including drug loading and delivery, bioimaging with variable fluorescence, and photodynamic activity, as evidenced by experiments in vitro and in vivo. These results demonstrate that the great potential of multicomponent reaction to customize versatile polymeric nanoparticles for biomedical applications.
Facile synthesis of size-tunable stable nanoparticles via click reaction for cancer drug delivery

Abstract:The stability and size of polymeric nanoparticles are two of the most important parameters determining their pharmacokinetics and tumor/drug accumulation efficiency in cancer-drug delivery. Herein, we report a facile one-pot synthesis of crosslinked nanoparticles (CNPs) with tunable sizes and polyethylene glycol (PEG) shells via click reactions. Simply by adjusting the contents of the macromonomer (PEG monoacrylate) in its reaction with ethylene diacrylate and a crosslinker containing hexa-thiols groups, the sizes of the resulting PEGylated crosslinked nanoparticles could be easily tuned from 10 to 90 nm. These nanoparticle cores could encapsulate hydrophobic drugs such as doxorubicin (DOX), and the unreacted thiol and acrylate groups could be used for drug conjugation or labeling. Thus, the nanoparticles provide a multifunctional platform for drug delivery. In vivo studies showed that the larger nanoparticles (about 83.7 nm) had a much longer blood-circulation time and better tumor-targeting efficiency. One of our most important findings was that the drug encapsulated in the crosslinked nanoparticles, even though little was released at pH 7.4 under in vitro conditions, had much faster blood clearance than the nanoparticles’ carrier, suggesting that drug release in the bloodstream was significant.
Particle size tailoring of ursolic acid nanosuspensions for improved anticancer activity by controlled antisolvent precipitation

Abstract:The present study was aimed at tailoring the particle size of ursolic acid (UA) nanosuspension for improved anticancer activity. UA nanosuspensions were prepared by antisolvent precipitation using a four-stream multi-inlet vortex mixer (MIVM) under defined conditions of varying solvent composition, drug feeding concentration or stream flow rate. The resulting products were characterized for particle size and polydispersity. Two of the UA nanosuspensions with mean particle sizes of 100 and 300 nm were further assessed for their in-vitro activity against MCF-7 breast cancer cells using fluorescence microscopy with 4',6-diamidino-2-phenylindole (DAPI) staining, as well as flow cytometry with propidium (PI) staining and with double staining by fluorescein isothiocyanate. It was revealed that the solvent composition, drug feeding concentration and stream flow rate were critical parameters for particle size control of the UA nanosuspensions generated with the MIVM. Specifically, decreasing the UA feeding concentration or increasing the stream flow rate or ethanol content resulted in a reduction of particle size. Excellent reproducibility for nanosuspension production was demonstrated for the 100 and 300 nm UA preparations with a deviation of not more than 5% in particle size from the mean value of three independent batches. Fluorescence microscopy and flow cytometry revealed that these two different sized UA nanosuspensions, particularly the 300 nm sample, exhibited a higher anti-proliferation activity against the MCF-7 cells and afforded a larger population of these cells in both early and late apoptotic phases. In conclusion, MIVM is a robust and pragmatic tool for tailoring the particle size of the UA nanosuspension. Particle size appears to be a critical determinant of the anticancer activity of the UA nanoparticles.
Enzyme/pH Dual-Responsive Size-Shrinkable Nanocomposites for Tumor Penetration and Enhanced Multimodal Cancer Therapy

Abstract:Combination therapy is emerging as a critical strategy in the pursuit of synergistic effects in cancer treatment due to the complicated pathogenesis and rapid progression of tumors. However, the limited tumor accumulation and penetration of nanomedicines within tumors are increasingly recognized as significant barriers to achieving the expected efficacy due to the complex physiological barriers of a solid tumor microenvironment. A size-shrinkable multistage nanocomposite CuS-DOX@MA/Gel was successfully fabricated by conjugating doxorubicin (DOX) onto copper sulfide nanoparticles (CuS NPs) through an imine bond (CuS-DOX), which endowed the system with pH-triggered cargo release. Subsequently, a capping layer composed of metalloproteinase-2 (MMP-2) degradable gelatin and autophagy inhibitor 3-methyladenine (3-MA) was grafted onto CuS-DOX through glutaraldehyde-mediated cross-linking. The size of CuS-DOX@MA/Gel can be effectively reduced from the initial 180.1 nm to small-sized CuS NPs of about 57.2 nm in response to MMP-2. This process of reducing particle size not only facilitated effective retention around the tumor periphery but also significantly enhanced deep penetration into both multicellular spheroids and solid tumors. Simultaneously, the MMP-2-dependent release of 3-MA could block autophagosome formation and augment the sensitivity of chemotherapy. Then, pH sensitivity was confirmed, and CuS-DOX@MA/Gel exhibited proficient drug release in vitro upon exposure to acidic conditions. CuS NPs exhibited the capacity to absorb near-infrared (NIR) light, thereby enabling a sophisticated photodynamic and photothermal therapy that synergistically enhanced chemotherapy. In vivo results demonstrated that CuS-DOX@MA/Gel treated with intravenous administration and an 808 nm laser irradiation achieved excellent tumor inhibition efficacy on 4T1 tumor-bearing mice. This size-adjustable multifunctional strategy should be a potential candidate in the anticancer field due to the outstanding synergistic therapeutic effect.

Abstract:Nanoparticles are widely used in biomedical applications and cancer treatments due to their minute scale, multi-function, and long retention time. Among the various nanoparticles, the unique optical property derived from the localized surface plasmon resonance effect of metallic nanoparticles is a primary reason that metallic nanoparticles are researched and applied. Copper and Iron nanoparticles have the potential to generate hydroxyl radicals in excess H 2 O 2 via Fenton or Fenton-like reactions. On the other hand, gold nanoparticles equipped with a photosensitizer can transfer the energy of photons to chemical energy and enhance the production of singlet oxygen, which is suitable for cancer treatment. With the actions of these two reactive oxygen species in the tumor microenvironment, cell apoptosis can further be induced. In this work, we first synthesized dual metal nanoparticles with poly[styrene-alt-(maleic acid, sodium salt)(Cu ferrite oxide-polymer) by a simple one-step hydrothermal reduction reaction. Then, gold(III) was reduced and doped into the structure, which formed a triple metal structure, Au-doped Cu ferrite nanoparticles (Au/Cu ferrite oxide-polymer NPs). The metal ratio of the product could be controlled by manipulating the Fe/Cu ratio of reactants and the sequence of addition of reactants. The core–shell structure was verified by transmission electron microscopy. Moreover, the hydroxyl radical and singlet oxygen generation ability of Au/Cu ferrite oxide-polymer was proved. The chemodynamic and photodynamic effect was measured, and the in vitro ROS generation was observed. Furthermore, the behavior of endocytosis by cancer cells could be controlled by the magnetic field. The result indicated that Au/Cu ferrite oxide-polymer core–shell nanoreactor is a potential agent for chemodynamic/photodynamic synergetic therapy.

Abstract:A supramolecular approach has been developed for the preparation of supramolecular nanoparticles (SNPs) with variable sizes (30-450 nm) from three different molecular building blocks using a cyclodextrin/adamantane recognition system. Positron emission tomography (PET) was employed to study the biodistribution and lymph node drainage of the SNPs in mice. The sizes of the SNPs affect their in vivo characteristics (see picture).

Abstract:To study the influence of self-assembled morphologies on drug delivery, four different nano-objects, spheres, nanorods, nanowires, and vesicles having aldehdye-based polymer as core, were successfully prepared via alcoholic RAFT dispersion polymerization of p-(methacryloxyethoxy)benzaldehyde (MAEBA) using poly((N,N'-dimethylamino)ethyl methacrylate) (PDMAEMA) as a macro chain transfer agent (macro-CTA) for the first time. The morphologies and sizes of the four nano-objects were characterized by TEM and DLS, and the spheres with average diameter (D) of 70 nm, the nanorods with D of 19 nm and length of 140 nm, and the vesicles with D of 137 nm were used in the subsequent cellular internalization, in vitro release, and intracellular release of the drug. The anticancer drug doxorubicin (DOX) was conjugated onto the core polymers of nano-objects through condensation reaction between aldehyde groups of the PMAEBA with primary amine groups in the DOX. Because the aromatic imine is stable under neutral conditions, but is decomposed in a weakly acidic solution, in vitro release of the DOX from the DOX-loaded nano-objects was investigated in the different acidic solutions. All of the block copolymer nano-objects show very low cytotoxicity to HeLa cells up to the concentration of 1.2 mg/mL, but the DOX-loaded nano-objects reveal different cell viability and their IC50s increase as the following order: nanorods-DOX < vesicles-DOX < spheres-DOX. The IC50 of nanowires-DOX is the biggest among the four nano-objects owing to their too large size to be internalized. Endocytosis tests demonstrate that the internalization of vesicles-DOX by the HeLa cells is faster than that of the nanorods-DOX, and the spheres-DOX are the slowest to internalize among the studied nano-objects. Relatively more nanorods localized in the acidic organelles of the HeLa cells lead to faster intracellular release of the DOX, so the IC50 of nanorods is lower than that of the vesicles-DOX.
Size-Controlled Preparation of Docetaxel- and Curcumin-Loaded Nanoemulsions for Potential Pulmonary Delivery

Abstract:Lung cancer is one of the deadliest pulmonary diseases in the world. Although docetaxel (DTX) has exhibited superior efficacy in lung cancer treatment, it has demonstrated numerous adverse effects and poor bioavailability. The natural product extract, curcumin (CCM), has reportedly reduced toxicity and synergistically improved DTX bioavailability. Nonetheless, the hydrophobic nature of DTX and CCM limits their clinical use. Nanoemulsion pulmonary delivery of DTX and CCM has demonstrated potential as a drug carrier to alleviate these drawbacks. The controlled preparation of inhalable DTX- and CCM-loaded nanoemulsions within the 100 to 200 nm range was explored in this study. A response surface methodology (RSM) based on a central composite design (CCD) was utilized to fabricate the desired size of the nanoemulsion under optimized conditions. Different process parameters were employed to control the size of the nanoemulsions procured through a high-energy emulsification technique. The size of the resultant nanoemulsions decreased with increasing energy input. The actual response according to the targeted sizes for DTX- and CCM-loaded nanoemulsion models exhibited excellent agreement with the predicted value at below 5% residual standard error under optimized conditions. The nanoemulsion of 100 nm particle size demonstrated better membrane permeability than their larger counterparts. Moreover, the formulations documented favorable physicochemical and aerodynamic pulmonary delivery properties and reduced toxicity in human lung fibroblast (MRC-5) cells. Hence, this tunable size of nanoemulsions could be a suitable alternative drug delivery for pulmonary diseases with increased local lung concentration.

Linear-dendritic Drug Conjugates Forming Long-Circulating Nanorods for Cancer-Drug Delivery

Abstract:Elongated micelles have many desirable characteristics for cancer-drug delivery, but they are difficult to obtain since amphiphilic polymers form such nanostructures only within narrow composition ranges depending on their own structures. Herein, we demonstrated a facile fabrication of different nanostructures via drug content-controlled self-assembly of amphiphilic linear-dendritic drug conjugates - using the number of the conjugated hydrophobic drug molecule camptothecin (CPT) to tailor the hydrophobicity of amphiphilic PEG-block-dendritic polylysine-CPT (PEG-xCPT) conjugates and thereby control their self-assembled nanostructures - nanospheres or nanorods of different diameters and lengths. The shape and size of the nanostructures were found to strongly affect their in vitro and in vivo properties, particularly the blood clearance kinetics, biodistribution and tumor targeting. The nanorods with medium lengths (<500 nm) had a much longer blood circulation and faster cellular uptake than the nanospheres or long nanorods. Thus, polymeric nanorods with proper lengths may be ideal nanocarriers capable of uniting the opposite requirements in cancer-drug delivery. (C) 2013 Elsevier Ltd. All rights reserved.

Abstract:The continual generation of drug resistance is a major challenge in clinical cancer chemotherapy. Combination drug therapy may overcome such limitations by minimizing the required dosage of each drug whilst achieving an enhancement of therapeutic efficacy. Regenerated silk fibroin (RSF) is a commonly used drug delivery platform due to its suitable biocompatibility, controllable biodegradability, and diverse material formats. Herein, dual drug-loaded RSF nanospheres were prepared using a facile and green method to deliver two anti-cancer drugs with distinct physical properties, namely hydrophobic paclitaxel (PTX) and hydrophilic doxorubicin (DOX). The particle size of the PTX/DOX-loaded RSF nanospheres was easily regulated, by varying the RSF and ethanol concentrations during the formation process, from approximately 100 to 600 nm, with subsequent applications in both intravenous and lymphatic chemotherapy. The drug release profile of both PTX and DOX in the PTX/DOX-loaded RSF nanospheres was found to be well controlled and sustained for over 7 days. Further, the dual drug-loaded RSF nanospheres exhibited a high cellular uptake via endocytosis. Importantly, the dual drug-loaded RSF nanospheres showed more efficient suppression of cancer cell growth than the single drug-loaded RSF nanospheres with either drug or the free drugs at the same concentration, particularly at a DOX/PTX ratio of 1:1. Thus, the prepared dual drug nanocarrier with a controllable particle size may have important clinical implications for combination chemotherapy.
Hollow Copper Sulfide Nanocubes Loaded with Pt(IV) Complexes for Cancer Multimodal Therapy

Abstract:Chemotherapy (CT) can significantly inhibit tumor growth, metastasis, and recurrence during cancer therapy. People have widely used platinum drugs in cancer treatment. However, as most chemotherapeutic drugs, platinum drugs still have shortcomings such as poor solubility, low cell uptake, nonspecific distribution, multidrug resistance, and adverse side effects. Therefore, we synthesized hollow copper sulfide (CuS) nanocubes with photothermal and photodynamic properties as carriers for Pt(IV) drugs. Hollow CuS nanocubes have attracted considerable interest in the field of cancer photothermal therapy (PTT) using multiple biological windows. Under near-infrared (NIR) laser irradiation, Cu2+ can be reduced into Cu+ in the presence of hydrogen peroxide in the tumor microenvironment. The resulting Cu+ can be used for photodynamic therapy (PDT), which can perform a Fenton-like reaction under acidic conditions (pH 5.5-6.5) and catalyze hydrogen peroxide to produce ·OH in the tumor microenvironment. In addition, compared with Pt(II) drugs, Pt(IV) drugs not only have lower systemic toxicity but also consume glutathione (GSH), thereby increasing reactive oxygen species (ROS) levels in tumor cells and effectively promoting PDT. In this study, we oxidized ethylenediamine platinum chloride to its tetravalent state, loaded the Pt(IV) complexes using hollow CuS nanocubes, and modified the surfaces of the nanoparticles with PEG to improve the EPR effect. The Pt(IV)-loaded hollow CuS nanocubes modified with PEG (Pt(IV)-CuS@PEG) are expected to be used for tumor chemo/photothermal/photodynamic therapy.

Abstract:Poor tumor accumulation remains a serious challenge for nanomedicines to achieve ideal antitumor outcomes. The different size preferences for systematic circulation, tumor retention and deep permeation have attracted great attention when designing antineoplastic nano-delivery system. Herein, we developed a nano-system which can shrink its size in tumor microenvironment to achieve better tumor retention and penetration. In this work, the cationic bovine serum albumin-protected, doxorubicin-loaded gold nanoclusters (CAuNC-DOX) and amino-functionalized mesoporous silica nanoparticles (MSN) are connected by Fe 2+ and are further coated with hyaluronic acid (HA) to obtain a core-satellite nano system (MFADH). When reaching the tumor site, the HA shell, which endows the system with both good biocompatibility and preferable tumor targeting ability, was disintegrated, followed by acid-stimulated release of small-sized pharmacological unit—CAuNC-DOX for further tumor penetration. As demonstrated in both in vitro and in vivo results, MFADH showed excellent antitumor effect, providing a proof of concept for the feasibility of shrinkable nanoplatforms in tumor treatment.

Abstract:In this paper, we studied the synergistic effect of sodium citrate dispersant and polyvinylpyrrolidone or polyvinylalcol capping polymer on controlling the size of copper nanoparticles (CuNPs) that were prepared by the chemical reduction method. The CuNPs were characterised by using ultraviolet–visible (UV–Vis) spectroscopy, transmission electron microscopy, and X-ray diffraction techniques. The size of the nanoparticles could be predicted via their UV–Vis absorbance. In the presence of capping polymers, the size of CuNPs was significantly changed, ranging from 2 ± 1 to 20 ± 7 nm with and without sodium citrate dispersant, respectively. Moreover, surface plasmon resonance of ultrafine CuNPs (2 nm in diameter) could be observed in the short wavelength region by using UV–Vis spectra. Our finding, could offer a new synthetic approach to the production of highly stable, small-sized CuNPs.
