Abstract:Tumors pose a significant threat to human health, and their occurrence and fatality rates are on the rise each year. Accurate tumor diagnosis is crucial in preventing untimely treatment and late-stage metastasis, thereby reducing mortality. To address this, we have developed a novel type of hybrid nanogels called γ-Fe2O3@PNIPAM/PAm/CTS, which contain iron oxide nanoparticles and poly(N-isopropyl acrylamide)/poly(acrylamide)/chitosan. The rational for this study relies on the concept that thermosensitive PNIPAM has the ability to contract when exposed to the elevated temperature conditions found within tumors. This contraction leads to a dense clustering of the high-loading γ-Fe2O3 nanoparticles within the nanogel, thus greatly enhancing the capabilities of MRI imaging. Additionally, the amino groups in chitosan on the particle surface can be converted into ammonium salts under mildly acidic conditions, allowing for an increase in the charge of the nanogel specifically at the slightly acidic tumor site. Consequently, it promotes the phagocytosis of tumor cells and effectively enhances the accumulation and retention of nanogels at the tumor site. The synthesis of the hybrid nanogels involves a surfactant-free emulsion copolymerization process, where vinyl-modified γ-Fe2O3 superparamagnetic nanoparticles are copolymerized with the monomers in the presence of chitosan. We have optimized various reaction parameters to achieve a high loading content of the superparamagnetic nanoparticles, reaching up to 60%. The achieved r2 value of 517.74 mM-1S-1 significantly surpasses that of the clinical imaging contrast agent Resovist (approximately 151 mM-1S-1). To assess the performance of these magnetic nanogels, we conducted experiments using Cal27 oral tumors and 4T1 breast tumors in animal models. The nanogels exhibited temperature- and pH-sensitivity, enabling magnetic targeting and enhancing diagnosis through MRI imaging. The results demonstrated the potential of these hybrid nanogels as contrast agents for magnetic targeting in biomedical applications.

Abstract:Despite advances in surgery and drug discovery, brain tumors remain fatal diseases. Early detection and diagnosis of brain tumors is of great importance for improving treatment outcomes. Magnetic resonance imaging (MRI) is a prominent, clinically-relevant imaging modality because of its excellent tissue contrast resolution, direct multiplanar imaging and increased sensitivity to edema. MRI utility is further enhanced with the use of magnetic iron oxide nanoparticles, which can function as both a contrast agent for imaging and as a drug delivery vehicle for treating brain cancer. In this review, the principles of various imaging modalities for brain tumors are discussed with focus on monocrystalline iron oxide nanoparticle (MION)-based MRI contrast agents. A summary is given on the mechanism of contrast effect, magnetophoretic mobility and magnetic retention, and strategies to enhance tumor selectivity, increase spatial resolution and reduce nonspecific uptake of MION.
An Adaptable Nanoplatform for Integrating Anatomic and Functional Magnetic Resonance Imaging under a 3.0 T Magnetic Field

Abstract:It is well known that acidic tumor microenvironment (TME) is very important for tumor' growth, metastasis, and drug resistance. In addition, accurate diagnosis of the solid tumors' acidic microenviroment based on the precise location of their site is especially essential for the further treatment and prognostic evaluation of cancer. Although magnetic resonance imaging (MRI) is widely used for clinical cancer diagnosis, there are still enormous challenges left for developing MRI agents which integrate anatomic imaging for tumor site location with functional imaging for pH evaluation, especially for medium or high magnetic field machines. Herein, an innovative strategy is proposed to incorporate anatomic imaging (T-1-weighted imaging, T1WI) with functional imaging (susceptibility weighted imaging, SWI) using one 3.0 T MRI scanner on an adaptable nanoplatform namely, NaGdF4@polydopamine@polyethylene glycol (PEG), which can self-aggregate under the specific low pH in TME to realize regulatory susceptibility. Moreover, the nanoplatform performs well in tumor accurately location as well as pH sensitively perception. Most importantly, the strategy not only fuses two MRI models on one nanoplatform using one 3.0 T machine to extend the application range but also provides a new insight for the design of other novel imaging agents.
Ligand‐Mediated Magnetism‐Conversion Nanoprobes for Activatable Ultra‐High Field Magnetic Resonance Imaging

Abstract:Ultra-high field (UHF) magnetic resonance imaging (MRI) has emerged as a focal point of interest in the field of cancer diagnosis. Despite the ability of current paramagnetic or superparamagnetic smart MRI contrast agents to selectively enhance tumor signals in low-field MRI, their effectiveness at UHF remains inadequate due to inherent magnetism. Here, we report a ligand-mediated magnetism-conversion nanoprobe (MCNP) composed of 3-mercaptopropionic acid ligand-coated silver-gadolinium bimetallic nanoparticles. The MCNP exhibits a pH-dependent magnetism conversion from ferromagnetism to diamagnetism, facilitating tunable nanomagnetism for pH-activatable UHF MRI. Under neutral pH, the thiolate (-S-) ligands lead to short τ'm and increased magnetization of the MCNPs. Conversely, in the acidic tumor microenvironment, the thiolate ligands are protonated and transform into thiol (-SH) ligands, resulting in prolonged τ'm and decreased magnetization of the MCNP, thereby enhancing longitudinal relaxivity (r1) values at UHF MRI. Notably, under a 9 T MRI field, the pH-sensitive changes in Ag-S binding affinity of the MCNP lead to a remarkable (>10-fold) r1 increase in an acidic medium (pH 5.0). In vivo studies demonstrate the capability of MCNPs to amplify MRI signal of hepatic tumors, suggesting their potential as a next-generation UHF-tailored smart MRI contrast agent.

Abstract:Chemodynamic therapy (CDT), as a reactive oxygen species (ROS)-based therapeutic modality, has attracted much attention in recent years. However, the insufficient therapeutic effect of CDT is due to antioxidant system in the tumor microenvironment, such as high levels of glutathione (GSH). In this study, we develop a biological/physical dual-targeting nanotheranostic agent (relaxation rate, r1:6.3 mM-1s-1 and r2:13.11 mM-1s-1) for enhanced CDT of SMCC-7721 tumors. This nanotheranostic agent is composed of a homologous tumor cell membrane (TCM), magnetic ferric oxide, and manganese oxide, and is denoted as FM@TCM nanoparticles (NPs). A favorable in vitro CDT effect on SMCC-7721 cells (IC50: 20 μg/mL) is demonstrated owing to Fenton reaction and oxidative stress by reduction of GSH level. In vivo T1/T2 magnetic resonance imaging (MRI) confirms that the tumor accumulation of FM@TCM NPs is promoted by concurrent bioactive targeting of homologous TCM and physico-magnetic targeting of tumor tissues with an external magnetic field. Impressive chemodynamic therapeutic effects on SMCC-7721 tumors are demonstrated through catalyzing endogenous hydrogen peroxide and depletion of GSH to generate high level of ROS. Dual-targeting FM@TCM NPs inhibit SMCC-7721 tumor growth (~90.9%) in vivo without any biotoxicity. This nanotheranostic agent has great potential for use in MRI-guided CDT.

Abstract:Introduction and objective: Precisely and sensitively diagnosing diseases especially early and accurate tumor diagnosis in clinical magnetic resonance (MR) scanner is a highly demanding but challenging task. Gadolinium (Gd) chelate is the most common T-1 magnetic resonance imaging (MRI) contrast agent at present. However, traditional Gd-chelates are suffering from low relaxivity, which hampers its application in clinical diagnosis. Currently, the development of nano-sized Gd based T-1 contrast agent, such as incorporating gadolinium chelate into nanocarriers, is an attractive and feasible strategy to enhance the T-1 contrast capacity of Gd chelate. The objective of this study is to improve the T-1 contrast ability of Gd-chelate by synthesizing nanoparticles (NPs) for accurate and early diagnosis in clinical diseases. Methods: Reverse microemulsion method was used to coat iron oxide (IO) with tunable silica shell and form cores of NPs IO@SiO2 at step one, then Gd-chelate was loaded on the surface of silica-coated iron oxide NPs. Finally, Gd-based silica coating magnetite NPs IO@SiO2-DTPA-Gd was developed and tested the ability to detect tumor cells on the cellular and in vivo level. Results: The r(1) value of IO@SiO2-DTPA-Gd NPs with the silica shell thickness of 12 nm was about 33.6 mM(-1)s(-1), which was approximately 6 times higher than Gd-DTPA, and based on its high T-1 contrast ability, IO@SiO2-DTPA-Gd NPs could effectively detect tumor cells on the cellular and in vivo level. Conclusion: Our findings revealed the improvement of T-1 relaxation was not only because of the increase of molecular tumbling time caused by the IO@SiO2 nanocarrier but also the generated magnetic field caused by the JO core. This nanostructure with high T-1 contrast ability may open a new approach to construct high-performance T-1 contrast agent.

Abstract:The development of high‐performance contrast agents in magnetic resonance imaging (MRI) has recently received considerable attention, as they hold great promise and potential as a powerful tool for cancer diagnosis. Despite substantial achievements, it remains challenging to develop nanostructure‐based biocompatible platforms that can generate on‐demand MRI signals with high signal‐to‐noise ratios and good tumor specificity. Here, the design and synthesis of a new class of nanoparticle‐based contrast agents comprising self‐assembled NaGdF4 and CaCO3 nanoconjugates is reported. In this design, the spatial confinement of the T1 source (Gd3+ ions) leads to an “OFF” MRI signal due to insufficient interaction between the protons and the crystal lattices. However, when immersed in the mildly acidic tumor microenvironment, the embedded CaCO3 nanoparticles generate CO2 bubbles and subsequently disconnect the nanoconjugate, thus resulting in an “ON” MRI signal. The in vivo performance of these nanoconjugates shows more than 60‐fold contrast enhancement in tumor visualization relative to the commercially used contrast agent Magnevist. This work presents a significant advance in the construction of smart MRI nanoprobes ideally suited for deep‐tissue imaging and target‐specific cancer diagnosis.

Abstract:A type of nanoparticle with three functional modalities was prepared with the aim of providing a multifunctional drug delivery system. The nanoparticle was 50 nm in size, with 2.7 nm mesopores and a magnetic nanocrystal core, which was further doped with FITC to enable the tracking of cellular uptake. We demonstrated that the internalization of the nanoparticles in tumor cells could be enhanced by applying an external magnetic field and furthermore, this kind of nanoparticle could be used in magnetic targeted drug delivery. With high transverse relaxivity, the magnetic nanoparticles shortened proton relaxation time and induced high magnetic resonance imaging contrast in tumor cells. Studies on anticancer drug loading and delivery capacity of anticancer drugs also showed that this type of nanoparticles could load water-soluble doxorubicin, and produce a prominent inhibitive effect against tumor cells. Taken together, the presented nanoparticles could become a promising agent in cancer theranostics.
Multi-modal Mn-Zn Ferrite Nanocrystals for Magnetically-Induced Cancer Targeted Hyperthermia: a Comparison of Passive and Active Targeting Effects.

Abstract:The high performance and increased tumor-targeting accumulation of magnetic nanocrystals (MNCs) are the most important considerations in cancer targeted magnetic hyperthermia (TMH). To achieve these goals, our study was firstly done using well-established fluorescence/magnetic Mn-Zn ferrite MNCs (core size: 14 nm) as multi-modal imaging contrast agents and highly-efficient "heat generators", which were coated with a biocompatible PEG-phospholipid (DSPE-PEG2000) and further modified by a cyclic tripeptide of arginine-glycine-aspartic acid (RGD). By using a mouse model bearing breast carcinoma (4T1), we then systematically compared PEGylated MNCs (MNCs@PEG)- and RGD-PEGylated MNCs (MNCs@RGD)-mediated tumor targeting abilities by intravenous administration. The MNCs@PEG-based passive targeting could successfully accumulate at the tumor due to the enhanced permeability and retention (EPR) effects, but the non-targeted localization might make the MNCs@PEG "leaking" from larger pores of tumor fenestrated vascular networks. Our designed MNCs@RGD, simultaneously functionalized with PEG and RGD ligands, might promote a synergistic effect including efficient tumor vasculature active targeting and EPR-mediated passive targeting, improving total MNC concentration and retention time in tumor tissues. By combining fluorescence/magnetic resonance (MR)/thermal multi-modal imaging-guided diagnostics and continuous TMH treatment under an alternating current magnetic field (ACMF, 2.58 kA m(-1), 390 kHz), the tumor surface could be heated to approximately 43-44 °C based on the MNC-mediated repeated injections. Sufficient temperature elevation induced the apoptosis of tumor cells, and inhibited the tumor angiogenesis. Compared with MNCs@PEG, the active MNCs@RGD-based tumor targeting MR image was significantly more efficient due to both the higher and long-lasting tumor accumulation, but its antitumor efficacy was not obviously improved in the TMH treatments. To achieve a singularly promising tumor TMH therapy, a greatly increased MNC content in tumor was needed. This insight indicated that not only the tumor vasculature targeting, but also the active tumor cells targeting of MNCs should receive considerable attention in future clinical TMH therapy application.
A Multifunctional Nanotheranostic for the Intelligent MRI Diagnosis and Synergistic Treatment of Hypoxic Tumor.

Abstract:Hypoxia, as an inevitable characteristic of solid tumors, has been regarded as a noticeably causative factor to therapeutic resistance and metastatic variants. Exploring novel theranostics to realize the accurate diagnosis of hypoxia and the simultaneous implementation of effective therapy is a promising prospect for the successful treatment of tumors. In the present study, we designed and synthesized a multifunctional rattle-structured nanotheranostic, with the inner core coated by hollow mesoporous silica for chemical drug Doxorubicin (DOX) storage and hypoxia-sensitive MnO2 enrichment. In various acidic micro-environments caused by hypoxia, MnO2 nanosheets could be degraded into manganese ions (Mn2+), which were chelated by the modified Tetraxetanum (DOTA) ligands for real-time T1-magnetic resonance imaging (T1-MRI), with on-demand DOX release to realize both normoxia and hypoxia-sensitive chemotherapy by overcoming hypoxia. Nanotheranostics integrating hypoxia-driven T1-MRI with synergetic chemotherapy have tremendous potential in the intelligent diagnosis, personalized treatment and excellent prognosis of solid tumors in the future.
Injectable thermosensitive magnetic nanoemulsion hydrogel for multimodal-imaging-guided accurate thermoablative cancer therapy.

Abstract:Ferrofluid-based magnetic hyperthermia of cancers has gained significant attention in recent years due to its excellent efficacy, few deleterious side effects and unlimited tissue penetration capacity. However, the high tumor osmotic pressure causes injection leakage and thus position imprecision because of the fluidity of the ferrofluid and the absence of multimodal imaging guidance, which create tremendous challenges for clinical application. Here, a body temperature-induced gelation strategy is constructed for accurate localized magnetic tumor regression based on the unique behaviors of a magnetic nanoemulsion hydrogel (MNH) within tumors. The rapid intra-tumor gelation can securely restrict the MNH in tumor tissue without diffusion and leakage. The magnetically induced nanoparticle assembly-enhanced heating in the hydrogel and the heat accumulation caused by crosslinking among the nanoemulsion droplets further increased the heating efficiency. Meanwhile, US/MR/NIR multimodal imaging can guide the whole therapeutic process, achieving excellent magnetic hyperthermia therapeutic efficiency. This work highlights the great promise for improving the magnetic hyperthermia efficiency and the precision of the injection site for localized tumor therapy.
Macrophage-Mediated Porous Magnetic Nanoparticles for Multimodal Imaging and Postoperative Photothermal Therapy of Gliomas

Abstract:Because of the blood-brain barrier and the high infiltration of glioma cells, the diagnostic accuracy and treatment efficiency of gliomas are still facing challenges. There is an urgent need to explore the integration of diagnostic and therapeutic methods to achieve an accurate diagnosis, guide surgery, and inhibit postoperative recurrence. In this work, we developed a macrophage loaded with a photothermal nanoprobe (MFe3O4-Cy5.5), which is able to cross the blood-brain barrier and accumulate into deep gliomas to achieve multimodal imaging and guided glioma surgery purposes. With desirable probing depth and high signal-to-noise ratio, Fe3O4-Cy5.5 can perform fluorescence, photoacoustic, and magnetic resonance imaging, which can distinguish brain tumors from the surrounding normal tissues and accurately guide glioma resection. Meanwhile, Fe3O4-Cy5.5 can effectively induce local photothermal therapy and inhibit the recurrence of glioma after surgery. These results demonstrate that the macrophage-mediated Fe3O4-Cy5.5, which can achieve a multimodal diagnosis, accurate imaging-guided surgery, and effective photothermal therapy, is a promising nanoplatform for gliomas.

Tumor Microenvironment Responsive Biodegradable Fe‐Doped MoO x Nanowires for Magnetic Resonance Imaging Guided Photothermal‐Enhanced Chemodynamic Synergistic Antitumor Therapy

Abstract:Rational design of nanosystems that target tumor microenvironment have attracted widespread attention. However, it is still a great challenge to make a multifunctional nanoplatform that actively and selectively interacts with tumor microenvironment, without causing toxicity to surrounding normal tissues. Herein, the biodegradable Fe‐doped MoOx (FMO) nanowires are designed as an anti‐tumor nanoreagent that possesses great photothermal conversion ability (48.5%) and magnetic properties for T1 weighted magnetic resonance imaging (MRI). Also, FMO can be used as a chemodynamic therapy (CDT) reagent to effectively catalyze the decomposition of H2O2 and produce hydroxyl radical (·OH). At the same time, the consumption of glutathione will also enhance the CDT effect. More importantly, FMO presents pH‐dependent degradation behavior: rapid degradation at physiological pH, but relatively stable at acidic pH. In vivo anti‐tumor experiment demonstrates that the FMO is able to effectively inhibit the tumor growth with minimal side effects. Generally speaking, these results indicate that the FMO has huge potential for MRI image‐guided cancer therapy and promotes the clinical translation of nanodrugs.

Abstract:Multifunctional nanoparticles, bearing low toxicity and tumor-targeting properties, coupled with multifunctional diagnostic imaging and enhanced treatment efficacy, have drawn tremendous attention due to their enormous potential for medical applications. Herein, we report a new kind of biocompatible and tumor-targeting magneto-gold@fluorescent polymer nanoparticle (MGFs-LyP-1), which is based on ultra-small magneto-gold (Fe3O4-Au) nanoparticles and NIR emissive fluorescent polymers by a solvent-mediated method. This kind of nanoparticle could be taken up efficiently and simultaneously serve for in vivo tumor targeting T1&T2-MRI/CT/near infrared (NIR) fluorescence bioimaging. Furthermore, the nanoparticles exhibit small size, higher tumor targeting accumulation, excellent cytocompatibility for long-term tracking, and no disturbing cell proliferation and differentiation. Moreover, clear and convincing evidence proves that as-synthesized MGFs-LyP-1 could elicit genuine autophagy via inducing autophagosome formation, which offers a definite synergistic effect to enhance cancer therapy with doxorubicin (DOX) at a nontoxic concentration through enhancement of the autophagy flux. Meanwhile, the as-prepared nanoparticles could be rapidly cleared from mice without any obvious organ impairment. The results indeed reveal a promising prospect of an MGFs-LyP-1 contrast agent with low toxicity and high efficiency for promising application in biomedicine.
Time-Dependent T-1-T-2 Switchable Magnetic Resonance Imaging Realized by C(rgdyk) Modified Ultrasmall Fe3O4 Nanoprobes

Abstract:To achieve the accurate diagnosis of tumor with the magnetic resonance imaging (MRI), nanomaterials-based contrast agents are developed rapidly. Here, a tumor targeting nanoprobe of c(RGDyK) modified ultrasmall sized iron oxide is reported with high saturation magnetization and high T-1-weighted imaging capability, attributed to a large number of paramagnetic centers on the surface of nanoprobes and rapid water proton exchange rate (inner sphere model), as well as strong superparamagnetism (outer sphere model). These nanoprobes could actively target and gradually accumulate at the tumor site with a time-dependent T-1-T-2 contrast enhancement imaging effect. In in vivo MRI experiments, the nanoprobes exhibit the best T-1 contrast enhancement at 30 min after intravenous administration, followed by gradually vanishing and generating T-2 contrast enhancement with increasing time at tumor site. This is likely due to time-dependent nanoprobes aggregation in tumor, in good agreement with in vitro experiment where aggregated nanoprobes display larger r(2)/r(1) value (19.1) than that of the dispersed nanoprobes (2.8). This dynamic property is completely different from other T-1-T-2 dual-modal nanoprobes which commonly exhibit the T-1- and T-2-weighted enhancement effect at the same time. To sum up, these c(RGDyK) modified ultrasmall Fe3O4 nanoprobes have significant potential to improve the diagnostic accuracy and sensitivity in MRI.

Abstract:Precise localization and visualization of early-stage prostate cancer (PCa) is critical to improve the success of focal ablation and reduce cancer mortality. However, it remains challenging under the current imaging techniques due to the heterogeneous nature of PCa and the suboptimal sensitivity of the techniques themselves. Herein, a novel genetic amplified nanoparticle tumor-homing strategy to enhance the MRI accuracy of ultrasmall PCa lesions is reported. This strategy could specifically drive TfR expressions in PCa under PCa-specific DD3 promoter, and thus remarkably increase Tf-USPIONs concentrations in a highly accurate manner while minimizing their non-specific off-target effects on normal tissues. Consequently, this strategy can pinpoint an ultrasmall PCa lesion, which is otherwise blurred in the current MRI, and thereby addresses the unmet key need in MRI imaging for focal therapy. With this proof-of-concept experiment, the synergistic gene-nano strategy holds great promise to boost the MRI effects of a wide variety of commonly used nanoscale and molecular probes that are otherwise limited. In addition, such a strategy may also be translated and applied to MR-specific imaging of other types of cancers by using their respective tumor-specific promoters.

Abstract:Personalized cancer medicine requires measurement of therapeutic efficacy as early as possible, which is optimally achieved by three-dimensional imaging given the heterogeneity of cancer. Magnetic resonance imaging (MRI) can obtain images of both anatomy and cellular responses, if acquired with a molecular imaging contrast agent. The poor sensitivity of MRI has limited the development of activatable molecular MR contrast agents. To overcome this limitation of molecular MRI, a novel implementation of our caspase-3-sensitive nanoaggregation MRI (C-SNAM) contrast agent is reported. C-SNAM is triggered to self-assemble into nanoparticles in apoptotic tumor cells and effectively amplifies molecular level changes through nanoaggregation, enhancing tissue retention and spin-lattice relaxivity. At one-tenth the current clinical dose of contrast agent and following a single imaging session, C-SNAM MRI accurately measured the response of tumors to either metronomic chemotherapy or radiation therapy, where the degree of signal enhancement is prognostic of long-term therapeutic efficacy. Importantly, C-SNAM is inert to immune activation, permitting radiation therapy monitoring.

Abstract:Gadolinium-based nanoscale magnetic resonance imaging (MRI) contrast agents (CAs) have gained significant momentum as a promising nanoplatform for detecting tumor tissue in medical diagnosis, due to their favorable capability of enhancing the longitudinal relaxivity (r(1)) of individual gadolinium ions, delivering to the region of interest a large number of gadolinium ions, and incorporating different functionalities. This mini-review highlights the latest developments and applications, and simultaneously gives some perspectives for their future development.

Abstract:In this review, we discussed the establishment of a so-called "theranostic" system by instituting the basic principles including the use of: [1] magnetic iron oxide nanoparticles (MION)-based drug carrier; [2] intra-arterial (I.A.) magnetic targeting; [3] macromolecular drugs with unmatched therapeutic potency and a repetitive reaction mechanism; [4] cell-penetrating peptide-mediated cellular drug uptake; and [5] heparin/protamine-regulated prodrug protection and tumor-specific drug re-activation into one single drug delivery system to overcome all possible obstacles, thereby achieving a potentially non-invasive, magnetic resonance imaging-guided, clinically enabled yet minimally toxic brain tumor drug therapy. By applying a topography-optimized I.A. magnetic targeting to dodge rapid organ clearance of the carrier during its first passage into the circulation, tumor capture of MION was enriched by >350 folds over that by conventional passive enhanced permeability and retention targeting. By adopting the prodrug strategy, we observed by far the first experimental success in a rat model of delivering micro-gram quantity of the large β-galactosidase model protein selectively into a brain tumor but not to the ipsi- or contra-lateral normal brain regions. With the therapeutic regimens of most toxin/siRNA drugs to fully (>99.9%) eradicate a tumor being in the nano-molar range, the prospects of reaching this threshold become practically accomplishable.
