Abstract:A one-pot two-step reaction of 2-(trifluoromethyl)-2-hydroxy-2H-chromene with aryl ketone afforded α-trifluoromethylated tertiary alcohols bearing coumarins. The atomic economic and simply manipulative reaction involved premier treatment of reaction mixtures with CH 3 COOH and NH 4 OAc in one-pot process. Abstract:A one-pot two-step reaction of 4-aminocoumarin with arylglyoxal monohydrates and p-toluenesulfonates has been developed, which offers 3-alkoxy-substituted chromeno[4,3-b]pyrrol-4(1H)-ones in moderate to good yields. At first, arylglyoxal monohydrates and 4-aminocoumarin were converted into 3-hydroxychromeno[4,3-b]pyrrol-4(1H)-one intermediates in AcOH under reflux. Then the title compounds in an in situ reaction of the intermediates with p-toluenesulfonates catalyzed by DBU in refluxing toluene were obtained.

Abstract:Here we report a mild Ag-catalyzed domino acetalization reaction using quinoline-2-carboxylates, which were substructures in a number of natural products. Methanol was directly utilized as the source of the acetal part. The main features of the domino reaction include the Minisci reaction, oxidation, and acetalization. Hydrogen fluoride generated in-situ participates in the Minisci reaction, and no external acids were needed. This domino reaction provides a simple and versatile route to heteroaromatic acetals under mild condition with good functional group tolerance using methanol. (C) 2021 Elsevier Ltd. Abstract:A facile and efficient domino cyclization between 3-aminocoumarins and arylglyoxal monohydrates in the presence of para-toluenesulfonic acid (p-TSA) as a promoter was employed for the synthesis of two libraries of chromeno[3,4-b]pyrrol-4(3H)-one derivatives. Simple workup procedures were used to obtain the products in moderate to excellent yields. The solvents toluene and ethanol served important roles in the selective formation of the pyrrolocoumarin derivatives characterized by their different substitution patterns.

Abstract:A concise and efficient synthetic approach was developed for the synthesis of functionalized chromeno[4,3-b]pyrrol-4(1H)-ones via a one-pot, three-component reaction of 4-aminocoumarin, arylglyoxal monohydrates, and aromatic amines promoted by KHSO4 in toluene under reflux condition. Abstract:A facile silver-catalyzed oxidative decarboxylation of arylthiodifluoroacetic acids or aryloxydifluoroacetic acids with coumarins/quinoxalin-2(1H)-ones was developed. This transformation provided a series of C-3 aryloxydifluoromethylated or arylthiodifluoromethylated coumarins/quinoxalin-2(1H)-ones containing various functional groups in moderate to good yields, featuring good functional group tolerance, easily accessible starting materials and operational simplicity.

Abstract:Sulfocoumarins are key structural motifs in several bioactive molecules. Herein, we describe a simple, one-pot procedure for the synthesis of structurally diverse sulfonocoumarin-3-carboxylates by heating 2-hydroxyaryl aldehydes with an active sulfonyl chloride in the presence of pyridine. The process tolerates numerous functional groups including alkoxy, alkyl, halogen, nitro, and even nucleophilic phenolic hydroxy. Additionally, reactions of 2-hydroxyaryl ketones and 2-methylaminoaryl aldehydes give 4-substituted sulfocoumarins and 1-aza-2-sulfocoumarins, respectively. A gram-scale synthesis and further derivatizations are also reported. The ester group is easily removed via Happer’s decarboxylation.

Abstract:Coumarins and 2H-pyran derivatives are among the most commonly found structural units in natural products. Therefore, the introduction of 2H-pyran moiety into the coumarin structural unit, i.e., dihydrocoumarin-fused dihydropyranones, is a potentially successful route for the identification of novel bioactive structures, and the synthesis of these structures has attracted continuing research interest. Herein, a chiral tertiary amine catalyzed [4 + 2] cyclization of 3-aroylcoumarines with benzyl 2,3-butadienoate was reported. In the presence of Kumar’s 6’-(4-biphenyl)-β-iso-cinchonine, the desired dihydrocoumarin-fused dihydropyranone products could be obtained in up to 97% yield and 90% ee values.

Abstract:An efficient tandem Michael addition/aromatization/cyclization reaction of pyrazolones with alpha,beta-unsaturated trifluoromethyl ketones is described. The notable features of this tandem process are its operational simplicity, easily accessible starting materials, and mild reaction conditions. The corresponding trifluoromethylated pyranopyrazoles were obtained in excellent yields (85-99%) with good to excellent diastereoselectivities (6:1-30:1). In addition, the stereochemical assignment of the major isomer by X-ray crystallographic analysis indicated that the trans-products were predominantly formed. (C) 2014 Elsevier B.V. Abstract:A concise and efficient synthetic approach to 3-arylaminopyrroline-2-ones from anilines and beta,gamma-unsaturated alpha-ketoesters in boiling dichloromethane has been developed. This protocol possesses many advantages such as short reaction time, high isolated yields, and expanding substrate scopes. According to the isolated intermediates and controlled reactions, the reaction was tentatively proposed to involve the Michael addition/condensation and subsequent intramolecular cyclization. The structures of the title compounds were unambiguously confirmed by various spectral data such as X-ray crystallographic analysis.

Abstract:3-Alkylated coumarins have many applications in medicinal chemistry, however, methods access to such structures are still limited. Herein, we report a site-selective photocatalytic deaminative alkylation of coumarins utilizing pyridinium-activated aliphatic primary amines as alkylation reagents. The protocol was highlighted by its mild reaction conditions, operational simplicity, and broad functional group compatibility. Moreover, this strategy enables late-stage modification of some pharmaceuticals and natural products, thus providing an appealing approach to valuable molecules in medicinal chemistry.

Abstract:A facile route to 3-((2,3-dihydrobenzofuran-3-yl)methyl)sulfonyl coumarins through the reaction of 2-(allyloxy)anilines, DABCO-bis(sulfur dioxide), and aryl propiolates is achieved. During the reaction process, a 2-(allyloxy) aryl radical would be generated in situ, which would undergo the intramolecular addition of a double bond to afford an alkyl radical intermediate. Further insertion of sulfur dioxide would produce an alkylsulfonyl radical. The subsequent combination with aryl propiolates would provide sulfonyl-bridged dihydrobenzofuran and coumarin derivatives through radical cyclization and rearrangement.

Abstract:An efficient approach for the direct synthesis of alkylated 4-hydroxycoumarin derivatives via a Cu-catalyzed cascade dehydrogenation/conjugate addition sequence starting from simple saturated ketones and 4-hydroxycoumarins has been developed. This protocol features excellent functional-group tolerance, easy scale-up, and a broad substrate scope including bioactive molecules. More importantly, a series of marketed drugs, such as warfarin, acenocoumarol, coumachlor, and coumafuryl, can be obtained by this method.

Abstract:Naphthols and 3-trifluoroethylidene oxindoles were found to undergo an asymmetric Friedel-Crafts alkylation/lactonization reaction, catalyzed by only 2.5 mol % of a quinine-derived squaramide catalyst, to afford the corresponding α-aryl-β-trifluoromethyl dihydrocoumarin derivatives in high yields (up to 99 %) with excellent enantio- and diastereoselectivities (up to 98 % ee, >20:1 d.r.). Importantly, the lactonization proceeded by nucleophilic attack of the naphthol hydroxy group at the amide motif of the oxindoles under mild reaction conditions. This protocol represents a new strategy for the formation of dihydrocoumarins by an efficient intramolecular amide C-N bond-cleavage and esterification process.
