Abstract:A photoinduced copper-catalyzed alkoxyl triggered C−C bond cleavage/aminocarbonylation cascade is presented. Through adjusting the structure of alkoxyl radical precursors, functionalized lactones and keto-amides were synthesized with good yields and excellent functional group tolerance under redox-neutral conditions. Notably, this protocol enables the integration of lactone fragments with many amine drugs and drug fragments.

Abstract:In contrast with abundant methods for the asymmetric functionalization of alkyl radicals to generate stereogenic centres at reaction sites, catalytic enantioselective desymmetrizing functionalization of alkyl radicals for forging multiple stereocentres—including positions that are remote from the reaction sites—with both high enantio- and diastereoselectivity remains largely unexplored. The major challenge for such reactions is the high reactivity of open-shell alkyl radicals. Here, we describe a strategy to address this challenge: the use of Cu( ii ) phosphate to immediately associate with the in situ-generated reactive alkyl radical species, creating a compact and confined chiral microenvironment for effective stereocontrol. With this strategy, we have developed a general and efficient catalytic enantioselective desymmetrizing functionalization of alkene-tethered 1,3-diols. It provides various tetrahydrofurans and analogues bearing multiple stereocentres with remarkably high levels of enantio- and diastereocontrol. Density functional theory calculations and mechanistic experiments revealed a reaction mechanism involving an enantiodetermining outer-sphere C–O bond formation step.

Abstract:Carbohydrates, one of the three primary macromolecules of living organisms, play significant roles in various biological processes such as intercellular communication, cell recognition, and immune activity. While the majority of established methods for the installation of carbohydrates through the anomeric carbon rely on nucleophilic displacement, anomeric radicals represent an attractive alternative because of their functional group compatibility and high anomeric selectivities. Herein, we demonstrate that anomeric nucleophiles such as C1 stannanes can be converted into anomeric radicals by merging Cu(I) catalysis with blue light irradiation to achieve highly stereoselective C(sp3)-S cross-coupling reactions. Mechanistic studies and DFT calculations revealed that the C-S bond-forming step occurs via the transfer of the anomeric radical directly to a sulfur electrophile bound to Cu(II) species. This pathway complements a radical chain observed for photochemical metal-free conditions where a disulfide initiator can be activated by a Lewis base additive. Both strategies utilize anomeric nucleophiles as efficient radical donors and achieve a switch from an ionic to a radical pathway. Taken together, the stability of glycosyl nucleophiles, a broad substrate scope, and high anomeric selectivities observed for the thermal and photochemical protocols make this novel C-S cross coupling a practical tool for late-stage glycodiversification of bioactive natural products and drug candidates.
Supramolecular Free Radicals: Fabrication, Modulation and Functions

Abstract:Modulating the activity of radicals is of great importance for the applications in radical-based materials and radical-mediated reactions. To this end, we have proposed a new concept of "supramolecular free radicals", which refers to the free radicals stabilized or activated through supramolecular approaches. Based on the host-guest chemistry of cucurbiturils (CB), we have fabricated three kinds of supramolecular free radicals to modulate the activity and realize diverse functions. Firstly, radical anions can be stabilized by the steric effect and electrostatic effect of CB. As a result, we have constructed a highly efficient near-infrared photothermal conversion system, which displays selective antibacterial performance. Secondly, owing to the electrostatic effect of CB, radical cations can be activated to induce a significant acceleration of Fenton oxidation reaction. Thirdly, by taking advantage of the dynamic nature of host-guest interactions, we can endow the reaction intermediate with adaptive reactivity, which greatly improves the catalytic efficiency of alcohol oxidation. It is highly anticipated that this series of research opens a new horizon in supramolecular materials and supramolecular catalysis.

Abstract:A Fe(III)-promoted highly selective photo-oxidation of benzylic C–H bond delivering relative carbonyl products is reported. By altering the concentration of ferric salt, methylarenes can be selectively oxidized under UV irradiation to furnish aromatic aldehydes or acids, respectively. By this protocol, the oxidation of ethylarenes provides the corresponding acetophenones. The reaction is inferred to involve divergent pathways in different concentrations of catalyst for the alternative selectivity between aldehydes and aicds. The reusable catalyst, high conversion and selectivity make this oxidation a green and economic protocol for the synthesis of aromatic carbonyl compounds.

The copper-mediated bond formation step of radicals plays a critical role in controlling chemo- and stereoselectivity of copper-catalyzed radical transformation reactions. This bond formation step involves three possible pathways: ion-type bond formation, radical substitution, and reductive elimination. This review highlights the recent advances in theoretical studies on mechanisms and controlling models on the selectivity of Cu-mediated radical-involved bond formation, providing a general mechanistic comprehension of this key elementary step in copper catalysis.

Abstract:Benefiting from the regulation of ROS species through a simple and feasible strategy, an efficient and switchable photocatalytic protocol for the chemoselective synthesis of aldehydes and carboxylic acids via oxidation of primary alcohols under mild conditions has been developed. Using the pyridinium derivative as a metal-free organic photocatalyst, a variety of benzyl alcohols can be converted into aldehydes or carboxylic acids under an air atmosphere at room temperature with high selectivity and yield via simple solvent regulation. A feasible tandem reaction combining the pyridinium-catalyzed alcohol-to-aldehyde conversion and catalyst-free photoinduced oxidation of aromatic aldehydes provides a powerful and efficient approach for the one-pot oxidation of benzyl alcohols to carboxylic acids, while a small amount of water can effectively regulate the competition of electron transfer and energy transfer and inhibit the generation of carboxylic acids, facilitating highly efficient and selective oxidation of alcohols to aldehydes.

Abstract:We report a supramolecular strategy to promote radical-mediated Fenton oxidation by the rational design of a folded host-guest complex based on cucurbit[8]uril (CB[8]). In the supramolecular complex between CB[8] and a derivative of 1,4-diketopyrrolo[3,4-c]pyrrole (DPP), the carbonyl groups of CB[8] and the DPP moiety are brought together through the formation of a folded conformation. In this way, the electrostatic effect of the carbonyl groups of CB[8] is fully applied to highly improve the reactivity of the DPP radical cation, which is the key intermediate of Fenton oxidation. As a result, the Fenton oxidation is extraordinarily accelerated by over 100 times. It is anticipated that this strategy could be applied to other radical reactions and enrich the field of supramolecular radical chemistry in radical polymerization, photocatalysis, and organic radical battery and holds potential in supramolecular catalysis and biocatalysis.

Abstract:Tuning the activity of radicals is crucial for radical reactions and radical-based materials. Herein, we report a supramolecular strategy to accelerate the Fenton reaction through the construction of supramolecularly activated radical cations. As a proof of the concept, cucurbit[7]uril (CB[7]) was introduced, through host-guest interactions, onto each side of a derivative of 1,4-diketopyrrolo[3,4-c]pyrrole (DPP), a model dye for Fenton oxidation. The DPP radical cation, the key intermediate in the oxidation process, was activated by the electrostatically negative carbonyl groups of CB[7]. The activation induced a drastic decrease in the apparent activation energy and greatly increased the reaction rate. This facile supramolecular strategy is a promising method for promoting radical reactions. It may also open up a new route for the catalytic oxidation of organic pollutants for water purification and widen the realm of supramolecular catalysis.

Abstract:The selective oxidation of benzylic C-H bonds is a pivotal transformation in organic synthesis. Undoubtedly, achieving efficient and highly selective aerobic oxidation of methylarenes to benzaldehydes has been highly challenging due to the propensity of benzaldehyde to undergo overoxidation under typical aerobic conditions. Herein, we propose an innovative approach to address this issue by leveraging electrocatalytic processes, facilitated by ion-pair mediators [Ph3C]+[B(C6F5)4]-. By harnessing the power of electrochemistry, we successfully demonstrated the effectiveness of our strategy, which enables the selective oxidation of benzylic C-H bonds in benzylic molecules and toluene derivatives. Notably, our approach exhibited high efficiency, excellent selectivity, and compatibility with various functional groups, underscoring the broad applicability of our methodology.

Abstract:Synthesizing benzyl skeleton derivatives via direct oxidation of functionalized benzylic C-H bonds has received extensive research attention. Herein, a method was developed to prepare carbonyl compounds via photoinduced aerobic oxidation of ubiquitous benzylic C-H bonds mediated by bromine radicals and tribromomethane radicals. This method employed commercially available CBr4 as a hydrogen atom transfer reagent precursor, air as an oxidant, water as a reaction solvent, and tetrabutylammonium perchlorate (TBAPC) as an additive under mild conditions. A series of substrates bearing different functional groups was converted to aromatic carbonyls in moderate to good yields. Moreover, a low environmental factor (E-factor value=0.45) showed that the proposed method is ecofriendly and environmentally sustainable. Photocatalytic strategy for synthesizing aromatic ketones. The reaction is performed in H2O using CBr4 as a mediator and using air as an oxidant under 400 nm irradiation. Abstract:Directly asymmetric oxidation of C(sp3)–H bonds of organic molecules would radically alter current methods of synthesizing chiral alcohols and esters, which are widely existed in the structures of natural products, pharmaceuticals, and fine chemicals. The Kharasch–Sosnovsky reaction is commonly used process for oxidation of allylic C(sp3)–H bonds of alkenes to yield allyl esters. However, its asymmetric variant is limited to cycloalkenes, and the enantioselectivity and efficiency of the reactions of open-chain alkenes are low. Moreover, asymmetric Kharasch–Sosnovsky reactions of alkanes remain a seemingly insurmountable challenge. However, we herein report a method for highly enantioselective oxidation of benzylic C(sp3)–H bonds of arylalkanes by dual photoredox and copper catalysis. The method extends the scope of the Kharasch–Sosnovsky reaction to alkanes and, more importantly, uses alcohols or carboxylic acids instead of peroxides as oxygenated coupling partners. With this method, we obtained various chiral alcohols and esters with high enantioselectivity directly from readily available arylalkanes.

Abstract:The mild catalytic generation of ketyl radicals for organic transformations remains an unsolved issue, although it facilitates the discovery of metal-catalyzed reactions in feature of high functional group tolerance. We report here the generation of ketyl radicals and coupling with alkynes that were enabled by cost-effective chromium catalysis, allowing for the formation of valuable E-exocyclic allyl alcohols in high stereo- and chemoselectivity. A broad range of synthetically useful functional groups that are sensitive to strong reductants are compatible by the catalytic system, providing access to diverse substituted E-exocyclic allyl alcohols under mild conditions. The appended hydroxyl groups in the products are facilely late-stage functionalized in accessing numerous derivatives, as well as the enantioenrichement of exocyclic allyl alcohol using chiral ligands. Mechanistic studies suggest that bipyridine-ligated Cr(II) complex serves as reactive catalyst with enabling the generation of ketyl radical for the coupling, giving vinyl radical and followed by the combination of Cr and transmetalation with Cp2ZrCl moiety in affording oxazirconiumacycle. This reaction provides a new entry for the mild formation of transient ketyl radicals from widely accessible aliphatic aldehydes for couplings by earth-abundant metal catalysis.

Abstract:An intermolecular addition of carbon radicals enabled by a cascade radical coupling strategy is developed. It includes an intermolecular alkyl radical addition to a carbonyl group followed by an intramolecular alkoxy radical addition to haloarenes and produces substituted benzofurans in high yields. The radical nature of this reaction is explored by radical trapping experiments and EPR analysis. The mechanism is investigated by KIE experiments and control experiments. This method could provide rapid and practical access to the key intermediate of TAM-16, a safe and potent antibacterial agent for treating tuberculosis, and, therefore, is of great importance for organic synthesis and the pharmaceutical industry.

Abstract:Multi-site functionalization of molecules provides a potent approach to accessing intricate compounds. However, simultaneous functionalization of the reactive site and the inert remote C(sp3)-H poses a formidable challenge, as chemical reactions conventionally occur at the most active site. In addition, achieving precise control over site selectivity for remote C(sp3)-H activation presents an additional hurdle. Here we report an alternative modular method for alkene difunctionalization, encompassing radical-triggered translocation of functional groups and remote C(sp3)-H desaturation via photo/cobalt dual catalysis. By systematically combining radical addition, functional group migration and cobalt-promoted hydrogen atom transfer, we successfully effectuate the translocation of the carbon-carbon double bond and another functional group with precise site selectivity and remarkable E/Z selectivity. This redox-neutral approach shows good compatibility with diverse fluoroalkyl and sulfonyl radical precursors, enabling the migration of benzoyloxy, acetoxy, formyl, cyano and heteroaryl groups. This protocol offers a resolution for the simultaneous transformation of manifold sites.

The strategically designed arylbenzothiazolylether diazonium salts are harnessed as dual-function reagents. A vast array of alkenes are proven to be suitable substrates.
