Abstract:Abstract Compound light is required for plant growth and development, but the response mechanisms of plants are undercharacterized and not fully understood. The present study was undertaken to evaluate the effect of supplemental light (green light, G; white light, W; yellow light, Y) added to red–blue light (RB) and sole W on the growth and photosynthesis of rapeseed seedlings. The results revealed that supplemental G/W improved the growth and photosynthesis of seedlings, but supplemental Y significantly reduced the photosynthetic rate and palisade tissue layer. Sole W caused similar responses in terms of growth, leaf development, oxidative damage, and antioxidant capability as supplemental Y. In total, 449, 367, 813, and 751 differentially expressed genes (DEGs) were identified under supplemental G, Y, and W and sole W, respectively, compared to RB. The DEGs under different lights were closely associated with pathways such as light stimulus and high-light response, root growth, leaf development, photosynthesis, photosynthesis-antenna proteins, carbohydrate synthesis and degradation, secondary metabolism, plant hormones, and antioxidant capacity, which contributed to the distinct growth and photosynthesis under different treatments. Our results suggest that Y is more likely substituted by other wavelengths to achieve certain effects similar to those of supplemental Y, while G has a more distinctive effect on rapeseed. Taken together, supplementation RB with G/W promotes the growth of rapeseed seedlings in a controlled environment.

Abstract:Under different red (R):blue (B) photon flux ratios, the growth performance of rapeseed (Brassica napus L.) is significantly different. Rapeseed under high R ratios shows shade response, while under high B ratios it shows sun-type morphology. Rapeseed under monochromatic red or blue light is seriously stressed. Transcriptomic and proteomic methods were used to analyze the metabolic pathway change of rapeseed (cv. “Zhongshuang 11”) leaves under different R:B photon flux ratios (including 100R:0B%, 75R:25B%, 25R:75B%, and 0R:100B%), based on digital gene expression (DGE) and two-dimensional gel electrophoresis (2-DE). For DGE analysis, 2054 differentially expressed transcripts (|log2(fold change)|≥1, q<0.005) were detected among the treatments. High R ratios (100R:0B% and 75R:25B%) enhanced the expression of cellular structural components, mainly the cell wall and cell membrane. These components participated in plant epidermis development and anatomical structure morphogenesis. This might be related to the shade response induced by red light. High B ratios (25R:75B% and 0R:100B%) promoted the expression of chloroplast-related components, which might be involved in the formation of sun-type chloroplast induced by blue light. For 2-DE analysis, 37 protein spots showed more than a 2-fold difference in expression among the treatments. Monochromatic light (ML; 100R:0B% and 0R:100B%) stimulated accumulation of proteins associated with antioxidation, photosystem II (PSII), DNA and ribosome repairs, while compound light (CL; 75R:25B% and 25R:75B%) accelerated accumulation of proteins associated with carbohydrate, nucleic acid, amino acid, vitamin, and xanthophyll metabolisms. These findings can be useful in understanding the response mechanisms of rapeseed leaves to different R:B photon flux ratios.
RNA-Seq-Based Profiling of pl Mutant Reveals Transcriptional Regulation of Anthocyanin Biosynthesis in Rice (Oryza sativa L.)

Abstract:Purple-colored leaves in plants attain much interest for their important biological functions and could be a potential source of phenotypic marker in selecting individuals in breeding. The transcriptional profiling helps to precisely identify mechanisms of leaf pigmentation in crop plants. In this study, two genetically unlike rice genotypes, the mutant purple leaf (pl) and wild (WT) were selected for RNA-sequencing and identifying the differentially expressed genes (DEGs) that are regulating purple leaf color. In total, 609 DEGs were identified, of which 513 and 96 genes were up- and down-regulated, respectively. The identified DEGs are categorized into metabolic process, carboxylic acid biosynthesis, phenylpropanoids, and phenylpropanoid biosynthesis process enrichment by GO analysis. Kyoto Encyclopedia of Genes and Genomes (KEGG) confirmed their association with phenylpropanoid synthesis, flavonoid synthesis, and phenylalanine metabolism. To explore molecular mechanism of purple leaf color, a set of anthocyanin biosynthetic and regulatory gene expression patterns were checked by qPCR. We found that OsPAL (Os02g0626100, Os02g0626400, Os04g0518400, Os05g0427400 and Os02g0627100), OsF3H (Os03g0122300), OsC4HL (Os05g0320700), and Os4CL5 (Os08g0448000) are associated with anthocyanin biosynthesis, and they were up-regulated in pl leaves. Two members of regulatory MYB genes (OsMYB55; Os05g0553400 and Os08g0428200), two bHLH genes (Os01g0196300 and Os04g0300600), and two WD40 genes (Os11g0132700 and Os11g0610700) also showed up-regulation in pl mutant. These genes might have significant and vital roles in pl leaf coloration and could provide reference materials for further experimentation to confirm the molecular mechanisms of anthocyanin biosynthesis in rice.
The regulation of photosynthesis and growth of rapeseed seedling by the interaction of red and yellow lights with blue light

Abstract:The interactive regulation among different lights on plant life-activities has not yet been fully understood. In the study, we investigated the interaction of red light (R) and yellow light (Y) with blue light (B) on growth and photosynthesis of rapeseed seedlings. The results showed Y and R as well as their combination of RY were not beneficial for plant photosynthesis and growth, but a quarter addition of B (RB, YB and RBY) obviously reversed these negative effects, improving stomatal behavior, leaf and root developments, photosynthetic traits, as well as synthesis and transport of photosynthetic products. Available light energy and B intervention together decreased the ratio of chlorophyll b (Chl b) to carotenoid (Car), while the opposite effect was observed in the absence of B treatments. Among the various lights, RY and Y significantly upregulated the majority of genes associated with chlorophyll metabolism, while RB predominantly upregulated genes related to the two photosystems (PS). Conversely, B and YB treatments generally suppressed the expression of these genes. Despite these differences in gene expression, the ratios of PSI/PSII to Chl b/Car were consistently maintained around 0.56 in the RB, YB, and RBY treatments. In contrast, this ratio was higher at 0.64 in the B treatment and lower, at less than 0.44, in the R, Y, and RY treatments. These ratios reflect the balance between light supply and utilization in plants under different lights, where plants under RB, YB, and RBY might achieve optimal light environments, while plants under B and under R, Y, and RY experienced high and low light stress, respectively. In conclusion, all combinations of B with R, Y or RY can be used for rapeseed cultivation, and the ratio of Chl b/Car to PSI/PSII might be an important reference index to select optimal light combinations for crop production.

Abstract:Rapeseed (Brassica napus L.) is sensitive to light quality. The factory production of rapeseed seedlings for vegetable use and for transplanting in the field requires an investigation of the responses of rapeseed to light quality. This study evaluated the responses of the leaf of rapeseed (cv. ‘Zhongshuang 11’) to different ratios of red-photonflux (RPF) and blue-photonflux (BPF) from light emitting diodes (LEDs). The treatments were set as monochromatic lights, including 100R:0B% and 0R:100B%, and compound lights (CLs), including 75R:25B%, 50R:50B% and 25R:75B%. The total photonflux in all of the treatments was set as 550 μmolm-2s-1. With an increase of BPF, the rapeseed leaves changed from wrinkled blades and down-rolled margins to flat blades and slightly up-rolled margins, and the compact degree of palisade tissue increased. One layer of the cells of palisade tissue was present under 100R:0B%, whereas two layers were present under the other treatments. Compared to 100R:0B%, 0R:100B% enhanced the indexes of leaf thickness, leaf mass per area (LMA), stomatal density, chlorophyll (Chl) content per weight and photosynthetic capacity (Pmax), and the CLs with high BPF ratios enhanced these indexes. However, the 100R:0B% and CLs with high RPF ratios enhanced the net photosynthetic rate (Pn). The leaves under the CLs showed growth vigor, whereas the leaves under 100R:0B% or 0R:100B% were stressed with a low Fv/Fm (photosynthetic maximum quantum yield) and a high content of O2.- and H2O2. The top second leaves under 100R:0B% or 0R:100B% showed stress resistance responses with a high activity of antioxidase, but the top third leaves showed irreversible damage and inactivity of antioxidase. Our results showed that the rapeseed leaves grown under 0R:100B% or CLs with a high BPF ratio showed higher ability to utilize high photonflux, while the leaves grown under 100R:0B% or CLs with a low BPF ratio showed higher efficiency in utilizing low photonflux. Under different R:B photonflux ratios, red and blue lights may play mutual roles in Pn. When the blue light dominated, the Pn showed a B-preference. When the red light dominated, the Pn showed an R-preference. Furthermore, CLs were suitable for the Pn of rapeseed seedlings.

Abstract:Here, we analyzed the transcriptomes of Brassica campestris L. ssp. chinensis Makino under the treatment of a combination of red (R) and blue (B) light. In total, 5,643 differentially expressed genes (DEGs) were found under the R and B combination compared with the white light. An analysis of the DEGs enriched in photosynthesis and antenna proteins showed that there were many more upregulated subunits in PSII than those in PSI. The DEGs encoding the cytochrome b(6)f complex, photosynthetic electron transport, ATP synthases, and key genes involved in chlorophyll (Chl) and carotenoids (Car) biosynthesis were also upregulated. In addition, real-time PCR indicated that 13 of 15 genes showed the similar expression patterns with the RNA-seq data. The R and B combination might promote photosynthesis by specifically regulating Chl biosynthesis, thus accelerating the growth and development of plants and providing a valuable reference for vegetable production in factories and variety breeding.
An RNA-Seq Analysis of Grape Plantlets Grown in vitro Reveals Different Responses to Blue, Green, Red LED Light, and White Fluorescent Light

Abstract:Using an RNA sequencing (RNA-seq) approach, we analyzed the differentially expressed genes (DEGs) and physiological behaviors of "Manicure Finger" grape plantlets grown in vitro under white, blue, green, and red light. A total of 670, 1601, and 746 DEGs were identified in plants exposed to blue, green, and red light, respectively, compared to the control (white light). By comparing the gene expression patterns with the growth and physiological responses of the grape plantlets, we were able to link the responses of the plants to light of different spectral wavelengths and the expression of particular sets of genes. Exposure to red and green light primarily triggered responses associated with the shade-avoidance syndrome (SAS), such as enhanced elongation of stems, reduced investment in leaf growth, and decreased chlorophyll levels accompanied by the expression of genes encoding histone H3, auxin repressed protein, xyloglucan endotransglycosylase/hydrolase, the ELIP protein, and microtubule proteins. Furthermore, specific light treatments were associated with the expression of a large number of genes, including those involved in the glucan metabolic pathway and the starch and sucrose metabolic pathways; these genes were up/down-regulated in ways that may explain the increase in the starch, sucrose, and total sugar contents in the plants. Moreover, the enhanced root growth and up-regulation of the expression of defense genes accompanied with SAS after exposure to red and green light may be related to the addition of 30 g/L sucrose to the culture medium of plantlets grown in vitro. In contrast, blue light induced the up-regulation of genes related to microtubules, serine carboxypeptidase, chlorophyll synthesis, and sugar degradation and the down-regulation of auxin-repressed protein as well as a large number of resistance-related genes that may promote leaf growth, improve chlorophyll synthesis and chloroplast development, increase the ratio of chlorophyll a (chla)/chlorophyll b (chlb), and decrease the ratio of carbohydrates to proteins in plants. Although exposure to red and green light seems to impose "shade stress" on the plantlets, growth under blue light is comparable to growth observed under white or broad-spectrum light.

Abstract:Efficient photosynthesis is critical for plant survival and growth. When plant-absorbed light exceeds the overall rate of energy conversion, it will trigger photooxidation. In this study, we selected a photooxidation mutant 812HS, it was isolated from the progeny of japonica rice ( Oryza sativa L.) 812S and shows leaf yellowing and hypersensitive to photooxidation. Chloroplast ultrastructure in the leaves of 812HS showed that photooxidation resulted in significant chloroplast damage compared with 812S for changes in gene expressions in response to photooxidation stress using next-generation sequencing technologies on an Illumina HiSeq 2000 platform. A total of 88508 and 88495 genes were identified from 812S and 812HS, respectively. Expressions of 1199 genes were significantly upregulated, while 1342 genes were remarkably downregulated in 812HS. These genes were notably enriched in the 21 KEGG pathways. Based on their expression patterns, several key pathways were identified to be involved in the photooxidation of 812HS. qRT-PCR analysis further confirmed the results of RNA-Seq. This study enabled us to integrate analysis of RNA-Seq in rice and offered a deeper insight into the molecular mechanisms in response to photo-oxidative stress and provided clues for further critical gene identification in the protective mechanisms against photooxidation.

Abstract:The purpose of this study is to determine the effects of red and blue lights on the photomorphogenesis and photosynthetic traits of rice seedlings. The rice seedlings were cultured with red light (R), blue light (B), combined red and blue lights (R3B1/R1B1/R1B3), and white light (CK) as the control. The combined application of red and blue lights could promote the growth of rice seedlings to varying degrees; enhance photosynthesis by increasing the seedling leaf area, chlorophyll content, and chlorophyll fluorescence; improve root characteristics by increasing root number, root volume, and root activity; and thus increase the dry matter accumulation of rice seedlings. In addition, the combination of red and blue lights could regulate the expression of genes related to photosynthesis in rice leaves, affect the activity of the Rubisco enzyme, and then affect the photosynthesis of rice seedlings. These results indicate that red and blue lights have direct synergistic effects, which can regulate the growth of rice seedlings and promote the morphogenesis of rice seedlings. The combined application of red and blue lights can be used to supplement the light in rice-factory seedling raising.
Whole-transcriptome Analysis Reveals Genetic Factors Underlying Flowering Time Regulation in Rapeseed (brassica Napus L.).

Abstract:Rapeseed (Brassica napus L.), one of the most important sources of vegetable oil and protein-rich meals worldwide, is adapted to different geographical regions by modification of flowering time. Rapeseed cultivars have different day length and vernalization requirements, which categorize them into winter, spring, and semiwinter ecotypes. To gain a deeper insight into genetic factors controlling floral transition in B.napus, we performed RNA sequencing (RNA-seq) in the semiwinter doubled haploid line, Ningyou7, at different developmental stages and temperature regimes. The expression profiles of more than 54,000 gene models were compared between different treatments and developmental stages, and the differentially expressed genes were considered as targets for association analysis and genetic mapping to confirm their role in floral transition. Consequently, 36 genes with association to flowering time, seed yield, or both were identified. We found novel indications for neofunctionalization in homologs of known flowering time regulators like VIN3 and FUL. Our study proved the potential of RNA-seq along with association analysis and genetic mapping to identify candidate genes for floral transition in rapeseed. The candidate genes identified in this study could be subjected to genetic modification or targeted mutagenesis and genotype building to breed rapeseed adapted to certain environments.
Simultaneous Application of Red and Blue Light Regulate Carbon and Nitrogen Metabolism, Induces Antioxidant Defense System and Promote Growth in Rice Seedlings under Low Light Stress

Abstract:The purpose of this study is to determine the effect of light quality on growth, carbon and nitrogen metabolism, and antioxidant defense system of rice seedlings. Six light conditions were employed, including white (W), red (R), blue (B), combined LED of R and B at 3:1 (R3B1), combined LED of R and B at 1:1 (R1B1), as well as combined LED of R and B at 1:3 (R1B3). Combined application of red light and blue light could promote the growth of rice seedling leaves and roots under low light stress to varying degrees, increase the photosynthetic area by increasing the leaf area, improve the root characteristics by increasing the root volume, and increase the dry matter accumulation of rice seedlings. In addition, the combination of red light and blue light could increase carbon and nitrogen metabolites in rice seedling leaves, regulate the expression of genes related to carbon and nitrogen metabolism and enzyme activity, and enhance the antioxidant enzyme activity of rice seedlings. These results indicate that red light and blue light directly have synergistic effects which can regulate the carbon and nitrogen metabolism of rice seedlings, promote the morphogenesis of rice seedlings under low light stress, and promote growth, which has never been reported in previous studies. This study is a new discovery in the application of light quality in crop production and provides new avenues to enhance crop stress resistance. However, further study is needed to explore the physio-biochemical and molecular mechanisms of light quality in crop production.

The RNA-seq approach to discriminate gene expression profiles in response to melatonin on cucumber lateral root formation.

Abstract:Cucumber is a model cucurbitaceous plant with a known genome sequence which is important for studying molecular mechanisms of root development. In this study, RNA sequencing was employed to explore the mechanism of melatonin-induced lateral root formation in cucumber under salt stress. Three groups of seeds were examined, that is, seeds primed without melatonin (CK), seeds primed in a solution containing 10 or 500mol/L melatonin (M10 and M500, respectively). These seeds were then germinated in NaCl solution. The RNA-seq analysis generated 16,866,670 sequence reads aligned with 17,920 genes, which provided abundant data for the analysis of lateral root formation. A total of 17,552, 17,450, and 17,393 genes were identified from roots of the three treatments (CK, M10 and M500, respectively). The expression of 121 genes was significantly up-regulated, and 196 genes were significantly down-regulated in M500 which showed an obvious increase on the number of lateral roots. These genes were significantly enriched in 57 KEGG pathways and 16 GO terms (M500 versus CK). Based on their expression pattern, peroxidase-related genes were selected as the candidates to be involved in the melatonin response. Several transcription factor families might play important roles in lateral root formation processes. A number of genes related to cell wall formation, carbohydrate metabolic processes, oxidation/reduction processes, and catalytic activity also showed different expression patterns as a result of melatonin treatments. This RNA-sequencing study will enable the scientific community to better define the molecular processes that affect lateral root formation in response to melatonin treatment.
Comparative Transcriptome Analyses of Gene Response to Different Light Conditions of Camellia oleifera Leaf Using Illumina and Single-Molecule Real-Time-Based RNA-Sequencing

Abstract:Camellia oleifera Abel. is a critical oil tree species. Camellia oil, which is extracted from the seeds, is widely regarded as a premium cooking oil, with the content of oleic acid being over 80%. Light is thought to be one of the largest essential natural components in the regulation of plant developmental processes, and different light qualities can considerably influence plant physiological and phenotypic traits. In this research, we examined the growth and physiological responses of C. oleifera "MIN 43" cultivar plantlets to three different wavelengths of light, containing white, red, and blue light, and we utilized the combination of the PacBio single-molecule real-time (SMRT) and Illumina HiSeq RNA sequencing to obtain the mRNA expression profiles. The results showed that plantlets growing under blue light conditions displayed superior growth performance, including stimulated enhancement of the leaf area, increased leaf number, increased chlorophyll synthesis, and improved photosynthesis. Furthermore, SMAT sequencing created 429,955 reads of inserts, where 406,722 of them were full-length non-chimeric reads, and 131,357 non-redundant isoforms were produced. Abundant differentially expressed genes were found in leaves under different light qualities by RNA-sequencing. Gene expression profiles of actin, dynein, tubulin, defectively organized tributaries 3 (DOT3), and ADP ribosylation factor 5 (ARF5) were associated with the greatest leaf performance occurring under blue light conditions. Moreover, Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis identified hundreds of pathways involved in different light conditions. The pathways of the plant circadian rhythm and hormone signal transduction were associated with different light quality responses in C. oleifera. Phytochrome B (PHYB), constitutively photomorphogenic 1 (COP1), long hypocotyl 5 (HY5), auxin/indole-3-acetic acid (AUX/IAA), Gretchen Hagen 3 (GH3), and small auxin-up RNA (SAUR), which were differentially expressed genes involved in these two pathways, play a vital role in responses to different wavelengths of light in C. oleifera. In addition, blue light significantly promotes flavonoid biosynthesis via changing expression of related genes.

Abstract:Chloroplast-to-nucleus retrograde signalling (RS) is known to impact plant growth and development. In Arabidopsis , we and others have shown that RS affects seedling establishment by inhibiting deetiolation. In the presence of lincomycin, a chloroplast protein synthesis inhibitor that triggers RS, Arabidopsis light-grown seedlings display partial skotomorphogenesis with undeveloped plastids and closed cotyledons. By contrast, RS in monocotyledonous has been much less studied. Here, we show that emerging rice seedlings exposed to lincomycin do not accumulate chlorophyll but otherwise remain remarkably unaffected. However, by using high red (R) and blue (B) monochromatic lights in combination with lincomycin, we have uncovered a RS inhibition of length and a reduction in the B light-induced declination of the second leaf. Furthermore, we present data showing that seedlings grown in high B and R light display different non-photochemical quenching capacity. Our findings support the view that excess B and R light impact seedling photomorphogenesis differently to photoprotect and optimize the response to high-light stress. Abstract:Light, as an energy source for plant photosynthesis, can not only affect the growth and development of plants, but also affect their leaf color. This study used white (WL), red (RL), and blue light (BL) to treat tea cuttings, aiming to investigate the effect of light quality on the color of tea leaves. The results showed that tea leaves turned yellow under red light, the SPAD and Fv/Fm values were significantly lower than WL and BL. Full-length transcriptome was analyzed, photosynthesis and chlorophyll biosynthesis related genes such as PsbS , Psb28 , HemL , and POR had the lowest expression levels under RLCarotenoid biosynthesis related genes ZEP , ABA2 , and CRTISO had the higher expression levels under RL. This study revealed the molecular mechanism of RL induced leaf yellowing in tea plants, providing new insights for the application of light quality in tea plants.

Abstract:Grape (Vitis vinifera L.) is an important fruit for which light quality plays a significant role in plant growth and development. However, the regulatory mechanisms between light quality and plant growth at the physiological and molecular levels are poorly understood. Therefore, in the present study, we performed multivariate sequencing analysis to investigate the effects of four light qualities: blue (B), red (R), green (G), and white (W, used as a control group) to investigate grapevine morphology at the molecular level, which revealed the possible networking pathway controlling grapevine growth. Our results showed that morphological and physiological parameters, such as plant height, stem diameter, leaf area, and the contents of total chlorophyll, chlorophyll a and b, and carotenoid, were significantly improved under blue and red light compared to those under white light. Photosynthetic measurements showed that the total conductance to water vapor, total conductance to water CO2, maximum quantum yield of photosystem (PS)II, apparent electron transfer chain, and net photosynthetic rate were elevated by blue and red light compared to white light. However, there were notable reductions in the intercellular CO2 concentration, transpiration rate, and non-photochemical quenching coefficient under blue light, which showed an irreversible trend compared to other light qualities. The contents of minerals, such as nitrogen, phosphorous, potassium, magnesium, iron, and zinc, were also improved by blue and red light compared to white light. Multivariate sequencing analysis revealed a total of 1065 metabolites, of which 747 were positive and 318 were negative. Gene ontology and Kyoto Encyclopedia of Genes and Genomes analyses showed that many differentially expressed genes (DEGs) were associated with the biosynthesis of secondary metabolites, including flavones, flavonols, and alkaloids, and metabolic and phenylpropanoid pathways. Further, numerous transcription factors, including MYB (37 DEGs), bHLH (32 DEGs), NAC (31 DEGs), and WRKY (29 DEGs), were reported. Photosynthesis-related genes (PsaD, PsaO, PsbB, PetC, PetE, PetF, PetH, PetJ, and Lhca) played pivotal roles in the photosynthetic pathways. In addition, using weighed gene correlation network analysis, 7 module relationships (MEblack, MEgreenyellow, MEred, MEturquoise, MEgreen, MEyellow, and MEmagneta), 4 metabolites, 14 structural genes, and 36 transcription factor-related genes were found. Conclusively, the present research provides new insights for viticulture in response to light quality and offers new strategies for agriculture.
Transcriptome Analysis Reveals that Red and Blue Light Regulate Growth and Phytohormone Metabolism in Norway Spruce [Picea abies (L.) Karst]

Abstract:The mechanisms by which different light spectra regulate plant shoot elongation vary, and phytohormones respond differently to such spectrum-associated regulatory effects. Light supplementation can effectively control seedling growth in Norway spruce. However, knowledge of the effective spectrum for promoting growth and phytohormone metabolism in this species is lacking. In this study, 3-year-old Norway spruce clones were illuminated for 12 h after sunset under blue or red light-emitting diode (LED) light for 90 d, and stem increments and other growth traits were determined. Endogenous hormone levels and transcriptome differences in the current needles were assessed to identify genes related to the red and blue light regulatory responses. The results showed that the stem increment and gibberellin (GA) levels of the seedlings illuminated by red light were 8.6% and 29.0% higher, respectively, than those of the seedlings illuminated by blue light. The indoleacetic acid (IAA) level of the seedlings illuminated by red light was 54.6% lower than that of the seedlings illuminated by blue light, and there were no significant differences in abscisic acid (ABA) or zeatin riboside [ZR] between the two groups of seedlings. The transcriptome results revealed 58,736,166 and 60,555,192 clean reads for the blue-light- and red-light-illuminated samples, respectively. Illumina sequencing revealed 21,923 unigenes, and 2744 (approximately 93.8%) out of 2926 differentially expressed genes (DEGs) were found to be upregulated under blue light. The main KEGG classifications of the DEGs were metabolic pathway (29%), biosynthesis of secondary metabolites (20.49%) and hormone signal transduction (8.39%). With regard to hormone signal transduction, AUXIN-RESISTANT1 (AUX1), AUX/IAA genes, auxin-inducible genes, and early auxin-responsive genes [(auxin response factor (ARF) and small auxin-up RNA (SAUR)] were all upregulated under blue light compared with red light, which might have yielded the higher IAA level. DELLA and phytochrome-interacting factor 3 (PIF3), involved in negative GA signaling, were also upregulated under blue light, which may be related to the lower GA level. Light quality also affects endogenous hormones by influencing secondary metabolism. Blue light promoted phenylpropanoid biosynthesis, phenylalanine metabolism, flavonoid biosynthesis and flavone and flavonol biosynthesis, accompanied by upregulation of most of the genes in their pathways. In conclusion, red light may promote stem growth by regulating biosynthesis of GAs, and blue light may promote flavonoid, lignin, phenylpropanoid and some hormones (such as jasmonic acid) which were related to plant defense in Norway spruce, which might reduce the primary metabolites available for plant growth.
Effect of Supplemental Light for Leaves Development and Seed Oil Content in Brassica napus

Abstract:Rapeseed is an important commercial crop globally, used for both animal fodder and human consumption. Varied insolation duration and intensity are among the main factors affecting the seed yield and quality of Brassica napus (B. napus) worldwide. In this study, the high-oil-content rapeseed cultivar "Qingyou 3" was subjected to a light supplementation trial during both the vegetative growth period and the seed productive stage. Different light intensity conditions were stimulated using light-emitting diodes (LEDs). The main plot factor was land condition, with LED treatment (Treatment) and without LED treatment (Control) under natural conditions. The results showed that the leaf size and thickness, photosynthesis efficiency, and seed oil content of B. napus increased significantly after light supplementation. Then, 18 cDNA libraries were constructed from leaf segments (30 days after transplanting—DAT) and seeds 30 and 40 days after pollination (DPA) for RNA transcriptome sequencing. It was found that genes encoding lipid transfer protein, phenylpropanoid biosynthesis, photosynthesis, and plant hormone signal transduction were enriched in differentially expressed genes (DEGs). The qRT-PCR analysis showed that eight key genes had significant variations, a finding also consistent with the RNA-seq results. The aim of this study was to identify the DEGs and signaling pathways in the leaves and seeds of B. napus during the vegetative and seed productive stages under different light intensities. The results provide insight into how sufficient light plays a critical role in promoting photosynthesis and serves as the foundation for material accumulation and yield formation.

Abstract:Abstract Cucumber ( Cucumis sativus L.) is a typical monoecism vegetable with individual male and female flowers, which has been used as a plant model for sex determination. It is well known that light is one of the most important environmental stimuli, which control the timing of the transition from vegetative growth to reproductive development. However, whether light controls sex determination remains elusive. To unravel this problem, we performed high-throughput RNA-Seq analyses, which compared the transcriptomes of shoot apices between R2B1(Red light:Blue light = 2:1)-treated and R4B1(Red light:Blue light = 4:1)-treated cucumber seedlings. Results showed that the higher proportion of blue light in the R2B1 treatment significantly induced the formation of female flowers and accelerated female flowering time in this whole study. The genes related to flowering time, such as flowering locus T ( FT ) and SUPPRESSOR OF OVEREXPRESSION OF CO1 ( SOC1 ), were up-regulated after R2B1 treatment. Furthermore, the transcriptome analysis showed that up-regulation and down-regulation of specific DEGs (the differentially expressed genes) were primarily the result of plant hormone signal transduction after treatments. The specific DEGs related with auxin formed the highest percentage of DEGs in the plant hormone signal transduction. In addition, the expression levels of transcription factors also changed after R2B1 treatment. Thus, sex differentiation affected by light quality might be induced by plant hormone signal transduction and transcription factors. These results provide a theoretical basis for further investigation of the regulatory mechanism of female flower formation under different light qualities in cucumber seedlings.
