Capturing the Conformational Heterogeneity of the Β-Amyloid Peptide Sequence Using the Engineered Aerolysin Confinement

Abstract:The accumulation of β-amyloid peptides is a widely recognized key factor in the pathophysiology of Alzheimer's disease. The dysregulation of metal homeostasis and post-translational modifications (PTMs) can disrupt the normal folding and clearance of amyloid-forming proteins, making them more prone to aggregation. However, differ from well-folded peptide, β-amyloid peptide is classified as an intrinsically disordered protein (IDP), making it challenging to study using conventional methods. Herein, we used engineered aerolysin confinement to capture the conformation heterogeneity of an intrinsically disordered β-amyloid peptide fragment at the single-molecule level. By monitoring the fast ionic current responses, the multiple dynamic conformations of the β-amyloid peptide fragment were snapshotted. The stochastical results of current blockages illustrated that the structural flexibility of β-amyloid peptide is largely enhanced after treating with metal ions, resulting in new conformations. The impact of sequence changes with PTMs is also studied. The various distribution of current blockages revealed that the serine phosphorylation (S8) and tyrosine nitration (Y10) could change the structural flexibility of β-amyloid peptide fragment and result in different conformations. These findings highlight the advantage of our approach in elucidating the conformational heterogeneity of IDPs, that might be not detected by conventional averaging measurement techniques.
