Abstract:In the field of contemporary medicine, inflammation has emerged as a significant concern in global public health. Among the current anti-inflammatory strategies, nanozymes possess distinctive advantages and demonstrate unexpected efficacy in combating inflammation. However, the indeterminate structures and limited enzyme-like activity exhibited by most developed nanozymes impede their clinical translation and therapeutic effectiveness. In this paper, we developed a nanozyme derived from a well-defined metal-organic cage (MOC). The oxidized MOC (MOC-O), containing pyridine nitrogen oxide moieties, exhibited effective cascade superoxide dismutase (SOD) and catalase (CAT)-like activities for scavenging reactive oxygen species (ROS). This ROS scavenging ability was confirmed through flow cytometry analysis using DCFH-DA in a hypoxia/reoxygenation (H/R) model, where MOC-O significantly alleviated oxidative stress. Furthermore, the administration of MOC-O resulted in preserved renal function during renal ischemia-reperfusion (I/R) injury due to downregulated oxidative stress levels and reduced cell apoptosis.

Abstract:Cascade biocatalytic reactions involving multiple antioxidative enzymes are necessary in living cells to regulate cellular metabolism and redox homeostasis. Substantial efforts have been made to construct cascade reactions through engineered enzyme mimics to improve intracellular metabolic flux, especially under pathophysiological conditions. Here, we show that MoS2 nanozymes exhibit activities of four major cellular cascade antioxidant enzymes, including superoxide dismutase, catalase, peroxidase, and glutathione peroxidase. Meanwhile, MoS2 nanozymes attenuate electron transfer in cytochrome c/H2O2 to ameliorate the inherent antioxidant defense system under stress conditions. Thus, MoS2 nanozymes function as a self-cascade platform to inhibit intracellular reactive oxygen species (ROS) production by modulating mitochondrial function and scavenging abundant ROS through their intrinsic antioxidant capacity. Density functional theory calculations reveal the underlying mechanisms of the intracellular environment-dependent catalase-like activity of MoS2 nanozymes. Furthermore, we find that the MoS2 nanozymes play a cytoprotective role in cells and significantly improve the treatment outcomes in a hepatic fibrosis mouse model. These results demonstrate the ROS-scavenging capacity of a single-component MoS2 nanozyme-based cascade reaction system and reveal the in-depth mechanism, which may advance the development of nanozyme-based antioxidative agents.
Spatially Organized Functional Bioreactors in Nanoscale Mesoporous MOFs for Cascade Scavenging of Intracellular ROS

Abstract:The creation of artificial microenvironments with specific functionalities to mimic multiple cascade biocatalytic processes in cells is highly desirable. Here, Zr-based large-pore mesoporous metal-organic frameworks (mesoMOFs) were successfully developed for the construction of such spatially organized cascade bioreactors. Thanks to the mediating effect of the Hofmeister ion and the flexibility of Zr-MOFs, large mesopores and functional microporous mesopore walls were integrated into a single nanoplatform, ensuring the efficient mass exchange between the interconnected pore units with different sizes. As a proof of concept, natural superoxide dismutase and catalase mimic were spatially positioned in these hierarchical mesoMOFs, which formed a cascade antioxidant defense system and presented an efficient intracellular reactive oxygen species scavenging ability. This model is expected to set a conceptual guideline for the design of compartmentalized bioreactors in nanoscale MOFs to mediate the complex intracellular catalytic networks.

Abstract:Abstract Nanozymes play a pivotal role in mitigating excessive oxidative stress, however, determining their specific enzyme-mimicking activities for intracellular free radical scavenging is challenging due to endo-lysosomal entrapment. In this study, we employed a genetic engineering strategy to generate ionizable ferritin nanocages (iFTn), enabling their escape from endo-lysosomes and entry into the cytoplasm. Specifically, ionizable repeated Histidine-Histidine-Glutamic acid (9H2E) sequences were genetically incorporated into the outer surface of human heavy chain FTn, followed by the assembly of various chain-like nanostructures via a two-armed polyethylene glycol (PEG). Utilizing endosome-escaping ability, we designed iFTn-based tetrameric cascade nanozymes with high superoxide dismutase- and catalase-mimicking activities. The in vivo protective effects of these ionizable cascade nanozymes against cardiac oxidative injury were demonstrated in mouse models of cardiac ischemia-reperfusion (IR). RNA-sequencing analysis highlighted the crucial role of these nanozymes in modulating superoxide anions-, hydrogen peroxide- and mitochondrial functions-relevant genes in IR injured cardiac tissue. These genetically engineered ionizable protein nanocarriers provide opportunities for developing ionizable drug delivery systems.

Abstract:The pursuit of single-assembled molecular cage reactors for complex tandem reactions is a long-standing target in biomimetic catalysis but still a grand challenge. Herein, nanozyme-like organic cages are reported by engineering air-stable radicals into the skeleton upon photoinduced electron transfer. The generation of radicals is accompanied by single-crystal structural transformation and exhibits superior stability over six months in air. Impressively, the radicals throughout the cage skeleton can mimic the peroxidase of natural enzymes to decompose H2O2 into OH center dot and facilitate oxidation reactions. Furthermore, an integrated catalyst by encapsulating Au clusters (glucose oxidase mimics) into the cage has been developed, in which the dual active sites (Au cluster and radical) are spatially isolated and can work as cascade nanozymes to prominently promote the enzyme-like tandem reaction via a substrate channeling effect.

Abstract:Development of enzyme mimics for the scavenging of excessive mitochondrial superoxide (O2•−) can serve as an effective strategy in the treatment of many diseases. Here, protein reconstruction technology and nanotechnology is taken advantage of to biomimetically create an artificial hybrid nanozyme. These nanozymes consist of ferritin‐heavy‐chain‐based protein as the enzyme scaffold and a metal nanoparticle core as the enzyme active center. This artificial cascade nanozyme possesses superoxide dismutase‐ and catalase‐like activities and also targets mitochondria by overcoming multiple biological barriers. Using cardiac ischemia‐reperfusion animal models, the protective advantages of the hybrid nanozymes are demonstrated in vivo during mitochondrial oxidative injury and in the recovery of heart functionality following infarction via systemic delivery and localized release from adhesive hydrogels (i.e., cardiac patch), respectively. This study illustrates a de novo design strategy in the development of enzyme mimics and provides a promising therapeutic option for alleviating oxidative damage in regenerative medicine.

Abstract:Cellular redox homeostasis is highly dependent on endogenous antioxidant systems, including superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx). The nanozymes mimicking the catalytic activities of natural antioxidant systems serve as potential candidates for the therapy of reactive oxygen species (ROS)-related diseases. However, previously reported metal-containing nanozymes possessed unsatisfactory catalytic activity and inevitable peroxidase activity, limiting their translational applications. Therefore, inspired by the superior ROS scavenging activities of natural polyphenols and metal-free carbon dots (CDs) with ultra-small size, phenol-functionalized CDs that mimic endogenous antioxidant system have been fabricated. The SOD activity was determined at an extremely high level (18187 U/mg), which was substantially comparable to natural SOD. Notably, this study reveals for the first time that CDs, as selenium-free nanomaterials, show robust GPx activity in vitro . The theory calculation suggests that the bond dissociation of phenols on CDs is considerably lower than selenocysteine of natural GPx, which facilitates the formation of peroxide intermediate to initiate GPx-like reactions. Furthermore, the kidney-targeting and intracellular antioxidant mimicking properties contribute to the effective alleviating cisplatin-induced acute kidney injury via suppressing oxidative stress-mediated ferroptosis in vivo . These results provide an effective polyphenol-functionalized strategy for synthesizing metal-free multifunctional antioxidant nanozyme to treat ROS-related diseases.

Abstract:Application of antioxidant enzymes in medical or industrial processes is limited due to their high sensitivity to environmental conditions. Incorporation of such enzymes in nanostructures provides a promising route to obtain highly efficient and robust biocatalytic system to scavenge reactive oxygen species (ROS). Here, this question was addressed by confinement of superoxide dismutase (SOD), horseradish peroxidase (HRP), and catalase (CAT) enzymes into nanostructures containing polyelectrolyte building blocks (alginate (Alg) and trimethyl chitosan (TMC)) and delaminated layered double hydroxide (dLDH) nanoparticle support. The nanocomposite possessed excellent structural and colloidal stability, while antioxidant tests revealed that the enzymes remained active upon immobilization and the developed composite greatly reduced intracellular oxidative stress in two-dimensional cell cultures. Moreover, it effectively prevented hydrogen peroxide-induced double stranded DNA breaks, which is a common consequence of oxidative stress. The results provide important tools to design complex nanostructures with multienzymatic antioxidant activities for ROS scavenging.

Abstract:Cascade nanoreactors (NRs) which can be activated in tumor microenvironment and catalyze the local substance to toxic species for killing cancer cells is an appealing emerging agent for cancer treatment in recent years. Peroxidase-like nanozymes are often integrated into such kind of systems for cancer therapy. In this study, magnetic metal-organic framework (MOF: zeolitic imidazolate framework 8, ZIF-8) nanoparticles (NPs) with ferrimagnetic structures inside are fabricated via the mineralization of iron during the growth of ZIF-8. The Fe-base magnetic structures can serve as peroxidase-like nanozyme. After the loading of GOx on the magnetic MOF NPs, magnetic cascade MOF/enzyme hybrid NRs (Fe@ZIF-8@GOx NRs) are successfully constructed. In the presence of glucose, the NRs can perform high cascade reaction activity to produce hydroxyl radicals center dot OH) under mild acidic condition at physiological temperature. The NRs can be more internalized by cancer cells under a magnetic field and take advantage of acidic environment in lysosomes to produce high toxic center dot OH by cascade reaction to kill cancer cells. Magnetic targeting to kill cancer cells at the specified location can be achieved by applying a magnetic field to the targeted area and then damage to normal cells will be minimized.

Abstract:Rationale: Myocardial injury triggers intense oxidative stress, inflammatory response, and cytokine release, which are essential for myocardial repair and remodeling. Excess reactive oxygen species (ROS) scavenging and inflammation elimination have long been considered to reverse myocardial injuries. However, the efficacy of traditional treatments (antioxidant, anti-inflammatory drugs and natural enzymes) is still poor due to their intrinsic defects such as unfavorable pharmacokinetics and bioavailability, low biological stability, and potential side effects. Nanozyme represents a candidate to effectively modulate redox homeostasis for the treatment of ROS related inflammation diseases. Methods: We develop an integrated bimetallic nanozyme derived from metal-organic framework (MOF) to eliminate ROS and alleviate inflammation. The bimetallic nanozyme (Cu-TCPP-Mn) is synthesized by embedding manganese and copper into the porphyrin followed by sonication, which could mimic the cascade activities of superoxide dismutase (SOD) and catalase (CAT) to transform oxygen radicals to hydrogen peroxide, followed by the catalysis of hydrogen peroxide into oxygen and water. Enzyme kinetic analysis and oxygen-production velocities analysis were performed to evaluate the enzymatic activities of Cu-TCPP-Mn. We also established myocardial infarction (MI) and myocardial ischemia-reperfusion (I/R) injury animal models to verify the ROS scavenging and anti-inflammation effect of Cu-TCPP-Mn. Results: As demonstrated by kinetic analysis and oxygen-production velocities analysis, Cu-TCPP-Mn nanozyme possesses good performance in both SOD- and CAT-like activities to achieve synergistic ROS scavenging effect and provide protection for myocardial injury. In both MI and I/R injury animal models, this bimetallic nanozyme represents a promising and reliable technology to protect the heart tissue from oxidative stress and inflammation-induced injury, and enables the myocardial function to recover from otherwise severe damage. Conclusions: This research provides a facile and applicable method to develop a bimetallic MOF nanozyme, which represents a promising alternative to the treatment of myocardial injuries.

Abstract:Aim: To target the reactive oxygen species (ROS) accumulation and renal tubular epithelial cell (rTEC) death in renal ischemia-reperfusion injury (IRI), we constructed a nanoparticle that offers ROS scavenging and rTEC-death inhibition: mesoporous zinc-tannic acid nanozyme (ZnTA). Materials & methods: After successfully constructing ZnTA, we proceeded to examine its effect on ROS accumulation, cellular ferroptosis and apoptosis, as well as injury severity. Results: Malondialdehyde, Fe2+ amounts and 4-HNE staining demonstrated that ZnTA effectively attenuated rTEC ferroptosis. TUNEL staining confirmed that Zn2+ carried by ZnTA could effectively inhibit caspase 3 and caspase 9, mitigating apoptosis. Finally, it reduced renal IRI through the synergistic effect of ROS scavenging and cell-death inhibition. Conclusion: This study is expected to provide a paradigm for a combined therapeutic strategy for renal IRI.

Abstract:Abstract A broad-spectrum reactive oxygen species (ROS)-scavenging hybrid material (CASCADE) was developed by sequential adsorption of heparin (HEP) and poly(L-lysine) (PLL) polyelectrolytes together with superoxide dismutase (SOD) and horseradish peroxidase (HRP) antioxidant enzymes on layered double hydroxide (LDH) nanoclay support. The synthetic conditions were optimized so that CASCADE possessed remarkable structural (no enzyme leakage) and colloidal (excellent resistance against salt-induced aggregation) stability. The obtained composite was active in decomposition of both superoxide radical anions and hydrogen peroxide in biochemical assays revealing that the strong electrostatic interaction with the functionalized support led to high enzyme loadings, nevertheless, it did not interfere with the native enzyme conformation. In vitro tests demonstrated that ROS generated in human cervical adenocarcinoma cells were successfully consumed by the hybrid material. The cellular uptake was not accompanied with any toxicity effects, which makes the developed CASCADE a promising candidate for treatment of oxidative stress-related diseases.

Abstract:Oxidative stress is associated with many acute and chronic inflammatory diseases. Development of nanomaterial-based enzyme mimetics for reactive oxygen species (ROS) scavenging is challenging, but holds great promise for the treatment of inflammatory diseases. Herein, we report the highly ordered manganese dioxide encapsulated selenium-melanin (Se@Me@MnO2) nanozyme with high efficiency for intracellular antioxidation and anti-inflammation. The Se@Me@MnO2 nanozyme is sequentially fabricated through the radical polymerization and the in-situ oxidation-reduction. In vitro experimental results demonstrated that the Se@Me@MnO2 nanozyme exhibits multiple enzyme activities to scavenge ROS, including catalase (CAT), glutathione peroxidase (GPx) and superoxide dismutase (SOD). Mechanism researches illustrated that the Se core possesses GPx-like catalytic activity, the Me and the MnO2 possess both the SOD-like and the CAT-like activities. What’s more, due to the stable unpaired electrons existing in the nanozyme, the Se, Me and MnO2 provide synergistic and fast electron transfer effect to achieve the quickly scavenging of hydrogen peroxide, hydroxyl radical, and superoxide anion. Further in vivo experimental results showed that this biocompatible nanozyme exhibits cytoprotective effects by resisting ROS-mediated damage, thereby alleviating the inflammation. This multienzyme mimetics is believed to be an excellent ROS scavenger and have a good potential in clinical therapy for ROS-related diseases.

Abstract:Significant advances have been made in recent years for the utilization of natural enzymes with antioxidant properties to treat acute kidney injury (AKI). However, these enzymes have been of limited clinical utility because of their limited cellular uptake, poor pharmacokinetic properties, and suboptimal stability. We employed a novel biomimetic mineralization approach to encapsulate catalase (CAT) and superoxide dismutase (SOD) in a zeolitic imidazolate framework-8 (ZIF-8). Next, this SOD@CAT@ZIF-8 complex was anchored with MPEG 2000 -COOH to yield an MPEG 2000 -SOD@CAT@ZIF-8 (PSCZ) composite. The composite was then used as a stable tool with antioxidant properties for the integrated cascade-based treatment of AKI, remarkably improved intracellular enzyme delivery. This dual-enzyme-embedded metal-organic framework could effectively scavenge reactive oxygen species. In conclusion, the ZIF-8-based “armor plating” represents an effective means of shielding enzymes with improved therapeutic utility to guide the precision medicine-based treatment of AKI.

M-TA NMs show adjustable broad-spectrum antioxidative activities toward multiple reactive nitrogen and oxygen species (RNOS) due to the adjustable catalytic active centers. Importantly, M-TA NMs could mimic the cascade processes of superoxide dismutase and catalase to maintain intracellular redox balance. Detailed structural and spectral analyses reveal that the existence of a transition metal could decrease the electronic energy band gaps of M-TA NMs to offer better electron transfers for RNOS scavenging. Notably, dynamic blood experiments demonstrate that Cu-TAn@PMS could serve as an antioxidant defense system for blood in hemoperfusion to scavenge intracellular reactive oxygen species (ROS) effectively even in the complex blood environment and further protect endogenous antioxidative enzymes and molecules. In general, this work developed antioxidative cascade nanozymes engineered microspheres with excellent therapeutic efficacy for the treatment of oxidative stress-related diseases, which exhibited potential for clinical blood purification and extended the biomedical applications of nanozymes.

Abstract:Reactive oxygen species (ROS) play important roles in regulating various physiological functions in the human body, however, excessive ROS can cause serious damage to the human body, considering the various limitations of natural enzymes as scavengers of ROS in the body, the development of better materials for the scavenging of ROS is of great significance to the biomedical field, and nanozymes, as a kind of nanomaterials which can show the activity of natural enzymes. have a good potential for the development in the area of ROS scavenging. Metal-organic frameworks (MOFs), which are porous crystalline materials with a periodic network structure composed of metal nodes and organic ligands, have been developed with a variety of active nanozymes including catalase-like, superoxide dismutase-like, and glutathione peroxidase-like enzymes due to the adjustability of active sites, structural diversity, and excellent biocompatibility, and they have shown a wide range of applications and prospects. The present review, we firstly introduce three representative natural enzymes for ROS scavenging in the human body, methods for the detection of relevant enzyme-like activities and mechanisms of enzyme-like clearance are discussed, meanwhile, we systematically summarize the progress of the research on MOF-based nanozymes, including the design strategy, mechanism of action, and medical application, etc. Finally, the current challenges of MOF-based nanozymes are summarized, and the future development direction is anticipated. We hope that this review can contribute to the research of MOF-based nanozymes in the medical field related to the scavenging of ROS.

Integrating Pt Nanoparticles with Carbon Nanodots to Achieve Robust Cascade Superoxide Dismutase-Catalase Nanozyme for Antioxidant Therapy

Abstract:Nanozymes, nanomaterials with intrinsic enzyme mimetic activity, have emerged and been used in a broad range of applications yet are constrained with limited catalytic efficiency. Herein, by integrating carbon nanodots (CNDs) with platinum nanoparticles (PtNPs), a novel Pt@CNDs nanocomposite was engineered as an efficient nanozyme with superoxide dismutase (SOD)-and catalase (CAT)-like specific activities of 12,605 U/mg and 3172 U/mg, respectively. The PtNPs were deposited on the surface of CNDs by in situ chemical decomposition of Pt4+ to produce Pt@CNDs, in which CNDs served as a SOD mimic while PtNPs could mimic CAT, thereby forming a cascade antioxidant nanozyme system. Of note, the Pt@CNDs could significantly scavenge hydroxyl radicals as well. The carbonyl and hydroxyl groups of CNDs could bind with PtNPs, which promoted the electron transfer between PtNPs and CNDs that endowed the Pt@CNDs with excellent catalytic performance. Furthermore, Pt@CNDs could enter living cells and target mitochondria, thereby reducing the upregulated reactive oxygen species (ROS) level. Moreover, in vivo experimental re-sults indicated that Pt@CNDs could effectively relieve ROS-induced inflammation in living mice.
