Acquisition of Information Encoded in Poly(phosphoester)s by a Biological Nanopore

Abstract:In the era of data explosion, there is an urgent need for new storage media with ultra-high storage density and long-term stability. Poly(phosphoester)s (PPEs) have the advantages of chemical stability and monomer diversity, meet the requirements of new storage media. In this study, we designed eight different monomers as the storage units and used their sequences in the PPE polymer to store information. Nowadays, nanopore techniques have been developed as a promising sequencing method for both the natural and the nonnatural polymers, with the merits of low cost, long read, and high throughput. Herein, we tailored a biological nanopore of matching sensing volume with the informational PPE chain, which can quickly and accurately measure the polymer and read the information. By using the tailored nanopore, we obtain the electrical signals of single poly(phosphoester)s and then decode the information based on their monomers sequence. This work demonstrated that a higher theoretical storage density than natural DNA can be achieved by using the abiotic PPEs, and a nanopore sequencing method feasible for accurately and efficiently decoding the nonnatural polymers would promote the application of this new storage media. These proof-of-concept experiments offer new insights for improving the performance of polymer-based data storage. We believe that the rapid development of nanopore techniques for the informational polymer sequencing will pave the way towards the high-density and low-cost information storage technology.
