Studies on Identification of Putative SSR Markers Linked with Hardseededness in Mung Bean (Vigna radiata L.) Using Bulk Segregant Analysis

Abstract:Hardseededness is a widely spread trait across mung bean (green gram) genotypes and among the released varieties in India and beyond. Larger proportion of hard seeds at harvest lowers the grain quality and impairs in proper cooking and digestion. If used as seed there is a risk of reduced yield, due to uneven germination and poor crop stand. In the present study identification of putative SSR markers linked with hardseededness in mung bean using Bulk Segregant Analysis was attempted. The predominantly hard seeded (TM 96/25) and one predominantly non-hard seeded genotype (IPM 02/3) were used as the parent to make the cross and further analysis. The phenotyping and trait stability was assessed for four seasons before the crosses were made and a F 2 population of 727 plants was examined for the study. The PCR result showed that among 144 SSR markers; 17 were polymorphic between the two parents. In bulk DNA made from hard and non hard seeded parents in F 2 generation 2 SSR markers (viz. Vr-1-550 and LOC-106-768) were found polymorphic among the identified 17 polymorphic SSR markers. These two putative SSR markers showing polymorphism in bulk DNA of segregating plants confirmed its tight linkage to hardseededness trait and may be used for early identification of hard seeded genotypes for further use in breeding programme.
