Abstract:In vitro micrografting is an important technique supporting the micropropagation of a range of plant species, particularly woody plant species. Over the past several decades, in vitro micrografting has become a strategy to facilitate shoot recovery and acclimatization of in vitro-grown horticultural species. This review focuses on studies on horticultural crops over the past two decades that cover the establishment of in vitro micrografting, discusses factors affecting the success of in vitro micrografting, and provides commentary on the contribution of micrografting applications to the field of micropropagation. Considering the important roles of micrografting in the restoration of vigor and rooting competence, in promotion of shoot recovery following somatic embryogenesis and organogenesis, and in facilitation of shoot regrowth after cryopreservation, the potential use of this technique in facilitation of genetic engineering and safe conservation of horticultural species are specially highlighted.

Abstract:Plant tissue culture technology has introduced a new phase into plant multiplication and is being increasingly utilized in plant breeding. This paper reviews the current and developing biotechnological applications of invitro methods for plant improvement and outlines the present state of the art of the regeneration of plants from cells and tissues. Also, the problems associated with the transfer of tissue culture derived plantlets to soil conditions are addressed. Methods are described for the invitro acclimatization of cultured plantlets.

Abstract:Several tissue culture techniques can be used to generate genetic variation in plants. These in vitro procedures have been applied successfully in many species and have been used to create plants that are disease and insect resistant and tolerant to water stress and demonstrate enhanced growth and improved yields and much more. Traditional plant breeding is enhanced with techniques such as embryo culture to rescue recalcitrant hybrids. Technologies as protoplast fusion and somaclonal variation are used to exploit existing genetic diversity or to produce new genetic variability. Once new plants are developed, micropropagation can be used to efficiently clonally propagate large numbers for the commercial market. Genetic engineering would not be possible without the involvement of in vitro plant tissue culture regeneration procedures.

Abstract:Today, the conservation of ornamental germplasm can take advantage of innovative techniques which allow preservation in vitro (slow growth storage) or in liquid nitrogen (cryopreservation) of plant material. Slow growth storage refers to the techniques enabling the in vitro conservation of shoot cultures in aseptic conditions by reducing markedly the frequency of periodic subculturing, without affecting the viability and regrowth of shoot cultures. Cryopreservation refers to the storage of explants from tissue culture at ultra-low temperature (-196 degrees C). At such temperature, all the biological reactions within the cells are hampered, hence the technique makes available the storage of plant material for theoretically unlimited periods of time. An exhaustive review of papers dealing with the slow growth storage and the cryopreservation of ornamental species is reported here. Step-by-step protocols for the slow growth storage of rose germplasm, the production of synthetic seeds for the in vitro conservation of ornamentals, and the cryopreservation of Chrysanthemum morifolium are included.

Abstract:For horticulture to meet the needs of global markets, the reproduction and breeding of ornamental plants is allimportant. Nevertheless, certain ornamental plants exhibit a lower capacity for explant regeneration when compared to model plants. These challenges hinder the rapid propagation, virus-free breeding, and molecular breeding of ornamental plants. This paper examines both traditional and emerging plant regeneration technologies and discusses the difficulties ornamental plants encounter during the regeneration process. It also provides an outlook on the applications of emerging technologies in ornamental plant regeneration. This study will provide insights into the industrialization and practical application of molecular tools in ornamental plant breeding.

Abstract:Plant tissue culture technique employed for the identification and isolation of bioactive phytocompounds has numerous industrial applications. It provides potential benefits for different industries which include food, pharmaceutical and cosmetics. Various agronomic crops i.e., cereals, fruits, vegetables, ornamental plants and forest trees are currently being used for in vitro propagation. Plant tissue culture coupled with biotechnological approaches leads towards sustainable agricultural development providing solutions to major food security issues. Plants are the rich source of phytochemicals with medicinal properties rendering them useful for the industrial production of pharmaceuticals and nutraceuticals. Furthermore, there are numerous plant compounds with application in the cosmetics industry. In addition to having moisturizing, anti-ageing, anti-wrinkle effects; plant-derived compounds also possess pharmacological properties such as antiviral, antimicrobial, antifungal, anticancer, antioxidant, anti-inflammatory, and anti-allergy characteristics. The in vitro propagation of industrially significant flora is gaining attention because of its several advantages over conventional plant propagation methods. One of the major advantages of this technique is the quick availability of food throughout the year, irrespective of the growing season, thus opening new opportunities to the producers and farmers. The sterile or endangered flora can also be conserved by plant micro propagation methods. Hence, plant tissue culture is an extremely efficient and cost-effective technique for biosynthetic studies and bio-production, biotransformation, or bioconversion of plant-derived compounds. However, there are certain limitations of in-vitro plant regeneration system including difficulties with continuous operation, product removal, and aseptic conditions. For sustainable industrial applications of in-vitro regenerated plants on a large scale, these constraints need to be addressed in future studies.

Abstract:Shipping of in vitro micro-cuttings in tubes or jars is a frequently used method as the plants are more likely to quickly reproduce and comply with quarantine regulations in plant germplasm distribution. However, these containers are fragile during transportation. To diminish the risk associated with the long-distance shipping of in vitro plants, a safe and widely applicable packing and conservation technique based on microplate and slow growth was developed in this study. Potato cultivar ZHB and ginger cultivar G-2 were used to optimize the system with microplates (96 wells), vacuum-sealed packaging, and slow-growth techniques. Under regular culture conditions, packing in vacuum-sealed microplates reduced the survival of ZHB and G-2 micro-cuttings to 85.8% and 20.0%, respectively, and regeneration to 61.8% and 0%, respectively. Reducing the temperature to 10 °C maintained the survival of ZHB and G-2 micro-cuttings in the range of 83.3–100% after 60 days. Exposure to darkness decreased the survival of G-2 and inhibited regrowth. Thus, conservation in darkness at 10 °C is suggested. The effects of iron concentration and plant growth retardants were further assessed. The addition of 1/4 MS medium combined with 100 mg/L chlormequat chloride (CCC) resulted in full survival and growth inhibition of plantlets, without malformation identified. Finally, incubation with 1/4 MS medium supplemented with 100 mg/L CCC in vacuum-sealed microplates at 10 °C in the dark resulted in high survival and suppressed germination. Sweet potato HXS was incubated as well to test the broad-spectrum applications of the technique; 100% survival and 6.7% germination was gained. Morphological indices of released cuttings recovered to control levels after two cycles of subculture in MS medium. A 0.1–0.2% genetic variation was detected by SSR and ISSR, suggesting genetic stability of the conserved samples. Finally, micro-cuttings were safely transported to cities located thousands of kilometers away without package and sample damage. Our results enable easy distribution of in vitro plant germplasms.

Abstract:Cotton (Gossypium hirsutum L.) is an important oilseed and fiber crop. Advanced biotechnology not only provides an innovation method for cotton breeding and germplasm multiply but also greatly accelerates the process of cotton breeding. The key to successful application of biotechnology in plant breeding is the establishment of an efficient regeneration system. The techniques of tissue culture and regeneration for cotton plants can be applied to cotton variety improvement, propagation and preservation of germplasm, inheritance of heterosis and genetic transformation. Therefore, in recent years, there has been increasing focus on the research of cotton tissue culture as the source of tissue explants for transforming cotton. In this paper, the recent progress on research of cotton tissue culture was reviewed including somatic cell culture, anther culture, shoot-tip culture, in vitro ovule culture and protoplast culture, and the future research works of this field were discussed.

Abstract:Purpose. Analysis of plant micropropagation technologies for the creation of viable interspecific hybrids and varieties of Actinidia Lindl. Methods. General scientific – hypothesis, experiment, observation, analysis, synthesis method for drawing conclusions. Results. The introduction of in vitro technologies is now becoming the dominant commercial method of large-scale and rapid production of seedlings with stable inheritance of variety traits, high multiplication rate, preservation of economically valuable traits in the absence of production seasonality and time constraints. In addition to reproduction, the breeding process is also accelerated, including mutagenesis and hybridization. It is important to obtain not only a sterile explant, but also a morphogenically active one, that is, a plant that takes roots and subsequently regenerates in vitro. The best in terms of decontamination efficiency is the method of treatment with hypochlorite and the addition of PPM biocide to the nutrient medium, but under these conditions, the lowest survival of explants in all samples was noted. The efficiency of introduction into aseptic culture at the first stage of micropropagation is also affected by the biological characteristics of the primary explants. In studies with nutrient media for A. arguta, it was found that of the elements of mineral nutrition, only 11 ions are necessary for life: five macro- (N, K, P, Mg, S) and six microelements (Cl, Fe, B, Mo, Na, I). Plants in vitro have a lower dry matter content and a greater amount of moisture, including free moisture, which is quickly lost when the water balance is disturbed. Conclusions. The ability to regenerate is more pronounced in the species A. chinensis and A. deliciosa, and to a lesser extent in A. arguta. For A. chinensis, the use of hydroponic technology for the adaptation of regenerants at the ex vitro stage is effective.

Abstract:Agronomic crops are responsible for the majority of the world’s food supply, either through direct consumption or as animal feed. Until recently, however, the importance of these crops has not been reflected by research efforts and progress in biotechnology. Many agronomic crops have been difficult to manipulate in vitro, and regeneration has been problematic. Currently, progress in regeneration has been significant, such that most crops can now be regenerated. For example, it is estimated that over 130 species of angiosperms and gymnosperms have been induced to form somatic embryos (Thorpe, 1988). As a result of progress in regeneration, a number of laboratories have reported success in stable transformation of many important agronomic crops (Deak et al., 1986; Everett et al., 1987; Firoozabady et al., 1987; Chabaud et al., 1988; Hinchee et al., 1988; Zhang et al., 1988; Gasser & Fraley, 1989). There are numerous applications of biotechnology to crop improvement, such as production of doubled haploids to produce homozygous plants rapidly, propagation of genetic marker stocks, clonal propagation for genetic and physiological studies, induction of variation, mutagenesis and cell selection for specific traits, protoplast fusion for wide hybridization, and genetic transformation. For this review, the focus is micropropagation.

Abstract:Plant tissue culture is an important agricultural biotechnological tool that contributes in the production of crops with improved food, fiber, fuel, and feed. It is one way toward commercialization to face the food availability challenge in developing countries and allow them to cope with their fast-growing population in a restricted area of land. In addition, plant tissue culture enables some rare and nearly extinct plant species to be rescued and propagated. Conventional methods of propagation thus need to be supplemented with modern breeding techniques. In this way, higher levels of agriculture, afforestation, plant improvement as well as in vitro production of metabolites and plant secondary products can be reached and fulfilled on a year-round basis and under disease-free conditions. The main applications of plant tissue culture in the agricultural field, plant micropropagation, inducing new varieties and constrains of plant tissue culture and challenges this technique is facing as an industry helping the agricultural field, are discussed in this chapter.

Abstract:Cryopreservation is considered an ideal strategy for the long-term preservation of plant genetic resources. Significant progress was achieved over the past several decades, resulting in the successful cryopreservation of the genetic resources of diverse plant species. Cryopreservation procedures often employ in vitro culture techniques and require the precise control of several steps, such as the excision of explants, preculture, osmo- and cryoprotection, dehydration, freeze-thaw cycle, unloading, and post-culture for the recovery of plants. These processes create a stressful environment and cause reactive oxygen species (ROS)-induced oxidative stress, which is detrimental to the growth and regeneration of tissues and plants from cryopreserved tissues. ROS-induced oxidative stresses were documented to induce (epi)genetic and somatic variations. Therefore, the development of true-to-type regenerants of the source germplasm is of primary concern in the application of plant cryopreservation technology. The present article provides a comprehensive assessment of epigenetic and genetic integrity, metabolic stability, and field performance of cryopreserved plants developed in the past decade. Potential areas and the directions of future research in plant cryopreservation are also proposed.

Abstract:The growing demand for native planting material in ecological restoration and rehabilitation for agro-silvo-pastoral ecosystems has resulted in a major global industry in their sourcing, multiplication, and sale. Plant tissue culture is used for producing high-quality, disease-free, and true-to-type plants at a fast rate. Micropropagation can help to meet the increasing demand for planting material and afforestation programs. However, in vitro plant propagation is an expensive technique compared to conventional methods using suckers, seeds, and cuttings. Therefore, adopting measures to lower production costs without compromising plant quality is essential. This can be achieved by improving the culture media composition. Incorporating organic growth additives can stimulate tissue growth and increase the number of shoots, leaves, and roots in culture media. Organic growth supplementation speeds up the formation and development of cultures and yields vigorous plants. Plant regeneration from meristems (shoot tips and axillary buds) is a reliable way to produce true-to-type plants compared with callus and somatic embryogenesis regeneration, but in vitro culture environments can be mutagenic. Therefore, detecting somaclonal variations at an early stage of development is considered crucial in propagating plants. The genetic stability of in vitro regenerated plants needs to be ascertained by using DNA-based molecular markers. This review aims to provide up-to-date research progress on incorporating organic growth additives to enhance in vitro tissue culture protocols and to emphasize the importance of using PCR-based molecular markers such as RAPD, ISSR, SSR, and SCoT. The review was assessed based on the peer-reviewed works published in scientific databases including Science Direct, Scopus, Springer, JSTOR, onlinelibrary, and Google Scholar.

Abstract:Abstract Despite the transformative power of gene editing for crop improvement, its widespread application across species and varieties is limited by the transformation bottleneck that exists for many crops. The genetic transformation of plants is hindered by a general reliance on in vitro regeneration through plant tissue culture. Tissue culture requires empirically-determined conditions and aseptic techniques, and cannot easily be translated to recalcitrant species and genotypes. Both Agrobacterium-mediated and alternative transformation protocols are limited by a dependency on in vitro regeneration, which also limits their use by non-experts and hinders research into non-model species such as those of possible novel biopharmaceutical or nutraceutical use, as well as novel ornamental varieties. Hence, there is significant interest in developing tissue culture-independent plant transformation and gene editing approaches which can circumvent the bottlenecks associated with in vitro plant regeneration recalcitrance. Compared to tissue culture-based transformations, tissue culture-independent approaches offer advantages such as avoidance of somaclonal variation effects, with more streamlined and expeditious methodological processes. The ease of use, dependability, and accessibility of tissue culture-independent procedures can make them attractive to non-experts, outperforming classic tissue culture-dependent systems. This review explores the diversity of tissue culture-independent transformation approaches and compares them to traditional tissue culture-dependent transformation strategies. We highlight their simplicity and provide examples of recent successful transformations accomplished using these systems. Our review also addresses current limitations and explores future perspectives, highlighting the significance of these techniques for advancing plant research and crop improvement.

Presently, its application holds immense promise and offers a glimpse into the future of agricultural practices. The multifaceted roles of plant tissue culture in vegetable crops encompass augmenting varietal production, safeguarding endangered species, and facilitating the large-scale production of secondary metabolites, plant-based vaccines, and antibodies through cell suspension culture. The effectiveness of plant tissue culture hinges on various factors, including the availability of nutrients, endogenous auxin synthesis, organic compounds, and environmental conditions. Additionally, it tackles the challenge of supplying planting materials for species that cannot be efficiently propagated through traditional seed methods, effectively meeting the ever-growing worldwide agricultural requirements [1]. Primary goal is to assist budding plant biotechnologists and amplify the global influence of the plant tissue culture industry.

Abstract:Temporary immersion systems (TIS) have been widely recognized as a promising technology for micropropagation of various plant species. The TIS provides a suitable environment for culture and allows intermittent contact of the explant with the culture medium at different immersion frequencies and aeration of the culture in each cycle. The frequency or immersion is one of the most critical parameters for the efficiency of these systems. The design, media volume, and container capacity substantially improve cultivation efficiency. Different TIS have been developed and successfully applied to micropropagation in various in vitro systems, such as sprout proliferation, microcuttings, and somatic embryos. TIS increases multiplication and conversion rates to plants and a better response during the ex vitro acclimatization phase. This article covers the use of different immersion systems and their applications in plant biotechnology, particularly in plant tissue culture, as well as its use in the massive propagation of plants of agroeconomic interest.

Abstract:In vitro fertilization (IVF) of isolated male and female gametes of flowering plants was first accomplished in the last decade. Successful isolation of male and female gametes, and culturing of in vitro zygotes to form new plants, is a prelude to the use of IVF for research into the cellular and molecular control of fertilization in higher plants and its application as a tool in biotechnology. Genes unique to male and female gametes and zygotes of higher plants, although currently incompletely characterized, are expected to permit direct molecular dissection of fertilization. By applying IVF and microculture to zygotes and endosperm obtained by both in vivo and in vitro methods, newly activated fusion products may be observed and manipulated in media where they are directly accessible to the techniques of molecular cell biology. IVF and zygote culture may also offer potential for creating new hybrid plants by fusing isolated gametes from different species to produce unique zygotes and ultimately plants that would be impossible to obtain using typical crossing techniques. Transformation and regeneration frequencies using IVF may also be high enough to avoid the necessity of adding controversial antibiotic and herbicide resistant genes to screen transformed products. This review describes advances using IVF in plant sexual reproduction and discusses its potential in the genetic improvement of flowering plants.
