Abstract:Heterotrophic-assisted photoautotrophic microalgae biofilm cultivation was an alternative way to realize CO2 reduction and wastewater treatment. Growth kinetics supplied a channel to better understand how the cultivation conditions affect microalgal growth and CO2 reduction. However, the two growth modes (heterotroph and photoautotroph) have different needs for organic and inorganic nutrients. Thus, combining the threshold theory and multiplication theory, an integral multifactorial kinetic model that looking insight into the comprehensive effect of glucose, CO2, light intensity, and nitrate was developed for heterotrophic-assisted photoautotrophic microalgae biofilm growth in this study. R2 between model and experiment was 0.99. It predicted the maximum specific growth rate and maximum CO2 consumption rate of heterotrophic-assisted photoautotrophic microalgae biofilm was 1.868 h-1 and 1.02 h-1, respectively. This model fully explained the influence of the main factors on microalgae biofilm growth and reasonably predicted the growth rate of microalgae biofilm under different growth conditions.

Abstract:Understanding and optimizing the microalgae growth process is an essential prerequisite for effective CO2 capture using microalgae in photobioreactors. In this study, the kinetic characteristics of microalgae Chlorella vulgaris growth in response to light intensity and dissolved inorganic carbon (DIC) concentration were investigated. The greatest values of maximum biomass concentration (X-max) and maximum specific growth rate (mu(max)) were obtained as 2.303 g L-1 and 0.078 h(-1), respectively, at a light intensity of 120 mu mol m(-2) s(-1) and DIC concentration of 17 mM. Based on the results, mathematical models describing the coupled effects of light intensity and DIC concentration on microalgae growth and CO2 biofixation are proposed. The models are able to predict the temporal evolution of C. vulgaris growth and CO2 biofixation rates from lag to stationary phases. Verification experiments confirmed that the model predictions agreed well with the experimental results. (C) 2016 Elsevier Ltd. All rights reserved.

Abstract:As photoautotrophic microorganisms, microalgae feature complex mechanisms of photosynthesis and light energy transfer and as such studying their intrinsic growth kinetics is fairly difficult. In this article, the quantum yield of photochemical reaction was introduced in a study of microalgal kinetics to establish an intrinsic kinetic model of photoautotrophic microalgal growth. The blue-green algae Synechococcus sp. PCC7942 was used to verify the kinetic model developed using chlorophyll fluorescence analysis and growth kinetics determination. Results indicate that the kinetic model can realistically reflect the light energy utilization efficiency of microalgae as well as their intrinsic growth kinetic characteristics. The model and method proposed in this article may be utilized in intrinsic kinetics studies of photoautotrophic microorganisms.
Growth-based Dynamic Light Transmission Modeling and Optimization in Microalgal Photobioreactors for High Efficiency CO2 Fixation

However, the continuously changing light transmission during microalgal growth poses a significant challenge for enhancing their performance from the light optimization perspective. In this study, the growth-dependent light transmission characteristics in microalgal suspensions were investigated. As microalgae grew, the light saturation regions within the microalgal suspension expanded initially and then contracted, while the light inhibition expanded and the light limitation regions continuously shrank. Although biomass accumulation was relatively slow during the early growth stage (time <50 h) with biomass concentrations below 0.5 g/L, rapid changes in light attenuation occurred. Furthermore, the light-dependent local growth kinetic was developed to predict microalgal growth and CO2 fixation potential during growth. A contraction of the local microalgal CO2 fixation and specific growth rate distribution curve towards the incident light source occurred with its growth. Finally, the performance-enhancing effects of light optimization strategies on flat plate photobioreactors were analyzed based on the above model. Microalgal growth potential and CO2 fixation performance in the flat plate reactor with 2 cm light path could be increased by up to 188% when the growth-based light intensity enhancement strategy was adopted. It could be further increased by approximately 31% when the light was changed from single-sided to double-sided illumination (same light energy input), and the maximum net profit from microalgal cultivation could be enhanced by 60%. In conclusion, this study provides a unique insight into promoting microalgal carbon sequestration and biomass production from the light optimization perspective.

Abstract:A mechanistic understanding of algal growth is essential for maintaining a sustainable environment in an era of climate change and population expansion. It is known that algal growth is tightly controlled by complex interactive physical and chemical conditions. Many mathematical models have been proposed to describe the relation of algal growth and environmental parameters, but experimental verification has been difficult due to the lack of tools to measure cell growth under precise physical and chemical conditions. As such, current models depend on the specific testing systems, and the fitted growth kinetic constants vary widely for the same organisms in the existing literature. Here, we present a microfluidic platform where both light intensity and nutrient gradients can be well controlled for algal cell growth studies. In particular, light shading is avoided, a common problem in macroscale assays. Our results revealed that light and nitrogen colimit the growth of algal cells, with each contributing a Monod growth kinetic term in a multiplicative model. We argue that the microfluidic platform can lead towards a general culture system independent algal growth model with systematic screening of many environmental parameters. Our work advances technology for algal cell growth studies and provides essential information for future bioreactor designs and ecological predictions.

Abstract:Scaling up microalgae cultivation for large-scale utilization is challenging. To solve this problem and to optimize energy conversion by predicting and regulating the microalgae growth process, a universal and integrated numerical model framework for PBR design and optimization was developed. The model integrates computational fluid dynamics, light transfer, and microalgae growth sub-models, and supports aeration and stirring (dynamic mesh) for mixing. In this work, the model was first used to optimize culture conditions such as stir rate and light intensity, and then verified them experimentally. The model estimated the growth curve and growth rate of microalgae with an average error of 4.59% and a maximum error of 7.14%, respectively. The model can capture the growth kinetics of microalgae under large-scale conditions, and provide guidance for large-scale microalgae cultivation. Besides, we obtained that the traditional viewpoint of the dynamic illumination strategy is not exact and some PBR improvements for enhancing mixing and illumination were suggested.

Abstract:Microalgae are considered as a promising biofuel resource to fix carbon dioxide (CO2) in the future. Modelling microalgae growth is an effective method of studying the performance of microalgae growth manifested by the parameters including light distribution, pigment dynamics, nitrogen uptake, growth rate, respiration rate, temperature dependence, and depth dependence, and helping to control the culture of microalgae in artificial bioreactors. In this paper, the models of microalgae growth in nitrogen-limited and light-limited culture system for estimating biomass productivity are investigated by comparing different expressions and coefficients used in several main models. The results show that there are some differences among the results of numerical modelling due to different expressions and coefficients. This paper will lay a foundation for the selective use of microalgae growth models for researchers.

Abstract:Organisms are composed of multiple chemical elements such as carbon, nitrogen, and phosphorus. The scarcity of any of these elements can severely restrict organismal and population growth. However, many trophic interaction models only consider carbon limitation via energy flow. In this paper, we construct an algal growth model with the explicit incorporation of light and nutrient availability to characterize both carbon and phosphorus limitations. We provide a global analysis of this model to illustrate how light and nutrient availability regulate algal dynamics.

Abstract:The ever-increasing concern for energy shortages and greenhouse effect has triggered the development of sustainable green technologies. Microalgae have received more attention due to the characteristics of biofuel production and CO2 fixation. From the perspective of autotrophic growth, the optimization of light quality has the potential to promote biomass production and bio-component accumulation in microalgae at low cost. In this study, bibliometric analysis was used to describe the basic features, identify the hotspots, and predict future trends of the research related to the light quality on microalgae cultivation. In addition, a mini-review referring to regulation methods of light quality was provided to optimize the framework of research. Results demonstrated that China has the greatest interest in this area. The destination of most research was to obtain biofuels and high-value-added products. Both blue and red lights were identified as the crucial spectrums for microalgae cultivation. However, sunlight is the most affordable light resource, which could not be fully utilized by microalgae through the photosynthetic process. Hence, some regulation approaches (e.g., dyes, plasmonic scattering, and carbon-based quantum dots) are proposed to increase the proportion of beneficial spectrum for enhancement of photosynthetic efficiency. In summary, this review introduces state-of-the-art research and provides theoretical guidance for light quality optimization in microalgae cultivation to obtain more benefits.
Revealing the Synergistic Effects of Cells, Pigments, and Light Spectra on Light Transfer During Microalgae Growth: A Comprehensive Light Attenuation Model.

Abstract:As the sole energy for photosynthesis, light decrease rapidly with path due to absorption by pigments and scattering by cells in microalgal suspensions. By comprehensively considering cell concentrations, pigment components, and light spectra, a modified Cornet model for light transmission in microalgal suspensions is established. The developed model better fits experimental data with a higher adjusted R-2, which is 5% higher than the model that is based only on cell concentration. The attenuation of blue light is the most severe, followed by red and green light. Among the three main pigments, total carotenoids contribute the most to the absorption of blue and green light (with contribution coefficients of 89.26 +/- 4.53% and 46.04 +/- 3.77%, respectively), and chlorophyll a contributes the most to the absorption of red light (with a contribution coefficient of 75.33 +/- 5.08%). This study provides a better understanding and prediction of light transmission during microalgal cultivation.

Abstract:A robust multinutrient kinetic model, addressing the intricacies of mixotrophic microalgae cultivation, is introduced for enhanced lutein and biomass production. This model, notable for its complexity with seven state variables and 13 unknown parameters, enables accurate predictions of rapid growth and high lutein yields. Bentahar and Deschênes' approach is biotechnologically relevant, offering outcomes readily transferable to large‐scale production and a generalizable model applicable across various microalgal species. Mixotrophic cultivation holds great promise to significantly enhance the productivities of biomass and valuable metabolites from microalgae. In this study, a new kinetic model is developed, explicitly describing the effect of the most influential environmental factors on both biomass growth and the production of the high‐value product lutein. This extensive study of multinutrient kinetics for Tetradesmus obliquus in a mixotrophic regime covers various nutritional conditions. Crucial nutrients governing the model include nitrate, phosphate, and glucose. Using seven state variables and 13 unknown parameters, the model's accuracy was ensured through a well‐designed two‐factor, four‐level experimental setup, providing ample data for reliable calibration and validation. Results accurately predict dynamic concentration profiles for all validation experiments, revealing broad applicability. Optimizing nitrogen availability led to significant increases in biomass (up to fourfold) and lutein production (up to 12‐fold), with observed maximum biomass concentration of 6.80 g L−1 and lutein reaching 25.58 mg L−1. Noticeably, the model exhibits a maximum specific growth rate of 4.03 day−1, surpassing reported values for photoautotrophic and heterotrophic conditions, suggesting synergistic effects. Valuable guidance is provided for applying the method to various microalgal species and results are large‐scale production‐ready. Future work will exploit these results to develop real‐time photobioreactor operation strategies.

Multi-layer model for algal growth under different behaviors of nutritious salt

Abstract:According to the fact that different behaviors of nutrition at different medium layers would make obviously different effects on rate of algal growth in water system, we established a new numerical model of the algae growth by considering these effects from various nutrition concentrations in water environment, on the surface and inside of algae cell, and especially the effect of adsorption/desorption of nutrition on the surface of algal cell. The validation of the model parameters are carried out by numerical iterative calculation with experimental data of literature, and computation results show that the mean relative error between the actual measured values cited from literature and the numerical results of this model is less than 6.9% . Furthermore, the maximum absolute values of cumulative relative error of the model and the original model which doesn't consider effect on adsorption/desorption of nutrition are 11.7% and 34.18% respectively. Obviously, the model would fit well the actual measured data. The concentration of nutrition on the surface from the model computation shows the real change status of the algae absorb nutrition under alternate condition of light/dark, and at same time, the concentration varying of ATP inside algae cell would correlate to the concentration of nutrition in water environment and to the nutrition's condition of algae cell so that the cooperating relation between molecular layer and cell layer that has theory significance has been built.

Abstract:Microalgae are key players in the global carbon cycle and emerging producers of biofuels. Algal growth is critically regulated by its complex microenvironment, including nitrogen and phosphorous levels, light intensity, and temperature. Mechanistic understanding of algal growth is important for maintaining a balanced ecosystem at a time of climate change and population expansion, as well as providing essential formulations for optimizing biofuel production. Current mathematical models for algal growth in complex environmental conditions are still in their infancy, due in part to the lack of experimental tools necessary to generate data amenable to theoretical modeling. Here, we present a high throughput microfluidic platform that allows for algal growth with precise control over light intensity and nutrient gradients, while also performing real-time microscopic imaging. We propose a general mathematical model that describes algal growth under multiple physical and chemical environments, which we have validated experimentally. We showed that light and nitrogen colimited the growth of the model alga Chlamydomonas reinhardtii following a multiplicative Monod kinetic model. The microfluidic platform presented here can be easily adapted to studies of other photosynthetic micro-organisms, and the algal growth model will be essential for future bioreactor designs and ecological predictions.

Abstract:How to improve the growth efficiency of microalgae is the bottleneck of microalgae large-scale application. The addition of trace substances can promote the growth of microalgae, but there is no suitable model that can be used to predict the effects of trace substance concentrations on the growth of microalgae. In the present study, a mathematical model based on hormesis is proposed to describe the effects produced by trace substances on the biomass of microalgae and applied to assess the dose-response of four phytohormones on Scenedesmus sp. LX1 with a high coefficient of determination (R 2 ≥ 0.90). Several new mathematical parameters, such as starting effective dose (SD), inflection point dose (PD), concentration for 0 % of maximal effect, end effective dose (ED), maximum stimulatory effect (MSE), and maximum inhibitory effect (MIE), were extracted and useful to help researchers in applying trace substances to assist in the production of microalgal biomass for data reference and prediction. In concrete terms, the above model parameters can be well applied to screen the trace substances, dominant algal species and determine the concentration range. This study provides valuable insights into the potential of using phytohormones to enhance the biomass production of microalgae and offers a new approach to optimizing the culture of microalgae.

Abstract:Microalgae is a highly photosynthetic efficiency autotrophic nutritious. It is widely distributed over land and ocean, and is extensively used in fields such as medicine, food, aerospace, biotechnology, environmental protection. As an important equipment, photobioreactor is mainly used in mass cultivation and high-density microalgae. In this paper, we identified and established the mathematical model and computer simulation model of the microalgae growth. The Runge-Kutta method was used to solve the equations on the Matlab platform, in order to verify the accuracy of the model. Based on the mathematical model of microalgae grew under different light intensity and temperature, three-dimensional visualization model was built and implemented in a number of three dimensional software such as 3ds max and Virtools. As a photosynthetic organism, Microalgae is able to efficiently produce oxygen and absorb carbon dioxide. The goal of the visual simulation is to intuitively visualize its change and impact on oxygen and carbon dioxide. We selected different temperatures and light intensities control the photobioreactor, and dynamic change of microalgal biomass, oxygen and carbon dioxide was observed with the aim of providing visualization support for microalgae and photobioreactor research. Abstract:The cultivation of microalgae is significantly influenced by light intensity and utilization efficiency. This study developed a modified Cornet (M−Cornet) model to assess the distribution of light intensity and flux in microalgae cultivation systems. Algal biofilm cultivation represents a more concentrated approach of algal suspension cultivation. Both follow the M−Cornet model and exhibit the same growth rates when cultured under identical conditions. Algal pigments and morphologies greatly impact the light absorption and scattering, resulting in light attenuation in intensity, penetration distance and light flux distribution. Algae varieties exhibit diverse light flux characteristics. 37% − 90% of the incident light is absorbed, of which, 80% to 90% is dissipated as heat. 10% to 63% of the incident light is scattered off the photobioreactor. The overall light utilization efficiency ranges 6% to 13%. The light footprint using the M−Cornet model offers valuable insights for photobioreactors designing and cultivation operating.

Abstract:In this paper, based on the mathematical models of microalgae growth, the performance of microalgae growth in nitrogen-limited and light-limited continuous culture is investigated and the effect of important factors on the growth is examined. The dilution rate and the influent inorganic nitrogen concentration have been shown to have a significant influence on the growth of microalgae in continuous culture. In order to obtain a maximum productivity of microalgae, lower dilution rate is better for a lower influent inorganic nitrogen concentration and an optimal dilution rate can be obtained for a higher influent inorganic nitrogen concentration. There is an optimal influent inorganic nitrogen concentration corresponding to maximum microalgae productivity, and the optimal value for lower dilution rate is far higher than that for higher dilution rate. This paper will lay a foundation for the design of the operational parameters of continuous culture PBR (photobioreactor).

Abstract:To comprehensively understand kinetic characteristics of microalgae growth and lipid synthesis in different phases, a phase-feeding strategy was proposed to simultaneously regulate light, carbon and nutrients in adaption, growth and stationary phases of microalgae cultivation. Physiological-phased kinetic characteristics of microalgae Chlorella vulgaris growth and lipid synthesis under synergistic effects of light, carbon and nutrients were investigated, and supply-demand relationships of electrons and energy between light and dark reactions of photosynthesis process were discussed. Finally, the optimized cultivation strategy for microalgae in various phases were obtained, under which the lipid productivity was significantly improved from 130.11 mg/L/d to 163.42 mg/L/d. The study provided some important guidance for the large-scale production of biofuels from microalgae.
