Abstract:The construction of chiral C(sp3)-rich molecules is of great interest in natural products synthesis and drug discovery.To fulfill the requirement of chiral C(sp3)-rich molecules,various approaches have been developed recently.Among these elegant approaches,nickel-catalyzed asymmetric C(sp3)-C(sp3)cross-cou-pling has emerged as one of the most appealing approaches,which enables the enantioselective coupling between divergent racemic alkyl electrophiles and racemic alkyl nucleophiles[1,2].Despite the advances,the reaction partners of nickel catalytic system are generally limited in stoichiometric amounts of organometallic reagents,such as Grignard and organozinc reagents,and suffered from limited functional group compatibility and the requirement of multiple-step synthesis(Scheme 1a).To overcome the draw-backs,enantioselective nickel-hydride(Ni-H)catalyzed hydroalkylation of alkenes has emerged as an alternative and con-venient entry to control the stereochemistry of the formation of C(sp3)-(sp3)bonds(Scheme 1b).Notably,the abundant,stable alkenes are typically more attractive and broadly applicable coupling partners than the organometallic reagents.
