Abstract:The fear responses to environmental threats play a fundamental role in survival. Little is known about the neural circuits specifically processing threat-relevant sensory information in the mammalian brain. We identified parvalbumin-positive (PV+) excitatory projection neurons in mouse superior colliculus (SC) as a key neuronal subtype for detecting looming objects and triggering fear responses. These neurons, distributed predominantly in the superficial SC, divergently projected to different brain areas, including the parabigeminal nucleus (PBGN), an intermediate station leading to the amygdala. Activation of the PV+ SC-PBGN pathway triggered fear responses, induced conditioned aversion, and caused depression-related behaviors. Approximately 20% of mice subjected to the fear-conditioning paradigm developed a generalized fear memory.

Abstract:Abstract The ability of animals to respond to life-threatening stimuli is essential for survival. Although vision provides one of the major sensory inputs for detecting threats across animal species, the circuitry underlying defensive responses to visual stimuli remains poorly defined. Here, we investigate the circuitry underlying innate defensive behaviours elicited by predator-like visual stimuli in mice. Our results demonstrate that neurons in the superior colliculus (SC) are essential for a variety of acute and persistent defensive responses to overhead looming stimuli. Optogenetic mapping revealed that SC projections to the lateral posterior nucleus (LP) of the thalamus, a non-canonical polymodal sensory relay, are sufficient to mimic visually evoked fear responses. In vivo electrophysiology experiments identified a di-synaptic circuit from SC through LP to the lateral amygdale (Amg), and lesions of the Amg blocked the full range of visually evoked defensive responses. Our results reveal a novel collicular–thalamic–Amg circuit important for innate defensive responses to visual threats.

The Superior Colliculus/Lateral Posterior Thalamic Nuclei in Mice Rapidly Transmit Fear Visual Information Through the Theta Frequency Band

Abstract:Animals perceive threat information mainly from vision, and the subcortical visual pathway plays a critical role in the rapid processing of fear visual information. The superior colliculus (SC) and lateral posterior (LP) nuclei of the thalamus are key components of the subcortical visual pathway; however, how animals encode and transmit fear visual information is unclear. To evaluate the response characteristics of neurons in SC and LP thalamic nuclei under fear visual stimuli, extracellular action potentials (spikes) and local field potential (LFP) signals were recorded under looming and dimming visual stimuli. The results showed that both SC and LP thalamic nuclei were strongly responsive to looming visual stimuli but not sensitive to dimming visual stimuli. Under the looming visual stimulus, the theta (θ) frequency bands of both nuclei showed obvious oscillations, which markedly enhanced the synchronization between neurons. The functional network characteristics also indicated that the network connection density and information transmission efficiency were higher under fear visual stimuli. These findings suggest that both SC and LP thalamic nuclei can effectively identify threatening fear visual information and rapidly transmit it between nuclei through the θ frequency band. This discovery can provide a basis for subsequent coding and decoding studies in the subcortical visual pathways.

Off Starburst Amacrine Cells in the Retina Trigger Looming-Evoked Fear Responses in Mice

Abstract:A rapidly approaching dark object evokes an evolutionarily conserved fear response in both vertebrates and invertebrates, young to old. A looming visual stimulus mimics an approaching object and triggers a similarly robust fear response in mice, resulting in freeze and flight. However, the retinal neural pathway responsible for this innate response has not been fully understood. We first explored a variety of visual stimuli that reliably induced these innate responses, and found that a looming stimulus with 2-d acclimation consistently evoked fear responses. Because the fear responses were triggered by the looming stimulus with moving edges, but not by a screen flipping from light to dark, we targeted the starburst amacrine cells (SACs), crucial neurons for retinal motion detection. We used intraocular injection of diphtheria toxin (DT) in mutant mice expressing diphtheria toxin receptors (DTR) in SACs. The looming-evoked fear responses disappeared in half of the DT-injected mice, and the other mice still exhibited the fear responses. The optomotor responses (OMRs) were reduced or eliminated, which occurred independent of the disappearance of the fear responses. A histologic examination revealed that ON SACs were reduced in both mouse groups preserved or absent fear responses. In contrast, the number of OFF SACs was different among two groups. The OFF SACs were relatively preserved in mice exhibiting continued fear responses, whereas they were ablated in mice lacking fear response to looming stimulation. These results indicate that OFF SACs and the direction-selective pathway in the retina play a role in looming-induced fear behaviors.

Abstract:Understanding how threats in environments drive fear memory formation is crucial to understanding how organisms adapt to changing environments and treat threat-related disorders such as PTSD. Despite decades of pioneering work using the Pavlovian conditioning model, our understanding has been limited because only one type (an electric shock) among diverse threats has been exclusively used. We developed a threat conditioning paradigm by utilizing a looming visual threat as an unconditioned stimulus (US) in mice and identified a distinct threat conditioning circuit. Parabrachial CGRP neurons were required for conditioning and memory retrieval. Their upstream neurons in the posterior insular cortex (pIC) responded to looming stimuli and projections of these neurons to the parabrachial nucleus induced aversive affective states and drove conditioning. But the pIC to PBN pathway was dispensable for foot-shock conditioning. Our findings provide insights into how a simple pattern of non-nociceptive visual stimuli can induce aversive states and drive fear memory formation.

Abstract:Conditioned fear requires neural activity in the basolateral amygdala (BLA) and medial prefrontal cortex (mPFC), structures that are densely interconnected at the synaptic level. Previous work has suggested that anatomical subdivisions of mPFC make distinct contributions to fear expression and inhibition, and that the functional output of this processing is relayed to the BLA complex. However, it remains unknown whether synaptic plasticity in mPFC-BLA networks contributes to fear memory encoding. Here we use optogenetics and ex vivo electrophysiology to reveal the impact of fear conditioning on BLA excitatory and feedforward inhibitory circuits formed by projections from infralimbic (IL) and prelimbic (PL) cortices. In naive mice, these pathways recruit equivalent excitation and feedforward inhibition in BLA principal neurons. However, fear learning leads to a selective decrease in inhibition:excitation balance in PL circuits that is attributable to a postsynaptic increase in AMPA receptor function. These data suggest a pathway-specific mechanism for fear memory encoding by adjustment of mPFC-BLA transmission. Upon reengagement of PL by conditioned cues, these modifications may serve to amplify emotional responses.

Abstract:The neural basis of fear of heights remains largely unknown. In this study, we investigated the fear response to heights in mice and observed characteristic aversive behaviors resembling human height vertigo. We identified visual input as a critical factor in mouse reactions to heights, while peripheral vestibular input was found to be nonessential for fear of heights. Unexpectedly, we found that fear of heights in naïve mice does not rely on image-forming visual processing by the primary visual cortex. Instead, a subset of neurons in the ventral lateral geniculate nucleus (vLGN), which connects to the lateral/ventrolateral periaqueductal gray (l/vlPAG), drives the expression of fear associated with heights. Additionally, we observed that a subcortical visual pathway linking the superior colliculus to the lateral posterior thalamic nucleus inhibits the defensive response to height threats. These findings highlight a rapid fear response to heights threat through a subcortical visual and defensive pathway from the vLGN to the l/vlPAG.

Abstract:It has been reported that threatening and non-threatening visual stimuli can be distinguished based on the multi-voxel patterns of haemodynamic activity in the human ventral visual stream. Do these findings mean that there may be evolutionarily hardwired mechanisms within early perception, for the fast and automatic detection of threat, and maybe even for the generation of the subjective experience of fear? In this human neuroimaging study, we presented participants ('fear' group: N = 30; 'no fear' group: N = 30) with 2700 images of animals that could trigger subjective fear or not as a function of the individual's idiosyncratic ‘fear profiles’ (i.e. fear ratings of animals reported by a given participant). We provide evidence that the ventral visual stream may represent affectively neutral visual features that are statistically associated with fear ratings of participants, without representing the subjective experience of fear itself. More specifically, we show that patterns of haemodynamic activity predictive of a specific ‘fear profile’ can be observed in the ventral visual stream whether a participant reports being afraid of the stimuli or not. Further, we found that the multivariate information synchronization between ventral visual areas and prefrontal regions distinguished participants who reported being subjectively afraid of the stimuli from those who did not. Together, these findings support the view that the subjective experience of fear may depend on the relevant visual information triggering implicit metacognitive mechanisms in the prefrontal cortex. Abstract:The human brain has evolved a multifaceted fear system, allowing threat detection to enable rapid adaptive responses crucial for survival. Although many cortical and subcortical brain areas are believed to be involved in the survival circuits detecting and responding to threat, the amygdala has reportedly a crucial role in the fear system. Here, we review evidence demonstrating that fearful faces, a specific category of salient stimuli indicating the presence of threat in the surrounding, are preferentially processed in the fear system and in the connected sensory cortices, even when they are presented outside of awareness or are irrelevant to the task. In the visual domain, we discuss evidence showing in hemianopic patients that fearful faces, via a subcortical colliculo-pulvinar-amygdala pathway, have a privileged visual processing even in the absence of awareness and facilitate responses towards visual stimuli in the intact visual field. Moreover, evidence showing that somatosensory cortices prioritise fearful-related signals, to the extent that tactile processing is enhanced in the presence of fearful faces, will be also reported. Finally, we will review evidence revealing that fearful faces have a pivotal role in modulating responses in peripersonal space, in line with the defensive functional definition of PPS.

Abstract:Neuronal responses in the primary visual cortex (V1) are driven by simple stimuli, but these stimulus-evoked responses can be markedly modulated by non-sensory factors, such as attention and reward [1], and shaped by perceptual training [2]. In real-life situations, neutral visual stimuli can become emotionally tagged by experience, resulting in altered perceptual abilities to detect and discriminate these stimuli [3-5]. Human imaging [4] and electroencephalography (EEG) studies [6-9] have shown that visual fear learning (the acquisition of aversive emotion associated with a visual stimulus) affects the activities in visual cortical areas as early as in V1. However, it remains elusive as to whether the fear-related activities seen in the early visual cortex have to do with feedback influences from other cortical areas; it is also unclear whether and how the response properties of V1 cells are modified during the fear learning. In the current study, we addressed these issues by recording from V1 of awake monkeys implanted with an array of microelectrodes. We found that responses of V1 neurons were rapidly modified when a given orientation of grating stimulus was repeatedly associated with an aversive stimulus. The output visual signals from V1 cells conveyed, from their response outset, fear-related signals that were specific to the fear-associated grating orientation and visual-field location. The specific fear signals were independent of neurons' orientation preferences and were present even though the fear-associated stimuli were rendered invisible. Our findings suggest a bottom-up mechanism that allows for proactive labeling of visual inputs that are predictive of imminent danger.

Abstract:Fear is defined as a fundamental emotion promptly arising in the context of threat and when danger is perceived. Fear can be innate or learned. Examples of innate fear include fears that are triggered by predators, pain, heights, rapidly approaching objects, and ancestral threats such as snakes and spiders. Animals and humans detect and respond more rapidly to threatening stimuli than to nonthreatening stimuli in the natural world. The threatening stimuli for most animals are predators, and most predators are themselves prey to other animals. Predatory avoidance is of crucial importance for survival of animals. Although humans are rarely affected by predators, we are constantly challenged by social threats such as a fearful or angry facial expression. This chapter will summarize the current knowledge on brain circuits processing innate fear responses to visual stimuli derived from studies conducted in mice and humans.

State- and circuit-dependent opponent-processing of fear

Abstract:The presence of valence coding neurons in the basolateral amygdala (BLA) that form distinct projections to other brain regions implies functional opposition between aversion and reward during learning. However, evidence for opponent interactions in fear learning is sparse and may only be apparent under certain conditions. Here we test this possibility by studying the roles of the BLA➔central amygdala (CeA) and BLA➔nucleus accumbens (Acb) pathways in fear learning in male rats. First, we assessed the organisation of these pathways in the rat brain. BLA➔CeA and BLA➔Acb pathways were largely segregated in the BLA but shared overlapping molecular profiles. Then we assessed activity of the BLA➔CeA and BLA➔Acb pathways during two different forms of fear learning - fear learning in a neutral context and fear learning in a reward context. BLA ➔ CeA neurons were robustly recruited by footshock regardless of where fear learning occurred whereas recruitment of BLA➔Acb neurons was state-dependent because footshock only recruited this pathway in a reward context. Finally, we assessed the causal roles of activity in these pathways in fear learning. Photoinhibition of the BLA➔CeA pathway during the footshock US impaired fear learning, regardless of where fear learning occurred. In contrast, photoinhibition of the BLA➔Acb pathway augmented fear learning, but only in the reward context. Taken together, our findings show circuit- and state-dependent opponent processing of fear. Footshock-driven activity in the BLA➔Acb pathway can functionally oppose the BLA➔CeA pathway to limit how much fear is learned.

Early Sensory Pathways for Detection of Fearful Conditioned Stimuli: Tectal and Thalamic Relays

Abstract:Sensory stimuli acquire significance through learning. A neutral sensory stimulus can become a fearful conditioned stimulus (CS) through conditioning. Here we report that the sensory pathways used to detect the CS depend on the conditioning paradigm. Animals trained to detect an electrical somatosensory stimulus delivered to the whisker pad in an active avoidance task were able to detect this CS and perform the task when a reversible or irreversible lesion was placed in either the somatosensory thalamus or the superior colliculus contralateral to the CS. However, simultaneous lesions of the somatosensory thalamus and superior colliculus contralateral to the CS blocked performance in the active avoidance task. In contrast, a lesion only of the somatosensory thalamus contralateral to the same CS, but not of the superior colliculus, blocked performance in a pavlovian fear conditioning task. In conclusion, during pavlovian fear conditioning, which is a situation in which the aversive outcome is not contingent on the behavior of the animal, the sensory thalamus is a critical relay for the detection of the CS. During active avoidance conditioning, a situation in which the aversive outcome is contingent on the behavior of the animal (i.e., the animal can avoid the aversive event), the sensory thalamus and the superior colliculus function as alternative routes for CS detection. Thus, even from early stages of sensory processing, the neural signals representing a CS are highly distributed in parallel and redundant sensory circuits, each of which can accomplish CS detection effectively depending on the conditioned behavior.

Abstract:Generalization allows for experience to flexibly guide behavior when conditions change. A basic physical unit of memory storage and expression in the brain are sparse, distributed groups of neurons known as ensembles (i.e., the engram). The infralimbic (IL) subregion of the ventral medial prefrontal cortex plays a key role in modulating conditioned defensive responses. How IL neuronal ensembles established during learning contribute to generalized responses is unknown. In this set of experiments, generalization was tested in male and female mice by presenting a novel, ambiguous, tone generalization stimulus following Pavlovian defensive (fear) conditioning. The first experiment was designed to test a role for IL in generalization using chemogenetic manipulations. Results show IL bidirectionally regulates defensive behavior. IL silencing promotes a switch in defensive state from vigilant scanning to generalized freezing, while IL stimulation reduces freezing in favor of scanning. Leveraging activity-dependent tagging technology (ArcCreER T2 x eYFP system), a neuronal ensemble, preferentially located in IL superficial layer 2/3, was associated with the generalization stimulus. Remarkably, in the identical discrete location, fewer reactivated neurons were associated with the generalization stimulus at the remote timepoint (30 days) following learning. When an IL neuronal ensemble established during learning was selectively chemogenetically silenced, generalization increased. Conversely, IL neuronal ensemble stimulation reduced generalization. Overall, these data identify a crucial role for IL in suppressing generalized responses. Further, we uncover an IL neuronal ensemble, formed during learning, functions to later attenuate the expression of generalization in the presence of ambiguous threat stimuli.

Abstract:The brain’s ability to appraise threats and execute appropriate defensive responses is essential for survival in a dynamic environment. Humans studies have implicated the anterior insular cortex (aIC) in subjective fear regulation and its abnormal activity in fear/anxiety disorders. However, the complex aIC connectivity patterns involved in regulating fear remain under investigated. To address this, we recorded single units in the aIC of freely moving male mice that had previously undergone auditory fear conditioning, assessed the effect of optogenetically activating specific aIC output structures in fear, and examined the organization of aIC neurons projecting to the specific structures with retrograde tracing. Single-unit recordings revealed that a balanced number of aIC pyramidal neurons’ activity either positively or negatively correlated with a conditioned tone-induced freezing (fear) response. Optogenetic manipulations of aIC pyramidal neuronal activity during conditioned tone presentation altered the expression of conditioned freezing. Neural tracing showed that non-overlapping populations of aIC neurons project to the amygdala or the medial thalamus, and the pathway bidirectionally modulated conditioned fear. Specifically, optogenetic stimulation of the aIC-amygdala pathway increased conditioned freezing, while optogenetic stimulation of the aIC-medial thalamus pathway decreased it. Our findings suggest that the balance of freezing-excited and freezing-inhibited neuronal activity in the aIC and the distinct efferent circuits interact collectively to modulate fear behavior.
