Abstract:Three new resveratrol oligomers, hopeahainanphenol (1), neohopeaphenol A (2), and neoisohopeaphenol A (3), were isolated front the stern bark of Hopea hainanensis (Dipterocarpaccae).Their structures were elucidatcd by in-depth spectroscopic analyses, including 1D- and 2D-NMR techniques, and by HR-ESI-MS. All the three phytochemicals were tested in vitro for acertylcholinesterase (AChE) inhibitory, and antitumor activity. The dimeric compound 2, which corresponds to (1S*,6S*,7S*,11bS*)- 1,6:7,11b-tetrahydro-1,7-bis(4-hydroxyplienyl)6-[(1R-,6R*,7R*.11bR*)-1,6,7,11b-benzofuran-4.8.10-triliydroxy-1,7-bis(4-hydroxyphenyl)[6,7]cyclohepta[1,2,3-cd]benzofuran-6-yl]benzo[6,7]cycloheptal[1,2,3-cd]benzofuran-4,8,10-triol. was found to be significantly active against AChE, with an IC50 value of 7.66 +/- 0.13 mu m.

Abstract:Bioassay-guided investigation of the stem bark of Hopea chinensis led to the isolation of two new polyphenols, hopeachinols C (1) and D (2), together with ten known compounds (3-12). Compounds 1 and 2 were determined by extensive analysis of spectroscopic data and computational methods. All of these phyto-chemicals were tested for acetylcholinesterase inhibitory activity, and five resveratrol-derived compounds (1 and 7-10) exhibited significant activity with their IC50 values ranging from 4.81 to 11.71 mu M.

Abstract:Phytochemical investigation of the stem wood of Hopea hainanensis, a Dipterocarpaceae tree endemic to Hainan Island of China, leads to four new oligostilbenoids, hopeahainols C-F (1-4), and eight known ones (5-12). All of the structures were elucidated by comprehensive analysis of spectroscopic data including UV, IR, MS, and NMR after purification of individual compounds through a combination of chromatographic methods. A resveratrol glucoside, eight resveratrol dimers, two trimers, and a tetramer were identified. All of the polyphenols were tested for their radical scavenging and total reducing capacities by measuring their capacity to scavenge the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical, anion superoxide radical, and also to induce the reduction of Mo (VI) to Mo (V). Most of the compounds exhibited potent antioxidant and radical scavenger capacity compared with the positive controls (resveratrol, ascorbic acid, and butylated hydroxyanisole (BHA)) at 0.4 mM concentration.

Abstract:Five new stilbenolignans, hopeachinols E-I (1-5), and two new stilbenoids, hopeachinols J and K (6 and 7), together with the known resveratrol trimer, vaticanol A (8), were isolated from the stem bark of Hopea chinensis. The structures and relative configurations of these natural products were elucidated by using spectroscopic and spectrometric methods. The (8R) absolute configuration of 1 was assigned by using the modified Mosher ester method. Hopeachinols E-I (1-5) possess unprecedented stilbenolignan skeletons, in which a stilbene trimer is linked with a phenylpropanoid unit through a pyran bridge. Hopeachinols J and K (6 and 7) have new skeletons in which a stilbene trimer is connected by an additional two-carbon unit through a furan moiety. An evaluation of the cytotoxicities against HCT116, MDA-MB-231, SMMC-7721, and HepG2 cell lines of the new hopeachinols showed that 1, 2, 4, and 6 have moderate activities with IC50 values in the 10.39-18.72 mu M range.

Abstract:Five resveratrol tetramers, amurensins I-M, were isolated from the roots of Vitis amurensis (Rupr.), together with five known resveratrol tetramers, (+)-hopeaphenol, isohopeaphenol, vitisin A, (+)-vitisifuran A, and heyneanol A. Their structures and stereochemistry were determined by chemical and spectroscopic methods, especially by use of 2D NMR analysis. Some of them had an ampelopsin A or a balanocarpol unit, in which the conformations of the seven-membered carbon ring were described for the first time. The anti-inflammatory activities of the tetramers were also tested. Among them, (+)-hopeaphenol, isohopeaphenol, vitisin A, (+)-vitisifuran A and heyneanol A showed potent inhibition on the biosynthesis of leukotriene B4 (LTB4), and amurensins I and L showed strong antagonism of the histamine acceptor.

Abstract:A phytochemical study of Hopea hainanensis has led to the isolation of three new polyphenols and one known compound. The most important of these compounds are hopeahainols A (2) and B (3), which contain an unprecedented carbon skeleton. The structures were elucidated by analysis of the spectroscopic data including single-crystal X-ray spectroscopy and computational methods. Hopeahainol A was an acetylcholinesterase inhibitor with an IC50 value of 4.33 mu m, which is comparable to that of huperzine A, a presently prescribed drug for the treatment of Alzheimer, while other similar structures were inactive. This observation was complemented by a 3D interaction model of the inhibitor with active sites.

Abstract:Hopeanolin (1), an unusual resveratral trimer with an ortho-quinone nucleus, was isolated and characterized from the stem bark of Hopea exalata. Also obtained were six known stibenoids, shoreaphenol (2), vaticanol G (3), alpha-viniferin (4), pauciflorol A (5), vaticanol A (6), and trans-3,5,4'-trihydroxystilbene 2-C-glucoside (7). The structure of 1 was determined by spectroscopic data interpretation. Compounds 1-7 were tested for antifungal activity and inhibitory effects against jack bean urease. Hopeanolin (1) demonstrated antifungal activity in the MIC value range 0.1-22.5 microg/mL.

Abstract:Two new flavonoids, Alpinetin-7-O-[3''- O -galloyl-4'',6''-hexahydroxydiphenoyl]- β - D -glucose ( 1 ), Pinocembrin-7- O -[4''- O -galloyl]- β - D -glucose ( 2 ), and three known compound ( 3-5 ) were isolated from Penthorum chinense Pursh (Saxifragaceae). The structures of all compounds were deduced from their comprehensive spectroscopic analysis including IR, HR-ESI-MS, 1 H NMR, 13 C NMR, DEPT, COSY, HMBC and HMQC. Molecular docking model was used to test the anti-cholestatic liver activities of the isolated compounds, and compounds 1 and 2 showed higher docking scores (-9.90 and −11.27 kcal/mol, respectively) binding with FXR than 3 , 4 and 5 (-8.00, −9.77, −9.10 kcal/mol, respectively), suggesting 1 and 2 exhibited potential anti-cholestatic activities. The present results show that P. chinense is a potential source of 2 new lead compounds that can be utilised to produce therapeutic drugs for liver diseases upon further studies.

Abstract:Four new tetrahydrofuran lignans, schpropinrins A-D ( 1–4 ), together with five known ones, were isolated from the leaves and stems of Schisandra propinqua var. sinensis . Their structures, including absolute configurations, were characterized by means of spectroscopic analysis and ECD calculation. Compounds 1–4 featured a ketal or hemiketal substructure at C-7 and all of the isolates were tested for their anti-HIV integrase activity.

Abstract:Chunganenol (1), an unusual resveratrol hexamer, has been isolated from Vitis chunganensis. Its unprecedented structure has been assigned through detailed analyses of 2D NMR data and consideration of oligostilbene biosynthetic pathway, representing the first highly condensed stilbene oligomer from Vitaceous plants. It also featured a novel interunit lingkage where two stilbene units are connected by a methylene bridge. Moreover, 1 exhibited potent and selective quenching effects on singlet oxygen, and might play a role in the antioxidative defense system of the plant.
[Non-alkaloid chemical constituents from Coptis chinensis]

Abstract:Objective: To separate and identify chemical constituents from Coptis chinensis. Method: The compounds were separated and purified by various chromatographic techniques. Their structures were identified on the basis of their physicochemical properties using spectral techniques such as NMR and MS. Result: Thirteen compounds were separated from ethanol extracts of C. chinensis, including seven lignans, three simple phenylpropanoids, two flavones and one phenolic acid, and identified as erythro-guaiacylglycerol-8-O-4'-(coniferyl alcohol) ether (1), threo-guaiacylglycerol-8-O-4'-(coniferyl alcohol) ether (2), (+)-pinoresinol (3), (+)-medioresinol (4), (+)-lariciresinol (5), (+)-5'-methoxylariciresinol (6), (+)-isolariciresinol (7), chlorogenic acid (8), ferulic acid (9), Z-octadecyl caffeate (10), rhamnetin (11), wogonin (12), and vanillic acid (13). Conclusion: Compounds 1, 2, 4, 6, 10-13 were separated from the genus Coptis for the first time.

Abstract:In this study, a new chromone, six coumarins, and a phenolic compound were isolated from a MeOH extract of an Angelica dahurica stems. Through the use of various spectral techniques, the structures of the isolated compounds were determined to be scopoletin ( 1 ), xanthyletin ( 2 ), xanthotoxin ( 3 ), hopeyhopin ( 4 ), thamnosmonin ( 5 ), 6-[(1S, 2R)-2,3-dihydroxy-1-methoxy-3-methylbutyl]-7-methoxycoumarin ( 6 ), decursidate ( 7 ), and new compound 8 , named dahuricalol. Among the isolatled and identified compounds, (S)-5-hydroxy-8-(hydroxymethyl)-2,2-dimethyl-6-oxo-3,4-dihydro-2H,6H-pyrano[3,2-g]chromen-3-yl (Z)-2-(hydroxymethyl)but-2-enoate ( 8 ) was isolated from plant for the first time.

Abstract:Six flavonoids named cryptogiones A–F, and nine known compounds were isolated from an ethanol extract of stems of Cryptocarya chingii. The structures of the compounds were elucidated by interpretation of comprehensive spectroscopic data and X-ray analysis. A majority of these flavonoids contained an acetic acid/lactone moiety, a possible taxonomic marker. Anti-inflammatory effects of the compounds were evaluated using in vitro assays. At 20μM concentration, three compounds significantly inhibited TNFα-induced NF-кB activation and LPS-induced IL-1β expression.

Abstract:Phytochemical investigation of the stem wood of Dipterocarpus alatus led to the isolation and characterization of four new oligostilbenoids, dipterocarpols A-D (1-4), together with two known resveratrol oligomers, hopeahainol (5) and hopeafuran (6). The structures of the new compounds were determined by comprehensive spectral analysis including 1D and 2DNMR, and high-resolution MS. The absolute configurations were determined by NOESY and CD spectra. Dipterocarpol A (1) and hopeahainol A (5) showed moderate acetylcholinesterase inhibitory activity with IC50 values of 8.28 mu M and 11.28 mu M, respectively. Furthermore, the discovery of compound 3 gave the first evidence that the biosynthetic origin of resveratrol aneuploids is related to the loss of a half resveratrol unit by oxidative cleavage.
Isolation and structural identification of the main anthocyanin monomer in Vitis amurensis Rupr

Abstract:Many types of anthocyanins are present in Vitis amurensis Rupr of 'Beibinghong', which is grown in North-east China and has high antioxidant activity. However, the anthocyanin with the highest content in V. amurensis Rupr has not yet been identified. In this study, pulsed electric field extraction and semi-preparative liquid phase separation were used to isolate the anthocyanin monomer from 'Beibinghong'. UV-vis spectroscopy, FTIR spectroscopy, mass spectra and nuclear magnetic resonance were used to identify the anthocyanin monomer. The antioxidant activities of the anthocyanin monomer were also analysed. Malvidin-3,5-O-diglucoside was identified as the main anthocyanin in V. amurensis Rupr, which could be used as a raw material for its extraction. Furthermore, malvidin-3,5-O-diglucoside can be potentially used as a functional food, and a novel therapeutic and preventive agent for oxidative stress-related diseases. This study provides technical information for the future purification and structural identification of anthocyanins.
