Abstract:The IMDA cycloadditions of 10 different ester-tethered 1,3,8-nonatrienes have been examined under controlled microwave heating (MeCN, 180 degrees C, 30 min), giving 90-99% yields, and the stereochemical outcome of the exo and endo adducts established together with X-ray crystal structural analysis. A microwave-assisted tandem Wittig-IMDA cycloaddition protocol has been established for a modular synthesis of the bicyclic lactones starting from alpha-bromoacetates of 2,4-pentadien-1-ols and alpha-oxo carbonyl compounds in the presence of PPh3 and 2,6-lutidine (MeCN, 180 degrees C, 30 min). The overall yields of the tandem reactions are 68-80% and the stereoselectivity of the major adducts formed from E-substituted 1,3,8-nonatriene is the same as that observed for the purified 1,3,8-nonatrienes. Abstract:Intramolecular Diels Alder (IMDA) cycloaddition of the ester-tethered 1,3,9,-decatrienes possessing a carbonyl substituent at C10 has been investigated under controlled microwave heating (MeCN, 180 degrees C) to afford a variety of 3,4,4a,7,8,8a-hexahydroisochromen-1-ones in 53-89% yields and in 64:36-79:21 ratios for the cis and trans isomers. Under the same microwave heating conditions, a tandem Wittig-IMDA cycloaddition, starting from the alpha-bromoacetates of 3,5-hexadien-1-ols and glyoxalate/phenylglyoxal hydrates in the presence of PPh3 and 2,6-lutidine, has been demonstrated, furnishing 3,4,4a,7,8,8a-hexahydroisochromen-1-one adducts in 73-91% yields in favor of the cis isomers. During this tandem process, three consecutive carbon carbon bonds in the end products were efficiently formed with the aid of microwave irradiation within short reaction times. (C) 2010 Elsevier Ltd. Abstract:To study the intramolecular Diels-Alder cycloadditon of 2H-chromen-2-one as a diene, a series of chiral N-allyl-N-benzylamides that contain a 2H-chromen-2-one moiety were designed for the synthesis of benzo[f]isoindol-1-ones via an intramolecular Diels-Alder and a subsequent retro-Diels-Alder cycloaddition with the expulsion of CO2. Both the yield (80%-89%) and absolute stereocontrol of the tandem reaction were high when an electron-withdrawing group was attached to the dienophile. The double bond in the styrene substructure remained in the products could be further derivatized by dihydroxylation. Abstract:Intramolecular Diets-Alder (IMDA) reaction of the hydroxamate-tethered 1,3,9-decatrienes has been investigated under both thermal and microwave heating, and the stereoselectivity has been revisited. It was found that a temperature-dependent rearrangement of (2E,4E)-hexadien-1-yl N-benzylhydroxamates into the corresponding (2E,4)-1-methylpentadien-1-yl derivatives took place prior to IMDA reaction upon heating, and up to four cycloadducts were identified by analysis of the crude reaction mixtures. The IMDA reaction could be accelerated with microwave irradiation at 180 °C in MeCN. Stereoselectivity of the microwave-assisted IMDA reaction has been examined by using the substituents appended at C1, C5, and C 10 of the hydroxamate-tethered 1,3,9-decatrienes.

Abstract:A highly stereoselective tandem Michael addition-Wittig reaction of (3-carboxy-2-oxopropylidene)triphenylphosphorane and alpha,beta-unsaturated aldehydes has been developed by employing the combined catalysis of a newly designed bulky chiral secondary amine 1g, LiClO(4), and DABCO. The multifunctional 6-carboxycyclohex-2-en-1-ones were generally obtained in excellent diastereo- and enantioselectivities (dr up to >50:1, 86-99% ee).

Abstract:The one-pot U-4CR and intramolecular O-alkylation sequence starting from 2-aminophenols in combination with α-bromoalkanoic acids, aldehydes, and isocyanides under controlled microwave heating has been established for a rapid access to highly functionalized 3,4-dihydro-3-oxo-2H-1,4-benzoxazines. With appropriate substitutions on the 1,4-benzoxazines, a microwave-assisted Cu-catalyzed intramolecular amidation was performed to furnish a novel class of heterocyclic conjugates of 1,4-benzoxazines with a 2-oxindole linked through a C–N single bond.

Abstract:Intramolecular Diels–Alder (IMDA) reaction of two sorbate-related 1,3,8-nonatrienes has been investigated in MeCN at 180°C for 1.5h under controlled microwave heating. On checking the crude product mixture before purification, partial epimerization of the major adduct was found during the reaction. After column chromatographic purification over silica gel, only two cis adducts were obtained and their structures have been thoroughly established by X-ray crystallographic analysis. It is concluded that the putative major trans adduct predicted by the IMDA reaction mechanism undergoes facile epimerization at high temperature or in the presence of silica gel. Structural revision on the adducts has been made.
