Abstract:Aggregation generally quenches the light emissions of chromophoric molecules. In this review, we demonstrate that 1,1-disubstituted 2,3,4,5-tetraphenyl siloles and 2,5-difunctionalized siloles as well as their polymers exhibit the opposite behaviors. Instead of quenching, aggregation has greatly boosted their photoluminescence quantum yields by up to two orders of magnitude, turning them from faint fluorophores into strong emitters. Such “abnormal” phenomenon of “aggregation-induced emission (AIE)” is attributed to restricted intramolecular rotations of the peripheral phenyl rings against the central silole core, which block the nonradiative channel via the rotational energy relaxation processes and effectively populate the radiative decay of the excitons. Utilizing such a novel effect, siloles and their polymers find an array of applications as: sensors for chemicals, explosives, pH, and biomacromolecules (proteins, DNAs and RNAs), indicators for determining CMC and monitoring layer-by-layer self-assembling, biocompatible fluorogens for cell imaging, visualizing agent for DNA gel electrophoresis, biolabels for immunoassay, stimuli-responsive organic nanomaterials, magnetic fluorescent nanoparticles for potential bio-imaging and -separation, and outstanding materials for efficient OLEDs and PV cells.

Abstract:Aggregation-induced emission (AIE) is a novel photophysical phenomenon coined in 2001 by our group and describes the enhanced light emission of some luminogens in the aggregate or solid state. The combination of AIE research and polymer science is a smart approach to produce functional luminescent materials with mechanical strength and excellent processability for real-world applications. In this feature article, recent progress in AIE polymeric systems, including chemical synthesis and physical blending strategies, is summarized. Through chemical synthesis, various AIE-active polymers, such as covalently bonded polymers, supramolecular polymers, and nonconjugated luminescent polymers, can be obtained. Serving as environmentally sensitive probes, AIE luminogens can also be physically doped into polymers to generate interesting systems. Finally, outlooks and perspectives on the future direction of AIE polymeric systems are discussed.

Abstract:Polymeric nanomaterials with aggregation-induced emission (AIE) characteristics have attracted significant attention from the scientific community because of their extensive biomedical applications. Nanoparticles prepared from AIE materials possess favorable advantages over fluorescent molecular dyes, including higher photostability, brightness, turn-on emission, easy functionalization, and tunable size/topology. In this review, we systematically summarize the preparation strategies of various polymeric AIE nanomaterials, including the encapsulation method, free radical copolymerization, controlled radical polymerization, click polymerization, supramolecular assembly, as well as post-polymerization reaction. Some special polymer topologies, such as star-shaped, crosslinked, and 2D polymers, are discussed in detail. Last but not least, perspectives on AIE polymeric nanomaterials are provided to stimulate future development.

Abstract:A series of naphthalimide-tetrazines were developed as bioorthogonal fluorogenic probes, which could produce significant fluorescence enhancement, notable aggregation-induced emission (AIE) characters and multicolor emissions after bioorthogonal reaction with strained dienophiles. Manipulating the pi-bridge in the fluorophore skeleton allows fine-tuning of the emission wavelength and influences the AIE-active properties. With these probes, we succeeded in no-wash fluorogenic protein labeling and mitochondria-selective bioorthogonal imaging in live cells.

Abstract:Self-assembly of amphiphilic luminescent copolymers is a general route to fabricate fluorescent polymeric microparticles (FPMs). In this work, the FPMs with aggregation-induced emission (AIE) feature were fabricated via the combination of the chain transfer free radical polymerization and "one-pot" multicomponent reaction, which conjugated the aldehyde-containing AIE active dye AIE (CHO-An-CHO) and amino-terminated hydrophilic polymer (ATPPEGMA) using mercaptoacetic acid (MTA) as the "lock" molecule. The structure, chemical compositions, optical properties as well as biological properties of the PPEGMA-An-PPEGMA FPMs were characterized and investigated by means of a series of techniques and experiments in detail. We demonstrated the final copolymers showed amphiphilic properties, strong yellow fluorescence and high water dispersibility. Biological evaluation suggested that PPEGMA-An-PPEGMA FPMs possess low cytotoxicity and can be used for cell imaging. More importantly, many other AIE active FPMs are expected to be fabricated using the similar strategy because of the good substrate and monomer applicability of the multicomponent reaction and chain transfer living radical polymerization. Therefore, we could conclude that the strategy described in this work should be of great interest for fabrication of multifunctional AIE active nanoprobes for biomedical applications.

Abstract:Fluorescent probes have been widely investigated for their features of rapid response, easy operation and high sensitivity. Among them, reaction-based fluorescent probes, for their unique reaction-based nature, guarantee them with excellent selectivity, effectively avoiding the possible interference from other chemical and biological species in physiological environment. Conventional reaction-based fluorescent probes are aggregation-caused quenching (ACQ) fluorophores. The application of these kinds of probes are limited for their poor photostability and narrow Stokes shifts. Compared with ACQ fluorophores, aggregation-induced emission (AIE) fluorophores become emissive in aggregation states with higher signal-to-noise ratio, better photostability and larger Stokes shifts. In this review, we summarize the latest developed reaction-based AIE-active probes, including the design principle and application in various sensing systems and give an outlook for the future development of this kind of promising fluorescent probes.

Abstract:It is found that the fluorescence of aliphatic poly(amido amine)s including linear and hyperbranched ones can be dramatically enhanced by simple aggregation of polymer chains, attributing to the formation of a variety of intra- and interchain clusters with shared lone-pair electrons and the restriction of intramolecular motions. Thanks to the combination of strong solid fluorescence and excellent biocompatibility, these non-conjugated polymers become promising candidates for bioimaging such as bacterial detection. This finding not only extends the aggregation-induced emission (AIE) systems from conjugated compounds to non-conjugated materials, which expands the bioapplication range of AIE systems, but also sheds light on the exploration of novel unconventional luminogens.

Abstract:“United we stand, divided we fall.”–Aesop.Aggregation‐induced emission (AIE) refers to a photophysical phenomenon shown by a group of luminogenic materials that are non‐emissive when they are dissolved in good solvents as molecules but become highly luminescent when they are clustered in poor solvents or solid state as aggregates. In this Review we summarize the recent progresses made in the area of AIE research. We conduct mechanistic analyses of the AIE processes, unify the restriction of intramolecular motions (RIM) as the main cause for the AIE effects, and derive RIM‐based molecular engineering strategies for the design of new AIE luminogens (AIEgens). Typical examples of the newly developed AIEgens and their high‐tech applications as optoelectronic materials, chemical sensors and biomedical probes are presented and discussed.

Abstract:Novel fluorescent nanoparticle (FNP)-based bioprobes are expected to generate new medical diagnostic techniques in biomedical and biological areas for their superior brightness and photostability compared with conventional molecular probes including small organic dyes and fluorescent proteins. Potentially interesting nanoscale platforms for various biomedical applications are greatly attractive due to the potential to avoid exposure of human tissues to toxic drugs, enhancing delivery of hydrophobic therapeutics and fabricating multifunctional imaging, targeting and delivery system. In this review, recent progress in the area of novel aggregation induced emission (AIE)-based FNPs is summarized over the past few years (2007-2013), and the reported fabrication methodologies of these fluorescent systems including non-covalent and covalent strategies are mainly discussed, and the biomedical applications of AIE-based FNPs are also summarized.

Abstract:Aggregation-induced emission (AIE) has become a hot research topic in chemistry and materials science. AIE polymers integrate the advantages of polymeric materials and AIE luminogens and show unique properties compared with traditional fluorophores and low-mass AIE materials for practical applications. In this review, we briefly summarize the progress of AIE polymers in sensing, imaging and theranostic applications in the last 3 years. It is hoped that this review will accelerate the development of these promising materials and serve as a trigger for future innovation in related research.

Abstract:Water dispersion aggregation-induced emission (AIE) dyes based nanomaterials have recently attracted increasing attention in the biomedical fields because of their unique optical properties, outstanding performance as imaging and therapeutic agents. The methods to conjugate hydrophilic polymers with AIE dyes to solve the hydrophobic nature of AIE dyes and makeS them widely used in biomedicine, which have been extensively explored and paid great effort previously. Although great advance has been made in the fabrication and biomedical applications of AIE-active polymeric nanoprobes, facile and efficient strategies for fabrication of biodegradable AIE-active nanoprobes are still high desirable. In this work, amphiphilic biodegradable fluorescent organic nanoparticles (PLL-TPE-O-E FONs) have been fabricated for the first time by conjugation of AIE dye tetraphenylethene acrylate (TPE-O-E) with Poly-l-Lysine (PLL) through a facile one-step Michael addition reaction, which was carried out under rather mild conditions, included air atmosphere, near room temperature and absent of metal catalysts or hazardous reagents. Due to the unique AIE properties, these amphiphilic copolymers tend to self-assemble into high luminescent water dispersible nanoparticles with size range from 400 to 600nm. Laser scanning microscope and cytotoxicity results revealed that PLL-TPE-O-E FONs can be internalized into cytoplasm with negative cytotoxicity, which implied that PLL-TPE-O-E FONs are promising for biological applications.

Abstract:Fluorescence bioimaging is considered an indispensable technique in biological research due to its high sensitivity, excellent spatial and temporal resolution, non-invasiveness, rapid and real-time responsiveness, as well as its ease of accessibility. Among these, fluorescence probes play a pivotal role in fluorescence imaging technology thanks to their strong specificity, high sensitivity, rapid response, and straightforward implementation. Aggregation-Induced Emission (AIE) molecules exhibit a unique phenomenon: they emit weak or no fluorescence when dissolved in a solution but become highly luminescent under aggregated/clustering conditions. Leveraging this, fluorescent probes with AIE characteristics often demonstrate fluorescence activation under conditions of spontaneous aggregation or binding with analytes, showcasing high sensitivity and exceptional signal-to-noise ratios. Consequently, fluorescence probes based on the distinctive optical properties of AIE hold immense potential for applications in fluorescence bioimaging. This paper primarily investigates the synthesis properties and applications in biosensing of a series of novel organic fluorescence probes with aggregation-induced emission (AIE) characteristics. By synthesizing a range of compounds with AIE properties and experimentally determining their photophysical properties and molecular structures, the study explores the potential applications of these compounds in DNA recognition, protein detection, small molecule recognition, cellular pH detection, and bioimaging.

Abstract:Aggregation-induced emission (AIE) dyes have received wide-spread concern since their inception. Several types of AIE-based fluorescent nanoparticle (FNP) have been developed, and the potential applications of these FNPs have also been explored. Recent studies of AIE-based FNPs in biological areas have suggested that they show promise as bio-materials for cell imaging and other biomedical applications. This article reviews recent progress in the synthesis of AIE-based FNPs via non-covalent, covalent and novel one-pot strategies, and the subsequent cell-imaging of those AIE-based FNPs. Many successes have been achieved, and there is still plenty of space for the development of AIE-based FNPs as new bio-materials.

Abstract:With the ever increasing interest in biomedical applications of luminescent probes, the development of novel luminescent nanoparticles which showed high water dispersibility, strong luminescence and excellent biocompatibility has been extensively pursued. In this work, cross-linked luminescent polymeric nanoparticles based on aggregation induced emission (AIE) dyes were fabricated through combination of free radical polymerization and ring-opening reaction using AIE dye as the fluorogen and linker simultaneously. Our results demonstrated that these AIE dye based cross-inked polymeric nanoparticles showed high water dispersibility, remarkable luminescent properties and excellent biocompatibility, is highly potential for biomedical applications.

Abstract:The aggregation-induced emission (AIE) is a photochemical process described in 2001, where the aggregation of specific kinds of organic compounds enhances the emission of light performed by these organic compounds. Since then, this phenomenon had attracted much interest because of its potential application in optics, electronics, energy and bioscience. In this review, the main concepts of AIE are going to be explained through the mechanistic decipherment of these photophysical processes. Additionally, some AIE systems will be discussed, describing the phosphorescence enhancement induced in organic molecules by this effect, but we will be focusing on the cyano-containing AIEgens, its recent advances and the driving forces that lead to the AIE effect in these cyano-containing molecules.

Abstract:Aggregation-induced emission (AIE) luminogens with highly tunable properties show great potential for many applications. In this study, we synthesized a new family of AIE-type poly(ethylene glycol)-block-poly(9-anthrylmethyl lysine) (PEG-b-PLys-An) diblock copolymers by taking advantage of amphiphilic self-assembly and rigid helical backbones. These copolymers can self-assemble into various assemblies through nanoprecipitation methods. The micelles using N,N-dimethylformamide (DMF) as a cosolvent present brighter fluorescence than the vesicles prepared from tetrahydrofuran (THF). We demonstrate that the decreased solubility of copolymers in DMF results in the formation of more compact micelles with more excimer formation during the self-assembly process, while better solvent THF favors the formation of vesicles with stretched core chains. In addition, the secondary conformation of the polypeptide block shows pronounced effects on the fluorescence property. We further show the internalization of the assemblies using two types of cells by cellular uptake experiments. By the delicate design of the block copolymer, we successfully prepare the morphology- and conformation-dependent AIE materials for potential biomedical applications.

Abstract:Supramolecular polymers are typical self-assemblies, in which repeating monomer units are bonded together with dynamic and reversible noncovalent interactions. Supramolecular polymers can combine the advantages of polymer science and supramolecular chemistry. Aggregation-induced emission (AIE) means that a molecule remains faintly emissive in the dispersed state but intensively luminescent in a highly aggregated state. AIE has brought new opportunities and further development potential to the field of polymeric chemistry. The integration of AIE luminogens with supramolecular interactions can provide new vitality for supramolecular polymers. Therefore, it is essential for scientists to understand the preparation and applications of AIE-active supramolecular polymers. This review focuses on the recent advanced progress in the preparation of AIE-active supramolecular polymers. In addition, we summarize the newly developed supramolecular polymers with an AIE nature and their applications in chemical sensing, and in vitro and in vivo imaging, as well as the visualization of their structure and properties. Finally, the development trends and challenges of AIE-active supramolecular polymers are prospected.

Abstract:AbstractOwing to the outstanding photophysical properties, organic luminescent materials featuring aggregation‐induced emission (AIE) characteristics have attracted wide attention in various fields. Numerous researches focused on low‐mass AIE luminogens, and relatively less attention has been paid on AIE polymers and the related applications, in spite of the fact that AIE polymers exhibit excellent advantages of processability, multifunctional integration and synergistic effects. In this review, we briefly summarize and discuss the superiorities of AIE polymers in preparation, properties and bio‐applications, and the considerable progress in these aspects are introduced as well. Finally, the structure‐property relationship, challenges and opportunities are also discussed. Hopefully, this review will be a trigger for smart AIE polymer research and further broaden their applications.

Abstract:Fluorescent materials with aggregation-induced emission (AIE) have standout as one of the most interesting candidates for biomedical applications. The development of new methods for fabrication of AIE-active materials will surely advance their applications in related fields. In this work, we reported a novel method for preparation of AIE-active luminescent polymer (Lys-PEG-TPE) through a three-component reaction involving carbon dioxide (CO 2 ) using lysine, Polyethylene glycol (PEG) and AIEgen as the reactants. On account of the introduction of PEG, Lys-PEG-TPE could well disperse in aqueous solution, while hydrophobic TPE will aggregate in the core of self-assembled nanoparticles, endowing the obvious enhancement of fluorescence intensity. Results from biological assays suggested that Lys-PEG-TPE exhibits almost no toxicity after incubation with HepG2 cells and its fluorescence can be clearly observed in cytoplasm after cell internalization. More importantly, the AIE feature of Lys-PEG-TPE could not only effectively solve the aggregation-induced quenching effect of traditional fluorescent molecules, the carboxyl groups of lysine could also be used for loading functional cationic ions (e.g., anticancer drugs or imaging agents) to fabricate multifunctional theranostic and imaging composites. Considering the benign reaction conditions, high atom economy, and well tunability of reactants of multicomponent reactions, this strategy should be of broad interesting for fabrication of multifunctional AIE-active materials for biomedical applications.

Abstract:Fluorescent probes have long been regarded as tools for imaging living organisms with advantages such as high sensitivity, good designability and multifunctional potential. Many fluorescent probes, especially the probes based on aggregation-induced emission (AIE) dyes, have received increasing attention since the AIE phenomenon was discovered. These AIE dye-based fluorescent probes could elegantly overcome the notorious quenching effect caused by aggregation of conventional organic dyes. However, it is still difficult to directly apply these AIE-active dyes for biomedical applications owing to their hydrophobic nature. Therefore, the development of novel and facile strategies to endow them with water dispersibility is of critical importance. In this work, we exploit an efficient and simple strategy to fabricate an AIE dye-based fluorescent copolymer through the combination of reversible addition-fragmentation chain transfer and the Biginelli reaction. Moreover, the copolymer can self-assemble to fluorescent polymeric nanoparticles (FPNs) in water solution. Hydrophilic poly(PEGMA-co-AEMA) was reacted with the AIE-active dye 4',4‴-(1,2-diphenylethene-1,2-diyl)bis([1,1'-biphenyl]-4-carbaldehyde (CHO-TPE-CHO) to form amphiphilic luminescent polymers using urea as the connection bridge. The successful synthesis of the final products (poly(PEGMA-co-AEMA-TPE) FPNs) was confirmed by various instruments. Furthermore, Transmission electron microscopy (TEM) images manifest that poly(PEGMA-co-AEMA-TPE) copolymers will self-assemble into spherical nanoparticles in aqueous environments with sizes between 100 nm and 200 nm. The cell uptake and bioimaging experiment confirm that poly(PEGMA-co-AEMA-TPE) FPNs have excellent biocompatibility and emit strong green fluorescence in a cellular environment. Thus, poly(PEGMA-co-AEMA-TPE) FPNs are excellent candidates for biomedical applications.
