Abstract:Objective: To look f or a new kind of anti-tumor compounds with high activity and low toxicity. Methods: Piperazine was reacted w ith ［bi-(4-fluorophenyl)］methyl chloride in the presence of phase transfer c atalyst to form the key intermediate Ⅰ. The different kinds of antitumor groups were introduced into 4-position of Ⅰ via different react ions, including alkylation, alkyldithioformy lation, Mannich reaction, and acylation, to give the corresponding 1-［bi-(4- fluo rophenyl)methyl］-4-substituted piperazines Ⅱa-i. The tumor-inhibition of compounds Ⅱ a-iwas tested against the growth of eight kinds of cell s by SRB or MTT method. Resu lts: The synthetic method of key intermediate Ⅰwas improved and the yield was i ncreased from 54% to 74%. Preliminary result of biological activity showed that this kind of compounds exhibited higher tumor-inhibition against skov3 than against other kinds of teste d cells. In comparison with the structures and activities of compounds Ⅱc and Ⅱg, the presence of dithioester group in the molecules affected the biological activ ity obviously. The introduction of benzyl in the 4-position of piperazine could en hance the biological activity. Conclusion: 1-［Bi-(4-fluoroph enyl)methyl］-4 -substituted benzyl piperazine is a potential anti-tumor compounds.

Abstract:The title compound 1-(3-((benzo[d][1,3]dioxo1-5-yloxy)methyl)phenethyl)-4-phenylpiperidin-4-ol (4, C27H29NO4, M-r = 431.51) was synthesized by a four -step reaction and characterized by H-1 NMR, C-13 NMR, elemental analysis and single -crystal X-ray diffraction. The crystal is of triclinic, space group P1 with a = 5.8033(12), b = 10.705(2), c = 18.393(4) angstrom, alpha = 83.44(3), beta= 82.71(3), gamma = 88.88 (3)degrees, V= 1126.0(4) angstrom(3), Z= 1, Dc= 1.273 g/cm(3), S = 1.070, mu= 0.681 mm(-1), F(000) = 460, R = 0.0618 and wR = 0.1619 for 6452 observed reflections with I> 2 sigma(I). Geometry conformations revealed that the dihedral angle between benzodioxole and aromatic planes contributed to the formation of C-H center dot center dot center dot pi interactions. Hydrogen bonds and van der waals interactions were observed to stabilize the three-dimensional packing structure. The compound showed a moderate inhibitory activity against the prostate cancer cells.

Abstract:Seven new 1, 1-dialkyl-4-(3-bromopropionyl) piperazinium bromides were synthesized. Their structures were elucidated by IR, 1H NMR and elemental analysis. The compounds were screened for antitumor, analgesic and antioxide SOD activities. Though none of them showed antitumor activity, most of them showed distinct analgesic activity and antioxide SOD activity.

Abstract:Ten novel benzotriazole compounds were synthesized. Their chemical structures were confirmed by 1 H NMR, IR, and elemental analyses, coupled with three selected single-crystal structures (compounds A2 , B3 , and B5 ). Their antimycotic and antitumor activities were also investigated. The title compounds showed some antitumor activities, especially in the case of A3 and A4 , which showed the most potent activity of propagation inhibition in liver and galactophore cancer cells.
