Abstract:Aim To find a reasonable way to prepare the designed CPP32 inhibitors. Method Ugi four-component condensation reaction was used to synthesize peptide mimic CPP32 inhibitors; Results A key isocyanide component (aspartate-derived isocyanide 3) and one of the designed CPP32 inhibitors 4 (as a template) were synthesized; Conclusion The CPP32 inhibitor 4 was synthesized by the newly developed procedure, which is an Ugi four-component condensation reaction based on aspartate-derived isocyanide 3. This method can be used to build up the CPP32 inhibitor library.

Abstract:Two biocatalytic reactions, transamination catalyzed by transaminases and reductive amination catalyzed by amino acid dehydrogenases, can be used for asymmetric synthesis of optically pure unnatural amino acids. However, although transaminases show a great diversity and broad substrate spectrum, most transaminase reactions are reversible, while amino acid dehydrogenases catalyze reductive amination irreversibly but with strict substrate specificity. Accordingly, herein we developed a tri-enzyme one-pot reaction system to exploit the respective advantages of transaminases and amino acid dehydrogenases, while overcoming the disadvantages of each. In this work, representatives of all four subgroups of transaminases coupled with different amino acid dehydrogenases to produce five l- and four d- unnatural amino acid products, using ammonia and the co-enzyme NAD(P)H, which is regenerated by a robust alcohol dehydrogenase with 2-propanol as cheap cosubstrate. The complete conversion and high enantiopurity (ee > 99 %) of the products, demonstrated it as an ideal alternative for asymmetric synthesis of chiral amino acid compounds.

Abstract:Described is a facile helix-nucleating template based on a tethered aspartic acid at the N-terminus [terminal aspartic acid (TD)]. The nucleating effect of the template is subtly influenced by the substituent at the end of the side-chain-end tether as indicated by circular dichroism, nuclear magnetic resonance, and molecular dynamics simulations. Unlike most nucleating strategies, the N-terminal amine is preserved, thus enabling further modification. Peptidomimetic estrogen receptor modulators (PERMs) constructed using this strategy show improved therapeutic properties. The current strategy can be regarded as a good complement to existing helix-stabilizing methods.

Abstract:Peptide nucleic acids (PNA) have received great attention as tools in molecular biology and biotechnology, diagnostic agents. and potential antisense drugs. Many derivatives and analogues of PNA were designed and prepared to improve their physicochemical and biological properties. In most cases, PNA oligomers were synthesized by peptide synthesis from N-protected monomers. In this work. a new method to obtain PNA monomers and PNA chain prolongation by Ugi four-component condensation reaction was tested by synthesizing PNA monomers and dimers. This method differs from classical peptide chemistry in that no prior preparation of PNA monomers is needed as they are built up along with the extension of chain. The synthesis of the key component isocyanide is also presented.

Abstract:A general and efficient method for the synthesis of azabicyclo[4,3,0]nonanone and azabicyclo[5,3,0]clecanone amino acid derivatives was developed with the palladium-catalyzed coupling of intermediates 9 and 10 with BocNH(2) and Boc(2)NH and the following reduction of the C-C double bond by hydrogenation as the key steps. The exploration of the synthesis of C-6-substituted dipeptide mimetics from 9 and 10 using Suzuki coupling as the key reaction has also been performed.

Abstract:Unnatural polar -aminoxy acid residue with proteingenous hydroxymethyl side chain, a building block of the peptidomimetic foldamer of -aminoxy peptide, was synthesized starting from natural amino acid L-serine. The starting material, L-serine, undergoes a reaction sequence to produce compound 1 in three steps: (1) the neighboring carboxyl group participates in diazotization/bromination to transform the amino group to a bromo group, (2) the C-terminal carboxyl group is protected, and (3) bromide is SN2-displaced by N-hydroxyl phthalimide to introduce a N-O bond. After several conventional deprotection/coupling reactions, compound 1 is easily transformed to an -aminoxy diamide, which can be widely used in peptidomimetics design.

Abstract:Peptide nucleic acids (PNA) oligomers were synthesized in most cases by peptide synthesis from N-protected monomers. In this work a new method of obtaining PNA monomer by Ugi four-component condensation reaction was tested by solid-phase synthesis. The Fmoc protected PNA monomer was build up with thymin-1-yl acetic acid, 3-methylbutyl aldehyde, Fmoc protected aminoethyl isocyanide and Gly-Wang resin.
A straightforward method to prevent the under-estimated problem of aspartimide formation during chemical ligation-mediated protein synthesis

Abstract:Formation of a five-membered ring aspartimide through the attack of a backbone amide to the side chain of aspartate and asparagine residues is a long-known side-reaction in solid phase peptide synthesis, and is also associated with in vivo protein ageing and instability of purified proteins. Conversely, its possible occurrence during chemical ligation-based protein synthesis, in particular when using the gold-standard reaction NCL (native chemical ligation), is dubious. We herein report a systematic study which demonstrates that the prevalence of this side-reaction may have been overlooked, due to the difficulty to identify it through standard HPLC analytical methods, but also the in situ conversion of aspar-timide into other byproducts, having the same molecular mass as the parent aspartate residue. We show that the for-mation of aspartimide and related byproducts can be limited by adopting "good NCL practices", which involve restricting the ligation temperature and reaction times, as well as replacing the commonly used phosphate buffer with HEPES. How-ever, the efficiency of such precautions is expected to vary considerably depending on the sequence of the target protein, and the amount of byproducts is expected to grow with the length of the target protein, as a result of the number of NCL reactions and potential aspartimide hotspots. To overcome such limitations, we developed a novel straightforward and potentially generally applicable methodology based on the temporary protection of the backbone nitrogen by a 2-(4-aminobutanoyloxy)-4-methoxybenzyl (GABA-Hmb) group. This strategy was validated by the byproduct-free synthesis of SUMO-2 and a SUMOylated peptide mimic.

Abstract:Despite numerous optimizations in peptide synthesis, the formation of aspartimide remains a significant side reaction that needs to be addressed. Herein, we introduce an approach that utilizes hydrazide as a carboxylic-acid-protecting group to reduce the formation of aspartimide. The aspartic acid hydrazide effectively suppressed the formation of aspartimide, even under microwave conditions, and was readily converted to native aspartic acid using CuSO(4) in an aqueous medium.

Abstract:The antimicrobial peptide tachyplesin I was used as a model to apply the title strategy, which was developed for the preparation of peptidic macrocycles with double disulfide surrogates. The folding and activity of the tachyplesin I analogues were found to be sensitive to the structure of the disulfide surrogates, thus underlining the necessity of a flexible synthetic route for generating disulfide bond surrogates with high structural diversity. Abstract:A robust, practical, and sustainable isomerization‐suppressed peptide bond formation via acyl sulfonamide, a twisted amide, is disclosed. Tosyl isocyanate and pentafluorobenzyl bromide were applied in combination to activate the peptide C‐terminus, which then reacted with an amine to yield an elongated peptide with high stereochemical purity. Careful analysis of NMR spectra of the active intermediate revealed the presence of an intramolecular hydrogen bond, suggesting that the hydrogen bond suppressed Cα‐epimerization during amidation. The isomerization suppression by the intramolecular hydrogen bond is expected to be effective even under high dilution conditions, making the present method a powerful tool for the synthesis of complex macrocyclic peptides. In addition to peptide synthesis, the developed synthetic entry to twisted amides can be applied to the investigation of transition metal‐catalyzed N–C bond activation. Moreover, the application to the N–C bond activation returned insight into peptide synthesis, leading to the use of sulfonamide as a protecting group of carboxylic acid that can be orthogonally removed in the presence of other conventional protecting groups.

Abstract:Aim To synthesize the tripepide Weinreb amide Boc-Asp(OBzl)-β-Ala- Asp(OBzl)-N(OMe)Me (7) as a useful precursor of aspartyl peptide aldehyde derivatives; Methods DCC, IBCF method was used for preparation of Weinreb amide; N-hydroxysuccinimide activated ester was used in peptide synthesis; and Boc as N-protecting group of amino acid. Results Boc-Asp(OBzl)-N(OMe)Me (3), Boc-β-Ala-Asp(OBzl)-N(OMe)Me (5), and Boc-Asp(OBzl)-β-Ala-Asp (OBzl)-N(OMe)Me (7) were synthesized successfully. Abstract:Aim The PNA(peptide nucleic acids)as a new type of the antisense compound family,is being researched as the biological probe and applied to the diagnoses and treatment of diseases.However,it is necessary to optimize the PNA′s structure because it has some shortcomings.In this paper,the exploration and synthesis of the PNA analog monomer by the Ugi 4CC reaction were reported.Methods The general idea of synthesizing the PNA was changed and the PNA monomer was synthesized by the Ugi 4CC reaction in fewer steps with easy changes in the groups.Result It designed and synthesized the key component in the Ugi reaction:isocyanide,and PNA monomer.Conclusion The structures of the products were confirmed by MS,IR, 1H NMR spectra and C,H,N elemental analysis.

Abstract:A novel solubilizing tag system for Asp/Asn/Glu/Gln-containing peptides is described. In this method, an Asp/Glu[Dbz-Cys-NH2 ]-containing peptide (Dbz: 3,4-diaminobenzoic acid) is first synthesized through fluorenylmethyloxycarbonyl (Fmoc) solid-phase peptide synthesis. The solubilizing moiety containing an oligo-Lys group is then attached to the peptide in hexafluoroisopropanol through a trityl anchor to afford a hydrophilic tagged peptide. To detach the solubilizing tag, the Dbz moiety of the tagged peptide is activated with NaNO2 , and the Asp/Asn/Glu/Gln-containing peptide is obtained through hydrolysis or ammonolysis. This synthetic approach proved to be compatible with native chemical ligation, and amyloid β-protein 1-42 was successfully synthesized by the solubilizing-tag-aided native chemical ligation-desulfurization method.
