Abstract:Mild conditions have been developed to achieve a Pd(OAc)(2)-catalyzed cross-coupling between indoles and arylsiloxanes in the presence of TBAF and Ag2O in acidic medium. Electron-deficient arylsiloxanes presented high efficiency in this system to give the arylated indoles in excellent yields.

Abstract:An effective and practical strategy has been established for the direct and site-selective arylation of indoles at the C4 and C5 positions with the aid of a readily accessible, cheap, and removable pivaloyl directing group at the C3 position. This transformation shows good functional-group tolerance and could serve as a powerful synthetic tool for the synthesis of medicinally relevant compounds. This method and those developed in previous research together enable the regiocontrolled direct arylation of indole at each C-H bond without prefunctionalization of the reactive sites.

Abstract:A Rh(I)‐catalyzed chelation‐assisted C2‐selective C−H decarbonylative alkylation of indoles with readily available, cheap, safe and structurally diverse alkyl carboxylic acids or anhydrides has been developed. A wide variety of primary and secondary alkyl carboxylic acids and differently substituted indoles are compatible with this transformation, allowing facile synthesis of various C2‐alkylated indoles with high efficiency and broad tolerance of diverse functional groups. The reaction proceeds in the absence of any external oxidant, and the presence of readily installable and removable N‐pyrimidyl directing group is critical for catalysis. The process is convenient and scalable, and avoids the use of a dried solvent and an inert atmosphere. Moreover, selective C7‐alkylation and C2, C7‐dialkylation have also been achieved by slightly modifying the reaction conditions.

Abstract:Heteroarenes containing carbon–silicon (C–Si) bonds are important building blocks that play an important role in the construction of natural products, pharmaceuticals, and organic materials. In this context, the C–H silylation of heteroarenes is a topic of intense interest. Indole C–H silylation can preferentially occur at the nucleophilic C3 and C2 position (pyrrole core), while accessing the C4-C7 positions (benzene core) of the indole remains highly challenging. Here, we show a general strategy for the regioselective C7-H silylation of indole derivatives. Mainly, the regioselectivity is determined by strong coordination of the palladium catalyst with phosphorus (III) directing group. Using this expedient synthetic strategy, the diverse C7-silylated indoles are synthesized effectively which exhibits the broad functional group compatibility. Moreover, this protocol also been extended to other heteroarenes such as carbazoles. The obtained silylated indoles have been employed in various transformations to enable the corresponding differently functionalized indole derivatives. Significantly, a cyclopalladated intermediate is successfully synthesized to test the hypothesis about the P(III)-directed C–H metalation event. A series of mechanistic experiments and density functional theory (M06-2X) calculations has shown the preferred pathway of this directed C–H silylation process.

This selective CH activation reaction was implemented without the assistance of directing groups and took place under mild conditions. Abstract:Here, we report an practical method to selectively access silylated pyrrolo[1,2-a]indole using alkenyl indoles and readily available silanes through a radical cascade cyclization. The reaction proceeds through an unusual intermolecular polarity-mismatched addition of a nucleophilic silyl radical to e-rich alkene SOMOphiles via a coordination-assisted interaction. Abstract:A palladium-catalyzed direct C-H bond sulfonylation of indoles with the insertion of sulfur dioxide is achieved through a three-component reaction of 1-(pyridin-2-yl)indoles, DABCO·(SO2)2, and aryldiazonium tetrafluoroborates under mild conditions. Diverse 2-sulfonated indoles are generated by using 10 mol % of palladium(II) bromide as the catalyst at room temperature. This synthetic approach is efficient by merging palladium catalysis and insertion of sulfur dioxide via a radical process. 2-Pyrimidinyl can be used as the directing group well as for the C-H bond sulfonylation. Additionally, the directing group can be easily removed.

Abstract:The possibility of using phosphine-free ligands to promote Pd-catalyzed indolization of 2-bromoanilines with internal alkynes was examined for the first time.
