Abstract:BACKGROUND This study explored the effects of nano-hydroxyapatite/polyetheretherketone (n-HA/PEEK)- coated sandblasted, large-grit, and acid-etched (SLA) implants on inflammatory cytokines and osseointegration in peri-implantitis model beagle dogs. MATERIAL AND METHODS Peri-implantitis models were established. Eight beagle dogs were randomly and evenly assigned into SLA tied, SLA + n-HA/PEEK tied, SLA untied, or SLA + n-HA/PEEK untied groups. A special periodontal probe was used to detect the plaque index (PLI), probing depth (PD), and modified Sulcus Bleeding Index (mSBI). Gingival crevicular fluid was collected and an ELISA kit was utilized to detect IL-1, IL-6, and IL-17 levels. The colony-forming units were counted and the maximum shear strength of implants was tested using the axial pullout test. HE staining was used to detect the inflammation of peri-implant bone tissues. Osseointegration was observed through toluidine blue staining. Bone-to-implant contact (BIC) was obtained through histological observation and the mineral apposition rate (MAR) was calculated after immune fluorescent double staining. RESULTS The SLA tied group demonstrated higher levels of PLI, PD, mSBI, IL-1, IL-6, and IL-17 and a higher degree of inflammation than the SLA + n-HA/PEEK tied group. The tied groups also displayed similar results over the untied groups at the same time point. The maximum shear strength, BIC, and MAR in the SLA tied group were significantly lower than in the SLA + n-HA/PEEK tied group. CONCLUSIONS Our findings demonstrate that SLA + n-HA/PEEK implants can promote osseointegration and relieve the inflammation response of peri-implantitis in beagle dogs.

Abstract:Purpose. The purpose of this study was to investigate the effect of the thin nano-hydroxyapatite (nano-HA) coating on implant osseointegration in an ovariectomized rat model.Materials and Methods. Implants were divided into a control group and a test group (nano-HA-coated group). Surface morphology was examined using field-emission scanning electron microscopy (FSEM). Surface roughness of both groups was performed. Sixteen ovariectomized rats randomly received 2 implants in both tibiae. After 12 weeks of implantation, tibias were retrieved and prepared for histomorphometric evaluation and removal torque tests (RTQ).Results. Rodlike crystals uniformly covered the porous surfaces and the surface morphology of the implant was still clear. No significant differences were found in surface roughness between the 2 groups (P > .05). More bone tissue was formed around test implants compared with control implants. Test implants showed a significantly greater BIC, bone area within all threads, and RTQ values compared with control implants (P < .05).Conclusions. These results indicate the thin nano-HA coating by an electrochemical process has potential benefits to enhance implant osseointegration in ovariectomized rats. Abstract:Purpose: This study investigated the effects of a strontium-substituted nanohydroxyapatite (Sr-HA) coating, deposited onto porous implant surfaces using an electrochemical process, on implant osseointegration in a rabbit model. Materials and Methods: The surfaces were analyzed by field-emission scanning electron microscopy, x-ray diffractometry (XRD), Fourier transform infrared spectroscopy (FT-IR), a portable surface roughness tester, and inductively coupled plasma atomic emission spectroscopy (ICP-AES). Thirty implants (half HA-coated and half Sr-HA-coated) were inserted into femurs of 15 rabbits. After 2, 4, and 8 weeks, the femurs were retrieved and prepared for histomorphometric evaluation. Results: Microscopic examination showed a surface topography of rodlike crystals on both surfaces. XRD and FT-IR showed that the phase of the deposits was HA. No differences were found in surface roughness between the two groups. ICP-AES showed that the Sr/(Ca+Sr) molar ratio of Sr-HA coating was 10.1 mol%. Histologic observation showed that new bone appeared on both surfaces after 2 weeks and became mature after 8 weeks. Histomorphometric analysis showed no differences between the two groups in bone-to-implant contact at 2 weeks or in bone area within all threads at 2 and 4 weeks. The Sr-HA coated group had significantly higher bone-to-implant contact at 4 and 8 weeks. Significant differences were also found in bone area at 8 weeks. Conclusion: The present study showed that this Sr-HA coating, deposited using an electrochemical process, has the potential to enhance implant osseointegration. Abstract:Dental implants are considered a superior option for the replacement of missing teeth. However, the invasive nature of the surgical procedure often results in significant postoperative inflammation, and the prolonged healing period of 3-6 months presents a notable disadvantage. High mobility group box 1 (HMGB1) is a critical mediator of acute inflammation following surgical injury, which can hinder the onset of osseointegration. This study aims to examine whether the inhibition of HMGB1 can mitigate acute inflammation and subsequently enhance osseointegration. The findings indicate that HMGB1 inhibition markedly reduces inflammation and promotes bone repair in murine models. Further in vitro investigations into the regulatory mechanisms of HMGB1 in macrophages reveal its role in increasing Yes-associated protein (YAP) activity, which contributes to pro-inflammatory polarization. Additionally, conditioned media derived from macrophages influenced by HMGB1 significantly impair the migratory and osteogenic capabilities of bone marrow-derived mesenchymal stem cells, which are essential for bone regeneration. In vivo experiments further validate that the administration of exogenous HMGB1 exacerbates postoperative acute inflammation and obstructs osseointegration. The study concludes that inhibiting HMGB1 fosters an anti-inflammatory polarization of macrophages, leading to diminished postoperative acute inflammation and expedited osseointegration around dental implants in mice.
