Abstract:In this study, we describe a solution procedure for the preparation and surface modification of photostable colloidal silicon quantum dots (SiQDs) for imaging of cancer cells. Photoluminescent SiQDs were synthesized by reduction of halogenated silane precursors using a microemulsion process. It was shown that 1,8-nonadiyne molecules could be grafted onto the surface of hydrogen-terminated SiQDs via ultraviolet (UV)-promoted hydrosilylation, demonstrated by Fourier transform infrared spectroscopy (FTIR) measurements. In addition, various azide molecules were coupled onto nonadlyne-functionalized particles, rendering particles dispersible in selected polar and nonpolar solvents. The photoluminescence of functionalized SiQDs was stable against photobleaching and did not vary appreciably within biologically applicable pH and temperature ranges. To demonstrate compatibility with biological systems, water-soluble SiQDs were used for fluorescent imaging of HeLa cells. In addition, the SiQDs were shown to be non-cytotoxic at concentrations up to 240 mu g/mL. The results presented herein provide good evidence for the versatility of functionalized SiQDs for fluorescent bioimaging application.

Abstract:Fluorescent dyes such as rhodamines are widely used to assay the activity and image the location of otherwise invisible molecules. Si-rhodamines, in which the bridging oxygen of rhodamines is replaced with a dimethyl silyl group, are increasingly the dye scaffold of choice for biological applications, as fluorescence is shifted into the near-infrared while maintaining high brightness. Despite intense interest in Si-rhodamines, there has been no exploration of the scope of silicon functionalization in these dyes, a potential site of modification that does not exist in conventional rhodamines. Here we report a broad range of silyl modifications that enable brighter dyes, further red-shifting, new ways to modulate fluorescence, and the introduction of handles for dye attachment, including fluorogenic labeling agents for nuclear DNA, SNAP-tag and HaloTag labeling. Modifications to the bridging silicon are therefore of broad utility to improve and expand the applications of all Si-dyes.

Abstract:We report the synthesis, characterization, and photophysical and ion-binding properties of deep-red to near-infrared (NIR) fluorescent rhodamine derivatives, bearing two spirolactone rings and substitution of the oxygen atoms in the xanthene ring with sulfur atoms (1-S). The diastereoisomeric cis- and trans-forms of the rhodamine derivative were separated and the cis-form (cis-1-S) was structurally characterized by X-ray crystallography. Upon treatment with Hg2+ ion, cis-1-S was converted into the dual spirolactone ring-opened species, resulting in significant color change and fluorescence enhancement. Substitution of the oxygen atoms with sulfur and extended π-conjugation across the fused six-membered rings upon the two rings-opening processes in the presence of Hg2+ ion led to a significant red-shift of absorption (623 nm) and fluorescence (706 nm) peaks, compared to the ordinary rhodamine. Furthermore, the intracellular Hg2+ -sensing properties of the cis-1-S have been studied by confocal microscopy.

Abstract:Photostable and bright organic dyes emitting in the near-infrared region are highly desirable for long-term dynamic bioimaging. Herein, we report a synthetic approach to build novel methoxy modified Si-rhodamine (SiRMO) dyes by introducing the methoxybenzene on the xanthene moiety. The brightness of SiRMO increased from 2300 M-1 cm-1 (SiRMO-0) to 49000 M-1 cm-1 (SiRMO-2) when the substituent 2,5-dimethoxybenzene was replaced with 2,6-dimethoxybenzene. Moreover, the stability of SiRMO-2 was significantly improved due to the steric hindrance protection of the two methoxy groups on the ninth carbon atom of the xanthene. After fast cellular uptake, the SiRMO dyes selectively stained the mitochondria with a low background in live cultured cells and primary neurons. The high brightness and stability of SiRMO-2 significantly improved the capability of monitoring mitochondria dynamic processes in living cells under super-resolution conditions. Moreover, with the fluorescence nanoscopy techniques, we observed the structure of mitochondrial cristae and mitochondria fission, fusion, and apoptosis with a high temporal resolution. Under two-photon illumination, SiRMO-2 showed also enhanced two-photon brightness and stability, which are important for imaging in thick tissue.

Abstract:A general strategy for the efficient preparation of S-rhodamines from the condensation of diaryl thioether and 2-carboxybenzaldehydes was reported. We further took a morpholine containing spirolactam structure as an example to illustrate that these S-rhodamine dyes could be utilized to construct fluorescent probes based on the ring-opening process. This work provided a general approach for the synthesis of novel S-rhodamine dyes, thus possibly facilitating the development of fluorescence imaging.

Abstract:Si-rhodamine (SiR) is an ideal fluorophore because it possesses bright emission in the NIR region and can be implemented flexibly in living cells. Currently, several promising approaches for synthesizing SiR are being developed. However, challenges remain in the construction of SiR containing functional groups for bioimaging application. Herein, we introduce a general and simple approach by a condensation reaction of diarylsilylether and arylaldehyde in o-dichlorobenzene to synthesize a series of SiRs bearing various functional substituents. These SiRs have moderate to high quantum efficiency, tolerance to photobleaching, and high water solubility as well as NIR emitting, and their NIR fluorescence properties can be controlled through the photoinduced electron transfer (PET) mechanism. Fluorescence OFF-ON switching effect is observed for SiR 9 in the presence of acid, which is rationalized by DFT/TDDFT calculations. Moreover, reversible stimuli response toward temperature is achieved. Since positive charge enables mitochondrial targeting ability and chloromethyl unit can covalently immobilize the dyes onto the mitochondrial via click reaction between the benzyl choride and protein sulfhydryls, SiR 8 is identified as a valuable fluorescent marker to visualize the morphology and monitor the temperature change of mitochondria with high photostability.

Abstract:Near-infrared (NIR) dyes are of great interest in biomedicine due to diminished interfering absorption and fluorescence from biological samples, reduced scattering, and enhanced tissue penetration depth. In this context, we report the synthesis of rectilinearly pi-extended rhodamine dyes using a unique intramolecular nucleophilic substitution of aromatic hydrogen (SNArH) strategy. The strategy makes use of an SNArH reaction between a preorganized aromatic amino nitrogen and an electron-deficient carbon in the xanthylium ion. The SNArH reaction presented herein can be performed under mild conditions without a transition metal catalyst and can be expected to enable the preparation of a wide variety of pi-extended near-infrared fluorescent rhodamine dyes. Using this strategy, seven rectilinearly pi-extended rhodamines (RE1-RE7) that had fluorescence emission wavelengths in the near-infrared region were synthesized. RE1, RE3, and RE4 were lysosome targetable and showed good photostabilities. In addition, using dye RE1 as a precursor, we constructed a novel NIR fluorescent turn-on probe (RE1-Cu), which can be used for detecting Cu2+ in living cells, demonstrating the value of our NIR functional fluorescent dyes.

Abstract:The title compound, C33H34N4O2S·CH3OH, was prepared as a spirolactam ring formation of rhodamine 6 G dye for comparison with a ring-opened form. The xanthene and spirolactam rings are approximately planar [r.m.s. deviations from planarity = 0.122 (3) and 0.072 (6) Å, respectively]. The dihedral angles formed by the spirolactam and thiophene rings with the xanthene ring system are 89.7 (6) and 86.5 (2)°, respectively. The crystal structure features N—H...O and C—H...O hydrogen bonds.

Abstract:A new fluorescent probe 3 was synthesized, and it exhibited high selectivity for Fe-III over other commonly coexistent metal ions in both ethanol and water. Upon the addition of Fe-III, the spirocyclic ring of 3 was opened and a significant enhancement of visible color and fluorescence in the range of 500-600 nm was observed.

Abstract:Siloles represent an important emerging class of photoluminescent materials. Reported herein is a new synthetic strategy involving a tandem cyclization/Si-C activation reaction featuring high efficiency, wide substrate scope, and practical utility. This method enabled the first synthesis of benzofuran siloles as well as rapid access to conjugated siloles. During the course of the study we also uncovered an unusual yet general Si-C(sp(2)) activation in the presence of π acids.

Abstract:Fluorescence imaging is a powerful technique for visualizing biological events in living samples with high temporal and spatial resolution. Fluorescent probes emitting far-red to near infrared (NIR) fluorescence are particularly advantageous for in vivo imaging due to their high tissue permeability and low autofluorescence, as well as their suitability for multicolor imaging. Among the far-red to NIR fluorophores, Si-rhodamine is one of the most practical fluorophores for the development of tailor-made NIR fluorescent probes because of the relative ease of synthesis of various derivatives, the unique intramolecular spirocyclization behavior, and the relatively high water solubility and high photostability of the probes. This review summarizes these features of Si-rhodamines and presents recent advances in the synthesis and applications of far-red to NIR fluorescent probes based on Si-rhodamines, focusing on live-cell imaging applications such as fluorogenic probes, super-resolution imaging and dye–protein hybrid-based indicators.

Abstract:Rhodamine spirolactam based photoswitches have been extensively applied in super-resolution single-molecule localization microscopy (SMLM). However, the ring-opening reactions of spirolactams are cross-sensitive to acid, limiting their photoswitch use to neutral pH conditions. In addition, the ring-closing reactions of spirolactams are environment-sensitive and slow (up to hours), virtually making rhodamine spirolactams caged fluorescent dyes instead of reversible photoswitches in SMLM. Herein, by introducing hydrogen bonds to stabilize spirolactams, we report a series of acid-resistant rhodamine spirolactams with accelerated ring-closing reactions from fluorescent xanthyliums to non-fluorescent spirolactams, endowing them with good photoswitchable properties even in acidic environments. By further substitution of 6-phenylethynyl naphthalimide on the spirolactam, we shifted the photoactivation wavelength into the visible region (>400 nm). Subsequently, we have successfully applied these dyes in labeling and imaging the cell surface of Bacillus subtilis at pH 4.5 using SMLM.

Abstract:Herein, we figured out a novel ratiometric fluorescent probe based on Janus silica nanoparticles (SiO2 NPs) where fluorescein isothiocyanate (FITC) and rhodamine B (RB) were asymmetrically functionalized for intracellular pH detection. With this unique strategy, the probe exhibited dual emission bands at 530 and 580 nm upon excitation at a single wavelength of 488 nm. The pH titrations of the probe showed a remarkable increase in ratiometric intensities ratio (I-530nm/I-580nm) when the pH was switched from 3.0 to 8.0 with a acid dissociation constant (pKa) value of 6.64. The probe was successfully applied to imaging and detection of pH in living cells without noticeable cytotoxicity, indicating that it is a promising candidate for bioanalysis and bioimaging.
