Abstract:Nanopesticides are becoming more popular in modern agriculture to improve crop protection product efficacy. However, information on their implications for crop plants is needed. In this study, 3-week-old maize plants were exposed to different doses of Cu(OH)(2) nanopesticide (0, 10, and 100 mg) for 7 days via foliar application. Gene expression of 9 antioxidant-related enzymes (CAT1, POD1, GST1, SOD-1A, SOD-B, GPX, APX1, HSP1, PER1) was determined using a quantitative real-time polymerase chain reaction. Using gas chromatography time-of-flight mass spectrometry, levels of 12 low-molecular-weight antioxidant compounds were measured. Results showed that a dose of 100 mg of Cu(OH)(2) nanopesticide significantly decreased leaf chlorophyll content and biomass by 17-20%. In addition, potassium and phosphorus were up-regulated (14% and 13%, respectively) in response to this dose. Gene expression of POD1 and GST1 was significantly (p < 0.05) increased by 42.6% and 71.8%, respectively, at a dose of 10 mg, but declined at high dose (100 mg). Precursors of phenolic acids (phenylalanine and tyrosine), 4-hydroxycinnamic acid, and total phenolic content were significantly increased (24-122%) in response to 100 mg, indicating that phenolic acids may also play an important role in antioxidant defense. This study provides important information on maize plant responses to the Cu(OH)(2) nanopesticide at genetic, metabolic, and physiological levels and may be applied to other nanoparticle/plant interaction studies.

Abstract:Due to their unique properties, copper-based nanopesticides are emerging in the market. Thus, understanding their effect on crop plants is very important. Metabolomics can capture a snapshot of cellular metabolic responses to a stressor. We selected maize and cucumber as model plants for exposure to different doses of Cu(OH)2 nanopesticide. GC-TOF-MS-based metabolomics was employed to determine the metabolic responses of these two species. Results revealed significant differences in metabolite profile changes between maize and cucumber. Furthermore, the Cu(OH)2 nanopesticide induced metabolic reprogramming in both species, but in different manners. In maize, several intermediate metabolites of the glycolysis pathway and tricarboxylic acid cycle (TCA) were up-regulated, indicating the energy metabolism was activated. In addition, the levels of aromatic compounds (4-hydroxycinnamic acid and 1,2,4-benzenetriol) and their precursors (phenylalanine, tyrosine) were enhanced, indicating the activation of shikimate-phenylpropanoid biosynthesis in maize leaves, which is an antioxidant defense-related pathway. In cucumber, arginine and proline metabolic pathways were the most significantly altered pathway. Both species exhibited altered levels of fatty acids and polysaccharides, suggesting the cell membrane and cell wall composition may change in response to Cu(OH)2 nanopesticide. Thus, metabolomics helps to deeply understand the differential response of these plants to the same nanopesticide stressor.
Proteomic Analysis Unravels Response and Antioxidation Defense Mechanism of Rice Plants to Copper Oxide Nanoparticles: Comparison with Bulk Particles and Dissolved Cu Ions

Abstract:The increasing application of nanomaterials in agriculture raises environmental risks, but the information on how nanomaterials impact plant proteins is still limited. Here, we exposed the roots of rice seedlings to CuO nanoparticles (NPs, 100 mg/L), CuO bulk particles (BPs, 100 mg/L), and dissolved Cu ions (0.5 mg/L) for 14 days. The results show that CuO NPs caused more Cu accumulation, higher ascorbate peroxidase activity, and a higher ratio of GSH and GSSG but lower activity of Mn-SOD than BPs and Cu ions. Isotopic multilabel relative quantitative proteomics results indicate that NPs induced double differentially expressed proteins compared to BPs and Cu ions, especially phosphoglycerate kinase (fold change (FC) > 10) and glycosyltransferase (FC < 0.1). Unlike BPs and Cu ions, the proteins that responded to NPs were primarily connected with the C2 domain and glutathione S-transferase and significantly enriched for drug metabolism and monovalent inorganic cation transport processes and oxidative phosphorylation and phagosome pathways. Furthermore, almost 20% of proteins in the protein-protein interaction belonged to the ribosomal protein family. The parallel reaction monitoring further proved the defense response to NPs involving in redox, signal transduction, and heavy metal detoxification. Thus, proteomic analysis suggests that CuO NPs caused higher phytotoxicity mainly due to the nanospecific effect but elevated cysteine synthesis and GSSG reduction for detoxification.

The root exudate metabolic changes revealed an active defense mechanism against nano-Cu stress: up-regulation of amino acids to sequester/exclude Cu/nano-Cu; down-regulation of citric acid to reduce the mobilization of Cu ions; ascorbic acid up-regulation to combat reactive oxygen species; and up-regulation of phenolic compounds to improve antioxidant system. Thus, we demonstrate that nontargeted (1)H NMR and GC-MS based metabolomics can successfully identify physiological responses induced by nanoparticles. Root exudates metabolomics revealed important detoxification mechanisms.

Abstract:There has been an increasing influx of nanopesticides into agriculture in recent years. Understanding the interaction between nanopesticides and edible plants is crucial in evaluating the potential impact of nanotechnology on the environment and agriculture. Here we exposed lettuce plants to Cu(OH)2 nanopesticides (1050-2100 mg/L) through foliar spray for one month. Inductively coupled plasma-mass spectrometry (ICP-MS) results indicate that 97-99% (1353-2501 mg/kg) of copper was sequestered in the leaves and only a small percentage (1-3%) (17.5-56.9 mg/kg) was translocated to root tissues through phloem loading. Gas chromatography-time-of-flight mass spectrometry (GC-TOF-MS) based metabolomics combined with partial least squares-discriminant analysis (PLS-DA) multivariate analysis revealed that Cu(OH)2 nanopesticides altered metabolite levels of lettuce leaves. Tricarboxylic (TCA) cycle and a number of amino acid-related biological pathways were disturbed. Some antioxidant levels (cis-caffeic acid, chlorogenic acid, 3,4-dihydroxycinnamic acid, dehydroascorbic acid) were significantly decreased compared to the control, indicating that oxidative stress and a defense response occurred. Nicotianamine, a copper chelator, increased by 12-27 fold compared to the control, which may represent a detoxification mechanism. The up-regulation of polyamines (spermidine and putrescine) and potassium may mitigate oxidative stress and enhance tolerance. The data presented here provide a molecular-scale perspective on the response of plants to copper nanopesticides.
Metabolomics Reveals How Cucumber (cucumis Sativus) Reprograms Metabolites to Cope with Silver Ions and Silver Nanoparticle-Induced Oxidative Stress

Abstract:Due to their well-known antifungal activity, the intentional use of silver nanoparticles (AgNPs) as sustainable nanofungicides is expected to increase in agriculture. However, the impacts of AgNPs on plants must be critically evaluated to guarantee their safe use in food production. In this study, 4-week-old cucumber (Cucumis sativus) plants received a foliar application of AgNPs (4 or 40 mg/plant) or Ag+ (0.04 or 0.4 mg/plant) for 7 days. Gas chromatography-mass spectrometry (GCMS)=based nontarget metabolomics enabled the identification and quantification of 268 metabolites in cucumber leaves. Multivariate analysis revealed that all the treatments significantly altered the metabolite profile. Exposure to AgNPs resulted in metabolic reprogramming, including activation of antioxidant defense systems (upregulation of phenolic compounds) and downregulation of photosynthesis (upregulation of phytol). Additionally, AgNPs enhanced respiration (upregulation of tricarboxylic acid cycle intermediates), inhibited photorespiration (downregulation of glycine/serine ratio), altered membrane properties (upregulation of pentadecanoic and arachidonic acids, downregulation of linoleic and linolenic acids), and reduced inorganic nitrogen fixation (downregulation of glutamine and asparagine). Although Ag ions induced some of the same metabolic changes, alterations in the levels of carbazole, lactulose, raffinose, citraconic acid, lactamide, acetanilide, and pbenzoquinone were AgNP-specific. The results of this study offer new insight into the molecular mechanisms by which cucumber responds to AgNP exposure and provide important information to support the sustainable use of AgNPs in agriculture.

Abstract:During the senescence of cucumber (Cucumis sativus L.) cotyledons, declined activities of superoxide dismutase (SOD), catalase (CAT) and ascorbate-peroxidase (ASA-POD) were observed, but peroxidase (POD) activity was observed increasing. Triadimefon at the concentration of 20 mg/L promoted the activities of POD. On the other hand, the levers of malondialdehyde (MDA) were increased and triadimefon treatment decreased the content of MDA. These results suggested that triadimefon retarded the senescence of cucumber cotyledons by reducing the level of membrane lipid peroxidation.

Abstract:Targeted metabolomics aims to provide a new approach to investigate metabolites and gather both qualitative and quantitative information. We describe a protocol for extraction and analysis of plant metabolites, specifically 13 secondary metabolites (antioxidants) using liquid chromatography coupled to triple quadrupole mass spectrometry (LC-MS/MS), with high linearity (R2 > 0.99) and reproducibility (0.23-6.23 R%) with low limits of detection (>0.001 ng/mL) and quantification (>0.2 ng/mL). The protocol was applied to study the antioxidant response of cucumber plants exposed to nanocopper pesticide. Dose-dependent changes in antioxidant concentrations were found, and 10 antioxidants were significantly consumed to scavenge reactive oxygen species, protecting plants from damage. Levels of three antioxidants were up-regulated, as a response to the depletion of the other antioxidants, signaling activation of the defense system. We demonstrated that the reported LC-MS/MS method provides a quantitative analysis of antioxidants in plant tissues, for example to investigate interactions between plants and nanomaterials.

Abstract:Programmed cell death (PCD) in root border cells (RBCs) induced by Copper (Cu) has been little studied. This study explored whether Cu induced PCD in RBCs of cucumber or not and investigated the possible mechanisms. The results showed that the percentage of apoptotic and necrotic RBCs increased with increasing concentration of Cu treatment. A quick burst of ROS in RBCs was detected, while mitochondrial membrane potential (ΔΨm) decreased sharply with Cu treatment. Caspase-3 like protease activity showed a tendency of increase with Cu treatment. The potential of Cu to induce PCD in RBCs of cucumber was first proved. Our results showed that ROS generation and mitochondrial membrane potential loss played important roles in Cu-induced caspase-3-like activation and PCD in RBCs of cucumber, which provided new insight into the signaling cascades that modulate Cu phytotoxicity mechanism.

Abstract:With the increased use of metal nanoparticles (NPs), their access to the food chain has become a main concern to scientists and holds controversial social implications. This research particularly sheds light on copper nanoparticles (CuNP), as they have been commonly used in several industries nowadays. In this study, we investigated the phytotoxicity of CuNP on cucumber (Cucumis sativus) plants grown hydroponically. Atomic Absorption Spectroscopy (AAS), X-Ray Fluorescence (XRF), and Scanning Electron Microscopy (SEM) analysis confirmed that C. sativus treated with CuNP accumulated CuNP in the plant tissues, with higher levels in roots, with amounts that were concentration dependent. Furthermore, genotoxicity was assessed using Random amplified polymorphic DNA (RAPD) technique, and our results showed that CuNP caused genomic alterations in C. sativus. Phenotypical, physiological, and biochemical changes were assessed by determining the CuNP treated plant's total biomass, chlorophyll, H2O2 and MDA contents, and electrolyte leakage percentage. The results revealed notable adverse phenotypical changes along with decreased biomass and decreased levels of the photosynthetic pigments (Chlorophyll a and b) in a concentration-dependent manner. Moreover, CuNP induced damage to the root plasma membrane as determined by the increased electrolyte leakage. A significant increase in H2O2 and MDA contents were detected in C. sativus CuNP treated plants. Additionally, copper-zinc superoxide dismutase (Cu-Zn SOD) gene expression was induced under CuNP treatment. Overall, our results demonstrated that CuNP of 10-30 nm size were toxic to C. sativus plants. This finding will encourage the safe production and disposal NPs. Thus, reducing nano-metallic bioaccumulation into our food chain through crop plants; that possesses a threat to the ecological system.

Abstract:The effects of root exudates and aqueous root extracts of cucumber (Cucumis sativus) and allelochemicals on root antioxidant enzymes and leaf photosynthesis, transpiration and stomatal conductance were investigated in cucumber. Cucumber seedlings were incubated in solutions containing root exudates at 50 or 100 mg/l, root extracts at 1:100, 1:50, 1:25 and 1:10 (root dry weight:distilled water), and derivatives of benzoic and cinnamic acids at 0.25 mM, respectively. Root peroxidase and superoxide dismutase activities increased significantly after exposure to allelopathic agents. Membrane peroxidation was also enhanced by root exudates, root extracts and some of the tested acids. Allelopathic agents reduced leaf stomatal conductance from 1.28 mol m−2 s−1 to 0.02–0.33 mol m−2 s−1. Leaf transpiration was reduced by 67% by root exudates at 100 mg/l, by 85–95% by root extracts, and by 51–87% by tested acids, respectively. Net assimilate rate was also inhibited by 29% by root exudates at 100 mg/l, by 49–83% by root extracts, and by 7–43% by tested acids, respectively. The intercellular CO2 concentrations for the treated seedlings were only 47–89% of the control. However, leaf temperature was increased by 2–4°C by the tested agents.
Application of Metabolomics to Assess the Impact of Cu(OH)2 Nanopesticide on the Nutritional Value of Lettuce (lactuca Sativa): Enhanced Cu Intake and Reduced Antioxidants

Abstract:Copper-based nanopesticides are increasingly being used in the agriculture, especially in organic farming. This has triggered some concerns about their risk to environmental and human health. In this study, 24-day-old lettuce plants grown in soil were exposed via the leaves to different concentrations (0, 1050 and 1555mg/L) of Cu(OH)2 nanopesticides for one month. Results showed Cu was mainly localized in lettuce leaves (823–1111 and 1353–2008mg/kg in vascular and photosynthetic tissues), which may potentially increase Cu intake and impact human health. In addition, foliar application of Cu(OH)2 nanopesticide significantly increased potassium concentration in lettuce leaves by 6–7% and 21–28%, in vascular and photosynthetic tissues. A Gas Chromatography-Time-of-Flight Mass Spectrometry (GC–TOF-MS) based metabolomics approach was applied to determine hundreds of organic compounds simultaneously. Using relative quantitation and Partial Least Squares-Discriminant Analysis (PLS-DA) clustering of all compounds, clear differences were observed in the metabolite profiles of control and Cu(OH)2 nanopesticides treated leaves. Discriminating compounds include amino acids, organic acids, polyamines, vitamin C and polyphenols. Dehydroascorbic acid and cis-caffeic acid, which are important antioxidants, were significantly decreased (19–33% and 5–8%) due to foliar exposure to the nanopesticide. Total antioxidant capacity was significantly decreased 20–23% after exposure to Cu(OH)2 nanopesticides. There was also up- and down-regulation of a number of amino acids, particularly 4-hydroxybutytic acid (GABA) which decreased 50% compared to the control, potentially affecting the overall nutritional value of lettuce leaves exposed to the Cu(OH)2 nanopesticides. In future work, determining an appropriate level of nanopesticide will be important to obtain the antifungal benefits without resulting in a significant decrease in nutritional value.

A gas chromatography-time-of-flight-mass spectrometry metabolomics approach was applied to assess metabolic alterations induced by Cu(OH)2 nanopesticide in spinach leaves. Exposure to Cu(OH)2 nanopesticide and copper ions induced alterations in the metabolite profiles of spinach leaves. Compared to the control, exposure to 18 mg of Cu(OH)2 nanopesticide induced significant reduction (29-85%) in antioxidant or defense-associated metabolites including ascorbic acid, α-tocopherol, threonic acid, β-sitosterol, 4-hydroxybutyric acid, ferulic acid, and total phenolics. The metabolic pathway for ascorbate and aldarate was disturbed in all exposed spinach plants (nanopesticide and Cu2+). Cu2+ is responsible for the reduction in antioxidants and perturbation of the ascorbate and aldarate metabolism. However, nitrogen metabolism perturbation was nanopesticide-specific. Spinach biomass and photosynthetic pigments were not altered, indicating that metabolomics can be a rapid and sensitive tool for the detection og earlier nanopesticide effects. Consumption of antioxidants during the antioxidant defense process resulted in reduction of the nutritional value of exposed spinach.

Abstract:Numerous studies on short term effects of copper-based nanomaterials on plants have been published, however investigations with plants grown in a complex soil medium are lacking. In this study Grey Zucchini (Cucurbita pepo) was grown in an environmental growth chamber using a 1:1 (v/v) potting mix native soil mixture amended with Kocide 3000, nCuO, bCuO, or Cu NPs. After 3 weeks Cu concentrations in the root, stem, and leaves of treated plants were significantly higher than control plants. This increase in Cu concentration did not adversely affect plant growth or chlorophyll production. The activity ascorbate peroxidase (APX) in the roots tissues of plants treated with Kocide 3000, nCuO, and bCuO decreased by at least 45%. Catalase (CAT) activity in root tissues of plants treated with 50 mg/kg of Cu NP decreased by 77%, while those treated at 200 mg/kg were reduced by 80%, compared to controls. The activity of APX and CAT in the leaves of all treated plants remained similar to control plants. Based on the endpoints used in this study, with the exception of a decrease in the accumulation of Zn and B in the roots, the exposure of zucchini to the tested copper compounds resulted in no negative effects.

Abstract:Copper oxide nanoparticles (CuO NPs) have received considerable attention for their toxic effects on crops and potential application in agriculture. In order to investigate the biological effects of CuO NPs on plants, we exposed cucumber (Cucumis sativus) to two sizes of CuO NPs (510 nm, μCuO and 43 nm, nCuO). Results indicated that with concentration increased, the available Cu content in soil increased significantly. The addition of CuO NPs increased Cu content and other nutrient element (e.g., K, P, Mn, and Zn) content in plants. However, diverse particle sizes had different effects. The nCuO treatment had larger translocation factor, higher nutrient element content in fruits, and lower oxidative damage than μCuO treatment. Moreover, nCuO of 100 mg/kg could stimulate cucumber growth, while μCuO had no obvious effects on growth. Conclusively, CuO NPs could be used as copper fertilizer to supply copper to cucumber. The nCuO had better effects on improving the bioavailability of Cu and nutritional value of fruits. These results can help develop strategies for safe disposal of CuO NPs as agricultural fertilizer.
Effect of short-term water deficit stress on antioxidative systems in cucumber seedling roots

Abstract:Abstract Background Cucumber is one of the most popular vegetables, and have little tolerance for water stress. The antioxidant defense system is one of major drought defense and adaptive mechanisms in plants, however, relatively few data are available regarding antioxidant systems in responses of cucumber to water deficit. The effect of short-term drought stress on the antioxidant system, lipid peroxidation and water content in cucumber seedlings roots was investigated. Results The results showed that polyethylene glycol (PEG) induced water stress markedly decreased water content of cucumber seedling roots after treatment of 36 h, and caused excessive generation of reactive oxygen species (ROS) including superoxide (O 2 .− ), hydrogen peroxide (H 2 O 2 ). Meanwhile, malondialdehyde (MDA) content increased. Antioxidant enzymes including superoxide dismutases (SOD), peroxidases (POD), catalase (CAT) and ascorbate peroxidase (APX) activities increased in different time and different extent under water stress, while ascorbate (AsA) and glutathione (GSH) content, glutathione reductase (GR), dehydroascorbate reductase (DHAR) and monodehydroascorbate reductase (MDHAR) activities all decreased when compared to control. Conclusions Therefore, it can be concluded that water stress strongly disrupted the normal metabolism of roots and restrained water absorption, and seemingly enzymatic system played more important roles in protecting cucumber seedling roots against oxidative damage than non-enzymatic system in short-term water deficit stress.

Abstract:Abiotic and biotic stress induce the production of reactive oxygen species (ROS), which limit crop production. Little is known about ROS reduction through the application of exogenous scavengers. In this study, C60 fullerol, a free radical scavenger, was foliar applied to three-week-old cucumber plants (1 or 2 mg/plant) before exposure to copper ions (5 mg/plant). Results showed that C60 fullerols augmented Cu toxicity by increasing the influx of Cu ions into cells (170% and 511%, respectively, for 1 and 2 mg of C60 fullerols/plant). We further use metabolomics and proteomics to investigate the mechanism of plant response to C60 fullerols. Metabolomics revealed that C60 fullerols up-regulated antioxidant metabolites including 3-hydroxyflavone, 1,2,4-benzenetriol, and methyl trans-cinnamate, among others, while it down-regulated cell membrane metabolites (linolenic and palmitoleic acid). Proteomics analysis revealed that C60 fullerols up-regulated chloroplast proteins involved in water photolysis (PSII protein), light-harvesting (CAB), ATP production (ATP synthase), pigment fixation (Mg-PPIX), and electron transport (Cyt b6f). Chlorophyll fluorescence measurement showed that C60 fullerols significantly accelerated the electron transport rate in leaves (13.3% and 9.4%, respectively, for 1 and 2 mg C60 fullerols/plant). The global view of the metabolic pathway network suggests that C60 fullerols accelerated electron transport rate, which induced ROS overproduction in chloroplast thylakoids. Plant activated antioxidant and defense pathways to protect the cell from ROS damaging. The revealed benefit (enhance electron transport) and risk (alter membrane composition) suggest a cautious use of C60 fullerols for agricultural application.
