Abstract:Continuous hyperglycemia is considered to be the most significant pathogenesis of diabetic cardiomyopathy, which manifests as cardiac hypertrophy and subsequent heart failure. O-GlcNAcylation has attracted attention as a post-translational protein modification in the past decade. The role of O-GlcNAcylation in high glucose-induced cardiomyocyte hypertrophy remains unclear. We studied the effect of O-GlcNAcylation on neonatal rat cardiomyocytes that were exposed to high glucose and myocardium in diabetic rats induced by streptozocin. High glucose (30 mM) incubation induced a greater than twofold increase in cell size and increased hypertrophy marker gene expression accompanied by elevated O-GlcNAcylation protein levels. High glucose increased ERK1/2 but not p38 MAPK or JNK activity, and cyclin D2 expression was also increased. PUGNAc, an inhibitor of β-N-acetylglucosaminidase, enhanced O-GlcNAcylation and imitated the effects of high glucose. OGT siRNA and ERK1/2 inhibition with PD98059 treatment blunted the hypertrophic response and cyclin D2 upregulation. OGT inhibition also prevented ERK1/2 activation. We also observed concentric hypertrophy and similar changes of O-GlcNAcylation level, ERK1/2 activation and cyclin D2 expression in myocardium of diabetic rats induced by streptozocin. In conclusion, O-GlcNAcylation plays a role in high glucose-induced cardiac hypertrophy via ERK1/2 and cyclin D2.
High Levels of Glucose Induce Apoptosis in Cardiomyocyte Via Epigenetic Regulation of the Insulin-Like Growth Factor Receptor

Abstract:Diabetic hyperglycemia result in cardiovascular complications, but the mechanisms by which high levels of glucose (HG) cause diabetic cardiomyopathy are not known. We investigate whether HG-induced repression of insulin-like growth factor 1 receptor (IGF-1R) mediated by epigenetic modifications is one potential mechanism. We found that HG resulted in decreased IGF-1 receptor (IGF-1R) mRNA levels, and IGF-1R protein when compared with H9C2 rat cardiomyocyte cells incubated in normal glucose. HG also induced apoptosis of H9C2 cells. The effects of HG on reduced expression of IGF-1R and increased apoptosis were blocked by silencing p53 with small interference RNA but not by non-targeting scrambled siRNA. Moreover, HG negatively regulated IGF-1R promoter activity as determined by ChIP analysis, which was dependent on p53 since siRNA-p53 attenuated the effects of HG on IGF-1R promoter activity. HG also increased the association of p53 with histone deacetylase 1 (HDAC1), and decreased the association of acetylated histone-4 with the IGF-1R promoter. Furthermore, HDAC inhibitor relieved the repression of IGF-1R following HG state. These results suggest that HG-induced repression of IGF-1R is mediated by the association of p53 with the IGF-1R promoter, and by the subsequent enhanced recruitment of chromatin-modifying proteins, such as HDAC1, to the IGF-1R promoter–p53 complex. In conclusion, our data demonstrate that HG decreases expression of IGF-1R and decreases the association of acetylated histone-4 with the IGF-1R promoter. These studies may help delineate the complex pathways regulating diabetic cardiomyopathy, and have implications for the development of novel therapeutic strategies to prevent diabetic cardiomyopathy by epigenetic regulation of IGF-1R.

Abstract:Background: Cardiac progenitor cells (CPC) can repair the myocardium injured by infarction. However, CPC may not function well under the diabetic condition. Exposure to high levels of glucose (HG) may induce cardiomyopathy and fibrosis in patients with diabetes. Signaling transduction via transforming growth factor-β (TGF-β) and its responding transcription factor, Snail, (TGF-β/Snail) is involved in fibrosis. Insulin-like growth factor (IGF-1) decreases TGF-β expression. Thus, it is important to investigate whether IGF-1 activation of the PI3 kinase/glycogen synthase kinase 3β (GSK-3β) signaling pathway prevents HG-induced transition of CPC to fibroblasts. The addition of Ro-31-8220 and BAY11-7082 could partly reverse these changes induced by high glucose level. Abstract:Vascular disease is the leading cause of morbidity and mortality in patients with diabetes. Persistent hyperglycemia – the dominant metabolic derangement of diabetes, can cause endothelial cell apoptosis. Diabetes is often associated with low insulin like growth factor-1 (IGF-1), and the latter state has been linked to adverse risk profile and increased cardiovascular disease incidence. Since IGF-1 acts as an important survival factor for multiple cell types, this study was to investigate whether IGF-1 exert regulatory effects on high glucose-induced apoptosis of vascular endothelial cells. Exposure to high glucose dose- and time-dependently induced apoptotic changes (e.g., DNA fragmentation, altered mitochondrial membrane potential, and cytochrome-c release) in human umbilical vein endothelial cells (HUVECs). Addition of IGF-1 blocked the high glucose effect in a manner dependent on expression of IGF-1 receptor (IGF-1R) since silencing IGF-1R with small interference RNA could diminish the IGF-1′ anti-apoptosis effect. Our findings show that enhanced IGF-1 signaling inhibits glucose-induced apoptosis in HUVECs by reducing mitochondrial dysfunction, and maintaining the mitochondrial retention of cytochrome-c. These results may have therapeutic implications in preventing/reducing diabetes associated endothelial dysfunction.
Hyperglycemia Altered the Fate of Cardiac Stem Cells to Adipogenesis Through Inhibiting the Β-Catenin/tcf-4 Pathway

Abstract:Background/Aims: Hyperglycemia is an important risk factor for the most severe cardiovascular diseases in patients with diabetes. It has been demonstrated that cardiac stem cells (CSCs) play a pivotal role in the maintenance of cardiac homeostasis and regeneration. However, the mechanism underlying the influence of diabetes on CSCs remains unclear. This study demonstrated that hyperglycemia might promote adipogenesis in CSCs, which induces a decline in myocardial regeneration capability in diabetes. Methods: CSCs were isolated and cultured in high-glucose medium. The levels of β-catenin and TCF-4 in CSCs were determined by immunofluorescence staining and western blot analysis. Adipogenic transcriptional factors and CSCs markers were also examined by flow cytometry and western blot analysis after adipogenesis induction. In addition, Oil Red O staining was performed to investigate lipid droplet formation during adipogenesis induction with or without LiCl, a potent activator of TCF/β-catenin-dependent transcription. Results: High-glucose conditions inhibited nuclear translocation of β-catenin/TCF-4 and promoted adipogenesis in CSCs. After adipogenesis induction, expression of adipogenic transcriptional factors (PPARγ, ADD1, and C/EBPα) were increased (P < 0.01) and that of CSCs markers (c-Kit, Sca-1, MDR-1, and isl-1) were decreased (P< 0.01) in CSCs in the high-glucose group. Furthermore, lipid droplet formation was increased in CSCs cultured with high glucose, while LiCl attenuated lipid droplet formation in these CSCs (P < 0.01). Conclusion: These results demonstrated that hyperglycemia inhibited the β-catenin/TCF-4 pathway and promoted CSCs adipogenesis. Our findings suggest a new opportunity for future interventional strategie for abnormal myocardial regeneration and epicardial fat in patients with diabetes.

Abstract:Glucose toxicity is an important initiator of cardiovascular disease, contributing to the development of cardiomyocyte death and diabetic complications. The present study investigated whether high glucose state could induce apoptosis of rat cardiomyocyte cell line H9C2 through microRNA regulated insulin-like growth factor (IGF-1) signaling pathway. Our data showed that H9C2 cells exposed to high glucose have increased miR-1 expression level, decreased mitochondrial membrane potential, increased cytochrome-c release, and increased apoptosis. Glucose induced mitochondrial dysfunction, cytochrome-c release and apoptosis was blocked by IGF-1. Using prediction algorithms, we identified 3′-untranslated regions of IGF-1 gene are the target of miR-1. miR-1 mimics, but not mutant miR-1, blocked the capacity of IGF-1 to prevent glucose-induced mitochondrial dysfunction, cytochrome-c release and apoptosis. In conclusion, our data demonstrate that IGF-1 inhibits glucose-induced mitochondrial dysfunction, cytochrome-c release and apoptosis and IGF-1’s effect is regulated by miR-1.

Abstract:High glucose-induced endothelial cell dysfunction is considered to be the main cause of the development of vascular complications in patients with diabetes. In this study, we examined the effect of high glucose (HG) on apoptosis of human umbilical vein endothelial cells (HUVEC) and investigated the molecular mechanisms involved. HUVEC was exposed to HG and apoptosis was assessed by Death ELISA. Our data showed that HG induced significant apoptosis in HUVEC cells after exposing to HG for 5 days (O.D. value = 0.36±0.07 in HG group vs. 0.13±0.05 in control, p<0.05). HG induced apoptosis in HUVEC is associated with decreased of insulin-like growth factor receptor (IGF-1R) mRNA (4.1±0.6 fold at 72 h, p<0.05) as assessed by real-time PCR and IGF-1R protein (3. 1±0.5 fold at 96 h, p<0.05) as assessed by western blot, which were dose- and time-dependent effects (25 mM of glucose rather than 5 mM at 72 h and 96 h). The effects of HG on reduced expression of IGF-1R and apoptosis were blocked by silencing histone deacetylase 1 (HDAC1) with small interference RNA (siRNA-HDAC1) compared to non-targeting scrambled siRNA. Moreover, HG negatively regulated IGF-1R promoter activity as determined by ChIP (chromatin immunoprecipitation) analysis, which was dependent on HDAC1 since siRNA-HDAC1 attenuated the effects of HG on IGF-1R promoter activity. HG also increased the association of IGF-1R promoter with HDAC1, and decreased the association of IGF-1R promoter with acetylated histone-4. Furthermore, trichostatin A, a specific HDAC inhibitor, relieved the repression of IGF-1R and prevented apoptosis of HUVEC following HG state. These results suggest that HG-induced repression of IGF-1R is mediated by the association of HDAC1 with the IGF-1R promoter, and by the subsequent reduced recruitment of chromatin-modifying proteins, such as histone 4 to the IGF-1R promoter. In conclusion, our data demonstrate that high glucose exposure decreases expression of IGF-1R and decreases the association of acetylated histone-4 with the IGF-1R promoter, and suggest that enhancing acetylation of histone 4 and IGF-1R expression may be a useful strategy to prevent diabetic-induced apoptosis of endothelial cells. SGLT1 Inhibition Attenuates Apoptosis in Diabetic Cardiomyopathy Via the JNK and P38 Pathway

Abstract:Background: Recent studies have revealed that a novel selective sodium-glucose cotransporter 1 (SGLT1) inhibiton has shown beneficial effects in cardiovascular diseases. However, the question of whether SGLT1 inhibition influences diabetic cardiomyopathy (DCM) remains unanswered. In this study, we investigated the influence and underlying mechanism of SGLTI inhibition on DCM. Methods: SGLT1 levels were measured in diabetic patients with similar conditions who visited our hospital from January to December 2019. Wistar male rats (n = 50) were divided into five groups: control, diabetes induced by streptozotocin infusion, and diabetes treated with 0.5, 1.0, or 1.5 mg/kg mizagliflozin via stomach gavage for 12 weeks. H9C2 cardiomyocytes were treated with mizagliflozin and then exposed to a high glucose concentration (30 mmol/L). TUNEL assays were performed, and bcl2, bax, p-p38, p-Erk, p-JNK and caspase-3 levels were measured. We used siRNA and an SGLT1 overexpression plasmid to detect the effects of SGLT1. Results: SGLT1 levels were significantly elevated in DCM patients, and receiver operating characteristic (ROC) curve analysis identified SGLT1 as influencing DCM. The area under the curve (AUC) was 0.705 (p < 0.05), with 65.8% sensitivity, and 62.2% specificity. SGLT1 inhibition appeared to attenuate apoptosis in DCM via the JNK and p38 pathway. Conclusion: SGLT1 can be used as a marker for the diagnosis of DCM, and SGLT1 inhibition can attenuate apoptosis, thereby suppressing DCM development via the JNK and p38 pathway.
Liraglutide Prevents High Glucose Induced HUVECs Dysfunction Via Inhibition of PINK1/Parkin-dependent Mitophagy

Abstract:Functional loss of endothelial cells will lead to development and progression of atherosclerosis in diabetic patients. However, dysfunction of endothelial cells in diabetes has yet to be fully understood. We aimed to characterize the potential effects of liraglutide, a glucagon-like peptide-1 (GLP-1) receptor agonist, on preventing high glucose-induced endothelial dysfunction and excessive mitophagic response. Pretreatment with liraglutide prevented downregulation of eNOS phosphorylation and NO secretion, and reduced apoptosis and oxidative stress of the human umbilical vein endothelial cells (HUVECs) exposed to high glucose. We further demonstrated that liraglutide likely mediated such protective effects by reducing PINK1/Parkin mediated mitophagy. Liraglutide markedly decreased high glucose-induced mitochondrial ROS, lessened PINK1 expression and mitochondrial accumulation of Parkin, but recovered SIRT1 expression. Seahorse analysis revealed that liraglutide mitigated high glucose-induced reduction of basal and maximum respiration rates as well as spare respiration capacity. Inhibition of Parkin by RNA silencing not only resulted in increased mitochondrial and cytosolic ROS and reduced mitochondrial mass and mitochondrial membrane potential, but also led to increased apoptotic responses in high glucose treated HUVECs which were not preventable by liraglutide. Together, our study reveals that liraglutide acts upstream of the PINK1/Parkin pathway to effectively counteract high glucose induced cell dysfunction by suppression of the PINK1/Parkin-dependent mitophagy. Therefore, its use as an adjunct therapy for type 2 diabetes mellitus is warranted to reduce the risk of atherosclerosis. Further research is required to examine the exact molecules, including SIRT1, upstream of the PINK1/parkin pathway that liraglutide targets to maintain the mitochondrial homeostasis.
Inhibition of PI-3 Kinase/akt/mtor, but Not Calcineurin Signaling, Reverses Insulin-Like Growth Factor I-induced Protection Against Glucose Toxicity in Cardiomyocyte Contractile Function

Abstract:Insulin-like growth factor-I (IGF-1) ameliorates cardiac dysfunction in diabetes although the mechanism of action remains poorly understood. This study examined the role of PI-3 kinase/Akt/mammalian target of rapamycin (mTOR) and calcineurin pathways in cardiac effects of IGF-1 against glucose toxicity. Adult rat ventricular myocytes were cultured for 8 h with either normal (NG, 5.5 mM) or high (HG, 25.5 mM) glucose, in the presence or absence of IGF-1 (10-500 nM), the PI-3 kinase/Akt inhibitor LY294002 (10 microM), the mTOR inhibitor rapamycin (20 microM) or the calcineurin inhibitors cyclosporin A (5 microM) or FK506 (10 mg/l). Mechanical properties were evaluated using an IonOptix MyoCam system. HG depressed peak shortening (PS), reduced maximal velocity of shortening/relengthening (+/- dl/dt) and prolongs time-to-90% relengthening (TR90), which were abolished by IGF-1 (100 and 500 nM). Interestingly, the IGF-1-elicited protective effect against HG was nullified by either LY294002 or rapamycin, but not by cyclosporine A or FK506. None of the inhibitors affected cell mechanics. Western blot analysis indicated that HG and IGF-1 stimulated phosphorylation of Akt and mTOR. HG also activated p70s6k and suppressed GSK-3beta phosphorylation. However, the HG-induced alterations in phosphorylation of Akt, mTOR, p70s6k and GSK-3beta were significantly reversed by IGF-1. Protein expression of Akt, mTOR, p70s6k, GSK-3beta, SERCA2a and phospholamban was unaffected by HG, IGF-1 or rapamycin. Rapamycin significantly enhanced Akt phosphorylation whereas it inhibited mTOR phosphorylation. Collectively, our data suggest that IGF-1 may provide cardiac protection against glucose in part through a PI-3 kinase/Akt/mTOR/ p70s6k-dependent and calcineurin-independent pathway.

Abstract:Objective: Cardiac resistance to IGF-I occurs in diabetes and is attributed to cardiac dysfunction in diabetes. However, the mechanism of action responsible for cardiac lGF-I resistance is Still unknown. This study was designed to examine the impact of high glucose on lGF-1-induced contractile response and activation of serine-threonine kinase Akt as well as extracellular signal-regulated kinase (ERK1/2) in cardiac myocytes. Methods: Isolated adult rat vermicular myocytes were Cultured for 12-18 h in a serum-free medium containing either normal (NG. 5.5 mM) or high (HG, 25.5 HIM) glucose. Mechanical properties were evaluated using an lonOptix MyoCam(R) system. Myocytes were electrically stimulated at 0.5 Hz and contractile properties analyzed included peak shortening (PS), time-to-PS (TPS) and time-to-90% relengthening (TR90). Intracellular Ca2+-induced Ca2+ release was measured as fura-2 fluorescence intensity change (DeltaFFI). Protein levels of total and phosphorylated Akt and ERK1/2, indicators of Akt and ERK1/2 activation, IGF- I receptors (pro-IGF-IR and IGF-IRalpha) as well as the glucose transporter GLUT4 were assessed by Western blot. Results: IGF-I (10(-10)-10(-6) M) elicited a close-dependent increase in PS and DeltaFF1 in myocytes maintained in NG medium. However, IGF-I induced a negative response on PS and DeltaFFI in HG myocytes. The lGF-1-induced responses in NG or HG myocytes were blunted by the IGF-I receptor antagonist H-1356. Western blot analysis revealed that IGF-1Ralpha but not pro-IGF-IR was reduced in HG myocytes. While IGF-I 10(-6) M) upregulated total Akt protein levels in both NG and HG myocytes, it only induced a significant activation of Akt in NG but not HG myocytes. IGF-I elicited comparable ERK1/2 activation in both NG and HG inyocytes. Conclusion: These results suggest that the cardiac IGF-I resistance in diabetes is likely attributed, at least in part. to reduced IGF-IR and attenuated IGF-I-induced Akt phosphorylation under elevated extracellular glucose. Abstract:The brain-type glycogen phosphorylase (pygb) is one of the rate-limiting enzyme in glycogenolysis that plays a crucial role in the pathogenesis of type 2 diabetes mellitus. Here we investigated the role of pygb in high glucose (HG)-induced cardiomyocyte apoptosis and explored the underlying mechanisms, by using the specific pygb inhibitors or pygb siRNA. Results showed that inhibition of pygb significantly attenuated cell apoptosis and oxidative stress induced by HG in H9c2 cardiomyocytes. Inhibition of pygb improved glucose metabolism in cardiacmyocytes, as evidenced by increased glycogen content, glucose consumption and glucose transport. Mechanismly, pygb inhibition activated Akt/GSK-3β signal pathway and suppressed NF-κB activation in H9c2 cells exposed to HG. Additionally, pygb inhibition promoted the expression and the translocation of hypoxia-inducible factor-1α (HIF-1α) after HG stimulation. However, the changes in glucose metabolism and HIF-1α activation mediated by pygb inhibition were significantly reversed in the presence of Akt inhibitor MK2206. In conclusion, the present study suggested that inhibition of pygb prevents HG-induced cardiomyocyte apoptosis via Akt-HIF-α activation.

Abstract:Aims: Insulin-like growth factor-1 (IGF-1) is a polypeptide protein hormone, similar in molecular structure to insulin, which plays an important role in cell migration, cell cycle progression, cell survival and proliferation. In this study, we investigated the possible mechanisms of IGF-1 mediated cell cycle redistribution and apoptosis of vascular endothelial cells.Method: Human umbilical vein endothelial cells (HUVECs) were pretreated with 0.1, 0.5, or 2.5 mu g/mL of IGF-1 for 30 min before the addition of Ang II. Cell cycle redistribution and apoptosis were examined by flow cytometry. Expression of Ang II type 1 (AT(1)) mRNA and cyclin E protein were determined by RT-PCR and Western blot, respectively.Results: Ang II (1 mu mol/L) induced HUVECs arrested at G(0)/G(1), enhanced the expression level of AT(1) mRNA in a time-dependent manner, reduced the enzymatic activity of nitric oxide synthase (NOS) and nitric oxide (NO) content as well as the expression level of cyclin E protein. However, IGF-1 enhanced NOS activity, NO content, and the expression level of cyclin E protein, and reduced the expression level of AT(1) mRNA. L-NAME significantly counteracted these effects of IGF-1.Conclusions: Our data suggests that IGF-1 can reverse vascular endothelial cells arrested at G(0)/G(1) and apoptosis induced by Ang II, which might be mediated via a NOS-NO signaling pathway and is likely associated with the expression levels of AT(1) mRNA and cyclin E proteins. Irisin ameliorates high glucose‐induced cardiomyocytes injury via AMPK/mTOR signal pathway

Abstract:<p>High glucose (HG)‐induced cardiomyocytes (CMs) injury is a leading cause of diabetic cardiomyopathy with little treatment options. Irisin, a new myokine, which is cleaved from its precursor fibronectin type III domain‐containing protein 5 (FNDC5), has aroused great attentions as an essential cardioprotective factor and glucose metabolism regulator but little was known on diabetic cardiomyopathy yet. Here, we aim to clarify the role of irisin in the HG‐induced CMs injury. Neonatal Sprague‐Dawley (SD) rat CMs were cultured in a normal or high glucose medium for 12, 24 and 48 h respectively before exposing to irisin. The apoptosis level was determined by Terminal‐deoxynucleotidyl transferase mediated‐dUTP Nick End Labeling (TUNEL) assay. Cell viability was measured with the conventional methylthiazolyl tetrazolium (MTT) assay. Moreover, reactive oxygen species (ROS) production was evaluated by dihydroethidium (DHE) staining. Inflammatory factors, namely TNF‐ɑ, IL‐6, IL‐1β were determined by enzyme‐linked immunosorbent assay (ELISA) kits. Furthermore, protein and mRNA expressions were measured by Western blot and qRT‐PCR respectively. HG increases the apoptosis of CMs and activated the inflammatory responses and oxidative stress in CMs. Meanwhile, the mRNA and protein expressions of FNDC5 are decreased after HG exposure. Nevertheless, the increased apoptosis is alleviated by irisin treatment. Notably, irisn suppresses the inflammatory responses and oxidative stress in injured CMs. Mechanically, after administration of Compound C, AMPK inhibitor, these cardioprotective effects resulting from irisin are reversed. Irisin plays a significant role in anti‐apoptosis, anti‐inflammation, anti‐oxidative stress in HG‐induced CMs via AMPK/mTOR signaling pathway.</p><p>This article is protected by copyright. High glucose induces apoptosis in AC16 human cardiomyocytes via macrophage migration inhibitory factor and c-Jun N-terminal kinase

Abstract:1. It is known that high glucose can induce cardiomyocyte apoptosis and that macrophage migration inhibitory factor (MIF) may be involved in the development of diabetes. However, the relationship between high glucose and MIF in diabetic cardiomyopathy remains unclear. 2. In the present study, AC16 human cardiomyocytes were cultured in the presence of 25 mmol/L glucose for 20, 30 and 60 min before being subjected to western blot analyses to determine MIF expression and c-Jun N-terminal kinase (JNK) activation. In addition, AC16 cells were pretreated with 2.5 μmol/L SP600125 (a JNK inhibitor), 40 μmol/L (s,r)-3-(4-hydroxyphenyl)-4,5-dihydro-5-isoxazole acetic acid methyl ester (ISO-1; an MIF antagonist) or 0.1% dimethylsulphoxide (DMSO; vehicle) for 1 h prior to exposure to 25 mmol/L glucose and culture for 72 h, followed by annexin V-fluorescein isothiocyanate/propidium iodide staining and flow cytometry analysis. Caspase 3 activity and phosphorylation of JNK were also analysed by western blotting. 3. The high concentration of glucose increased expression of endogenous MIF and JNK phosphorylation in AC16 cardiomyocytes. Pretreatment of cells with SP600125 and ISO-1 reduced glucose-induced apoptosis and caspase 3 activity. Furthermore, JNK phosphorylation was attenuated by inhibition of endogenous MIF. 4. In conclusion, myocardial cell apoptosis induced by high glucose involves the overexpression of MIF and activation of the JNK signalling pathway. The identification of a high glucose-MIF-JNK pathway will help determine potential new targets in the treatment of diabetic cardiomyopathy.

Abstract:The induction of hypoxia tolerance has emerged as a novel therapeutic strategy for the treatment of ischemic diseases. The disruption of hypoxic signaling by hyperglycemia has been shown to contribute to diabetic cardiomyopathy. In this study, we explored the potential molecular mechanisms by which high glucose (HG) impairs hypoxia-inducible factor-α (HIF-α) signaling in cardiomyocytes. The exposure of H9c2 cell lines to HG resulted in time- and concentration-dependent decreases in HIF-1α and HIF-2α expression together with an increase in prolyl hydroxylase-1,2 (PHD1 and PHD2) expression, the main regulators of HIF-α destabilization in the heart. The exposure of H9c2 cells to normal glucose (5.5 mM) and high glucose (15, 30, and 45 mM) led to dose-dependent increases in p53 and TIGAR and a decrease in SIRT3 expression. The pretreatment of H9c2 with p53 siRNA to knockdown p53 attenuated PHD1 and PHD2 expression, thus significantly enhancing HIF-1α and HIF-2α expression in H9c2 cells under HG conditions. Interestingly, pretreatment with p53 siRNA altered H9c2 cell metabolism by reducing oxygen consumption rate and increasing glycolysis. Similarly, pretreatment with TIGAR siRNA blunted HG-induced PHD1 and PHD2 expression. This was accompanied by an increase in HIF-1α and HIF-2α expression with a reduction in oxygen consumption rate in H9c2 cells. Furthermore, pretreatment with adenovirus-SIRT3 (Ad-SIRT3) significantly reduced the HG-induced expression of p53 and PHDs and increased HIF-1α levels in H9c2 cells. Ad-SIRT3 treatment also regulated PHDs-HIF-1α levels in the hearts of diabetic db/db mice. Our study revealed a novel role of the HG-induced disruption of PHDs-HIF-α signaling via upregulating p53 and TIGAR expression. Therefore, the p53/TIGAR signaling pathway may be a novel target for diabetic cardiomyopathy.

Abstract:Persistent hyperglycemia represents a major metabolic complication of diabetes, which may cause endothelial cell dysfunction and apoptosis. microRNAs are novel biomarkers, modulators and therapeutic targets for disease. The microRNA miR-221 is expressed in human umbilical vein endothelial cells (HUVEC). Insulin-like growth factor 1 (IGF-1) is an important survival factor for multiple cell types. This study was to investigate whether high glucose (HG) induces endothelial dysfunction and apoptosis through altering the microRNA expression and IGF-1 signaling. The mirVana qRT-PCR miRNA detection assay showed that exposure to glucose for 24 h dose-dependently (up to 25 mM) increased miR-221 expression by 2.6 ± 0.5 folds (p<0.05). HG treatment decreased c-kit protein expression (58 ± 7 % decrease vs. control at 48 h) assessed by flow cytometry. The treated HUVEC had a reduced capacity for wound healing responded to stem cell factor (SCF, the ligand of c-kit). SCF-induced HUVEC migration potential was also reduced significantly by exposure to high dose of glucose. As estimated by prediction algorithms, c-kit, but not IGF-1, was identified as the target of miR-221. Transfection of HUVEC with 2′-O-methyl antisense inhibitory oligonucleotide (AMO-221) downregulation of miR-221 led to attenuation of the glucose’s effect on the c-kit expression, SCF-induced migration and wound healing in HUVEC. Prolonged glucose exposure (25 mM, 96 h) induced apoptosis of HUVEC assessed by Death ELISA (O.D. values = 0.49 ± 0.06 in glucose group vs. 0.12 ± 0.07 in control, p<0.01). Addition of IGF-1 (30 ng/ml) blocked the glucose-induced apoptosis, whereas AMO-221 did not attenuate the apoptosis. In addition, the anti-apoptotic effects of IGF-1 was attenuated by PI3 kinase inhibitor LY294002 (15 uM) (IGF-1 + glucose + PI3 kinase inhibitor vs. IGF-1 + glucose = 0.45 ± 0.07 vs. 0.15 ± 0.05 O.D. values). Our data demonstrate that downregulation of miR-221 attenuates HG-induced reduction of c-kit, wound healing and migration of HUVEC, and enhanced IGF-1 signaling prevents HG-induced apoptosis. Thus, reducing miR-221 expression and activating IGF-1 signaling pathway may be a useful strategy to prevent diabetic-induced endothelial dysfunction and apoptosis.
