Cloning of Mice by Nuclear Transfer

Abstract:ABREAKTHROUGH was made by Wilmut and his colleagues in 1996. A sheep named Dolly was successfully cloned from an adult somatic cell (Wilmut et al., 1997). In this experiment, the donor cell and enucleated oocyte were fused together as the means of nuclear transfer. After that, in 1998, Wakayama et al. (1998) used a modified method described to produce Dolly and cloned mice with cumulus cell nuclei. A Piezo-drill micromanipulator was employed for the direct injection of cumulus cell nuclei into metaphase II (MII) oocytes. Reconstructed embryos were cultured in vitro to morulae/blastocyst stage before transfer back into surrogate mothers. Compared to electrofusion method used for large animal cloning, Piezo-drill microinjection omitted the fusion procedure and less donor cell cytoplasm was introduced into the reconstructed oocyte. Cloning by nuclear transfer offers exciting new opportunities for examining the nuclear-cytoplasmic interactions that occur in early embryos and other basic questions related to gene function and differentiation. In comparison to large animals, mice have short gestation and generation periods and more is known about mouse genetic background. So it is possible to use the mouse as a model to understand basic mechanisms of reprogramming following nuclear transfer and it is also applicable for producing transgenic or knockout mice. Up to date, cloned mice have been produced from both somatic cells and embryonic stem (ES) cells, using donor cells with either hybrid or inbred backgrounds (Wakayama et al., 1998, 1999; Wakayama and Yanagimachi, 1999, 2001a; Ogura et al., 2000; Eggan et al., 2001; Rideout et al., 2000; Gao et al., 2003a). Because ES cells could be differentiated into several types of cells in vitro, mice have been studied as a model to investigate whether therapeutic cloning may be applicable clinically to treat human diseases (Rideout et al., 2002). ES cells were successfully derived from somatic cell cloned blastocysts and then differentiated in vitro to produce neural or bone cells. These differentiated cells were transplanted back to mutant mice to test whether these transplanted cells could survive and help the host organ to be rescued. For clinical application, differentiated cells derived from human embryonic stem cells (isolated from cloned blastocysts in which the donor somatic cell is from the patient) could be transplanted to the patient where they would integrate into the target tissue and restore organ function, prevent or slowdown further deterioration. Since the initial report of cloning mice by Wakayama et al., many laboratories in the world tried to repeat his achievement, but so far only a few laboratories have succeeded. Micromanipulation technique is one of the failed reasons, but some other undefined factors in preparation of solutions and cells seem also to be important. In our studies, we have successfully produced cloned mice from both hybrid and inbred ES cells and cumulus cells by using the “Honolulu method” (Gao et al., 2003a), and more recently using a modified form of this method (Chung et al., 2002; Gao and Latham, unpublished data). The methods of preparing solutions and donor
