Abstract:Single scattering particles, especially noble metal (plasmonic) nanoparticles, based analytical techniques are attractive recently and becoming the research focus of the light scattering analytical techniques. In this mini review, we summarize the single scattering particles based analytical techniques in the past decade including single scattering particles counting, single plasmonic nanoparticles sensing, and single plasmonic nanoparticles tracking/imaging. We emphasize the discussion on the single plasmonic nanoparticles sensing that combines with dark-field microscopy and resonant Rayleigh scattering spectroscopy.

Abstract:Metallic nanoparticles are used in every stage of science and are still charming the researchers to explore the localized surface plasmon resonance (LSPR) properties for the promising application of sensors and catalysts. In this review, recent progresses on silver-based and gold-based single nanoparticle for resonance light scattering imaging analysis and applications are presented. Firstly, fundamental principles and technique of dark-field microscope (DFM) coupled the LSPR spectroscopy are described. Then, the species of silver-based and gold-based single nanoparticle are outlined, with an emphasis on the resonance light scattering imaging analysis of silver-based and gold-based single nanoparticle. Finally, recent achievements of the plasmonic single-particle imaging analysis technique in analytical determinations and reaction monitoring are also comprehensively introduced. We hope to shed some light on the forthcoming study by showing these advances and some future challenges of resonance light scattering imaging analysis.
Single Nanoparticle-Based Dark-Field Microscopic Imaging in Analytical Chemistry

Abstract:T light scattering of single nanoparticle, such as gold and silver nanoparticles, is stable and efficient, hence the applications of single nanoparticle-based dark-field microscopic imaging have attracted extensive attention. Both the changes of single nanoparticle scattering imaging color and scattering intensity can be used as effective detection signal in analytical chemistry. In the author’s group, to study the localized surface plasmon resonance (LSPR) light scattering of single nanoparticle, first the shape effect of single silver nanoparticle on the scattering light color and refractive index sensitivities was investigated. It was found that particles with a large radial ratio or a tip structure always had a higher sensitivity which could be due to the LSPR maxima at long wavelengths and the strong electric field intensity distribution. Then single nanoparticle-based RGB analytical method was established by coding the colors of the scattering light of individual nanoparticles with the RGB system, and the imaging date was manipulated with the IPP software. In addition, the scattering light intensity of single gold nanoparticle was digitized and expressed as digital information through the aforementioned software. Based on the RGB system and digitized method, it was established new scattering analytical method which had been certified in our other energy transfer experiment. Furthermore, the single nanoparticle-based dark-field microscopic imaging was also used to monitoring some chemical reactions. Real-time monitoring of the etching process of gold nanoparticles by iodine, in situ growth of single Ag@Hg nanoalloys and photochemical reaction were successfully achieved by dark-field microscopic imaging. In addition to the chemical reactions, the single nanoparticle-based dark-field microscopic imaging has a potential to real-time monitoring the intracellular biochemical reactions and infection process of bacteria and virus which may provide a new method for the diagnosis of the disease.

Abstract:Plasmonic nanoparticles have been widely applied in cell imaging, disease diagnosis, and photothermal therapy owing to their unique scattering and absorption spectra based on localized surface plasmon resonance (LSPR) property. Recently, it is still a big challenge to study the detailed scattering properties of single plasmonic nanoparticles in living cells and tissues, which have dynamic and complicated environment. The conventional approach for measuring the scattering light is based on a spectrograph coupled to dark-field microscopy (DFM), which is time-consuming and limited by the small sample capacity. Alternatively, RGB-based method is promising in high-throughput analysis of single plasmonic nanoparticles in dark-field images, but the limitation in recognition of nanoparticles hinders its application for intracellular analysis. In this paper, we developed an automatic and robust method for recognizing the plasmonic nanoparticles in dark-field image for RGB-based analysis. The method involves a bias-modified fuzzy C-means algorithm, through which biased illumination in the image could be eliminated. Thus, nearly all of the gold nanoparticles in the recorded image were recognized both on glass slide and in living cells. As confirmed, the distribution of peak wavelength obtained by our method is well agreed to the result measured by conventional method. Furthermore, we demonstrated that our method is profound in cell imaging studies, where its advantages in fast and high-throughput analysis of the plasmonic nanoparticles could be applied to confirm the presence and location of important biological molecules and provide efficiency information for cancer drug selection.

Abstract:Dark-field microscopy is a widely used tool for measuring the optical resonance of plasmonic nanostructures. However, current numerical methods for simulating the dark-field scattering spectra were carried out with plane wave illumination either at normal incidence or at an oblique angle from one direction. In actual experiments, light is focused onto the sample through an annular ring within a range of glancing angles. In this paper, we present a theoretical model capable of accurately simulating the dark-field light source with an annular ring. Simulations correctly reproduce a counterintuitive blue shift in the scattering spectra from gold nanodisks with a diameter beyond 140 nm. We believe that our proposed simulation method can be potentially applied as a general tool capable of simulating the dark-field scattering spectra of plasmonic nanostructures as well as other dielectric nanostructures with sizes beyond the quasi-static limit.

Abstract:Dark-field microscopy (DFM) and spectroscopy base on localized surface plasmon resonance (LSPR) have been widely applied in biological sensing and single-molecule imaging. Using plasmonic nanoparticles with controlled geometrical, optical, and surface chemical properties as the probes, the scattering light depending on the surrounding environment can be detected by DF microscope. Signal-to-noise radio and time resolution of the conventional DFM is not sufficient to identify single molecular dynamics. To break these limitations, significant improvements have been made in recent years. This critical review is focused on the developments of the DFM and the utilization of DFM as a powerful technology in the application of LSPR detection.

Abstract:Plasma nanoparticles offer promise in data storage, biosensing, optical imaging, photoelectric integration, etc. This review highlights the local surface plasmon resonance (LSPR) excitation mechanism of plasmonic nanoprobes and its critical significance in the control of dark-field sensing, as well as three main sensing strategies based on plasmonic nanomaterial dielectric environment modification, electromagnetic coupling, and charge transfer. This review then describes the component materials of plasmonic nanoprobes based on gold, silver, and other noble metals, as well as their applications. According to this summary, researchers raised the LSPR performance of composite plasmonic nanomaterials by combining noble metals with other metals or oxides and using them in process analysis and quantitative detection.

Abstract:Single plasmonic nanoparticles (PNPs) analysis with dark-field microscopic imaging (iDFM) has attracted much attention in recent years. The ability for quantitative analysis of iDFM is critical, but cumbersome, for characterizing and analyzing the scattered light of single PNPs. Here, a simple automatic HSI colour coding method is established for coding dark-field microscopic (DFM) images of single PNPs with localized surface plasmon resonance (LSPR) scattered light, showing that hue value in the HSI system can realize accurate quantitative analysis of iDFM and providing a novel approach for quantitative chemical and biochemical imaging at the single nanoparticle level.

Abstract:The mechanism for the photoluminescence (PL) emission from gold nanoparticles has attracted considerable attention for many years. However, there is an important gap between small nanoclusters (similar to 2 nm) and larger plasmonic particles (similar to 50 nm). In this work, using defocused imaging technique, we investigate the PL properties of gold nanospheres (15-20 nm in diameter) on a single particle level. Photoblinking and photobleaching phenomena are both observed. We notice that although these nanospheres can support surface plasmon resonance at similar to 515 nm, they emit at similar to 630 nm (excited by 532 nm), which is obviously plasmon-independent. The observed defocused images (DIs) exhibit isotropy first and then either transform into anisotropy or vanish rapidly. Surprisingly, the Dls can change their emission pattern within one single nanosphere during the tracing time. All these PL properties suggest a multiple-dipole emission model. High-resolution transmission electron microscopy images demonstrate that these nanospheres are polycrystalline containing multiple small crystal domains (similar to 3 nm). We believe that these small domains divide the particle into different clusters and give rise to the photoblinking behavior and rotary DIs. The presented PL mechanism links gold nanoclusters and single-crystal nanorods, which could be helpful in understanding the origins of photoblinking and the luminescent properties from metallic nanoparticles. In addition, these water-soluble gold nanospheres provide new opportunities for biological labels and light-emitting sources in nanophotonics.

Abstract:A dark and brightfield imaging technique explored plasmonic phenomena arising from bumps on a gold nanofilm coated on a silica substrate. The study employs darkfield polarization indirect microscopic imaging to investigate the scattering photon state and reveal the spatial distribution characteristics with distinct multipolar features, contrasting with those observed in the brightfield imaging configuration. Computational simulations utilizing the Finite-Difference Time-Domain method were conducted to understand this behavior further and consider the experimental setup's impact. The observations of varying multipolar scattering features with changes in the incident angle of the dark-field illumination suggest that the excitation of plasmonic effects differs for light beams incident at different angles.

A simple method to disentangle nanoparticle optical properties by darkfield microspectroscopy

Abstract:We present a darkfield optical microspectroscopy technique devoted to the disentangled measurement of the absorption and scattering cross sections of nanoparticle (NP) samples with variable concentration. The robustness of the method, including the needed instrumental calibrations, is examined in detail by analyzing and quantifying the major sources of statistic and systematic errors. As an exemplary case, results are presented on a gold NP colloid. The technique takes advantage of a simple inverted microscope, coupled with a spectrograph and equipped with a darkfield condenser and a variable numerical aperture objective to obtain spectra either in darkfield or brightfield optical configurations. By adopting the Lambert-Beer (LB) equation modeling, we were able to disentangle and measure with a single setup the absorption, scattering, and extinction coefficients of the same sample by combining three spectra, obtained by opportunely varying the objective numerical aperture. Typical plasmonic resonances were recognized at approximately 520 and 750 nm. Optical coefficients were measured as a function of particle number density (0.04-3.94 µm(-3), corresponding to 40 µM-4 mM nominal Au concentration) and good linearity was verified up to ∼1.5 µm(-3) (∼1 mM Au). Moreover, extinction and scattering cross sections were quantified and the validity of the LB approximation was reviewed. Besides its applications to plasmonic NPs, this method may be appropriate for any colloid, provided there exists a characteristic spectral feature in the ultraviolet-visible-near infrared range. This technique may be exploited to localize NPs in biological samples.
Correlation and Characterization of Three-Dimensional Morphologically Dependent Localized Surface Plasmon Resonance Spectra of Single Silver Nanoparticles Using Dark-Field Optical Microscopy and Spectroscopy and Atomic Force Microscopy

Abstract:We have developed a new and effective methodology to correlate optical and AFM images of single Ag nanoparticles (NPs), allowing us to study 3D-morphological dependent localized surface plasmon resonance (LSPR) spectra of individual Ag NPs. We fabricated arrays of distinctive microwindows on glass coverslips using photo-lithography method, and created well-isolated individual Ag NPs with a wide variety of shapes and morphologies on the glass coverslips using a modified nanosphere lithography method (NSL). Using distinctive geometries of microwindows, we located individual Ag NPs of interest in their optical and AFM images, enabling us to correlate and characterize the LSPR spectra and 3D morphologies of the same single NPs using dark-field optical microscopy and spectroscopy (DFOMS) and AFM, respectively. We found that LSPR spectra of single Ag NPs, with nearly equal volume [(8.6 ± 0.4) × 10(3) nm(3)], cross-section [(2.2 ± 0.2) × 10(2) nm(3)], and height (39.6 ± 3.6 nm), highly depend on their shapes, showing the red shift of peak wavelength to 629 nm (quasi trapezoidal cylindrical NP) from that of 506 nm (quasi circular cylindrical NP). LSPR spectra of single Ag NPs simulated using discrete dipole approximation (DDA) agree well with those measured experimentally when their shapes and morphologies can be accuractely described in both methods, but differ when they are not. Furthermore, we found location-dependent LSPR spectra on and around a single NP, offering a unique opportunity to characterize multi-mode plasmonic NPs at nanometer resolution for better understanding their plasmonic optical properties and for rational design of single NP optics.

Abstract:We proposed the estimation of the plasmonic near-field volume in the vicinity of a single gold nanoparticle, and observed experimentally the near-field variation due to a change in the polarization of the illuminating light. Under total-internal-reflection illumination, the plasmonic near-field volume is varied by tuning the polarization of the excitation light. The variation in the optical near-field around a single gold nanoparticle was simulated theoretically with a finite-difference time domain method, and was characterized experimentally employing a fluorescence correlation spectroscopy technique. The experimental results are in agreement quantitatively with the theoretical analysis. These results are highly relevant to important efforts to clarify the interaction between the emitter and the plasmonic antenna, and should be helpful in developing a plasmonic-enhanced total-internal-reflection fluorescence imaging microscope.

Abstract:Owing to the unique physical, chemical, optical, and biological properties, plasmonic nanoparticles (PNPs) have been widely used in various research fields such as materials science, biology, and medicine. The optical properties of PNPs can be regulated by changing their composition, shape, and size, so that a suitable light-scattering probe can be screened by means of controllable synthesis. Real-time study of the dynamic behavior of PNPs at the single molecule level is of great significance for understanding the biological behaviors of living cells and tissues, fabricating functional nanomaterials, and developing new chemical biosensors. Starting from the traditional dark-field microscopy (DFM), we have developed a series of plasmonic light-scattering imaging techniques with high sensitivity, high temporal-spatial resolution, and high throughput through optimizing the assembly of light sources, detectors, and other optical components, and applied these techniques to single molecule detection, multi-particle sensing, single cell imaging, biological process tracing, and so on. Based on the PNPs with optical anisotropy, we have also developed a three-dimensional scanning imaging system for living cells and asupercontinuum laser light-sheet imaging system coupled with high-speed capillary electrophoresis system, advancing the study of single molecule spectroscopy. This paper will summarize the work on PNP single particle analysis and imaging in our research group during the past ten years, and put forward some new ideas for further development in this field.
Sub-diffraction-limit localization imaging of a plasmonic nanoparticle pair with wavelength-resolved dark-field microscopy.

Abstract:In this work, with wavelength-resolved dark-field microscopy, the center-of-mass localization information from nanoparticle pairs (i.e., spherical (45 nm in diameter) and rod (45 × 70 nm) shaped gold nanoparticle pairs with different gap distances and orientations) was explored and compared with the results determined by scanning electron microscopy (SEM) measurements. When the gap distance was less than 20 nm, the scattering spectrum of the nanoparticle pair was seriously modulated by the plasmonic coupling effect. The measured coordinate information determined by the optical method (Gaussian fitting) was not consistent with the true results determined by SEM measurement. A good correlation between the optical and SEM measurements was achieved when the gap distance was further increased (e.g., 20, 40 and 60 nm). Under these conditions, well-defined scattering peaks assigned to the corresponding individual nanoparticles could be distinguished from the obtained scattering spectrum. These results would afford valuable information for the studies on single plasmonic nanoparticle imaging applications with the optical microscopy method such as super-localization imaging, high precision single particle tracking in a crowding environment and so on.

Abstract:Light scattered from nanoparticles (NPs) can easily be observed by the naked eye. Light-scattering signals from nanomaterials have been applied extensively for analysis (e.g., in biochemical and pharmaceutical sciences). Metal NPs with defined components, size and shape can be used as fluorescent analogs for labeling biomolecules in biochemical analysis and biological imaging, due to the colors of the scattered light, if excited with a white-light source. Enhanced light-scattering signals from aggregates of NPs induced by analytes can be used for monitoring molecular interaction or quantitative detection of analytes. Due to the advantages of simplicity and sensitivity, light-scattering spectrometric methods based on nanomaterials have shown great promise. In this contribution, we review applications of light-scattering signals from NPs in biochemical assay, pharmaceutical analysis and biological imaging.

Abstract:In this paper, light scattering by a chain of nanoparticles of different shapes including spheres and ellipsoids lis investigated. Characterized by surface plasmon resonances of metallic nanoparticles, these structures could behave like nanoantenna and they have potential practical applications in the microscopy, spectroscopy, and optoelectronic devices. As a result, they have drawn increasing interests in theoretical studies and experimental characterisations of the near-field properties of nanoparticles in the optical range. Based on the discussions on dielectric constant and the simulations of fields around spherical and ellipsoidal scatterers using plane wave excitation, various characteristics of the magnitudes of the scatter filed, the light intensities, power densities and energy densities are studied in the near-field region. Figures are proposed as an intuitive interpretation for better understanding in a microscopic view. The results are expected to be useful for studies on the local field enhancement properties of nanoantennas working in the optical region as well as the fundamentals of electromagnetic inverse problem.

Abstract:The development of optical imaging techniques has led to significant advancements in single-nanoparticle tracking and analysis, but these techniques are incapable of label-free selective nanoparticle recognition. A label-free plasmonic imaging technology that is able to identify different kinds of nanoparticles in water is now presented. It quantifies the plasmonic interferometric scattering patterns of nanoparticles and establishes relationships among the refractive index, particle size, and pattern both numerically and experimentally. Using this approach, metallic and metallic oxide particles with different radii were distinguished without any calibration. The ability to optically identify and size different kinds of nanoparticles can provide a promising platform for investigating nanoparticles in complex environments to facilitate nanoscience studies, such as single-nanoparticle catalysis and nanoparticle-based drug delivery.
