Abstract:Herein, a general strategy for chemo- and regioselective 1,2-reduction of chromium-bound arenes was developed, thus providing rapid access to 1,3-cyclohexadienes. Selective arene activation via π-complexation along with the use of mild hydride Ph3SiH can overcome the inherently low reactivity of arene π-bonds while tolerating various reduction-sensitive functional groups. Its versatility further enables a regiodivergent deuteration. Using different sequences of (non)deuterated hydride and acid reagents, the deuterated positions as well as the degrees of deuterium incorporation can be controlled precisely, which leads to a large and previously inaccessible chemical space for 1,3-cyclohexadiene isotopologues. A reasonable mechanism was proposed based on intermediate capture and control experiments. The synthetic value of this selective 1,2-reduction was demonstrated in the formal total synthesis of (±)-galanthamine and (±)-lycoramine.

Abstract:Iodine-mediated intermolecular C-N and C-I bond formation of alkenes is described. A series of alkenes could be selectively converted into the corresponding aminoiodination products, which are versatile building blocks in organic synthesis and medicinal chemistry. Wide substrate scope, broad functional group tolerance and easy scale-up operation make this protocol very practical and attractive in synthetic chemistry.

Abstract:An efficient palladium-catalyzed [2 + 2 + 1] annulation of 3-iodochromones, bridged olefins, and iodomethane is described, affording a range of chromone-containing polycyclic compounds. Additionally, the corresponding deuterated products were smoothly obtained with iodomethane- d 3 instead of iodomethane. Moreover, the synthetic utility of this method is further substantiated by gram scale preparation and application to late-stage modification of estrone.

Abstract:A highly regioselective iodosulfonylation of allenes in the presence of CuI and 1,10-phenanthroline has been developed for the synthesis of various useful ( E)-α-iodomethyl vinylsulfones in moderate to excellent yields. This practical reaction is fast, operationally simple, and in particular, proceeds under very mild conditions to afford the target products with high regio- and stereoselectivity. The selectivity was illustrated by a conceptual DFT analysis.

Abstract:Isodesmic reactions represent mild alternatives to other chemical transformations that require harsh oxidizing agents or highly reactive intermediates. However, enantioselective isodesmic C-H functionalization is unknown and enantioselective direct iodination of inert C-H bond is very rare. Rapid synthesis of chiral aromatic iodides is of significant importance for synthetic chemistry. Herein, we report an unprecedented highly enantioselective isodesmic C-H functionalization to access chiral iodinated phenylacetic Weinreb amides via desymmetrization and kinetic resolution with PdII catalysis. Importantly, further transformations of the enantioenriched products are readily available at the iodinated or the Weinreb amide position, paving the way of related studies for synthetic and medicinal chemists.

Abstract:A hypervalent iodine-reagent-based C-H functionalization strategy was utilized to synthesize diaryl ethers. This method directly transforms various arenes into their corresponding diaryliodonium salts, followed by a C-O coupling reaction to produce structurally diverse diaryl ethers. The efficacy of this approach in the late-stage structural modifications of complex molecules was demonstrated.

Abstract:Organomagnesium compounds (Grignard reagents) are among the most useful organometallic reagents and have greatly accelerated the advancement of synthetic chemistry and related sciences. Nevertheless, heavy Grignard reagents based on the metals calcium, strontium or barium are not widely used, mainly due to their rather inert heavy alkaline-earth metals and extremely high reactivity of their corresponding Grignard-type reagents. Here we report the generation and reaction chemistry of butadienyl heavy Grignard reagents whose extremely high reactivity is successfully tamed. Facile synthesis of perfluoro-π-extended pentalene and naphthalene derivatives is realized by the in situ generated heavy Grignard reagents via intramolecular C–F/C–H bond cleavage. These obtained perfluorodibenzopentalene and perfluorodinaphthopentalene derivatives show low-lying LUMO levels, with one being the lowest value so far among all pentalene derivatives. Our results set an exciting example for the meaningful synthetic application of heavy Grignard reagents. Grignard reagents are one of the mainstays of organic synthesis, but analogues with other Group 2 metals (heavy Grignards) are less easy to control. Here, the authors generate butadienyl calcium and barium heavy Grignards and control their reactivity in C–F and C–H cleavage reactions.

Abstract:Hypervalent iodine diazo reagents with electron‐withdrawing groups (EWG) connected to diazo methyl center were a type of donor‐acceptor diazo compounds with potential reaction abilities similar to ordinary diazo compounds. Although several hypervalent iodine diazo reagents were synthesized and used in the nucleophilic substitution reactions as early as 1994, the synthesis and application of new iodine(III) diazo reagents have only been reported to a certain extent in recent years. In the presence of rhodium catalyst, photocatalyst, or nucleophiles, hypervalent iodine diazo reagents can be converted into rhodium‐carbenes, diazomethyl radicals, ester radicals or nucleophilic intermediates, which can be used as key intermediates for the formation of chemical bonds. The aim of this review is to give an overview of hypervalent iodine diazo compounds in organic synthesis.

Abstract:Practical approaches for chemoselective mono-bromination, di-bromination, and tetra-bromination of terminal alkynes to generate 1-bromoalkynes, 1,2-dibromoalkenes, α,α-dibromoketones, and 1,1,2,2-tetrabromoalkanes based on efficient oxidative brominations mediated by a hypervalent iodine reagent have been developed. Chemoselective bromination can be realized under mild conditions by altering the bromine source. The tetrabutylammonium bromide (TBAB)/(diacetoxyiodo)benzene (PIDA) system is specific for mono-bromination to provide 1-bromoalkynes, while the NaBr/PIDA system is selective toward di-bromination to achieve 1,2-dibromoalkenes. When a certain amount of water was added to the NaBr/PIDA system, a different di-bromination product, α,α-dibromo ketones, was generated. Tetra-bromination of terminal alkynes provides an efficient protocol for the synthesis of 1,1,2,2-tetrabromoalkanes in a system with an excess loading of NaBr/PIDA in one pot. This bromination affords good yields (up to 99%) with excellent chemoselectivity (up to 100%). These methods can be applied to the efficient chemoselective synthesis of bromide derivatives, intermediates, and related biologically active compounds.

Abstract:A substrate-induced strategy for the bromochlorination and bromoazidation of diverse 1,3-dienes has been reported. This protocol offers straightforward and highly selective access to vicinal or allyl chlorobromides and azidobromides, which are ubiquitous moieties in many natural products and valuable synthetic intermediates. Additionally, a series of transformations of the products were performed to demonstrate their synthetic potential. Furthermore, control experiments were performed to elucidate the mutual conversion pathway between vicinal and allyl chlorobromides. Bromochlorination and bromoazidation of diverse 1,3-dienes to prepare various chlorobromides and azidobromides with high efficiency and selectivity using a general protocol with mild conditions and simple operation.

Abstract:The systematic synthesis of a series of N-acyl chloroformamidines is achieved by reaction of carbodiimides with acid chlorides; the structure of an N-acyl chloroformamidine was confirmed by X-ray diffraction. The reactivity of N-acyl chloroformamidines towards diethyl malonate, phenylethynyllithium, pentane-2,4-dione, methyl acetoacetate, water, 1,3-dimethylurea, 1,3-dimethylthiourea, and sodium hydrosulfide, resulted in unique protocols for the synthesis of N-acyl-substituted 1,1-diaminoethylenes, amidines, ureas, and thioureas, respectively. These results show that N-acyl chloroformamidines have a good ability to accept nucleophilic attack for the construction of useful N-containing compounds.

Abstract:Lithium chloride was found to accelerate formation of the Grignard reagent from inactive 3-bromo-2-methylthiophene (1) and commercial magnesium metal. Based on this finding, a convenient and potentially scalable preparation of ethyl 2-methylthiophene-3-carboxylate (3) was achieved. In addition, this process has been found to provide a new, general approach to 3-substituted 2-methylthiophenes.

Abstract:Iodine(III) reagents have gained importance in organic synthesis due to their strong oxidative properties in addition to several advantages over other related such as, non-toxic, green, easy to handle and in general safe for using. These characteristics become them useful tools and represent relevant alternatives to procedures which involves harsh and/or hazardous reaction conditions. They are versatile oxidants that has been largely used in the functionalization of a broad variety of aromatic nucleus, resulting in the introduction of several groups, including in the most general classification, organic and inorganics. In this sense, the metal-free arylation mediated by iodine(III) reagents is the most reported issue in the literature. Nevertheless, other relevant inorganic functionalities were also introduced. Herein, we review the most important procedures described to date for the aromatic functionalization with inorganic groups such as halogens, as well as the nitro-, azido, cyano-, sulfur- and selenium groups, under the iodine(III) chemistry.

Abstract:Alkenyl magnesium compounds have received much attention in synthetic organic chemistry because of their high reactivity. This review summarizes three types of alkenyl magnesium compounds which contain at least one [Mg-C=C] fragment, for example, (1) alkenyl Grignard reagents prepared by halogen-magnesium exchange reaction; (2) alkenyl magnesium carbenoids prepared by halogen-magnesium exchange reaction or sulfoxide-magnesium exchange reaction; (3) magnesacarbocycles containing at least one alkenyl magnesium bond prepared from Zr- or Ti-catalyzed cyclomagnesation or transmetalation, as well as their further reactions and applications.
