Abstract:This feature article focuses on the recent development of giant molecules, which has emerged at the interface among chemistry, physics, and bio-science. Their molecular designs are inspired by natural polymers like proteins and are modularly constructed from molecular nanoparticle building blocks via sequential “click” chemistry. Most important molecular parameters such as topology, composition, and molecular weight can be precisely controlled. Their hierarchical assembly reveals many features reminiscent of both small molecules and proteins yet unusual for conventional synthetic polymers. These features are summarized and compared along with synthetic polymers and proteins. Specifically, examples are given in each category of giant molecules to illustrate the characteristics of their hierarchical assembly across different length, time and energy scales. The idea of “artificial domain” is presented in analogy to the structural domains in proteins. By doing so, we aim to develop a rational and modular approach toward functional materials. The factors that dominate the materials functions are discussed with respect to the precision and dynamics of the assembly. The complexity of structure-function relationship is acknowledged, which suggests that there is still a long way to go toward the convergence of synthetic polymers and biopolymers.
Molecular Nanoparticles Are Unique Elements for Macromolecular Science: from “nanoatoms” to Giant Molecules

Abstract:In this Perspective, we present a unique approach to the design and synthesis of giant molecules based on "nanoatoms" for engineering structures across multiple length scales and controlling their macroscopic properties. Herein, "nanoatoms" refer to shape-persistent molecular nanoparticles (MNPs) with precisely defined chemical structures and surface functionalities that can serve as elemental building blocks for the precision synthesis of giant molecules by methods such as sequential "click" approach. Typical "nanoatoms" include those MNPs based on fullerenes, polyhedral oligomeric silsesquioxanes, polyoxometalates, and folded globular proteins. The resulting giant molecules are precisely defined macromolecules. They include, but are not limited to, giant surfactants, giant shape amphiphiles, and giant polyhedra. Giant surfactants are polymer tail-tethered "nanoatoms" where the two components have drastic chemical differences to impart amphiphilicity. Giant shape amphiphiles not only are built up by covalently bonded MNPs of distinct shapes where the self-assembly is driven by chemical interactions but also are largely influenced by the packing constraints of each individual shape. Giant polyhedra are either made of a large MNP or by deliberately placing "nanoatoms" at the vertices of a polyhedron. In general, giant molecules capture the essential structural features of their small-molecule counterparts in many ways but possess much larger sizes. They are recognized in certain cases as size-amplified versions of those counterparts, and often, they bridge the gap between small molecules and traditional macromolecules. Highly diverse, thermodynamically stable and metastable hierarchal structures are commonly observed in the bulk, thin film, and solution states of these giant molecules. Controlled structural variations by precision synthesis further reveal a remarkable sensitivity of their self-assembled structures to the primary chemical structures. Unconventional nanostructures can be obtained in confined environments or through directed self-assembly. All the results demonstrate that MNPs are unique elements for macromolecular science, providing a versatile platform for engineering nanostructures that are not only scientifically intriguing but also technologically relevant.

Abstract:With recent development in controlled/“living” polymerization and sequence-specific polymeric architecture design, polymeric materials with accurately designed primary structure can be to some extent designed and prepared. Such development has actuated the design of single chain polymer nanoparticles (SCPNs) with tunable high-order structures from well-defined polymeric precursors. Until now, various synthetic strategies for SCPNs have been proposed and some possible applications, especially protein mimicry, have demonstrated the SCPNs' attracting prospect. After briefly introducing the origin of SCPNs, this feature article reviewed the recent development of SCPNs in terms of different crosslinking strategies as well as their potential applications. Meanwhile, we highlighted some important characterization techniques for SCPNs study.

Abstract:The interactions between the nanoscale particles, such as the carbon nanotubes, and biomolecules are essential to the nanoscale particle based biotechnology and biomedical applications, such as gene delivery, cellular imaging, tumor therapy. However, how the structure changes and whether the functions of the biomolecules are affected due to the existence of nanoscale particles are still poorly understood. In this paper, we review some of our recent progresses, which are based on the large scale molecular dynamics simulations, towards this direction. In our studies, by using an all-α domain named HP35 and an all-β domain named YJQ8WW as the examples, we show that the single-walled carbon nanotube (SWCNT), a typical form of hydrophobic nanoscale particle, can considerably change both the secondary and the tertiary structures of the proteins and form the protein-SWCNT complexes, in which the unfolded part of the proteins wrap around the SWCNT. The hydrophobic interaction and π-π stacking interaction between the nanoscale particles and the hydrophobic residues are found to play important roles in our observation. Abstract:This paper illustrates the merits of convergence in nanobiology of two seemingly disparate fields, material science and computational biology. Traditionally, material science has been a discipline involving design and fabrication of synthetic polymers consisting of repeating units. Collaboration with synthetic organic chemists allowed design of new polymers, with a range of altered conformations. Yet, naturally occurring proteins are also materials. Their varied sequences and structures should enrich material science providing more complex shapes, scaffolds and chemical properties. For material scientists, the enhanced coverage of chemical space obtained by integrating proteins and synthetic organic chemistry through the introduction of non-natural residues allows a range of new useful potential applications.

Abstract:As size-amplified analogues of canonical macromolecules, polymeric chains built up by "giant" monomers represent an experimental realization of the "beads-on-a-string" model at larger length scales, which could provide insights into fundamental principles of polymer science. In this work, we modularly constructed discrete giant polymeric chains using nanosized building blocks (polyhedral oligomeric silsesquioxane, POSS) as basic repeat units through an efficient and robust iterative exponential growth approach, with precise control on molecular parameters, including size, composition, regioconfiguration, and surface functionalities. Their chemical structures were fully characterized by nuclear magnetic resonance spectroscopy, size-exclusion chromatography, and matrix-assisted laser desorption ionization time-of-flight mass spectrometry. With elaborately designed amphiphilic block POSS chains and their analogues made of conventional monomers, the size effects were delicately studied and highlighted. Interesting assembly behaviors emerge as a result of distinct interactions and molecular dynamics. This category of molecules shares general self-assembly characteristics as the conventional counterparts in terms of phase transition and evolution. Meanwhile, it turns out that the monomer size has profound impacts on phase stability, as a trade-off between entropic and enthalpic contributions. It may open up a door for modular and programmable design of interesting materials with complex structures and diverse functions.

Abstract:Precise control of molecular hierarchical organization has endowed biological systems with unique functions, which inspires great efforts in the synthesis of artificial mimics. Polymeric single-chain nanoparticles (SCNPs) with tunable microstructure and composition and distinct compartmentalization features are regarded as an ideal platform for biomimetic architectures. SCNPs represent an intermediate state between polymer and colloidal matter and are constructed by intramolecular crosslinking of linear polymers with tailored composition and topology. The microstructure of the SCNPs is broadly tunable from individual particle, to tadpole-shaped and dimer by using the polymers with different sequential distribution. It is noted that cyclic polymers are regarded as specific SCNPs with well-defined number of knotting (crosslinking). Functional composite hybrids are derived whose head and chain parts are greatly enriched in composition and microstructure via selective growth of functional materials or post-modification. Synergistic effects can emerge through hybridization of functionalities of the colloidal nanoparticles and characteristics of the polymer chains. These hybrids are attractive for applications such as the fabrication of novel superstructures that mimic catalytic functionalities of enzymes. This review summarizes recent advances in the synthesis of SCNP based functional hybrid nanoparticles. Large-scale synthetic approaches of the hybrid nanoparticles are highlighted. The use of SCNP as platform for functional hybrid nanoparticles is subsequently illustrated by applications such as biomimetic catalysis, interfacial engineering and biomedicine.

Abstract:Precise patterning of polymer-based biomaterials for functional bio-nanostructures has extensive applications including biosensing, tissue engineering, and regenerative medicine. Remarkable progress is made in both top-down (based on lithographic methods) and bottom-up (via self-assembly) approaches with natural and synthetic biopolymers. However, most methods only yield 2D and pseudo-3D structures with restricted geometries and functionalities. Here, it is reported that precise nanostructuring on genetically engineered spider silk by accurately directing ion and electron beam interactions with the protein's matrix at the nanoscale to create well-defined 2D bionanopatterns and further assemble 3D bionanoarchitectures with shape and function on demand, termed "Protein Bricks." The added control over protein sequence and molecular weight of recombinant spider silk via genetic engineering provides unprecedented lithographic resolution (approaching the molecular limit), sharpness, and biological functions compared to natural proteins. This approach provides a facile method for patterning and immobilizing functional molecules within nanoscopic, hierarchical protein structures, which sheds light on a wide range of biomedical applications such as structure-enhanced fluorescence and biomimetic microenvironments for controlling cell fate.

Abstract:Protein adsorption onto polymer surfaces is a very complex, ubiquitous, and integrated process, impacting essential areas of food processing and packaging, health devices, diagnostic tools, and medical products. The nature of protein-surface interactions is becoming much more complicated with continuous efforts toward miniaturization, especially for the development of highly compact protein detection and diagnostic devices. A large body of literature reports on protein adsorption from the perspective of ensemble-averaged behavior on macroscopic, chemically homogeneous, polymeric surfaces. However, protein-surface interactions governing the nanoscale size regime may not be effectively inferred from their macroscopic and microscopic characteristics. Recently, research efforts have been made to produce periodically arranged, nanoscopic protein patterns on diblock copolymer surfaces solely through self-assembly. Intriguing protein adsorption phenomena are directly probed on the individual biomolecule level for a fundamental understanding of protein adsorption on nanoscale surfaces exhibiting varying degrees of chemical heterogeneity. Insight gained from protein assembly on diblock copolymers can be effectively used to control the surface density, conformation, orientation, and biofunctionality of prebound proteins in highly miniaturized applications, now approaching the nanoscale. This feature article will highlight recent experimental and theoretical advances made on these fronts while focusing on single-biomolecule-level investigations of protein adsorption behavior combined with surface chemical heterogeneity on the length scale commensurate with a single protein. This article will also address advantages and challenges of the self-assembly-driven patterning technology used to produce protein nanoarrays and its implications for ultrahigh density, functional, and quantifiable protein detection in a highly miniaturized format.

Abstract:A framework is presented for understanding the common character of proteins. Proteins are linear chain molecules. However, the simple model of a polymer viewed as spheres tethered together does not account for many of the observed characteristics of protein structures. The authors show here that proteins may be regarded as tubes of nonzero thickness. This approach allows one to bridge the conventional compact polymer phase with a novel phase employed by Nature to house biomolecular structures. The continuum description of a tube (or a sheet) of arbitrary thickness entails using appropriately chosen many-body interactions rather than two-body interactions. The authors suggest that the structures of folded proteins are selected based on geometrical considerations and are poised at the edge of compaction, thus accounting for their versatility and flexibility. This approach also offers an explanation for why helices and sheets are the building blocks of protein structures.

Abstract:As an important component that constitutes all the cells and tissues of the human body, protein is involved in most of the biological processes. Inspired by natural protein systems, considerable efforts covering many discipline fields were made to design artificial protein assemblies and put them into application in recent decades. The rapid development of structural DNA nanotechnology offers significant means for protein assemblies and promotes their application. Owing to the programmability, addressability and accurate recognition ability of DNA, many protein assemblies with unprecedented structures and improved functions have been successfully fabricated, consequently creating many brand-new researching fields. In this review, we briefly introduced the DNA-based protein assemblies, and highlighted the limitations in application process and corresponding strategies in four aspects, including biological catalysis, protein detection, biomedicine treatment and other applications.

Abstract:The chain effect of polymer is the kernel of the precise synthesis of complex polymer compounds and the development of polymer science. Nucleic acids, a special type of polyester, can form supramolecular polymers via hydrogen bonds among corresponding bases. We focused on the precise control of polymer chains and proposed the concept of "nucleic acid-backboned chimeric polymers " (NCPs). There are three important elements of NCPs, namely nucleic acid backbone, side group and intra-/inter-chain weak interactions. In this review, we expound how to realize the construction of intelligent DNA structures through the regulation of these three elements and the applications of NCPs in various fields.

Abstract:Functional nanomaterials have found wide applications in diverse areas because of their intrinsically different properties compared to their bulk counterparts. To achieve the goal of preparing functional nanomaterials, various strategies have been successfully developed. Among them, the biomolecule-directed approach has been extensively explored to synthesize many functional nanomaterials owing to their programmability, self-assembly and recognition capabilities. This Feature Article highlights the use of lysozyme as a model protein to the direct synthesis of nanomaterials. Future advances in rational de novo design and synthesis of functional nanomaterials with proteins will depend on a deep understanding of the synthetic strategies and the formation mechanisms. This Feature Article discusses the synthesis of nanomaterials with lysozyme in both the solution phase and crystal form. The synthetic strategies, formation mechanisms and wide applications of several kinds of materials, such as metals, oxides, metal sulfides, and composites, are covered. The lessons from this case study will provide invaluable guidance in future materials design using proteins and other biomolecules. Rational design of personalized functional nanomaterials will be possible in the future (366 references).

Abstract:Conventional inorganic materials are usually brittle and hard, which limits their application in some fields. Great efforts have been made to endow the inorganic materials with flexibility; however, there is hardly any effective method. Because of the sub-1-nm size, sub-1-nm nanomaterials show some unique properties compared with nanomaterials with larger sizes, such as the polymer-like property. Thus, sub-1-nm nanomaterials are found to be a good entry point to bridge inorganic materials and polymer materials. Rational assembly and processing of the functional sub-1-nm nanomaterials will bring great opportunities for the construction of micro- or nano-devices and applications of inorganic nanomaterials. In this Viewpoint, we briefly introduce the significance and research of the sub-1-nm nanomaterials and envision the future research direction and challenge.

Abstract:Nanoparticles are among the most fascinating materials because of their unique size-dependent properties. The spatial positioning of the nanoparticles with a nanoscale precision will greatly enhance their potential for use in the fabrication of functional materials and devices. Recently, the development of DNA nanotechnology has enabled the construction of two- and three-dimensional, precisely addressable nanostructures and devices based on DNA sequence design and programmable assembly. Advances in conjugating DNA with nanoparticles can bridge these two technologies and provide scientists with a new tool to study material transportation and energy transfer at a nanometer scale. In this review, we summarize the recent progress in this emerging research field, which includes not only the static arrangements of inorganic nanoparticles but also the dynamic regulation of organic and biological units at a nanometer scale. With the rapid development in this field, new challenges and new possibilities will emerge and bring about fruitful achievements with a significant impact on future work.

Abstract:Artificial molecular machine is one of the most brilliant areas in the last half a century. They represent people's control abilities to the microscopic molecules in solution. Beside the hard works in the proof-of-concept research on the exploration of fundamental principles, the wide hybrid of them with diverse materials was confirmed. Through the coupling of these movable molecules with various materials, the microscopic motion of molecular machines could be easily translated and amplified to macroscopic world to produce useful work. The integration further improves the ability of us to control the molecules in solid state. Among the various materials, polymers have the widest applications in many areas. The incorporation of artificial molecular machines into the bulk polymer materials would produce more influence on our life in the near future. In this review, we summarized the proofed hybrid strategies of polymers with different molecular machines. We hope that the summarization could provide a research roadmap for the related researchers. Abstract:In this brief review, we look back on the conception of nano-atoms and their gradual evolutions into a new class of giant molecules in the context of soft matter science. The structural features and the characteristics about giant molecular self-assembly are summarized. It is found that these giant molecules with high conformational rigidities and precisely-defined shapes and symmetries can exhibit unusual phase structures and phase transition behaviors which are not commonly observed in conventional polymers. Their self-assembly is robust due to collective and cooperative interactions among nano-atoms, forming hierarchical structures that are sensitive to their primary structures. This modular feature is reminiscent to the domain concept in protein science. It is thus proposed that nano-atoms can serve as unique elements for macromolecular science.
