Abstract:The protein adsorption is a crucial but complex phenomenon in many applications, like biosensors, biomaterials. The amount, orientation and biological activity of adsorbed proteins are heavily dependent upon the structure of protein, like widely-applied Bovine Serum Albumin (BSA), and the properties of the substrate. At some extent, to explore the physicochemical properties and dynamics interaction process between protein / multi-proteins with crystal/biomaterial surface, it has been presented that how to dig out the potential ability of the apparatus and other methods/tools for removing the block of studying interactions and measuring these complex interactions with crystal environments efficiently. In our recent works, it was demonstrated how to detect the BSA under aqueous conditions by steering the AFM tip in contact mode to sample more abundant topographical clues. The results indicated that this approach should be available for exploring more complex interaction system and acquiring more useful interaction information without altering the current apparatus platforms.
In Situ Comparative Studies of Self-Assembly Adsorption of Bovine Serum Albumin on Nano Films by Atomic Force Microscopy

Abstract:The self-assembly adsorption of function protein on crystal surfaces, as a common phenomenon, broadly takes place in many applications of biosensors, biocapsules and bioMEMS/bioNEMS. To systematically investigate the different adsorption characteristic of the same function protein on two different crystal surfaces under the identical environment, a hybrid surface composing silica and discontinuous Gold Nano Film (GNF) was fabricated by Physical Vapor Deposition (PVD) and ultrasonic cleaning method, where the dynamic process of the self-assembly adsorption of Bovine Serum Albumin (BSA) was in situ observed by Atomic Force Microscope (AFM). The variations on the junction area of the two different surfaces were studied in the aqueous solution before and after injecting BSA with 0.05 mg/ml concentration. It was found that silica, compared with same hydrophilic GNF, took on a fairly weak adsorption force. The results indicated that the adsorption strength of BSA on the hydrophilic crystal surface was determined not only by hydrophilic property, but also other interaction forces, like Van der Waals and so on. Moreover, observed under the contact mode of AFM, BSA adsorbed on GNF had great tendency to forming a ridge-like topography. These results may be helpful in the application of immunosensors and other areas.

Abstract:The self-assembly adsorption of bovine serum albumin (BSA) on the hybrid surface including the discontinuous gold nano film and silica under liquid condition was studied in situ by atomic force microscope (AFM) at room temperature. To investigate the adsorption of BSA on silica and gold nano film simultaneously, the surface with discontinuous gold nano film and visible blank block on silica by physical vapor deposition (PVD) and the ultrasonic cleaning technology was fabricated. Then we observed the variation on the substrate surface before and after injecting BSA aqueous solution with a 0.05 mg/ml concentration at real time by AFM. The experimental results indicated that the adsorption strength of BSA on the surface of gold nano film is larger than that on silica surface, and further implied that the adsorption capability of the hydrophobic surface is larger than that of the hydrophilic, which is consistent with those reported. Moreover, it could be easily observed that the adsorption of BSA on gold nano film has great tendency to form a ridgeway-like topography.
Adsorption of Bovine Serum Albumin on Nano and Bulk Oxide Particles in Deionized Water.

Abstract:In this work, the influence of particle size and surface functional groups on the adsorption behavior of bovine serum albumin (BSA) by three types of oxide nanoparticles (NPs), TiO(2) (50±5 nm), SiO(2) (30±5 nm), and Al(2)O(3) (150±5 nm for α type and 60±5 nm for γ type) was investigated in deionized water, in order to explore their interaction mechanisms without competitive influence of other ions. Bulkparticles (BPs) were also used for comparison with NPs. BSA adsorption maxima on oxide particles were controlled by the surface area and hydrogen content, while adsorption process was primarily induced by electrostatic interaction, hydrophobic interaction and ligand exchange between BSA and oxide surfaces. With the increase of hydrogen content, the BSA adsorption mechanism switched from mainly hydrophobic interaction to hydrogen bonding and ligand exchange. Calculations, based on surface area and BSA size, suggested that a multilayer of BSA covered on α-Al(2)O(3), and single layer on the other oxide particle surfaces. BPs led to greater conformational change of BSA molecules after the adsorption on the surfaces of oxide particles though NPs adsorbed more BSA than BPs.

Abstract:The surface oxide films on titanium were characterized and the relationship between the characterization and the adsorption of bovine serum albumin (BSA) on titanium was studied. The surface oxide films on titanium were obtained by heat-treatment in different oxidizing atmospheres, such as air and water vapor. The surface roughness, energy, morphology, chemical composition and crystal structure were used to characterize the titanium surfaces. The characterization was performed using a profilometer, scanning electronic microscopy (SEM), a sessile drop method, X-ray photoelectron spectroscopy (XPS) and X-ray diffraction (XRD). Percentages of surface hydroxyl groups were determined by XPS analysis for the titanium plates and the densities were measured by a chemical method for titanium powders. After heat-treatment, the titanium surfaces were uniformly roughened and the surface titanium oxide was predominantly rutile TiO2. The crystal planes in the rutile films were preferentially orientated in the (110) plane with the highest density of titanium ions. Heat-treatment increased the surface energy and the amount of surface hydroxyl groups on the titanium. The different oxidizing atmospheres resulted in different percentages of oxygen species in the TiO2, in the physisorbed water and acidic hydroxyl groups and in the basic hydroxyl groups on the titanium surfaces. The analysis for the adsorption of BSA on titanium confirmed that the surface characterization of titanium has a strong effect on the bioactivity of titanium. The BSA chemically adsorbed onto the titanium surfaces. The adsorption of BSA on the titanium surfaces was positively related with the amounts of their surface hydroxyl groups, including basic hydroxyl groups and acidic hydroxyl groups, and the values of the polar component of the total surface energy.
Adsorption of BSA on Carbon-Coated Fe3O4 Microspheres Activated with 1-Ethyl-3-(3-dimethylaminopropyl)-carbodiimide Hydrochloride

Abstract:Carbon-coated Fe3O4 (Fe3O4/C) microspheres activated with 1-ethyl-3-(3-dimethylaminopropyl)-carbodiimide hydrochloride (EDC) were prepared, characterized and applied to adsorb bovine serum albumin (BSA). The prepared magnetic microspheres had spherical core-shell structure with a uniform and continuous carbon coating coupled with activation by EDC, and possessed superparamagnetic characteristics. The experimental results showed that the adsorption amount of BSA on the EDC-activated Fe3O4/C (Fe3O4/C-EDC) microspheres was higher than that on the Fe3O4/C microspheres. The maximum adsorption of BSA on Fe3O4/C-EDC microspheres occurred at pH 4.7, which was the isoelectric point of BSA. At low concentrations (below 1.0 M), salt had no noticeable effect on BSA adsorption. The BSA adsorption of Fe3O4/C-EDC microspheres had a better fit to the Langmuir model than the Freundlich isotherm and Temkin isotherm model, and the kinetic data were well described by the pseudo-second-order model. The adsorption equilibrium could be reached within 20 min. High desorption efficiency (97.6%) of BSA from Fe3O4/C-EDC microspheres was obtained with 0.5 M Na2HPO4 (pH 9.4) as the desorbent.

Abstract:Protein adsorption is driven by various interactions. The contribution of surface charge to bovine serum albumin (BSA) adsorption on hydroxyapatite (HA) ceramic was investigated by adjusting the liquid environment in which the solid particles dispersed. Zeta potentials of HA and the adsorption of BSA on the surface were tested as a function of pH, ionic strength, Ca2+ and PO43- concentrations in the aqueous solutions, and the results showed that both of them were greatly affected by those experimental variations. Besides, the amount of adsorbed BSA was related to the variation of zeta potential of HA, as could be well understood in terms of electrostatic interactions.

Abstract:<p>CoCrMo alloys have been used as biomedical materials for decades. During long-term service in the body environment, however, proteins adsorbing onto the surface of implant can cause their spontaneous corrosion and even the release of toxic ions, which may cause long-term damage to patients. It is essential to understand the key factors affecting protein adsorption and how it can alter corrosion reactions. In this study, the surface energy of a CoCrMo alloy was changed through various oxidation processes. Surface analysis techniques, such as XPS, AES, and GIXRD were used to characterise the chemical properties of the surfaces of modified CoCrMo alloys. Surface topography and potential images of samples after bovine serum albumin (BSA) adsorption were acquired by AFM/SKPFM. The results show that with the increase of the oxidation temperature, the hydrophilicity of the samples was enhanced. With the increase in oxidation time at the same temperature, the surface energy of the samples increased, the dispersion component increased, and the polar component decreased. As a result, the amount of BSA adsorbed on the surfaces decreased, and this was affected more by the polar component than by other components, which was due to the electrostatic and hydrophobic interactions.</p>

Abstract:Two kinds of pH-dependent magnetic nanoadsorbent based on silica-coated MnFe2O4 nanoparticles (SMNPs) and amino-modified silica-coated MnFe2O4 nanoparticles (AS-MNPs) have been synthesized using sol–gel method. X-ray diffraction, Fourier transform infrared spectroscopy and transmission electron microscopy have been employed to characterize the structure and morphology of the nanoadsorbents. The magnetic properties and zeta potential were also investigated. These nanoadsorbents were used to adsorb bovine serum albumin (BSA). The adsorption capacity of BSA largely depended on the pH and ionic strength of solution. In the best result, BSA was adsorbed on AS-MNPs at a high value of 164mgg−1, which is much higher than that of SMNPs (100mgg−1). This may due to the increased surface amino that can be conjugated to BSA by a chemical bond and the electrostatic attraction between BSA and magnetic nanoparticles.

Abstract:Magnetic sandwich structured mesoporous silica microspheres containing a silica-coated Fe3O4 core and a layered mesoporous silica shell have been successfully synthesized by multi-step reactions. The as-prepared Fe3O4/SiO2/mesoporous SiO2 (Fe3O4/SiO2/mSiO(2)) microspheres were characterized via X-ray diffractometry (XRD), transmission electron microscopy (TEM), nitrogen adsorption-desorption and vibrating sample magnetometry (VSM). The results indicate that these materials possess superior porosity with high surface areas and large pore volumes, while still maintaining the microspheres morphologies, which is beneficial for enrichment and purification applications. The microspheres were applied as adsorbents for bovine serum albumin (BSA). The adsorption equilibrium was well explained in the entire experimental region by the Langmuir isotherm model and the maximum adsorption amounts of BSA were calculated as high as 289 mg g(-1). This work provides a promising approach for the design and synthesis of multifunctional microspheres, which can be used for potential applications in a variety of biomedical fields including drug delivery and biosensors.
Surface Functionalization of Zirconium Dioxide Nano-Adsorbents with 3-Aminopropyl Triethoxysilane and Promoted Adsorption Activity for Bovine Serum Albumin

Abstract:Surface functionalization of zirconium dioxide (ZrO2) nano-adsorbents was carried out by using 3-aminopropyl triethoxysilane (APTES) as the modifier. The addition amount of APTES was varied to determine the optimum modification extent, and the bulk ZrO2 microparticles were also modified by APTES for comparison. Some means, such as TEM, XRD, FT-IR, XPS and TG-DSC were used to character these ZrO2 particles. The results showed that the APTES molecules were chemically immobilized on the surface of ZrO2 nanoparticles via ZrOSi bonds, and the nano-ZrO2 samples showed larger special surface area. In the adsorption of bovine serum albumin (BSA), nano-ZrO2 samples exhibited enhanced adsorption activity, and APTES modified nano-ZrO2 with proper APTES content presented the best adsorption property. Under the same adsorption conditions, the equilibrium adsorption capacity of BSA on APTES-ZrO2-2 was almost 2.3 times as that on pristine nano-ZrO2 and 3.0 times as on bulk ZrO2 microparticles. The increased adsorption capacity of APTES-ZrO2 nano-adsorbents can be attributed to the chemical interaction between amino and carboxyl groups at APTES-ZrO2/BSA interface. The pH-dependent experiments showed that the optimum pH value for the adsorption and desorption was 5.0 and 9.0, respectively, which suggested that the adsorption and release of BSA could be controlled simply by adjusting the solution pH condition.

Abstract:Studying on the interaction between proteins and calcium phosphate implants is one of the basic subjects in biomaterials science and engineering. In this work, zeta potentials and contact angles of hydroxyapatite (HA) and biphasic calcium phosphate (BCP) ceramics were measured, and the adsorption behaviors of bovine serum albumin (BSA) on the surfaces were investigated. The adsorption isotherms of BSA on both ceramics follow the Langmuir type, however, BCP shows higher BSA adsorption ability. The differences of contact angles and zeta potentials in HA and BCP are the key factors to determine their ability to bind BSA molecules.

Abstract:MnO2-impregnated carbon-coated Fe3O4 (Fe3O4/C/MnO2) nanocomposites with a good core–shell structure were prepared by direct oxidation of carbon-coated Fe3O4 (Fe3O4/C) microspheres with KMnO4 in alkaline solution and applied to adsorb bovine serum albumin (BSA). X-ray diffraction (XRD), transmission electron microscope (TEM), Fourier transform infrared spectrometer (FTIR), vibrating sample magnetometer (VSM) and thermogravimetric analyzer (TGA) tests show that Fe3O4/C microspheres were newly functionalized via the oxidation by KMnO4. Fe3O4/C/MnO2 nanocomposites exhibit a higher adsorption capacity for BSA than Fe3O4/C microspheres and the maximum adsorption of BSA on them occurs at pH 4.7, which is the isoelectric point of BSA. Langmuir isotherm model describes the adsorption of BSA better than Freundlich model and Temkin model, and the kinetics data fit well with the pseudo-second-order model.

Abstract:Attenuated total reflection (ATR) FT-IR spectroscopy was used to quantitatively characterize the extent of bovine serum albumin (BSA) adsorbed on the surface-coating-modified poly(ether urethane) (PEU) matrix. The two surface modifying additives (SMA) were respectively a tri-block-coupling-polymer of stearyl poly (ethylene oxide)-4,4'-methylene diphenyl diisocyanate-stearyl poly(ethylene oxide), for short MSPEO, and another similar block-coupling polymer with the Cibacron Blue F3G-A endgroups, for short cibaMPEO. The experiments of static BSA adsorption were composed by two parts. One was static isothermal adsorption, and the other was static adsorption kinetics. The quantitative characterization was based on the optical principles of FT-IR, method of experiment and index of the apparatus, by which the enhancement of BSA adsorption on the SMA-modified surfaces was confirmed.

Abstract:The magnetic carboxymethyl chitosan nanoparticles (MNPs-CMC) were developed as effective magnetic affinity adsorbent for Bovine serum albumin and the adsorption reactions were investigated. The obtained experimental data were compared with the adsorption kinetics models and equilibrium isotherms. The experimental kinetic data were modeled using Pseudo-first order, Pseudo-second order, Bangham's equation, Intra-particle diffusion model and Elovich equations. It was found that the adsorption reactions followed the Pseudo-second order kinetics equation. The experimental isotherm data were analyzed using Langmuir, Freundlich, Dubinin-Radushkevich and Temkin equations. By comparing the correlation coefficients determined for each linear transformation of isotherm analysis, it was found that the Langmuir equation was the best fit equilibrium model for the adsorption of BSA. Error functions have been used to determine the alternative single component parameters by nonlinear regression due to the inherent bias in using the correlation coefficient resulting from linearization. It showed that the Langmuir equation resulted in the lowest values for the error function and thus fitted the data better than the other isotherm. Various thermodynamic parameters such as enthalpy (Delta H degrees), entropy (Delta S degrees) and Gibbs free energy (Delta G degrees) were evaluated. MNPs-CMC nanoaprticles were shown to be a promising material for adsorption of BSA from aqueous solutions. Crown Copyright (C) 2013 Published by Elsevier B.V. All rights reserved.

Abstract:The mode of adsorption of bovine serum albumin (BSA) on porous polyethylene (PE) membrane was studied as a function of time and concentration, which may contribute to the surface coverage. An improved physical model for adsorption is initiated based on the results of the adsorptional and desorptional measurements, FTIR analysis, and AFM observations as well as streaming potential measurements. The results obtained indicate that the adsorptional mode depend on both time and concentration. It is shown that a critical concentration (about 1000 ppm here) exists in the adsorptional process. Below this concentration, the adsorption seems to be conducted in a normal side-on way but time elapse gives rise to greater conformational change than concentration increase; above this concentration, the aggregation of protein molecular plays a decisive role and the adsorption is in an aggregation way, which is similar to end-on, but a relative large gap between the adsorbed molecules exists due to aggregation. This conclusion is general and can be expected to apply in other globular protein–hydrophobic porous surface systems.
Spectroscopic Studies of Bovine Serum Albumin Adsorbed Onto Magnetic-Thermosensitive Carbon Microspheres.

Abstract:To investigate the influence of magnetic-thermosensitive carbon microspheres (MTCMSs) as a targeting drug carrier on serum albumins in vitro, in this study, bovine serum albumin (BSA) was chosen as a template protein to explore the interaction between serum proteins and MTCMSs. Fluorescence spectrophotometry, ultraviolet-visible absorbance (UV-vis) spectrophotometry and circular dichroism spectrometry were used to investigate the interaction between MTCMSs and BSA. Results indicate that BSA interacts with MTCMSs and the fluorescence intensity of BSA is quenched by 50% in a static quenching at 310 K when the concentration of MTCMSs reaches 30 mg/L. Thermodynamic parameters including free energy change (△Gθ ), enthalpy change (△Hθ ) and entropy change (△Sθ ) were calculated. The results (△Gθ < 0, △Hθ < 0 and △Sθ > 0) suggest a spontaneous process and the formation of a hydrogen bond between MTCMSs and BSA. UV-vis measurements reveal that the micro-environment of an amino acid residue is altered in the presence of MTCMSs. The α-helix content of BSA decreases by 4% and the β-sheet content increases by 3.2% with increasing concentrations of MTCMSs to 30 mg/L, illustrating a change in the skeletal structure of BSA. These results demonstrate that MTCMSs as a targeting drug carrier impact the structure of serum albumins. This work provides not only a theoretical basis of BSA adsorption onto MTCMSs, but also an understanding of safe drug carriers in biomedicine. Copyright © 2016 John Wiley & Sons, Ltd.

Abstract:Carboxymethyl chitosan (CMC) were prepared from the carboxymethylation reaction of chitosan under mild conditions, and magnetic carboxymethyl chitosan nanoparticles (MNPs-CMC) were developed by binding CMC on the surface of magnetic Fe3O4 nanoparticles (MNPs) via EDC/NHS activation. The characterization results indicated that MNPs were successfully modified by CMC and the modified nanoparticles had a good magnetic property. By investigating the adsorption of BSA, it was found that the maximum BSA immobilization capacity was up to 159.51 mg·g−1. The adsorption characteristics of BSA can fit the pseudo-second-order model and Langmuir equation well. Furthermore, a high desorption efficiency of 81.47% was achieved under an alkaline condition (pH 9.6).

Abstract:Butyl, octyl and hexadecyl moieties were introduced into chitosan. The adhesion of bovine serum albumin (BSA) onto glucose aldehyde-crosslinked alkylated chitosan membranes in pH media was investigated by probing the force-displacement curves with BSA-coated Atom force microscope (AFM) tips. The results indicated that, at the isoelectric point (IP), the sample membranes exhibited higher adhesion forces; and deviating from IP ie at pH 2, pH 6, the adhesion forces decreased. The adhesion forces at pH 2 are less than those at pH 6 due to the presence of electrostatic repulsive and attractive interactions, respectively. Measurements of the adhesion force confirmed quantitatively that the introduction of hydrophobic side-chains to chitosan can facilitate protein adsorption; however, longer flexible side-chains can depress protein adsorption to a certain degree. From an analyses of the adhesion forces, it is proposed that protein adsorption can be tuned by adjusting the lengths of the introduced side-alkyl moieties. (C) 2002 Society of Chemical Industry.
