Changes in SNK/SPAR Mrna and Protein Levels in Glutamate-Induced Rat Cultured Hippocampal Neurons' Excitotoxicity

Abstract:Objective To observe the change in serum-inducible kinase(SNK)/spine-associated Rap guanosine triphosphatase(GTPase)activating protein(SPAR)expression levels in glutamate-induced cultured hippocampal neurons' excitotoxicity.Methods Excitotoxicity was induced by glutamate in rat cultured hippocampal neurons.RT-PCR was used to detect the changes in SNK/SPAR mRNA levels.Western blot was used to detect the changes in SNK/SPAR protein levels.Double immunofluorescence staining was used to identify the distribution change in SNK/SPAR on hippocampal neurons.Results Compared with the basal level,1 h after 100 μmol/L glutamate exposure,SNK mRNA markedly elevated.This elevation continued,reached the peak in 6 h,and then back to the base level gradually.For the protein level,3 h after glutamate stimulation,SNK was detected to increase,and reached the peak in 36 h.the expression tendency of SPAR was contrary to that of SNK.Immunocytochemistry showed that the distribution changes in SNK and SPAR were specifically significant in the somatodendritic compartment.Conclusion SNK-SPAR pathway may play an important role in glutamate-induced excitotoxic damage in neurons.
