Abstract:Piriformospora indica(P. indica), a root-colonizing endophytic fungus, can improve the resistance of plants against biotic and abiotic stresses. In order to clarify the influences of P. indica on the disease resistance of Nicotiana tabacum, the resistance experiments against tobacco brown spot, anthracnose, damping off and sore shin were conducted wherein P. indica-colonized N. tabacum was inoculated with Alternaria longipes(A.longipes), Colletotrichum gloeosporioides(C. gloeosporioides), Pythium ultimum(P. ultimum) and Rhizoctonia solani(R. solani), respectively; and the contents of malondialdehyde(MDA) and proline, antioxidant enzyme activity and the expression levels of pathogenesis-related protein genes in tobacco leaves inoculated with A. longipes were analyzed. The results showed that comparing with the control, the size of disease spots and the MDA content in the leaves of P. indica-colonized N. tabacum inoculated with A. longipes reduced significantly, while the proline content and the expression levels of pathogenesis-related protein genes PR-1a, PR2, PR3 and PR5 increased significantly; the size of disease spots in the leaves of P. indica-colonized N. tabacum inoculated with C. gloeosporioides reduced; and damage symptoms of the leaves of P. indica-colonized N. tabacum inoculated with P. ultimum and R. solani significantly decreased. P. indica can improve the disease resistance of N.tabacum through maintaining the integrity of cell biomembrane system, keeping intracellular osmotic pressure and the peroxidation of membrane lipid at a lower level and regulating the expression of pathogenesis-related protein genes.
Piriformospora Indica Confers Cadmium Tolerance in Nicotiana Tabacum

Abstract:Piriformospora indica, a root-colonizing endophytic fungus of Sebacinales, promotes plant growth and confers resistance against biotic and abiotic stresses. In order to confirm the influence of P. indica on growth, proline, malondialdehyde (MDA), chlorophyll, and cadmium (Cd) amounts in Nicotiana tabacum under Cd stress, hydroponics, pot and field trials were conducted. The results showed that P. indica can store Cd in plant roots and reduce leaf Cd content, reduce the concentration of MDA, and increase the proline and chlorophyll content and the activities of catalase, peroxidase, and superoxide dismutase under hydroponic Cd stress. RT-PCR analysis showed that the relative expression level of genes Gsh2, TaPCS1, oas1, GPX, and Hsp70 in colonized plants was 4.3, 1.4, 2.9, 1.7, and 6.9 fold higher than in un-colonized plants respectively. Cd exposure significantly reduced un-colonized plants' agronomic traits compared to P. indica-colonized ones. Our results suggested that P. indica can sequester Cd in roots, so that much less cadmium was transported to leaves, and the increased concentrations of antioxidant enzymes, pigments and proline contents, as well as the higher expression of stress-related phytochelatin biosynthesis genes in P. indica-inoculated plants, may also serve to protect N. tabacum plants against oxidative damage, enhancing Cd tolerance.

Abstract:The beneficial endophytic microorganisms have received significant attention in agriculture because of their exceptional capabilities to facilitate functions like nutrient enrichment, water status, and stress tolerance (biotic and abiotic). This review signifies the molecular mechanisms to better understand the Piriformospora indica-mediated plants improvement or protection for sustainable agriculture. P. indica, an endophytic fungus, belonging to the order Sebacinales (Basidiomycota), is versatile in building mutualistic associations with a variety of plants including pteridophytes, bryophytes, gymnosperms, and angiosperms. P. indica has enormous potential to manipulate the hormonal pathway such as the production of indole-3-acetic acid which in turn increases root proliferation and subsequently improves plant nutrient acquisition. P. indica also enhances components of the antioxidant system and expression of stress-related genes which induce plant stress tolerance under adverse environmental conditions. P. indica has tremendous potential for crop improvement because of its multi-dimensional functions such as plant growth promotion, immunomodulatory effect, biofertilizer, obviates biotic (pathogens) and abiotic (metal toxicity, water stress, soil structure, salt, and pH) stresses, phytoremediator, and bio-herbicide. Considering the above points, herein, we reviewed the physiological and molecular mechanisms underlying P. indica-mediated plants improvement or protection under diverse agricultural environment. The first part of the review focuses on the symbiotic association of P. indica with special reference to biotic and abiotic stress tolerance and host plant root colonization mechanisms, respectively. Emphasis is given to the expression level of essential genes involved in the processes that induce changes at the cellular level. The last half emphasizes critical aspects related to the seed germination, plant yield, and nutrients acquisition.
Effects of Piriformospora Indica on Drought Resistance of Nicotiana Tabacum

Abstract:To investigate the effect of Piriformospora indica on drought resistance of Nicotiana tabacum, a pot experiment was conducted by using polyethylene glycol (PEG) 6000 to simulate drought and the malondialdehyde (MDA) and proline contents, relative electrical conductivity and expression levels of drought-related genes in tobacco leaves under drought stress were analyzed. The results showed that P. indica could colonize in the roots of N. tabacum and promote the growth of both root and shoot of the plants. When P. indica-inoculated plants were under drought stress, the symptoms of water shortage were slighter, the proline content in leaves was significantly up-regulated, while the MDA content and relative electrical conductivity of leaves were significantly decreased comparing with those of un-colonized treatment. Quantitative real-time polymerase chain reaction (qRT-PCR) analysis indicated that the relative expression levels of drought-related genes RD29A, ERD10A, RD26A, SDIR1, HAT, ERD1 and DREB2A were higher in leaves of colonized plants under drought stress. P. indica enhances the drought resistance of N. tabacum via reducing tobacco cell biological membrane damage degree, enhancing the antioxidant ability of tobacco, promoting the accumulation of osmotic substances proline and up-regulating the expression of drought-related genes.
Functional Analysis and Genome Mining Reveal High Potential of Biocontrol and Plant Growth Promotion in Nodule-Inhabiting Bacteria Within <i>Paenibacillus polymyxa</i> Complex

Abstract:Bacteria belonging to the genus Paenibacillus were frequently isolated from legume nodules. The nodule-inhabiting Paenibacillus as a resource of biocontrol and plant growth-promoting endophytes has rarely been explored. This study explored the nodule-inhabiting Paenibacillus' antifungal activities and biocontrol potentials against broad-spectrum important phytopathogenic fungi. We collected strains which were isolated from nodules of Robinia pseudoacacia, Dendrolobium triangulare, Ormosia semicastrata, Cicer arietinum, Acacia crassicarpa, or Acacia implexa and belong to P. peoriae, P. kribbensis, P. endophyticus, P. enshidis, P. puldeungensis, P. taichungensis, or closely related to P. kribbensis, or P. anseongense. These nodule-inhabiting Paenibacillus showed diverse antagonistic activities against five phytopathogenic fungi (Fusarium graminearum, Magnaporthe oryzae, Rhizoctonia solani, Sclerotinia sclerotiorum, and Botrytis cinerea). Six strains within the P. polymyxa complex showed broad-spectrum and potent activities against all the five pathogens, and produced multiple hydrolytic enzymes, siderophores, and lipopeptide fusaricidins. Fusaricidins are likely the key antimicrobials responsible for the broad-spectrum antifungal activities. The nodule-inhabiting strains within the P. polymyxa complex were able to epiphytically and endophytically colonize the non-host wheat plants, produce indole acetic acids (IAA), and dissolve calcium phosphate and calcium phytate. P. peoriae strains RP20, RP51, and RP62 could fix N-2. P. peoriae RP51 and Paenibacillus sp. RP31, which showed potent plant colonization and plant growth-promotion competence, effectively control fungal infection in planta. Genome mining revealed that all strains (n = 76) within the P. polymyxa complex contain ipdC gene encoding indole-3-pyruvate decarboxylase for biosynthesis of IAA, 96% (n = 73) contain the fus cluster for biosynthesis of fusaricidins, and 43% (n = 33) contain the nif cluster for nitrogen fixation. Together, our study highlights that endophytic strains within the P. polymyxa complex have a high probability to be effective biocontrol agents and biofertilizers and we propose an effective approach to screen strains within the P. polymyxa complex.

Abstract:Piriformospora indica (Serendipita indica), a mycorrhizal fungus, has garnered significant attention in recent decades owing to its distinctive capacity to stimulate plant growth and augment plant resilience against environmental stressors. As an axenically cultivable fungus, P. indica exhibits a remarkable ability to colonize varieties of plants and promote symbiotic processes by directly influencing nutrient acquisition and hormone metabolism. The interaction of plant and P. indica raises hormone production including ethylene (ET), jasmonic acid (JA), gibberellin (GA), salicylic acid (SA), and abscisic acid (ABA), which also promotes root proliferation, facilitating improved nutrient acquisition, and subsequently leading to enhanced plant growth and productivity. Additionally, the plant defense system was employed by P. indica colonization and the defense genes associated with oxidation resistance were activated subsequently. This fungus-mediated defense response elicits an elevation in the enzyme activity of antioxidant enzymes, including superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT), and, finally, bolsters plant tolerance. Furthermore, P. indica colonization can initiate local and systemic immune responses against fungal and viral plant diseases through signal transduction mechanisms and RNA interference by regulating defense gene expression and sRNA secretion. Consequently, P. indica can serve diverse roles such as plant promoter, biofertilizer, bioprotectant, bioregulator, and bioactivator. A comprehensive review of recent literature will facilitate the elucidation of the mechanistic foundations underlying P. indica-crop interactions. Such discussions will significantly contribute to an in-depth comprehension of the interaction mechanisms, potential applications, and the consequential effects of P. indica on crop protection, enhancement, and sustainable agricultural practices.

Abstract:Piriformospora indica is a wide-host root-colonizing endophytic fungus which allows the plants to grow under extreme physical and nutrient stress. The fungus can be cultivated on complex and minimal substrates. It belongs to the Sebacinales in Basidiomycota. P. indica has a vast geographical distribution and is reported from Asia, South America and Australia. The fungus is interesting for basic research as well as biotechnological applications because: (i) it functions as a plant promoter and biofertilizer in nutrient-deficient soils, (ii) as a bioprotector against biotic and abiotic stresses including root and leaf fungus pathogens and insect invaders, (iii) as a bioregulator for plant growth development, early flowering, enhanced seed production, and stimulation of active ingredients in medicinal plants (iv) as well as a bio-agent for the hardening of tissue-culture-raised plants. Positive interaction are established for many plants of economic importance in arboriculture, agro-forestry, flori-horticulture including Orchids, and those utilized for energy production and paper industry. P. indica also interacts with members of bryophyte, Aneura pinguis, pteridophyte, Pteris ensiormis, Gymnosperms (Pinus halepensis) and a large number of angiosperms (145 tested till date) including the model plant Arabidopsis thaliana and other members of the mustard family. Similar to arbuscular mycorrhizal fungi, P. indica stimulates nutrient uptake in the roots and solubilizes insoluble phosphatic and sulphur components in the soil. The interaction of P. indica with the model plants Arabidopsis thaliana and barley (Hordeum vulgare L.) is used to understand the molecular basis of this beneficial plant/microbe interaction. We describe the current knowledge about the molecular basis of the interaction of plants with P. indica. An attempt is made to compare it with pathogenic and mycorrhizal plant/microbe interactions and also propose possible biotechnological applications.

Proteomic Approach to Understand the Molecular Physiology of Symbiotic Interaction Between Piriformospora Indica and Brassica Napus.

Abstract:Many studies have been now focused on the promising approach of fungal endophytes to protect the plant from nutrient deficiency and environmental stresses along with better development and productivity. Quantitative and qualitative protein characteristics are regulated at genomic, transcriptomic, and posttranscriptional levels. Here, we used integrated in-depth proteome analyses to characterize the relationship between endophyte Piriformospora indica and Brassica napus plant highlighting its potential involvement in symbiosis and overall growth and development of the plant. An LC-MS/MS based label-free quantitative technique was used to evaluate the differential proteomics under P. indica treatment vs. control plants. In this study, 8,123 proteins were assessed, of which 46 showed significant abundance (34 downregulated and 12 upregulated) under high confidence conditions (p-value ≤ 0.05, fold change ≥2, confidence level 95%). Mapping of identified differentially expressed proteins with bioinformatics tools such as GO and KEGG pathway analysis showed significant enrichment of gene sets involves in metabolic processes, symbiotic signaling, stress/defense responses, energy production, nutrient acquisition, biosynthesis of essential metabolites. These proteins are responsible for root's architectural modification, cell remodeling, and cellular homeostasis during the symbiotic growth phase of plant's life. Analysis of growth dynamics in five different media and metabolic phenotypic characteristics of Piriformospora indica

Abstract:Piriformospora indica is an important endophytic fungus with broad potential for alleviating biotic and abiotic stress on host plants. This study monitored the growth dynamics of P. indica on five commonly used artificial media for microorganisms and analyzed its metabolic characteristics using Biolog Phenotype Microarray (PM) technology. The results showed that P. indica grew fastest on Potato Dextrose Agar (PDA), followed by Kidney Bean Agar (KBA), Alkyl Ester Agar (AEA), Oatmeal Agar (OA), and Luria-Bertani Agar (LB), and the most suitable medium for spore production was OA. Using Biolog PM1-10, 950 metabolic phenotypes of P. indica were obtained. P. indica could metabolize 87.89% of the tested carbon sources, 87.63% of the tested nitrogen sources, 96.61% of the tested phosphorus sources, and 100% of the tested sulfur sources. P. indica displayed 92 kinds of tested biosynthetic pathways, and it could grow under 92 kinds of tested osmotic pressures and 88 kinds of tested pH conditions. PM plates 1-2 revealed 43 efficient carbon sources, including M-Hydroxyphenyl acid, N-Acetyl-D-Glucosamine, Tyramine, Maltotrios, α-D-Glucosine, I-Erythritol, L-Valine, D-Melezitose, D-Tagatose, and Turanose. PM plates 3,6-8 indicated 170 efficient nitrogen sources, including Adenosine, Inosine Allantoin, D, L-Lactamide, Arg-Met, lle-Trp, Ala-Arg, Thr-Arg, Trp-Tyr, Val-Asn, Gly-Gly-D-Leu, Gly-Gly-Phe, and Leu-Leu-Leu. This study demonstrates that P. indica can metabolize a variety of substrates, such as carbon and nitrogen sources, and has a wide range of environmental adaptability. The growth dynamics on artificial culture media and metabolic phenotypes of P. indica can be used to investigate its biological characteristics, screen for more suitable growth and sporulation conditions, and elucidate the physiological mechanisms that enhance the stress resistance of host plants. This study provides a theoretical basis for its better application in agriculture.

Abstract:Seredipita indica (formerly Piriformospora indica ) is an endophytic fungus that establishes the symbiosis within the roots of various plants, exhibiting analogous functions to arbuscular mycorrhizal fungi. S . indica can proliferate in vitro in synthetic media, without the need of a host. Due to its isolation from desert environments, S . indica is particularly prominent in enhancing the host plant's tolerance to abiotic stresses. This review briefly analyzes the role of S . indica in plants exposed to abiotic stresses (e.g., drought, waterlogging, salt stress, low temperatures, and heavy metal stress). This review also elucidates the underlying mechanism regarding S . indica -enhanced tolerance of host plants in response to abiotic stress by regulating nutrient acquisition, osmoregulation (proline, soluble sugars, betaine, and K + ), phytohormone (auxins, abscisic acid, ethylene, and gibberellins) balance, antioxidant enzyme defense systems, polyamines (e.g., putrescine), and functional genes (e.g., aquaporins and phosphate transporter). Some of the fungus' own genes, such as transporters of polyamines, also respond to abiotic stresses, thereby assisting the host in co-resistance to abiotic stresses. The review further examines the application potential of S . indica to enhance stress tolerance in the field as well as proposes future prospects (e.g., omics, fungal self-response, reactive oxygen species signalling transduction, and its association with other microorganisms).

Abstract:The root-endophytic fungus Piriformospora indica (=Serendipita indica) has been revealed for its growth-promoting effects and its capacity to induce resistance in a broad spectrum of host plants. However, the bioefficacy of this fungus had not yet been tested against any pathogen affecting onion (Allium cepa). In this study, the biocontrol potency of P. indica against onion leaf blight, an impacting disease caused by the necrotrophic fungal pathogen Stemphylium vesicarium, was evaluated. First, it was proved that colonisation of onion roots by P. indica was beneficial for plant growth, as it increased leaf development and root biomass. Most relevantly, P. indica was also effective in reducing Stemphylium leaf blight (SLB) severity, as assessed under greenhouse conditions and confirmed in field trials in two consecutive years. These investigations could also provide some insight into the biochemical and molecular changes that treatment with P. indica induces in the main pathways associated with host defence response. It was possible to highlight the protective effect of P. indica colonisation against peroxidative damage, and its role in signalling oxidative stress, by assessing changes in malondialdehyde and H2O2 content. It was also showed that treatment with P. indica contributes to modulate the enzymatic activity of superoxide dismutase, catalase, phenylalanine ammonia-lyase and peroxidase, in the course of infection. qPCR-based expression analysis of defence-related genes AcLOX1, AcLOX2, AcPAL1, AcGST, AcCHI, AcWRKY1, and AcWRKY70 provided further indications on P. indica ability to induce onion systemic response. Based on the evidence gathered, this study aims to propose P. indica application as a sustainable tool for improving SLB control, which might not only enhance onion growth performance but also activate defence signalling mechanisms more effectively, involving different pathways.
Role of auxin in the symbiotic relationship between Piriformospora indica and rice plants

Abstract:Piriformospora indica (P. indica) is a beneficial endophytic fungus colonized with roots of a wide range of host plant species; however, the mechanisms of P. indica symbiosis with different plants could vary and need to be further addressed. Our study showed that root architecture and growth behaviors were changed in P. indica-colonized rice plants. The P. indica-induced primary root shorting and coiling phenomena were consistent with the auxin-responsive phenotype, suggesting that the auxin levels of P. indica-colonized roots were modulated. To investigate the changes in auxin levels, auxin-responsive gene expressions and promoter activities were used as indicators to monitor auxin levels in root tissues. The results showed that the auxin levels of primary roots were increased in P. indica-inoculated seedlings. Furthermore, an auxin inhibitor (p-chlorophenoxyisobutyric acid, PCIB) was applied to block auxin action to confirm whether auxin is a critical factor in regulating P. indica-induced root growth. The P. indica-induced root architecture modification was significantly repressed by PCIB which was added to the rice culture solutions at the initiation of P. indica inoculation. A proposed hypothesis was that the PCIB treatment might result in lower colonization and further reduce the influence of P. indica colonization on root architecture. The effect of PCIB on colonization was observed by counting the percent root colonization. The data showed that the colonization levels of P. indica in PCIB-treated seedlings were significantly decreased. Taken together, our data indicated that auxin levels would be increased in P. indica-colonized rice roots. Moreover, it was demonstrated that auxin has dual roles in the symbiotic relationship between rice and P. indica, which are a signal that triggers P. indica-induced root growth and a positive factor involved in the colonization mechanism.

The Beneficial Fungus Piriformospora Indica Protects Arabidopsis from Verticillium Dahliae Infection by Downregulation Plant Defense Responses

Abstract:BACKGROUND:Verticillium dahliae (Vd) is a soil-borne vascular pathogen which causes severe wilt symptoms in a wide range of plants. The microsclerotia produced by the pathogen survive in soil for more than 15 years.RESULTS:Here we demonstrate that an exudate preparation induces cytoplasmic calcium elevation in Arabidopsis roots, and the disease development requires the ethylene-activated transcription factor EIN3. Furthermore, the beneficial endophytic fungus Piriformospora indica (Pi) significantly reduced Vd-mediated disease development in Arabidopsis. Pi inhibited the growth of Vd in a dual culture on PDA agar plates and pretreatment of Arabidopsis roots with Pi protected plants from Vd infection. The Pi-pretreated plants grew better after Vd infection and the production of Vd microsclerotia was dramatically reduced, all without activating stress hormones and defense genes in the host.CONCLUSIONS:We conclude that Pi is an efficient biocontrol agent that protects Arabidopsis from Vd infection. Our data demonstrate that Vd growth is restricted in the presence of Pi and the additional signals from Pi must participate in the regulation of the immune response against Vd.
Piriformospora indica colonization increases the growth, development, and herbivory resistance of sweet potato (Ipomoea batatas L.)

Abstract:Piriformospora indica symbiosis promoted the growth and photosynthesis, and simultaneously enhanced the resistance against insect herbivory by regulating sporamin-dependent defense in sweet potato. Piriformospora indica (P. indica), a versatile endophytic fungus, promotes the growth and confers resistance against multiple stresses by root colonization in plant hosts. In this study, the effects of P. indica colonization on the growth, physiological change, and herbivore resistance of leaf-vegetable sweet potato cultivar were investigated. P. indica symbiosis significantly improved the biomass in both above- and under-ground parts of sweet potato plants. In comparison with the non-colonized plants, the content of photosynthetic pigments and the efficiency of photosynthesis were increased in P. indica-colonized sweet potato plants. Further investigation showed that the activity of catalase was enhanced in both leaves and roots of sweet potato plants after colonization, but ascorbate peroxidase, peroxidase, and superoxide dismutase were not enhanced. Furthermore, the interaction between P. indica and sweet potato plants also showed the biological function in jasmonic acid (JA)-mediated defense. The plants colonized by P. indica had greatly increased JA accumulation and defense gene expressions, including IbNAC1, IbbHLH3, IbpreproHypSys, and sporamin, leading to elevated trypsin inhibitory activity, which was consistent with a reduced Spodoptera litura performance when larvae fed on the leaves of P. indica-colonized sweet potato plants. The root symbiosis of P. indica is helpful for the plant promoting growth and development and has a strong function as resistance inducers against herbivore attack in sweet potato cultivation by regulating sporamin-dependent defense.
Comparative metabolomic profiling reveals molecular mechanisms underlying growth promotion and disease resistance in wheat conferred by Piriformospora indica in the field

Abstract:Piriformospora indica , a plant root-colonizing basidiomycete fungus, exhibits strong growth-promoting activity in symbiosis with a broad range of plants. Here, we report the potential of P. indica to improve growth, yield, and disease resistance in wheat in the field. In the present study, P. indica successfully colonized wheat through chlamydospores and formed dense mycelial networks that covered roots. Plants subjected to the seed soaking (SS) treatment with P. indica chlamydospore suspensions enhanced tillering 2.28-fold compared to the non-inoculated wheat in the tillering stage. In addition, P. indica colonization promoted vegetative growth significantly during the three-leaf, tillering, and jointing stages. Moreover, the P. indica -SS-treatment enhanced wheat yield by 16.37 ± 1.63%, by increasing grains per ear and panicle weight and decreased damage to wheat shoot and root architecture markedly, with high field control effects against Fusarium pseudograminearum (81.59 ± 1.32%), Bipolaris sorokiniana (82.19 ± 1.59%), and Rhizoctonia cerealis (75.98 ± 1.36%). Most of the primary metabolites, such as amino acids, nucleotides, and lipids, involved in vegetative reproduction were increased in P. indica -SS-treatment plants, whereas secondary metabolites, such as terpenoids, polyketides, and alkaloids, decreased following P. indica inoculation. The up-regulated processes of protein, carbohydrate, and lipid metabolism indicated that P. indica colonization increased growth, yield, and disease resistance via the acceleration of plant primary metabolism. In conclusion, P. indica improved morphological, physiological, and metabolic substance levels, and further promoted its growth, yield, and disease resistance in wheat.

Abstract:Endophytes are considered as plant mutualists which are living asymptomatically within plant tissues have been found in virtually all plant species. Endophytes receive nutrition and protection from the host plant while the host plant may benefit from enhanced competitive abilities and increased resistance to herbivores, pathogens and various abiotic stresses. This review focuses on the biology of endophytic fungi, their discovery, isolation, identification and diversity and their biological activities in environmental and agricultural sustainability. It also considers and their medicinal applications especially in the production of anticancer, antimicrobial, antioxidant and antiviral compounds. Endophytic fungi are one of the most creative groups of secondary metabolite producers that play important biological roles for human life. They are potential sources of novel natural agents for exploitation in the pharmaceutical industry, agriculture and in environmental applications. So, in this review we summarize their potential role against some important plant pathogens.
Piriformospora indica Increases Resistance to Fusarium pseudograminearum in Wheat by Inducing Phenylpropanoid Pathway

Abstract:Fusarium crown rot (FCR), mainly caused by Fusarium pseudograminearum, not only seriously threatens the yield and quality of wheat, but also endangers the health and safety of humans and livestock. Piriformospora indica is a root endophytic fungus that colonizes plant roots extensively and can effectively promote plant growth and improve plant resistance to biotic and abiotic stresses. In this study, the mechanism of FCR resistance mediated by P. indica in wheat was revealed from the phenylpropanoid metabolic pathway. The results showed that the colonization of P. indica significantly reduced the progression of wheat disease, the amount of F. pseudograminearum colonization, and the content of deoxynivalenol (DON) in wheat roots. RNA-seq suggested that P. indica colonization could reduce the number of differentially expressed genes (DEGs) in the transcriptome caused by F. pseudograminearum infection. The DEGs induced by the colonization of P. indica were partially enriched in phenylpropanoid biosynthesis. Transcriptome sequencing and qPCR indicated that the colonization of P. indica up-regulated the expression of genes involved in the phenylpropanoid biosynthesis pathway. The metabolome analysis indicated that the colonization of P. indica increased the metabolites’ accumulation in the phenylpropanoid biosynthesis. Consistent with transcriptome and metabolomic analysis, microscopic observations showed enhanced lignin accumulation in the roots of the Piri and Piri+Fp lines, most likely contributing to the arrested infection by F. pseudograminearum. These results suggested that P. indica increased resistance to F. pseudograminearum in wheat by inducing the phenylpropanoid pathway.

Abstract:Piriformospora indica , the mutualistic biotrophic root colonizing endosymbiotic fungus belonging to the order Sebacinales, offers host plants various benefits and enhances its growth and performance. The effect of colonization of P. indica in Piper nigrum L. cv. Panniyur1 on growth advantages, floral induction and evocation was investigated. Growth and yield benefits are credited to the alteration in the phytohormone levels fine-tuned by plants in response to the fungal colonization and perpetuation. The remarkable upregulation in the phytohormone levels, as estimated by LC- MS/MS and quantified by qRT-PCR, revealed the effectual contribution by the endophyte. qRT-PCR results revealed a significant shift in the expression of putative flowering regulatory genes in the photoperiod induction pathway ( FLOWERING LOCUS T, LEAFY, APETALA1, AGAMOUS, SUPPRESSOR OF CONSTANS 1, GIGANTEA, PHYTOCHROMEA , and CRYPTOCHROME1 ) gibberellin biosynthetic pathway genes ( GIBBERELLIN 20-OXIDASE2, GIBBERELLIN 2-OXIDASE, DELLA PROTEIN REPRESSOR OF GA1-3 1 ) autonomous ( FLOWERING LOCUS C, FLOWERING LOCUS VE, FLOWERING LOCUS CA ), and age pathway ( SQUAMOSA PROMOTER LIKE9, APETALA2 ). The endophytic colonization had no effect on vernalization ( FLOWERING LOCUS C ) or biotic stress pathways ( SALICYLIC ACID INDUCTION DEFICIENT 2, WRKY family transcription factor 22 ). The data suggest that P. nigrum responds positively to P. indica colonization, affecting preponement in floral induction as well as evocation, and thereby shortening the juvenile phase of the crop.

Abstract:Numerous studies have shown that the endophytic fungus Piriformospora indica has a broad range of promoting effects on root development and plant growth in host plants. However, there are currently no reports on the application of this fungus on Cerasus humilis. This study first compared the colonization ability of P. indica on 11 C. humilis varieties and found that the colonization rate of this fungus on these varieties ranged from 90% to 100%, with the colonization rate of the varieties '09-01' and 'Nongda 7' being as high as 100%. Subsequently, the effect of P. indica on root development and plant growth of C. humilis was investigated using cuttings of '09-01' and 'Nongda 7' as materials. P. indica colonization was found to increase the biomass of '09-01' and 'Nongda 7' plants; root activity, POD enzymes, and chlorophyll content were also significantly increased. In addition, indole-3-acetic acid (IAA) content in the roots of C. humilis plants increased after colonization, while jasmonic acid (JA) and 1-aminocyclopropane-1-car- boxylic acid (ACC) content decreased. In conclusion, it has been demonstrated that P. indica can promote the growth of C. humilis plants by accelerating biomass accumulation, promoting rooting, and enhancing the production of photosynthetic pigments, as well as regulating hormone synthesis.
