Abstract:The mutation strain was obtained from Paenibacillus sp. X534 of our lab, and was mutagenized with Ultraviolet+LiCl and Ultraviolet+8-mop, giving XUM-10 strain. The fermentation conditions of XUM-10 strain were also studied. Results showed that the enzyme activity was 221.20U/L, which was 2.5 times higher than parent strain.

Abstract:For improved production of cholesterol oxidase,Paenibacillus sp.X5 was selected as starting strain for mutagenesis.After treatment with ultraviolet ray and γ-ray(60Co),a mutant strain named P.sp.X534 was obtained.Production of enzyme by the strain could reach 300 U/L,which was two times as starting strain.The results indicated that the combined methods were effective on improvement of bacterial enzyme production.

Abstract:Objective: Taking 1444 muscular Candida mycoderma as initial strain,mutated with UV in order to obtain mutants with high lipase production.Methods: After repetition of UV mutation,screening and gene stability assay of mutants, two strains,Z6,Z8 were finally obtained.Results: Their enzyme activity were 22.6,25 U/ml respectively,separately increased by 220% and 250% respectively.The optimal pH and temperature of lipase of Z6 were pH 8.0 and 45℃ respectively. The optimal pH and temperature of lipase of Z8 were pH 8.0 and 50℃ respectively.Activity of lipase of Z6 and Z8 were no much change in 50℃ for 60minutes,and stable from pH 6.0 to pH 9.0.

Abstract:Objective:Breeding of Mucor protease strain with high enzyme activity and good high-temperaturre for fermentation experiments or production of sufu.Method: Mucor protease strains were picked out from sufu,its original strain was induced mutation by UV and fostered by temperature control,breeding of excellent mutant strain.Result:One high enzyme vitality and excellent high-temperature mutant strain named SCLG-mucor 16 was selected,the strain can strongly grow at 32℃.Its HE is 1.39,protease vitality is 180.375u/g,which was enhanced by 7.8% and 51.4% more than the original strain respectively.Conclusion: The excellent mutant strain was used for fermentation experiments or production of sufu, it has a certain using value in shortening the fermentation time,enhancing the quality of sufu and extending the season of production.

Abstract:Screening high yielding mutants of trichodermin production by protplast mutagenesis with resistance of target metabolite trichodermin was developed. Trichoderma taxi wild strain ZJUF0986 was grown in N15 medium for 20 h and mycelium was harvested. Mycelium was treated with mixed enzyme combined with 15 mg/mL driselase, 30 mg/mL helicase and 30 mg/mL lywallzyme at ratio of mycelium : enzyme = 1 : 2.5 (w/v), the optimal condition of protoplast release was at 30 ℃, 100 r/min for 90 min and protoplast production reached 9.42×10~8/mL. Protoplast of ZJUF0986 was mutated with lithium chloride solution and UV. The mutation rate and positive mutation rate were the highest on 60 s UV radiation dose. A high yield mutant strain UL60-11 was screened, which increased 67.3% of fermentation efficiency than wild strain ZJUF0986.

Abstract:Lactobacillus plantarum was treated with UV, the exhibited good stability obtained by a screening to tow mutants with cholesterol gradient plate and Cholesterol-degrading testing in vitro. Among which the rate of Cholesterol-degrading of Lp-UVs 29 and Lp-UVs 44 are respective 48.7% and 44.2%, which raise respectively by 97.97% and 79.67%.

Abstract:In order to obtain mutants with high lipase production, the Micrococcus SX-1 was taken as initial strain, which enzyme activity was 10 U/ml, and was treated with ultraviolet(UV) radiation. And the optimum treating conditions which was got by primary screening were as follows: the 15 W UV lamp was used as UV light with 30 cm radiation distance and 90 s treating time. Through UV mutation a mutant strain named SU-1 was obtained. As compared with the initial strain, the activity of lipase had reached 22.5 U/mL , which was 125% higher than that of the initial strain. Then an experiment for studying the genetic stability was took by means of passaging 5 times and its results showed that Micrococcus SU-1 had a better genetic stability.

Abstract:Abstract To obtain alkaline protease producing strain with high-yield, the strain Bacillus subtilis was treated by atmospheric and room temperature plasma (ARTP) mutagenesis. The results showed that the strain had a higher positive mutation rate when the mutagenesis time was 50s. Finally, a high-yield mutant strain A59 was obtained after repeated mutagenesis by ARTP and enzyme activity increased by 23.38% from 6835U/mL to 8433U/mL. Then fermentation conditions of mutant strain were optimized by single-factor method and response surface method, and the optimum fermentation conditions were as follows: 3.6% soybean meal, 5.2% corn meal, 0.9115% Na 2 HPO 4 , 0.1593% Mg 2+ , 0.09% Tween-80 and 5% inoculation volume. Under the conditions of the verification experiment, the enzyme activity of the A59 strain reached 14026 U/mL, which was 2.05 times that of the original strain.

Abstract:In order to screen out the excellent lactic acid bacteria strain used for producing ferment dairy,ultraviolet(UV) and nitrosoguanidine(NTG) were both employed to mutate the lactobacillus bulgaricus B-3.The results showed that a new high acid-producing strain with acid-producing 82.65 °T was screen out.The acid-producing of the new strain was improved 42.92% compared with the original strain B-3 with acid-producing 57.83 °T after radiation with UV for 120 s and mutation with NTG(0.3 g/L) for 60 min for three times.

Abstract:The unspecific peroxygenase (UPO) from Cyclocybe aegerita (AaeUPO) can selectively oxidize C–H bonds using hydrogen peroxide as an oxygen donor without cofactors, which has drawn significant industrial attention. Many studies have made efforts to enhance the overall activity of AaeUPO expressed in Komagataella phaffii by employing strategies such as enzyme-directed evolution, utilizing appropriate promoters, and screening secretion peptides. Building upon these previous studies, the objective of this study was to further enhance the expression of a mutant of AaeUPO with improved activity (PaDa-I) by increasing the gene copy number, co-expressing chaperones, and optimizing culture conditions. Our results demonstrated that a strain carrying approximately three copies of expression cassettes and co-expressing the protein disulfide isomerase showed an approximately 10.7-fold increase in volumetric enzyme activity, using the 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) as the substrate. After optimizing the culture conditions, the volumetric enzyme activity of this strain further increased by approximately 48.7%, reaching 117.3 U/mL. Additionally, the purified catalytic domain of PaDa-I displayed regioselective hydroxylation of R-2-phenoxypropionic acid. The results of this study may facilitate the industrial application of UPOs.

Breeding High Astaxanthin-Producing Superior Yeast Strains by Synergetic Mutation

Abstract:Phaffia rhodozyma (Pr) (wild strain GH1) and their growth characteristics were studied preliminarily in order to breed high astaxanthin-producing and superior strains from.Pr (wild strain GH1) was mutated with of UV and NTG synergetically and screened with antimycin A and obtained a high astaxanthin-producing mutant strain GH1-7.1.The results showed that the its yield of astaxanthin was as high as 1 200 μg/g DCW four times higher than that of wild strain GH1(271 μg/g DCW);furthermore it grew faster,24 hours shorter to reach the highest biomass and yield of astaxanthin than the GH1.In addition,the wild strain GH1 could only grew at 20 ℃,and stopped growing at over 28 ℃,while GH1-7.1 had optimal temperature at 28～30 ℃,8～10 ℃ higher than GH1.It was for the first time to use synergetic mutation successfully to breed high astaxanthin-producing yeast strains characterized by shorter growing cycle and higher optimal temperature.It is a superior strain suitable for fermentation engineering application in high temperature environment such as South China.

Abstract:Schizochytrium limacinum was mutated with ultraviolet and two elite strains,OUC002 and OUC007,were screened out with quizalofop-p-ethyl.Compared with original strain,the biomass and DHA content of OUC002 were increased by 8.08% and 13.74%,respectively,and the biomass and DHA content of OUC007 were increased by 11.57% and 28.75%,respectively.As determined with GC-MS,DHA content of OUC007 reached 37.28% of total fatty acids.These mutants promise to be used in S.limacinum fermentation.The method used in this study was effective for breeding new strains of S.limacinum.

Abstract:By compound mutation treatment with ultraviolet(UV), nitrosoguanidine(NTG) and laser irradiation to the initial strain(alkaliphilic bacillus)BC-A36,the mutant JG-57 with high alkaline amylase activity was obtained.In the shaking flask culture with fermentation medium at 35～(37 ℃) for 45 hours its enzyme activity reached 867 u/mL,which was 2.1 times greater than that of the initial strain.Enzyme synthesis occurred at temperatures between 25～43 ℃ with an optimum of 37 ℃.There was a variation in amylase synthesis within the pH range 7～12 with an optimum pH of 9.The alkaline amylase produced by the mutant JG-57 had strong adaptability to high pH of 9.5～10.The results showed that the mutant was genetically stable.It proves that laser irradiation is an effective method for breeding of alkaline amylase producing strain.The JG-57 alkaline amylase is suitable in liquefaction of starch,detergent and textile industries and other industrial applications.

Abstract:As a original strain, Streptomyces spp. Xinong No.24 was mutated by UV, chemicals, microwave irradiation and ultrasonic. A high producing antibiotic strain named Ms 24 was obtained through the microwave irradiating 50 s. Successively cultured ten times in the medium, heredity character of Ms-24 was stable.The result of fermentation test showed that the fermentation yield increased by 23.25%,The result of antifungal activity test indicated that the activity increased by 31.75%.
Obtaining Efficient Mutant from the Wild Type Bacillus subtilis E6-5 by Physical and Chemical Mutagenesis for High Efficiency Protease Production, Optimizing the Mutant's Culture Medium

Abstract:In this study, to enhance protease production, the wild type of Bacillus subtilis E6-5 was mutagenized by random mutagenesis using ultraviolet radiation and ethidium bromide. After combined treatment, several mutants were obtained. Among these mutants, the mutant strain with the largest proteolytic zone diameter (25 mm) was selected and named Bacillus subtilis ATA38. The enzyme production capacity of the obtained mutant was tested and the mutant strain (404 IU/mL at 24 hours) produced 6.7 times more enzyme than the parental strain (60 IU/mL at 32 hours). The effects of some important parameters in the growth medium on enzyme production were examined. The best carbon, organic nitrogen and metal ion were obtained with wheat starch (525 IU/mL), meat extract (850 IU/mL) and KCl+CaCl2 (548 IU/mL), respectively. pH 6.0, 37°C, 200 rpm, inoculum age 18 hours and inoculation amount 1% were obtained as the best physical factors.To further increase the yield, the best nutritional and physical parameters were combined to create a new modified medium. It was determined that the enzyme yield with mutant strain increased 2.7 times in the modified medium (1096 IU/mL) compared to the control (404 U/mL). The mutant strain (1096 IU/mL) showed an 18.2-fold increase in production compared to the wild type (60 IU/mL) in the modified medium. Protease enzyme obtained from ATA38 mutant strain may have great potential in industry for different purposes.

Abstract:[Objective] The aim was to screen the high yield trichodermin strain and increase the yield of trichodermin.[Method] With Trichoderma harzianum,an endophytic fungi producing trichodermin separated from Llex cornuta,as the original strain for the treatment of ultraviolet radiation compound mutation for two times,the 5 generations of the screened mutation strains were transferred continuously and its genetic stability was detected.[Result] The lethality rate of T.harzianum was increased with the prolonging of the irradiation time.The ultraviolet irradiation of 45 s with the lethality rate of 88.1% was selected as the optimum mutation dose for mutation breeding.The sporulation time of the strain through twice mutation was advanced and the growing strain was dense.A high yield mutation strain UV-5-3 was obtained at last through the primary screening and the secondary screening and its antibiotic level was the highest,being 164.75 μg/ml,which was 56.77% higher than that of UV-3-1 mutation strain obtained through the primary screening,and the 2.3 times of the original strain.The passage experiment showed that the high yield preference of the mutation strain UV-5-3 had stable genetic characteristic.[Conclusion] The high yield trichodermin strain could be obtained by treating T.harzianum through ultraviolet radiation compound mutation treatment for two times.

Abstract:The primitive strain Aspergillus niger AS-Y was used as startup strain to produce strain of higher amylase producing capability by UV mutation, EMS mutation and complex mutation. Results showed that using UV and EMS complex mutation could produce AS-EUⅢ stain. The amylase activity of AS-EUⅢ was increased from 64.3 U/g to 198.8 U/g.
Mutation Breeding of Monascus Purpureus with High Productivity of Orange Pigments by Microwave Irradiation

Abstract:Microwave irradiation with water-cooling device was used for the mutagenesis of Monascus purpureus. The optimal microwave power, irradiation duration and dosage for the mutagenesis of Monascus purpureus were explored by evaluating the lethal and mutation regularity of Monascus purpureus. Results indicated that the optimal mutation conditions were microwave power of 500 W and irradiation duration of 80 s. Under the optimal microwave irradiation conditions, a mutated strain W5S8 with the highest productivity of orange pigments was achieved and the color value of orange pigments from strain W5S8 was 16.38 U/mL, which was increased by 59.62% compared with the parent strain. Its productivity for orange pigments remained stable after subculture for 5 generations or storage.

Abstract:Rhodopseudomonas sphaeroides 1.1737T was used as an original strain in this study R.sphaeroides 1.1737T was mutagenized by UV andnitrosoguanidine(NTG) treatment.Then,a high coenzyme Q10-producting mutant R.sphaeroides PHBr-2 was screened according to the tolerance onroxithromycin and precursor p-hydroxyben-zoic acid(PHB).Using R.sphaeroides PHBr-2 for fermentation,the biomass reached 15.7 g/L at 108 h,and the coenzyme Q10 yield was 50.6 mg/L(90.9% higher than that of the parent strain).R.sphaeroides PHBr-2 was genetic stable and produced49.5 mg/L coenzyme Q10 after five-generation subculture.It was indicated that R.sphaeroides PHBr-2 was a good candidate for coenzyme Q10production.

Abstract:Pyrroloquinoline quinone (PQQ) is a natural antioxidant with diverse applications in food and pharmaceutical industries. A lot of effort has been devoted toward the discovery of PQQ high-producing microbial species and characterization of biosynthesis, but it is still challenging to achieve a high PQQ yield. In this study, a combined strategy of random mutagenesis and adaptive laboratory evolution (ALE) with fermentation optimization was applied to improve PQQ production in Hyphomicrobium denitrificans H4-45. A mutant strain AE-9 was obtained after nearly 400 generations of UV-LiCl mutagenesis, followed by an ALE process, which was conducted with a consecutive increase of oxidative stress generated by kanamycin, sodium sulfide, and potassium tellurite. In the flask culture condition, the PQQ production in mutant strain AE-9 had an 80.4% increase, and the cell density increased by 14.9% when compared with that of the initial strain H4-45. Moreover, batch and fed-batch fermentation processes were optimized to further improve PQQ production by pH control strategy, methanol and H 2 O 2 feed flow, and segmented fermentation process. Finally, the highest PQQ production and productivity of the mutant strain AE-9 reached 307 mg/L and 4.26 mg/L/h in a 3.7-L bioreactor, respectively. Whole genome sequencing analysis showed that genetic mutations in the ftfL gene and thiC gene might contribute to improving PQQ production by enhancing methanol consumption and cell growth in the AE-9 strain. Our study provided a systematic strategy to obtain a PQQ high-producing mutant strain and achieve high production of PQQ in fermentation. These practical methods could be applicable to improve the production of other antioxidant compounds with uncleared regulation mechanisms.
