Growth Inhibition Effect of Sodium Butyrate on Human Endometrial Cancer Cells in Vivo

Abstract:OBJECTIVE:To investigate the growth inhibition of sodium butyrate on human endometrial cancer and to gain an insight into the possible underlying pathways.METHODS:Human endometrial cancer cell line (HHUA) was seeded in the subcutaneous and implanted in a xenograft model by using nude mice. Then they were randomly divided into two groups and treated with sodium butyrate (for experimental groups) and PBS (for control groups) for up to 4-6 weeks. Tumor size was measured in 2 dimensions at regular time-intervals and tumors, hearts, livers and kidneys were harvested at the end of treatment. Tumor sections were examined by HE staining, electron microscope and TUNEL method. Heart sections, liver sections and kidney sections were examined by HE staining for assessment of toxicity.RESULTS:All the nude mice grew in a good condition. There were no abnormal changes in heart sections, liver sections and kidney sections examined under a light microscope and no toxic reactions during treatment. Tumors decreased in size during sodium butyrate-treatment, but increased in size in the control groups. There were a lot of cell underwent apoptosis in the sodium butyrate-treated groups. TUNEL method demonstrated that apoptotic rate in the sodium butyrate-treated groups was significantly higher than that in the untreated control groups,P0.01. CONCLUSIONS:NaB can significantly inhibit the tumor growth in nude mice bearing HHUA endometrial cancer cells. Its mechanism may be closely related to inducing apoptosis in tumor cells. There are no toxicity to heart, liver and kidney at a treatment dose.
