Pharmacokinetics of Berberine in Healthy Volunteers Studied by LC-MS

Abstract:Objective:To study the pharmacokinetics of berberine in healthy Chinese volunteers after oral dose of berberine hydrochloride tablets.Methods:An LC-MS method for the determination of berberine in human plasma using chlorobenzylidine as the internal standard (IS) has been developed.The berberine concentrations in plasma of 10 healthy male volunteers were determined and the pharmacokinetic parameters were calculated.Results:The mean pharmacokinetic parameters of 10 healthy male volunteers after a single oral dose of 0.4 g berberine hydrochloride tablets were as follows:t1/2=(35.5±11.8) h,tmax=(8.50±4.60) h,Cmax=(0.30±0.15) μg·L-1,AUC0-96h=(6.68±2.56) h·μg·L-1,AUC0-∞=(8.05±2.87) h·μg·L-1.Conclusions:The method was sensitive and accurate.The validated method has been successfully applied to study the pharmacokinetics of berberine in healthy volunteers.The t1/2 of berberine is longer than 30 h.So in order to ensure the safety of clinical use,it is worth going a step further to investige whether berberine would be accumulated if the administration tid.were for a long time.
