Abstract:RATIONALE: The fragmentation behavior of seven secoiridoid glycosides (SIGs) isolated from Gentiana triflora Pall., a traditional Chinese medicine, was investigated by electrospray ionization ion trap mass spectrometry (ESI-IT-MS). METHODS: Multistage electrospray ionization mass spectrometry (ESI-MSn) experiments both in positive-and negative-ion mode were used to elucidate the main fragmentation pathways of these compounds. RESULTS: In full scan mass spectra, different deprotonated or protonated molecules were observed for compounds with different basic structure. Ring cleavages of the aglycon moiety were observed in MS2 spectra and the characteristic ions provided valuable information on the basic structural skeletons. Their fragmentation pathways and fragment ion structures were proposed. CONCLUSIONS: The established fragmentation patterns have been successfully used to identify eleven SIGs in the extract of Gentiana triflora Pall. Copyright (C) 2014 John Wiley & Sons, Ltd.

Abstract:A systematic study of the fragmentation pattern of N-diisopropyloxyphosphoryl (DIPP) dipeptide methyl esters in an electrospray ionization (ESI) tandem mass spectrometry (MS/MS) was presented. A combination of accurate mass measurement and tandem mass spectrometry had been used to characterize the major fragment ions observed in the ESI mass spectrum. It was found that the alkali metal ions acted as a fixed charge site and expelled the DIPP group after transferring a proton to the amide nitrogen. For all the N-phosphoryl dipeptide methyl esters, under the activation of a metal ion, the rearrangement product ion at m/z 163 was observed and confirmed to be the sodium adduct of phosphoric acid mono-isopropyl esters (PAIE), via a specific five-membered penta-co-ordinated phosphorus intermediate. However, no rearrangement ion was observed when a beta-amino acid was at the N-terminal. This could be used to develop a novel method for differentiating isomeric compounds when either alpha- or beta-amino acid are at the N-terminus of peptides. From the [M+Na]+ ESI-MS/MS spectra of N-phosphoryl dipeptide methyl esters (DIPP Xaa1 Xaa2 OMe), the peaks corresponding to the [M+Na Xaa1 C3H6]+ were observed and explained. The [M+Na]+ ESI-MS/MS spectra of N-phosphoryl dipeptide methyl esters with Phe located in the C-terminal, such as DIPPValPheOMe, DIPPLeuPheOMe, DIPPIlePheOMe, DIPPAlaPheOMe and DIPPPhePheOMe, had characteristic fragmentation. Two unusual gas-phase intramolecular rearrangement mechanisms were first proposed for this fragmentation. These rearrangements were not observed in dipeptide methyl ester analogs which did not contain the DIPP at the N-terminal, suggesting that this moiety was critical for the rearrangement.

Abstract:Using the condensation of N-protected amino acids with 2,6-bishydroxylmethyl pyridine, we synthesized six di-esters and four mono-esters. All ten esters are new compounds. They are all important intermediate compounds for synthesis of macrocylic compounds containing amino acid and pyridyl units. Because there are several complex sites in these macrocycles, they may be useful as enzyme models or as templates in asymmetric resolution. This paper reports fast-atom bombardment (FAB) mass spectrometry of these two kinds of esterification products, and electron impact (EI) of four mono-esters. Their FAB mass spectrometric behavior was studied with the aid of both mass-analyzed ion kinetic energy spectrometry and high-resolution data. The EI fragmentation mechanisms of the mono-esters are also studied with the aid of mass-analyzed ion kinetic energy spectrometry and high-resolution data. We discovered that, under FAB conditions, each mono-ester yields (2M+H](+) ions, and the ionizing proton is connected with the nitrogen atom in the pyridine ring. Through the high-resolution analysis of compound 10 under EI, we found two kinds of ions at m/z 139. One is a 2,6-bishydroxylmethyl pyridine residue, which is more stable than the other residue formed by the loss of one oxygen atom from p-toluenesulfonyl ion. Also of importance is the fact that a H-atom rearrangement proceeds via a four-membered ring intermediate under both FAB and EI conditions. (C) 1998 John Wiley & Sons, Ltd.

Abstract:A series of cyclic esters, which are optically active as a consequence of their helical structures, were synthesized to investigate the relationships between their structures and their optical activities. This paper reports the electron impact fragmentation mechanisms of these six cyclic esters. Accurate mass measurements and mass analyzed ion kinetic energy spectrometry confirmed fragmentation patterns. The stability of the fragment ions has a great influence on the fragmentation pathways, but no correlation with the optical activity was found.

Abstract:AbstractA series of different types of wax esters (represented by RCOOR′) were systematically studied by using electrospray ionization (ESI) collision‐induced dissociation tandem mass spectrometry (MS/MS) along with pseudo MS3 (in‐source dissociation combined with MS/MS) on a quadrupole time‐of‐flight (Q‐TOF) mass spectrometer. The tandem mass spectra patterns resulting from dissociation of ammonium/proton adducts of these wax esters were influenced by the wax ester type and the collision energy applied. The product ions [RCOOH2]+, [RCO]+ and [RCO−H2O]+ that have been reported previously were detected; however, different primary product ions were demonstrated for the three wax ester types including: (1) [RCOOH2]+ for saturated wax esters, (2) [RCOOH2]+, [RCO]+ and [RCO−H2O]+ for unsaturated wax esters containing only one double bond in the fatty acid moiety or with one additional double bond in the fatty alcohol moiety, and (3) [RCOOH2]+ and [RCO]+ for unsaturated wax esters containing a double bond in the fatty alcohol moiety alone. Other fragments included [R′]+ and several series of product ions for all types of wax esters. Interestingly, unusual product ions were detected, such as neutral molecule (including water, methanol and ammonia) adducts of [RCOOH2]+ ions for all types of wax esters and [R′−2H]+ ions for unsaturated fatty acyl‐containing wax esters. The patterns of tandem mass spectra for different types of wax esters will inform future identification and quantification approaches of wax esters in biological samples as supported by a preliminary study of quantification of isomeric wax esters in human meibomian gland secretions.

Abstract:The fragmentation pathways of lithiated alpha,beta-unsaturated thioesters with different substituents were investigated by electrospray ionization tandem mass spectrometry (ESI-MS/MS) in positive ion mode. In mass spectrometry of the alpha,beta-unsaturated thioesters, Ar-CH=CH-CO-S-Ph, loss of PhSLi and elimination of a thiophenol were the two major fragmentation reactions of the lithiated molecules. The elemental compositions of all the ions were confirmed by high-resolution Fourier transform ion cyclotron resonance tandem mass spectrometry (FTICR-MS/MS). The thioesters studied here were para-monosubstituted on the phenyl ring of cinnamoyl and the electron-withdrawing groups favored loss of a thiophenol, whereas the electron-releasing groups strongly favored the competing reaction leading to the loss of PhSLi to form a cinnamoyl cation, Ar-CH=CHCO(+). The intensity ratios of the two competitive product ions were well correlated with the sigma(p) (+) substituent constants. The mechanisms of these two competing routes were further investigated by density functional theory (DFT) calculations.

Abstract:Ingenol esters, one type of diterpenoids isolated from genus Euphorbia, were reported to possess cytotoxic activities. In our previous study, a number of ingenol esters were isolated from Euphorbia esula L. (Ru-Jiang-Da-Ji in Chinese). And as a part of the ongoing program, a study of fragmentation pathway of ingenol esters was carried out using mass spectrometry. The fragmentation behaviors of 17 standard compounds were investigated with ion trap (IT-MSn) mass spectrometry and hybrid quadrupole-time of flight-mass spectrometer (Q-TOF-MS/MS) in positive ion mode. Electrospray ionization (ESI) was used as ion source in analysis. According to the IT-MSn spectra, the elimination order of the acyloxy was R1OH>R2H>R3H>R4H, which was hard to determine in Q-TOF-MS/MS spectra. The Q-TOF-MS preferred to eliminate all the acyloxys along with some hydroxyls and produce three characteristic ions (x0, y0, z0), which indicate the numbers of the oxygenated groups. The Q-TOF-MS also provide high resolution mass spectra of the parent and product ions, which allowed the determination of the chemical formulas and characterization of the ion structures. These two instruments gave complementary information, which were very useful for structural characterization of the studied compounds.
Studies on the Optical Stability of Chiral Compounds in Ei Mass Spectrometry Process

Abstract:The optical stability of chiral 2-hydroxy-2-phenylacetic acid in electron ionization mass spectrometry(EIMS) process has been detected directly by deuterium labelling technique. From the EI mass spectrum of deuterated 2-hydroxy-2-phenylacetic acid, the major characteristic fragment ion at mit 108 (the capture of the carbonyl group) and the corresponding isotopic ion (C-13) at m/z 109 can be observed, and the ratio of m/z = 109 to m/z = 108 is about 8% (the calculated value is 7.8%). As enolization of deuterated 2-hydroxy-2-phenylacetic acid in mass spectrometry can yield the characteristic fragment ion at m/z 109 and results in much higher ratio of 109/108 than 8%; this study shows that no enolization takes place during the ET-MS process of 2-hydroxy-2-phenylacetic acid. This results can be concluded that these compounds are optically stable in the process of instantaneous vaporization at high temperature and electron impact ionization.

Abstract:The fragmentation patterns of a novel kind of dithiocarbamic acid esters with excellent anticancer activity were analyzed by positive ion electrospray ionization mass spectrometry in conjunction with tandem mass spectrometry (ESI/MSn). The fragmentation patterns of sodium adduct ions [M+Na]+ were characterized with elimination of hydrogen cyanide and most of their counterparts could be observed. The fragmentation patterns of protonated molecular ions [M+H]+ were characterized with single bond cleavage between thiocarbonyl group and nitrogen atom, and between thiocarbonyl group and sulfur atom. The piperazine moiety of the molecular of [M+H]+ favors a rearrangement to expel azirane, and the suggested rearrangement mechanism is consistent with experimental observations.

Abstract:In this study, we developed an AC-induced interfacial electro-epoxidation reaction for localization of double-bonds in nega-tively and positively charged forms of lipids simultaneously. An AC voltage plays multiple roles in this method, including initiation of the electrochemical epoxidation of double-bonds in both charge states of lipids, and protonation and deprotona-tion of lipid for detection in both ion modes. The advantages of simultaneous detection of negatively changed and positively charged unsaturated lipids, voltage-controlled electrochemical derivatization, and the low sample consumption allow its wide applications in a broad range of lipid-related research.

Abstract:The electron impact mass spectra of several cyclic esters with helical structures have been studied. Their fragmentation pathways were proposed and confirmed by mass-analyzed ion kinetic energy (MIKE) and high-resolution data. In general, the dominant fragmentation pathways in the spectra of these compounds originate from a alpha-cleavage with loss of a hydrogen or methyl group. The difference between hydrogen and methyl group loss greatly affects the subsequent fragmentations. Although, due to their helicity, these cyclic esters are optically active no stereo-related fragmentation pathway was observed. Copyright 2000 John Wiley & Sons, Ltd.

Abstract:Reported here is the first on-demand electrochemical epoxidation incorporated into the standard nano-electrospray ionization mass spectrometry (nanoESI-MS) workflow for double-bond identification. The capability lies in a novel tunable electro-epoxidation of double bonds, where onset of the reaction can be controlled by simply tuning the spray voltage. On-demand formation of mono-/multiple epoxides is achieved at different voltages. The electro-epoxidized products are then fragmented by tandem MS to generate diagnostic ions, indicating the double bond position(s). The process is completed within seconds, holding great potential for high-throughput analysis. The rapid switch-on/off electro-epoxidation of a single sample, the low sample consumption, the demonstrated applicability to complex lipids containing multiple double bonds, and the advantage of not requiring extra apparatus make this method attractive for use in lipid-related biological studies.

Abstract:The negative-ions of N-phosphoryl amino acids were studied by electrospray ionization tandem mass spectrometry (ESI-MS/MS). The negative-ion ESI-MS/MS of N-phosphoryl amino acids showed characteristic fragmentation patterns different from those observed in the corresponding positive-ion ESI-MS/MS and negative-ion fast-atom bombardment mass spectra. For negative-ion EST-MS/MS, a unique fragmentation from the N-terminal of N-phosphoryl amino acids or peptides containing a free beta-OH or CO2H group was observed to yield the characteristic fragment ion (RO)(2)P(O)O. The ease of the rearrangement depended on the position of the hydroxyl group in amino acids or peptides, and the N --> O rearrangement mechanism was proposed to involve the participation of the hydroxyl group. From previous solution-phase experiments and theoretical calculations, it was found that the beta-OH group was more active than gamma-OH, and the corresponding difference in negative-ion ESI-MS/MS was consistent with those previous findings. Abstract:The mass spectrometric behavior of eleven 1-(N-benzyloxycarbonyl(Cbz)amino)alkylphosphonate diesters was studied under positive ion electrospray ionization (ESI) conditions. Their fragmentation pathways are depicted and supported by tandem mass spectrometry. Besides the common eliminations of ether, benzyl alcohol, phosphite and an ether plus benzyl alcohol from molecular ions, the title compounds show a tendency to undergo an interesting intramolecular benzyl rearrangement to yield benzylphosphonate ions. The fragmentation patterns do not depend on the substituent attached to the alpha-carbon atom.

Abstract:The mechanism of N, N-dimethylglycine ethyl ester pyrolysis is explored by means of ab initio calculations at the MP2/6-31 + G* level in the gas phase. The synchronicities have been calculated by using Pauling bond orders. The results show that the reaction is a stepwise process. The first process is the decomposition of N, N-dimethylglycine ethyl ester to form N, N-dimethylglycine and ethylene, while the second process is the decomposition of N, N-dimethylglycine to form trimethylamine and carbon dioxide. The first process is the rate-controlling step. The reaction mechanism is an asynchronous proton transfer process. The calculated results are in good agreement with the experimental results.

Abstract:AM1,PM3 semiempirical methods and Hartree Fock ab initio method of molecular orbital theory have been used to obtain the molecular greometry of N,Nbis(8 hydroxy 5 quinolinemethyl) glycine ethylester, which has been synthesized recently, by energy gradient completed optimization. The electronic structure and some basic molecular properties such as heat of formation, dipole moment are also calculated. These results are discussed relating the classical organic electronic theory. The geometry obtained with these methods is consistent with each other and in good accordance with the experimental structure. Normal mode vibrational frequency analyse has also been performed on the optimized geometry with AM1 method. The frequencies of C C double bond, C N single and double bonds, C O single and dluble bonds and hydroxyl are derived and in agreement with the characteristic peaks of experimental infrared spectra very well.

Abstract:In this research, the characteristic ions' abundance ratio between two isomers A and B in MS/MS mass spectra was defined as a parameter for discriminating diastereomers. Through this ratio, the discrimination of four pairs of cyclic peptide (CP) diastereomers was successfully achieved. Furthermore, in the analysis of diastereomers' mixtures, both calibration curve and calculational methods were substantiated to have high precision and accuracy. The average absolute errors of the two methods were 2.0 and 2.5% in the 48 measurements of 16 samples, respectively. This research provided a promising approach for the analysis of the CP diastereomers in the fields of asymmetrical synthesis, chiral natural products and structural biology by ESI-MS/MS. Electron ionization fragmentation studies for a series of 4-methoxymethylene benzoate esters

Abstract:Rationale: Product ion studies and stable isotope deuterium labeling experiments provide useful data for understanding the electron ionization (EI)-mass spectroscopy (MS) fragmentation of methoxymethylene substituted benzoate esters. The methoxymethylene ether is regioisomeric with the ethoxy group and represents the two possible ether substituents of a benzene ring of C2 H5 O. Structural confirmation of these synthetic precursor materials via gas chromatography (GC)-EI-MS revealed unexpected fragment ions. The synthesis and EI-MS evaluation of some homologs and deuterated derivatives allowed for the characterization of these unique ions and their fragmentation pathways. The relative effects of the position of the oxygen of the ether side chain are the subject of this investigation. Methods: The desired compounds were prepared from 4-chloromethylbenzoyl chloride by alkoxide displacement followed by transesterifications and the deuterated analogs were prepared similarly. The compounds were separated by capillary GC and their MS fragmentation evaluated in EI, MS/MS and chemical ionization experiments. Results: The methoxymethylene-substituted benzoate esters yield major fragment ions from the loss of the alkyl group from the ether as well as alkoxy group loss from the ester or ether portion of the molecule. The loss of the alkyl group from the ether followed by loss of the ester alkoxy group as the corresponding alcohol yielded the unique cation at m/z 133 for all compounds. The identity of the major ions was confirmed by product ion and deuterium labeling studies and possible mechanisms of fragment ion formation are described. Conclusions: The aliphatic oxygen of the alkoxymethylene group plays a much more active role in the EI-MS fragment formation profile than the direct aromatic ring linked oxygen of the ethoxy group. Thus, yielding a greater variety of characteristic fragments. The m/z 133 ion is unique to this class of compounds and does not have an equivalent pathway for the regioisomeric ethoxy series.
