In Vitro Study of the Effects of Wild-Type PIEN Gene Expression on the Biological Behavior of MBA-MB-468 Inflammatory Breast Cancer Cell Line

Abstract:[Objective] To study in vitro effects of wild-type PTEN gene expression on the biological behavior of a human inflammatory breast cancer cell line MDA-MB-468. [Methods] The recombinant eukaryotic expression plasmid pcDNA3.1-PTEN was constructed by use of recombinant DNA technique. They were transfected into an inflammatory breast cancer cell line MDA-MB-468 using lipofectamine 2000. Positive clones were selected by G418, target genes had expressed by RT-PCR, immunohistochemical method and Western blotting. Then we also detect the expression of phosphospecific protein kinase B (PKB/Akt) protein stimulated by epidermal growth factor (EGF), cell viability was determined by MTT assay. The apoptosis of the MDA-MB-468 cells was determined by flow cytometry with the double-staining method using FITC-conjugated Annexin V and propidium iodide. [Results] The recombinant eukaryotic expression plasmid for PTEN was digested with XbaI and BamHI, the electrophore sis of the digested products showed two fragments: 1.4 kb fragment and 5.4 kb fragment. The transfected PTEN gene MDA-MB-468 cells showed expression of PTEN mRNA and PTEN protein by RT-PCR, immunohistochemical methods and western blotting, PTEN decreased the phosphorylation level of AKT protein in MDA-MB-468 stimulated by EGF, there were significant declined between the cell viability of transfected PTEN cDNA and non-transfected PTEN cDNA in MTT assay (P<0.01). Flow cytometric analyses indicated that the expression of PTEN protein caused an increase in the rate of apoptosis when compared with other groups' cells (P<0.01). [Conclusion] The recombinant eukaryotic expression plasmid pcDNA3.1-PTEN was constructed corrected. Results of these studies provide evidence that the inhibitory effect of wild-type PTEN gene on breast cancer cell proliferation is mainly induced cell apoptosis by phosphatase activity-dependent.
