Abstract:In this research the atomic absorption spectroscopy method (the correlation coefficient was 0.9995) was employed for determination of the content of heavy metal-manganese in panax pseudo-ginseng var. notoginseng, which was chosen as the sample. The optimum conditions of two pretreatments were researched independently. In the dry ashing, the optimized temperature and time are 600°C and 4~6 hours respectively, and the manganese content determined is 53.16 ug/g and the recovery rate of manganese added in sample is 98.64%. In the wet digestion, The HNO3-H2O2 digestion system is the best one, and the content was 52.95ug/g and the recovery rate is 97.30%. So the two pretreatment methods of the sample are accurate enough on manganese content determination. But the dry ashing is better than the wet digestion relatively.

Abstract:To investigate the amount of metal element in the citrus juices flame atomic absorption spectrophotometry (FAAS).Was used to determine the content of K,Mg,Ca,Fe,Cu,Mn and Zn in the selected fifteen citrus juices of China after the digestion with V(HNO_3)+V(HClO_4)=4+1.The results indicated that citrus juices were abundant in major elements (K,Mg and Ca),and there were large variations among different varieties.Among the trace elements,Fe and Zn had relatively higher amounts.Meanwhile,Hybrid 439 achieved the highest value of K,Ca and Mn among the selected fifteen citrus fruits.These results were useful to the nutritional label of citrus juices and also valuable to the citrus juice processing technology.

Abstract:In This paper,the contents of metal elements in three kinds of desert plants Salix bordensis,Elaeagnus angustifolia and Salix psammophila from Inner Mongolia were determined by ICP-AES technique.The recoveries obtained by standard addition method is between 95.1% and 110.3%,and its RSD is lower than 3.4%.The results show that major elements such as Na,K,Mg,Ca,Al and plant vital activities essential trace elements Cu,Fe,Mn,Zn have a different contents orders in these desert plants,and 4 elements contents of Cu,Fe,Mn,Zn are lower than the average contents of other terrestrial plants.

Abstract:To ensure the quality and safety of herbs, the content of 54 elements in MOUTAN CORTEX (MC) was determined by the ICP-AES and ICP-MS, and the health risks of Cu, As, Cd, Pb, Hg and rare earth elements (REEs) were assessed. These herbs were collected from 5 producing areas in Anhui Province, China, namely Wuhu, Tongling, Bozhou, Xuancheng and Chizhou. The multi-elements fingerprint identification of MC in Anhui Province was established. The total amount of macro-elements from Wuhu and Tongling is significantly lower than Bozhou. Among all MC from 5 producing areas, the highest content is Ca. Except for Bozhou, the content of macro-elements and REES in the other 4 origins of MC is from highest to lowest: Ca > K > Mg > Al > Fe > Na and Ce > La > Nd > Y > Pr > Er > Yb > Eu > Ho > Tb > Tm > Lu. The chemical forms of Cd in MC from Bozhou with the highest percentage were P H2O of high toxicity and migration, while the other 4 regions were P NaCl of low activity and mobility. There was a great difference in the content of inorganic elements and chemical forms of Cd between the MC produced from the plain (Bozhou) and the hilly areas (Wuhu, Tongling, Chizhou and Xuancheng). Except for Cd, the content of Cu, As, Pb and Hg in MC did not exceed the limit. The results of PTWI Fact and ADI for Cd and REEs showed that MC herbs did not pose a risk to human health.

Abstract:This study assessed the concentrations of five heavy metals Pb, Cd, Cr, Cu and Zn in both soil and a traditional Chinese medicine Cortex Moutan collected from 6 typical sites, growing this medicine, in China. The potential health risks associated with those heavy metals were also assessed. It is shown that Cd content in soils from five of the six sites was higher than the environmental Quality Standard for Soils of China (EQSS). The cortex from a copper tailings area in Tongling (marked as TLC) accumulated much higher amounts of heavy metals than the other sites. Calculated pollution index implied that the herbs in TLC were mildly contaminated by Pb, Cd, and Cu. Hazard quotient (HQ) indicated the intake of Pb, Cd, Cr, Cu, and Zn was safe by cortex consumption, but noncarcinogenic and carcinogenic risks of all metals in Cortex Moutan in hillside near the copper tailings (marked as TLB) and TLC were higher. The results suggested more attention should be paid to the planting area near the mining tailing, and in order to ensure the quality of the herbal medicines, the regions where this herb is grown should be at distance from the mining tailing area.
Determination of Metallic Element in Soybean Coat by Fermentation-Microwave Digestion-Microwave after Semi-Ashing Plasma Torch Atomic Emission Spectrometry

Abstract:Soybean coat samples were digested by H2O2-HNO3 solution after semi-ashing in a closed microwave system and the determination of iron,nickel,magnesium,calcium zinc and copper in soybean coat has been performed by MPT-AES.Experimenta1 conditions were optimized and established.The effects of acid concentration and coexisting ions(K,Na,Fe,Ni,Al,Zn,M g,Ca)on determination of iron,nickel,magnesium,calcium zinc and copper were investigated.The results showed that the detection limits of iron,nickel,magnesium,calcium,zinc and copper were 22.94,7.55,0.36,0.92,16.27 and 2.1 ng·mL-1,respectively.The RSD was no more than 3.2%,it showed the adopted method had good precision.The linear ranges of iron,nickel,magnesium,calcium,zinc and copper were 0-12,0-12,0-35,0-32,0-7 and 0-12 μg·mL-1,respectively.The recoveries were 96.8%-101.3%,98.4%-103.7%,95.9%-99.6%,96.3%-103.5%,99.8%-103.5% and 97.6%-105.3%,respectively.Compared with conventional test methods,MPT combined with half ash and microwave digestion samples was efficient with high accuracy and repeatability and applicable to sample analysis.

Abstract:An high performance liquid chromatography (HPLC) coupled with diode array detector (DAD) and electrospray ionization tandem mass spectrometry (ESI/MS(n)) method was developed for quality evaluation of Cortex Moutan through identification of common constituents based on chromatographic fingerprints and determination of key pharmacological compounds. The representative chromatographic fingerprints of Cortex Moutan were obtained by analyzing 10 batches of samples under the optimized HPLC conditions and the results showed that the chromatographic profiles of the analyzed samples were very similar. Total of nineteen common peaks were detected and seventeen of them were identified rapidly by their characteristic UV profile and the information of molecular structure provided by ESI/MS(n) experiments. Simultaneously, five key pharmacological compounds, namely gallic acid, oxypaeoniflorin, paeoniflorin, benzoylpaeoniflorin and paeonol, were determined by the validated HPLC-DAD method. The linear calibration curves were acquired with correlation coefficients higher than 0.999. The precisions of intra-day and inter-day were not exceeding 3.1%, and the recoveries of five analytes were from 92.86 to 99.35%. This developed method that combined the chromatographic fingerprints and quantification assay ensured the phytoequivalence and pharmacological effects of Cortex Moutan and was successfully applied to the quality control of Cortex Moutan.

Abstract:A high performance liquid chromatography tandem mass spectrometric (HPLC/MS/MS) method was developed to establish the fingerprint of Cortex Moutan, a Chinese medicinal herb. The major constituents among Cortex Moutan from different origins were separated well by HPLC and the fingerprint had a good repeatability. Fifteen peaks in the fingerprint were identified by LC/MS/MS according to their retention time, molecular weight and fragmentation information. The result showed that the presented fingerprint of Cortex Moutan contained plenty of information, which was quite valuable for the quality evaluation as well as chemical characterization of this Chinese herb.

Abstract:Daphne altaica Pall. is an important plant in traditional Chinese medicine. Mineral elements play an important role in the formation of active ingredients in medicinal plants. In this study, 14 mineral elements were quantitatively determined by inductively coupled plasma emission spectrometry using the roots, stems and leaves of D. altaica. The distribution variance of mineral elements between different organs were analyzed by statistical analysis methods such as single factor analysis of variance (ANOVA), correlation analysis (CA), principal component analysis (PCA) and cluster heat map. The results show that the mineral element contents in corresponding parts of D. altaica exhibited significant variation, with the order from highest to lowest being K > Ca > Mg > Fe > Al > Mn > Na > Zn > Sr > Cr > Ni > Cu > Pb > Mo. Among them, the highest content of mineral elements are K, Ca, Mg, Mn, Cu and Mo in leaves, and the stems contains more Zn, Sr, Cr and Ni, while the content of mineral elements Fe, Al, Na and Pb in roots is much higher than the other two organs. The ANOVA revealed significant variations in the mineral element content across different regions of D. altaica (p < 0.05). Based on single factor analysis of variance, different parts of D. altaica had different fingerprint of mineral elements, and the concentration of the same mineral elements in different parts had significant difference (p < 0.05). The principal component analysis (PCA) model showed that the cumulative variance contribution rate of first two principal components was 99.10%, in which Mn, Mg and Ca have higher load values, which is consistent with the results of bi-plot and cluster analysis. These results indicated that there were great differences in absorption and utilization of mineral nutrient elements between roots, stems and leaves of D. altaica, and there was significant correlation among mineral elements, indicating that there was a complex synergistic and antagonistic effect among the elements. At the same time, the discriminant analysis of the characteristic elements provided a scientific basis for the quality control of D. altaica.

Abstract:In this work, a simple, easy, and fast ultrasound-assisted extraction procedure for the determination of Cu, Fe, Mn, and Zn in plant samples was developed. Sample preparation conditions were optimized in a multivariate manner using mixture design and Box-Behnken. The extraction conditions adequate to obtain the maximum response were 90% power, 10 minutes, 40 °C temperature, and using an extraction solution composed of HNO3 and HCl at a concentration of 0.5 and 1.1 mol L-1, respectively, for a volume of 10 mL and 250 mg of samples. Accuracy was verified from the analysis of a certified reference material (Apple leaves, NIST 1515) yielding recoveries between 89 and 98% for the procedure. Precision was evaluated in a repeatability study (n = 10), yielding RSDs less than 6.7%, and the limits of quantification (mg kg-1) were 0.95, 2.42, 1.31, and 1.18 for Cu, Fe, Mn, and Zn, respectively. The concentrations determined in the stem and leaf of the mangrove species varied according to the collection point and the tissue, being consistent with other studies. The proposed procedure was efficient and reliable for elemental determination in plant samples, with the advantages of low cost and reagent consumption besides short analysis time as a valid alternative to the conventional sample preparation procedure.

Abstract:An effective method was developed for sequential rapid determination of trace elements in some seasonings by microwave digestion-high resolution continuum source graphite furnace atomic absorption spectrophotometry.The interferences were eliminated by adding the corresponding matrix modifiers.Results showed that the instrument detection limits of Cu,Cd,Pb,Mn and Al were 1.22 ng/mL,0.077 ng/mL,1.53 ng/mL,0.614 ng/mL and 1.54 ng/mL,respectively by this method.The recoveries were 95.2-106.4%.The precision relative standard deviations were less than 5.22%.Therefore,this method was rapid,accurate,stable and less pollution,and had a high practical value.It provided scientific basis for sequential rapid determination of many trace elements in food.
Preparation, Characterization, Antimicrobial and Cytotoxicity Studies of Copper/Zinc-Loaded Montmorillonite

Abstract:Background: A series of modified montmorillonites (Mt) including zinc-loaded Mt (Zn-Mt), copper-loaded Mt (Cu-Mt), copper/zinc-loaded Mt with different Cu/Zn ratio (Cu/Zn-Mt-1, Cu/Zn-Mt-2, Cu/Zn-Mt-3) were prepared by an ion-exchange reaction, and characterized using X-ray diffraction (XRD), fourier transformed infrared spectroscopy (FTIR) and transmission electron microscopy (TEM). The specific surface areas, antimicrobial activity and cytotoxicity of the modified Mt were investigated.Results: In the modified Mt, hydrated Cu ions and Zn ions were exchanged in the interlayer space of Mt and the particles were irregular shapes. The results showed that Cu/Zn-Mt enhanced antibacterial and antifungal activity compared with Zn-Mt and Cu-Mt possibly due to the synergistic effect between Cu and Zn. Among the Cu/Zn-Mt with different Cu/Zn raitos, Cu/Zn-Mt with a Cu/Zn ratio of 0.98 or 0.51 showed higher antimicrobial activity against gram-negative bacteria (Escherichia coli), gram-positive bacteria (Staphylococcus aureus), fungi (Candida albicans). Moreover, the antimicrobial activity of Cu/Zn-Mt was correlated with its specific surface area. Cytotoxicity studies on IPEC-J2 cell showed a slight cytotoxicity of Cu/Zn-Mt.Conclusions: The current data provide clear evidence that in terms of its antimicrobial activity and relatively low toxicity, the Cu/Zn-Mt holds great promise for applications in animal husbandry.

Abstract:The question of trace metal elements (TME) is still relevant and causes several environmental problems. Moreover, the digestion methods of TME have a significant impact on ecosystems. Sample preparation is an important step of any analytical procedure. In fact, defining the levels of TME in vegetal tissues requires various steps: drying samples, crushing, extraction, and dosage. The use of chemical extraction solutions can be put into question. Other than their lack of specificity, they are susceptible to provoking the redistribution and/or re-fixation of a part of the metallic ions between the various vegetal components. Our study aims to test the procedures of extraction that are specific for the metals Cu, Zn, Fe, Ca, Cr and Ba. Our results show an outstanding difference in the levels of TME derived from the same vegetal sample, according to the various techniques of extraction. In fact, cold extraction by nitric acid diluted at 10% is the most efficient way to put the ions of Cu, Zn, Fe and Ca into a solution. Meanwhile, the use of heated extraction using different acid digestion approaches is the most efficient for Cr and Ba solubilization from Atriplex portulacoides, Arthrocnemum indicum, Olea europaea BCR-62, Ulva lactuca and Ulva lactuca BCR-279.

Abstract:Dendrobium officinale is an important medicinal plant in traditional Chinese medicine. The consumption of D. officinale has increased rapidly in recent years due to the health awareness among Chinese people. The present study aimed to determine trace elements levels in 42 D. officinale samples and to utilize the elemental data for identifying the cultivation technique. Eighteen trace elements (B, V, Cr, As, Mn, Fe, Mo, Cu, Zn, Se, Sr, Sn, Ti, Al, Co, As Pb, Cd, and Bi) were determined by inductively coupled plasma–mass spectrometry. Of the 11 essential elements, the mean concentrations ranged from 30.6 µg kg−1 (Mo) in D. officinale with greenhouse cultivation to 161 mg kg−1 (Fe) in D. officinale with rock cultivation, and the contributions of Mn may exceed the dietary reference intake value by the uptake of 20 g of D. officinale. Of the seven toxic elements, the mean concentrations ranged from 3.29 µg kg−1 (Bi) in the D. officinale with greenhouse cultivation to 342 mg kg−1 (Al) in the D. officinale with rock cultivation, and the contributions of Al may exceed the provisional tolerable daily intake value by the uptake of 20 g of D. officinale. The concentrations of seven metals (Bi, Pb, Fe, Al, Cr, As, and Mo) were utilized to identify the cultivation technique of the analyzed D. officinale samples through linear discriminant analysis.
[Determination of trace elements in new food sources by flame atomic absorption spectrophotometry]

Abstract:Samples were digested by HNO3 + HClO4. Flame atomic absorption spectrophotometry (FAAS) was successfully used to determine copper, zinc and iron in new resource food. Under our experimental conditions, the recovery ratio was 94.66%-108.80%; the precision was 0.71%-4.78%. This method of measuring elements is convenient, rapid and accurate. The results showed that there are profitable elements, such as copper, zinc and iron in new resourse food in Henan province. By F test and SNK test, the content sequence of metal elements was found as follows: copper, Chrysanthemum morifolium Ramat = Silkworm pupa > flowers of Pueraria lobata Ohwin = Wheat germ = Codonopsis lanceolata = roots of Pueraria lobata Ohwi > Opuntia dillenii Haw. Zinc, Opuntia dillenii Haw > Silkworm pupa = flowers of Pueraria lobata Ohwi = roots of Pueraria lobata Ohwi = Wheat germ = Codonopsis lanceolata = Chrysanthemum morifolium Ramat. Determination of Trace Elements in Lophatherum Gracile Brongn from Different Habitat by Microwave Digestion-Atomic Absorption Spectroscopy

Abstract:A method of microwave digestion technique was proposed to determine the content of Zn, Fe, Cu, Mn, K, Ca, Mg, Ni, Cd, Pb, Cr, Co, Al, Se and As in Lophatherum gracile brongn of different habitat by atomic absorption spectroscopy. The RSD of the method was between 1.23% and 3.32%, and the recovery rates obtained by standard addition method were between 95.8% and 104.20%. The results of the study indicate that the proposed method has the advantages of simplicity, speediness and sensitivity. It is suitable for the determination of the contents of metal elements in Lophatherum gracile brongn. The experimental results also indicated that different areas' Lophantherum gracile brongn had different trace elements content. The content of trace elements K, Mg, Ca, Fe and Mn beneficial to the human body was rich. The content of the heavy metal trace element Pb in Lophantherum gracile brongn of Hunan province was slightly high. The content of the heavy metal trace element Cu in Lophantherum gracile brongn of Guangdong province and Anhui province is also slightly higher. Beside, the contents of harmful trace heavy metal elements Cd, Cu, Cr, Pb and As in Lophatherum gracile brongn of different habitat are all lower than the limits of Chinese Pharmacopoeia and Green Trade Standard for Importing and Exporting Medicinal Plant and Preparation and National Food Sanitation Standard. These determination results provided the scientific data for further discussing the relationship between the content of trace elements in Lophantherum gracile brongn and the medicine efficacy.

Abstract:Objective To study the chemical speciation of heavy metals and arsenate (As) in Phellodendri chinensis cortex ,Moutan cortex and Dictamni cortex .Methods Sample preparation was carried out by Tessier sequence extraction ;Pb ,Cd ,Cr ,Cu ,Hg and As were measured by atomic absorption spectroscopy .Results The concentration of Pb in Phellodendri chinensis cortex was the highest and the concentration of Hg was the lowest among the metals tested .Pb was mainly in the form of ion exchange and Hg was mainly in the form of residual .The concentration of Cu in Moutan cortex was the highest and the concentration of Hg was the lowest among the metals tested .Cu was mainly in the form of organic .The concentration of Pb in Dictamni cortex was the highest and the concentration of Hg was the lowest among the metals tested .Pb was mainly in the form of ion exchange .The con‐centration of As was higher than that of Hg ,but lower than that of other heavy metals in Phellodendri chinensis cortex ,Moutan cortex and Dictamni cortex .Conclusion The total amount of all harmful elements as the indicators of evaluation for medicine safe‐ty w as biased .
Determination of Micro-elements in Cephalanoplos Segetum Bge with FAAS

Abstract:[Objective] The content of the trace elements in Cephalanoplos segetum Bge was studied,which would provide the reference for its future development and guidance for the medical utilization.[Method] 6 kinds of trace elements such as Cu,Zn,Fe,Mg,Mn and Ca at the different parts of Cephalanoplos segetum Bge were detected with the method of flame atomic absorption spectrophotometry(FAAS).[Results] The results showed that the content of six micro-element was distinct variation in the leaf,stem and other different parts of Cephalanoplos segetum Bge.The content of Ca,Mg,Fe,and Mn was high and the content of Cu and Zn was low in the plant.[Conclusion] These conclusion could provide a useful data for the reasonable utilization and development of Cephalanoplos segetum Bge in which there was necessary and rich micro-element for humen body.
Influence of different processing methods on contents and speciation of heavy metals in Cortex Eucommiae

Abstract:Objective To investigate the influence of three different processing methods on contents and speciation of heavy metals Pb,Cd,Hg and Cu and a harmful nonmetallic element As in Cortex Eucommiae. Methods The five elements were measured by atomic absorption spectrometry. The determination wavelengthes of Pb and Cd was 283.3 and 228.8 nm using graphite furnace atom?ic absorption spectrometry;the determination wavelength of Cu was 324.8 nm using flame atomic absorption spectrometry;the determi?nation wavelengthes of As and Hg were 193.7 and 253.6 nm using flow injection-hydride generation method. Results Heavy metal′s contents in Cortex Eucommiae stir-fried with salt were increased. Contents of heavy metals in Cortex Eucommiae stir-fried and baked were decreased. Heavy metal morphology had no differences between processed products and raw materials. The experimental results showed that main existing form of Pb was in organic matter-bound,and mercury existed in exchange condition and organic matter-bound. Copper existed in organic matter-bound and carbonate bound and arsenic mainly existed with residual forms. Cadmium mainly exited in carbonate exchange condition. Conclusion There is a certain effect on the heavy metal content but less effect on the form of heavy metals by different processing methods.
