Abstract:2-Amino-3-methyl pyridine was prepared with 2-cyano-methylpyridine as raw material via hydrolysis in dilute basic solution using H_2O_2 as oxidant and Hofmann degradation with NaBrO.The feasible reaction conditions were as follows:hydrolysis,(50°C),5% NaOH,3.5 h,10% of H_2O_2;Hofmann degradation,(60°C),12% NaOH,0.5 h.The overall yield was 74%(mol).The product obtained was identified by GC-MS、IR、()~1H NMR and elemental analysis.

Abstract:3-Nitropyrazole was prepared via two steps, nitration and rearrangement, using pyrazole as a raw material. Pyrazole was nitrated in a mixed acid of nitric acid and sulfuric acid below 15 ℃for 3.5 hours with stirring, giving N-nitropyrazole with a yield of 86.6%, which was then refluxed in n-octanol at 185-190℃, giving 3-nitropyrazole with ayield of 87.8%. This nitration reaction was innovated by using sulfuric acid as the proton donor instead of acetic acid and acetic anhydride, and solvent n-octanol could be recycled without further treatment.

Abstract:The preparation of a series of 2-pyridinamines and their alkyl derivatives is described.The target compounds are synthesized starting from corresponding 2-cyanopyridines, via incomplete hydrolysis in the presence of hydrogen peroxide in the dilute alkaline mixture of acetone-2% sodium hydroxide solution, and Hoffmann degradation reaction with freshly made NaBrO.This route is of industrial value because of cheap and readily available materials, moderate reaction conditions and convenient operations.

Abstract:An improved and commercially valuable process is developed for the scalable synthesis of 2-chloro-3-amino-4-methylpyridine (CAPIC), a key intermediate of Nevirapine. The synthesis was accomplished in four steps, featuring condensation starting from 4,4-dimethoxyl-2-butanone and cyanoacetamide with ammonium acetate and acetic acid as catalysts. The total yield of the process is 62.1%. The pure CAPIC sample was confirmed with FTIR, 1H NMR, and 13C NMR spectra.

Abstract:2-Chloromethyl-3,5-dimethyl-4-nitropyridine hydrochloride, an important intermediate of omeprazole which is a very effective drug for the treatment of gastric ulcer, was prepared by N-oxidation with phosphotungstic acid as catalyst without solvent、nitration、rearrangement with acetic anhydride、hydrolysis in acidic condition and chlorination. The overall yield was over 70% and the molecular structure of the title compound was confirmed by ()~1H NMR.

Abstract:A new and efficient method for the synthesis of 4-methoxyl-2,3,5-trimethylpyiidine with an overall yield of 76% from cheap and easily available 2,3,5-trimethylpyridine was developed. The structure of related compounds was identified by IR, H-1 NMR and C-13 NMR. The method could be carried out economically on a huge scale.

Abstract:A general synthetic route of aminopyridines which are useful organic intermediates was investigated.The target compounds were synthesized starting from corresponding cyanopyridines,via incomplete hydrolysis in the presence of hydrogen peroxide using mixture of acetone-water and dilute base as solvent,and Hoffman degradation reaction with freshly made NaBrO.The feasible conditions of the above synthetic process were discussed taking the preparation of 4-aminopyridine as example.The optimum concentration of sodium hydroxide solution in the transformation of cyanopyridine to pyridinecarboxamide was 1.0 %.And the Hoffman degradation would be efficiently carried out at 75 ℃ in 12.5 % sodium hydroxide solution.

Abstract:Reaction of pyridine and substituted pyridines with N 2 O 5 in an organic solvent gives the N-nitropyridinium ion. When this is reacted with SO 2 /HSO 3 – in water, 3-nitropyridine is obtained (77 % yield). With substituted pyridines, the method gives good yields for 4-substituted and moderate yields for 3-substituted pyridines. The reaction mechanism is not an electrophilic aromatic substitution, but one in which the nitro group migrates from the 1-position to the 3-position by a [1,5 ] sigmatropic shift. From 4-aminopyridine, imi- dazo [4,5- c ] pyridines have been synthesized. From 3-nitropyridine, 5-nitropyridine-2-sulfonic acid is formed in a two-step reaction. From this, a series of 2-substituted-5-nitro-pyridines has been synthesized. 3-Nitropyridine and 4-substituted-3-nitropyridines have been substituted with ammonia and amines by the vicarious substitution method and by the oxidative substitution method in the position para to the nitro group. High regioselectivities and yields have been obtained in both cases to afford a series of 4-substituted-2-alkylamino-5-nitropyridines.

Abstract:5,6-Diaminobenzimidazolone-2 is obtained in an overall yield of 62.1% by the nitration of benzimidazolone-2 and followed with hydrogenation.Single-factor experiments were conducted to optimize the reaction condition.5,6-Dinitrobenzimidazolone-2 was synthesized by the nitration of benzimidazolone-2 under 0 ℃,for 6 h,n(nitric acid)/n(benzimidazolone-2)=2.2 in 69% yield.5,6-Diaminobenzimidazolone-2 was prepared in the absolute alcohol in 90% yield by 9 h of the Raney-Ni catalytic hydrogenation of 5,6-dinitrobenzimidazolone-2 under the condition of 100 ℃、2.5 MPa、m(catalyst)/m(5,6-dinitrobenzimidazolone-2)=0.09.MS、HPLC、IR、1HNMR、13CNMR and elementary analysis are properly used for the characterization of the structures of the product.

Abstract:A procedure for synthesis of 2-methoxyphenothiazine (1) has been developed, starting from resorcinol and aniline by condensation, following methylation and cyclization. p-Toluenesulfonic acid and polysubstituted aromatics were employed as the catalyst of condensation and the solvent of cyclization, respectively, to improve the yield. The use of parallel experiments, statistical experimental design, and multivariate modeling made the total yield of the procedure as high as 74.2%. J. Heterocyclic Chem., (2012).

Abstract:N,N'-Bis(4-nitrophenyl)urea was prepared by reacting p-nitroaniline with urea in the presence of DMF as solvent at refluxing temperature. The prepared product was analyzed by liquid chromatography. The purity of the product was above 98.00% and the yield was 80.30%. The optimal ratio of the reactants was p-nitroaniline: urea: concentrated hydrochloric acid=1.00∶1.00∶1.25, and the optimal reaction time was 5 h for 41.4 g(0.3 mol) p-nitroaniline under reflux temperature(adding the urea in batches within 1.0 h).

Synthesis of Trinitropyridine and Its N-Oxide

Abstract:2,2-Dinitropropane-1,3-diol was synthesized from nitromethane in low temperature by oxidative nitration method.Silver was successfully recycled with a yield upon 93% and reused in the synthesis of 2,2-dinitropropane-1,3-diol.Potassium 2,2-dinitroethanol,the key intermediatecompound for synthesis of trinitropyridine(TNPy),was prepared through one-pot method with a yield upon 62%,which is 10% higher than conventional method.Controlling the pH value in the range of 1-2,trinitropyridine N-oxide(TNPyO) was synthesized by the cyclization of potassium 2,2-dinitroethanol in 68% nitric acid at 50-60℃.Maintaining the temperature between 80-90 ℃,TNPyO can be further reduced to produce TNPy by refluxing for 1 h;The yields of TNPyO and TNPy are 50% and 15%,respectively.TNPyO can also be prepared from potassium 2,2dinitroethanol under phosphoric acid and sulfuric acid.The products were characterized by IR,nuclear magnetic resonance,elemental analysis and EI-MS.

Abstract:1-(2-Cyan)-phenyl-3-methyl-5-pyrazolone is synthesized from 2-aminobenzonitrile by diazotization,reduction with stannous chloride in the presence of hydrochloric acid to give 2-cyanphenylhydrazine hydrochloride,which is subjected to neutralization with sodium methoxide and nucleophilic reaction with methyl acetoacetate.The method has been improved in the process of synthesis of 2-cyanphenylhydrazine hydrochloride and 1-(2-cyan)-phenyl-3-methyl-5-pyrazolone and the total yield can be increased to 79.4%.

Abstract:A novel tandem reductive amination/intermolecular nucleophilic aromatic substitution (SNAr) sequence has been established for the synthesis of amine containing pyrimidine in formation of one carbon-oxygen and one carbon-nitrogen bonds in a one-pot fashion. Treatment of aldehyde with arylamine, 2-methanesulfonyl-4,6-dimethoxypyrimidine and sodium borohydride provides good overall yield. The p-toluenesulfonic acid (PTSA) can be used as activator and is generally needed in the reaction. Dioxane is the preferred reaction solvent, but reactions can also be carried out in tetrahydrofuran (THF), MeCN, toluene and dichloromethane. The procedure is carried out effectively in the presence of K2CO3. The reaction proceeds smoothly with aromatic aldehydes and arylamines possessing electron-donating or -withdrawing groups. This method can be applied to the synthesis of the oilseed rape herbicide and is superior to the classical one in several aspects: cutting out several purification steps, minimizing solvent use and chemical waste, and saving time. Its advantages such as operational convenience, high-efficient synthesis, and starting material availability make it a desirable method for preparing amines with molecular diversity and biological activity.

Abstract:By utilizing the dual reactivity function of N-aminopyridinium ylides, we developed a direct [3 + 2]-cycloaddition of N-aminopyridinium ylides and ynals to build the pyrazolo[1, 5-a]pyridine core while introducing a cyano group. In the transformation, N-aminopyridinium ylide serves as a 1,3-dipole and a nitrogen source, which results in the production of cyanated pyrazolo[1,5-a]pyridines and pyrazolo[1,5-a]pyrazines with a wide scope.

Abstract:An efficient and straightforward procedure toward the synthesis of amides-substituted 2H-benzopyrans H-benzopyrans has been explored through palladium-catalyzed one-pot aminocarbonylation reaction. By using nitro compounds as attractive nitrogen sources, with Mo(CO)6 6 as both CO source and reductant, a variety of amides-containing 2H- H- benzopyrans were obtained in moderate to high yields with good functional group tolerance. This protocol has advantages including atom-economy, no manipulation of toxic CO gas, and broad substrate scope.

Abstract:A facile, green, synthetic protocol of several substituted N-(pyridin-2-yl)imidates from nitrostyrenes and 2-aminopyridines via the corresponding N-(pyridin-2-yl)iminonitriles as intermediates is reported. The reaction process involved the in situ formation of the corresponding α-iminontriles under heterogeneous Lewis acid catalysis in the presence of Al2O3. Subsequently, α-iminonitriles were selectively transformed into the desired N-(pyridin-2-yl)imidates under ambient conditions and in the presence of Cs2CO3 in alcoholic media. Under these conditions, 1,2- and 1,3-propanediols also led to the corresponding mono-substituted imidates at room temperature. The present synthetic protocol was also developed on one mmol scale, providing access to this important scaffold. A preliminary synthetic application of the present N-(pyridin-2-yl)imidates was carried out for their facile conversion into the N-heterocycles 2-(4-chlorophenyl)-4,5-dihydro-1H-imidazole and 2-(4-chlorophenyl)-1,4,5,6-tetrahydropyrimidine in the presence of the corresponding ethylenediamine and 1,3-diaminopropane.

Abstract:The multicomponent reaction of 2-nitroacetophenone (or nitroacetone), acetaldehyde diethyl acetal, β-dicarbonyl compound, and ammonium acetate in an acetic acid solution allowed the acquisition of previously undescribed 4-methyl-substituted derivatives of 5-nitro-1,4-dihydropyridine in satisfactory yields. The oxidation of the obtained 5-nitro-1,4-dihydropyridine derivatives resulted in the corresponding 2,4-dimethyl-5-nitropyridines. In addition, for the first time in the synthesis of unsymmetrical 1,4-dihydropyridines by the Hantzsch reaction acetaldehyde, diethyl acetal was used as a source of acetaldehyde. The use of more volatile and sufficiently reactive acetaldehyde in this reaction did not lead to a controlled synthesis of unsymmetrical 5-nitro-1,4-dihydropyridines. The proposed multicomponent approach to the synthesis of 4-methyl-substituted 5-nitro-1,4-dihydropyridines and their subsequent aromatization into pyridines made it possible to obtain previously undescribed and hardly accessible substituted 5(3)-nitropyridines.
