Abstract:We compared the bacterial communities in the larval midgut of field and laboratory populations of a polyphagous pest, the cotton bollworm (Helicoverpa armigera), using denaturing gradient gel electrophoresis (DGGE) of amplified 16S rDNA sequences and 16S library sequence analysis. DGGE profiles and 16S rDNA library sequence analysis indicated similar patterns of midgut microbial community structure and diversity: specific bacterial types existed in both populations, and a more diverse microbial community was observed in caterpillars obtained from the field. The laboratory population harbored a rather simple gut microflora consisting mostly of phylotypes belonging to Enterococcus (84%). For the field population, phylotypes belonging to Enterococcus (28%) and Lactococcus (11%), as well as Flavobacterium (10%), Acinetobacter (19%), and Stenotrophomonas (10%) were dominant members. These results provided the first comprehensive description of the microbial diversity of the midgut of the important pest cotton bollworm and suggested that the environment and food supply might influence the diversity of the gut bacterial community.

Abstract:Helicoverpa armigera single nucleocapsid nucleopolyhedrovirus(HaSNPV)is ahighly infectious and selective pathogen of cotton bollworm.The researches of its molecular bi-ology and genetic engineering mainly cover some parts as follows.The complete nucleotide se-quence of HaSNPV G4 and C1 isolates were determined and compared with HzSNPV,AcMN-PV,SeMNPV,LdMNPV and XcGV genomes.The structure,transcription,expression andfunction of some genes in HaSNPV were studied.HaSNPV Bac to Bac baculovirus expressionsystem was constructed.The recombinant viruses of HaSNPV provide expansive promise forcontrolling Helicoverpa armigera in the field.With the deep researches of HaSNPV molecularbiology and genetic engineering,HaSNPV recombinant virus insecticides will play an importantrole in cotton bollworm integrated pest management.

Abstract:Studies on toxicity and safety evaluation of pesticides to silkworm(Bombyx mori) constitute the basic work for early-warning of pesticides' potential hazard to silkworm raising in sericultural area.This article reviewed the research progresses of this field in the past 20 years.Mainly,it introduced the acute and chronic toxicity of frequently used pesticides in mulberry field and nearby crop field to silkworm and the determination methods of pesticides' residue toxicity,compared the characteristics and applicable scopes of toxicity grading methods such as reference method,toxicity ratio method,and grade setting method,and put forward recommendations for improving performance in such aspects as the coverage of determination targets,the normalization of determination protocols,and the comprehensiveness of evaluation indexes in toxicity and safety evaluation of pesticides to silkworm.Furthermore,a number of suggestions were put forward for establishing a collaborative research network in the field,formulating testing procedures and comprehensive evaluation technological system,and implementing comprehensive hazard-reduce measures to prevent silkworm from being poisoned by pesticides.

Effect of Bt Protein on Development of Different Strains of the Cotton Bollworm, Helicoverpa Armigera (hbner) and the Parasitoid, Microplitis Mediator (Haliday)

Abstract:Two strains of the cotton bollworm, Helicoverpa armigera (Hubner), one is a susceptible laboratory strain and the other is collected from Bt cotton fields, were used as the insect hosts to study the effect of Bt insecticidal protein on the development of the parasitoid, Microplitis mediator (Haliday). The results showed that the larval duration of the parasitoid was delayed, and the pupal weight, body weight of the newly emerged adult and adult longevity decreased significantly when the hosts larvae fed on diet containing Bt protoxin Cry1Ac at the concentrations of 0.5-8.0 μg/g in all time or from 12 h before parasitism till pupation of the parasitoid . Compared with the control, the larval weight and pupal weight of H. armigera decreased significantly when the larvae fed on diet containing cry1Ac. Pupation rate of H. armigera decreased significantly in the treatment with diet containing Cry1Ac at 4.0 μg/g. When H. armigera larvae fed on the transgenic cotton leaves expressing both Cry1A and CpTI proteins, the mortality of the laboratory strain was 48.5%-95.8% in 2-5 days after treatment, which was significantly higher than that of the field strain (30.9%-86.1%). It is concluded that feeding on diet containing Bt insecticidal protein, both strains of the cotton bollworm will have significantly negative effects on development of the parasitoid, M.mediator.
Current Research Status and Prospects of Genomes of Insects Important to Agriculture in China

Abstract:Insects are the largest class of all living things with overwhelming diversity in the earth. Many of them are of agricultural importance: serious crop pests or beneficial insects. Modern entomological researches are more and more relying on information obtained from different insect genomes and transcriptomes since Drosophila melanogaster genome being sequenced in 2000. To better understand insect biology and transform our ability to manage insects that threaten our health, food supply, and economic security and to use beneficial insects that are essential to the maintenance and productivity of natural and agricultural ecosystems or provide us with silk, honey, medicine and other insect products, we need to know their genomic and transcriptomic information. Up to date, genomes of more than 50 insect species have been sequenced and analyzed around the world, and genomes of five insects of agricultural importance, including the domestic silkworm (Bombyx mori), the diamondback moth (Plutella xylostella), the oriental migratory locust (Locusta migratoria manilensis), the fig wasp (Ceratosolen solmsi) and the brown planthopper (Nilaparvata lugens), have been analyzed during the past decade in China, and genome sequencing for several agricultural insects including the whitefly (Bemisia tabaci), the oriental leafworm moth (Spodoptera litura), the rice stem borer (Chilo suppressalis), the white backed planthopper (Sogatella furcifera), the small brown planthopper (Laodelphax striatellu) and several parasite wasps, are in progress. Transcriptomes of the whiteflyand brown planthopper were reported in China in 2010, first two insect transciptomes reported in the world. Hundreds of insect transcriptomes have been reported in China since that year. Many important progresses in the functional genomics of the silkworm, the locust and the brown planthopper have been achieved, including the resequencing of 40 varieties of silkworm genomes which revealed domestication events and genes in silkworm, the uncovering of the precise mechanism of phase changes of the migratory locust, and the finding of that two insulin receptors determine alternative wing morphs in planthoppers. Data mining of insect genomes and transcriptomes, together with newly developed targeted genome editing and RNAi technologies, will lead to a revolutionary change in insect pest control and beneficial insect utilization.

Abstract:Cotton (Gossypium spp.) is an economically important crop that is grown throughout the world. It is the main material in textile and the main economic resource for more than 100 millions of cotton farmers in China. Due to its main role in our economy, cotton biotechnology has been significantly improved in the last decades in China, which led to a significantly progress both in cotton research and production. Cotton biotechnology started from 1970's in China has been improved greatly in various aspects such as tissue culture, protoplast culture and somatic hybridization, and recombination technology or gene engineering. In this paper, the cotton biotechnology status in China has been taken into account by keeping in view the work done by Chinese scientist, especially in cotton somatic culture, cotton protoplast culture and somatic hybridization, and transgenic cotton with insect resistance, diseases resistance and fiber quality improvement in China.
Commercial Production of Transgenic Bt Insect-Resistant Cotton Varieties and the Resistance Management for Bollworm (helicoverpa Armigera Hubner)

Abstract:There are currently three kinds of transgenic Bt insect-resistant cotton germplasm lines, Shanxi 94-24, Zhongxin 94 and R19, in China. They showed high resistance to the neonate larvae of bollworm (Helicoverpa armigera). Transgenic Bt insect-resistant cotton varieties or hybrids have been bred using the three kinds of germplasm lines as parents. Our researches reveal that there exist different expressions in resistant level at different developmental stages in the three categories of germplasm lines. When neonate larvae are fed with leaves of cotton plant at the seeding stage with less than 10 leaves on the main stem, the mortality of the neonate larvae is 100%, but the resistance level will decline at later season. When Bt gene has been transferred to the cotton genome, it can be steadily transferred to the progeny, the level of resistance to bollworm keeps fundamentally uniform. Abstract:▪ Abstract The development of cotton pest management practices in China has followed a pattern seen for many crops that rely heavily on insecticides. Helicoverpa armigera resistance to chemical pesticides resulted in the unprecedented pest densities of the early 1990s. Transgenic cotton that expresses a gene derived from the bacterium Bacillus thuringiensis (Bt) has been deployed for combating H. armigera since 1997. The pest management tactics associated with Bt cotton have resulted in a drastic reduction in insecticide use, which usually results in a significant increase in populations of beneficial insects and thus contributes to the improvement of the natural control of some pests. Risk assessment analyses show that the natural refuges derived from the mixed-planting system of cotton, corn, soybean, and peanut on small-scale, single-family-owned farms play an important function in delaying evolution of cotton bollworm resistance, and that no trend toward Bt cotton resistance has been apparent despite intensive planting of Bt cotton over the past several years.

Abstract:A 25‐year dataset on climate, cropping planting structure, and the population dynamics of cotton bollworms was analyzed to assess the effects of changes in climate and crop planting structure on the population dynamics of Helicoverpa armigera. The three generations of H. armigera showed increasing trends in population size with climate warming, and the relative abundances of the first and second generations decreased, but that of the third generation increased. The population numbers of H. armigera increased with the increase in the non‐Bacillus thuringiensis (Bt) cotton‐planted area, and to reduce the harm to crops caused by H. armigera, Bt cotton should be widely planted.The interactions between plants and insects play an important role in ecosystems. Climate change and cropping patterns can affect herbivorous pest insect dynamics. Understanding the reasons for population fluctuations can help improve integrated pest management strategies. Here, a 25‐year dataset on climate, cropping planting structure, and the population dynamics of cotton bollworms (Helicoverpa armigera) from Bachu County, south Xinjiang, China, was analyzed to assess the effects of changes in climate and crop planting structure on the population dynamics of H. armigera. The three generations of H. armigera showed increasing trends in population size with climate warming, especially in the third generation. The relative abundances of the first and second generations decreased, but that of the third generation increased. Rising temperature and precipitation produced different impacts on the development of different generations. The population numbers of H. armigera increased with the increase in the non‐Bacillus thuringiensis (Bt) cotton‐planted area. Asynchrony of abrupt changes existed among climate change, crop flowering dates, and the phenology of H. armigera moths. The asynchronous responses in crop flowering dates and phenology of H. armigera to climate warming would expand in the future. The primary factors affecting the first, second, and third generations of moths were Tmean in June, the last appearance date of the second generation of moths, and the duration of the third generation of moths, respectively. To reduce the harm to crops caused by H. armigera, Bt cotton should be widely planted.

Managing Practical Resistance of Lepidopteran Pests to Bt Cotton in China

Abstract:China is one of the major cotton producers globally with small farmers. Lepidopteran pests have always been the main factor affecting cotton production. To reduce the occurrence of and damage caused by lepidopteran pests, China has employed a pest control method focused on planting Bt (Cry1Ac) cotton since 1997. Chinese resistance management tactics for the main target pests, the cotton bollworm and pink bollworm, were also implemented. For polyphagous (multiple hosts) and migratory pests such as the cotton bollworm (Helicoverpa armigera), the "natural refuge" strategy, consisting of non-Bt crops such as corn, soybean, vegetables, peanuts, and other host crops, was adopted in the Yellow River Region (YRR) and Northwest Region (NR). For a single host and weak migration ability pest, such as the pink bollworm (Pectinophora gossypiella), the seed mix refuge strategy yields a random mixture within fields of 25% non-Bt cotton by sowing second-generation (F2) seeds. According to field monitoring results for more than 20 years in China, practical resistance (Bt cotton failure) of target pests was avoided, and there were no cases of Bt (Cry1Ac) failure of pest control in cotton production. This indicated that this Chinese resistance management strategy was very successful. The Chinese government has decided to commercialize Bt corn, which will inevitably reduce the role of natural refuges; therefore, this paper also discusses adjustments and future directions of cotton pest resistance management strategies.

Abstract:With the rapid development of economic globalization, invasive species have become a far-reaching global issue. Impacts of invasive insects on agriculture, forestry, ecology, socio-economy and human health are becoming increasingly serious and cause for concern. In this paper, we introduce advances in theoretical and applied research on 14 invasive insects in China and summarize novel theory, methods and recent progress. Perspectives and directions for future research on invasive insects are discussed. The results provide new insights for the early warning, prevention, detection, monitoring, management, and control, of invasive insects.

Effects of Bt-cotton cultivation on Helicoverpa armigera activity-density in agricultural landscapes in northwestern China

Abstract:Background: Cotton bollworm, Helicoverpa armigera (Hübner), threatens many crops. Bacillus thuringiensis (Bt) cotton has been planted to control this severe pest in northern Xinjiang, China. In 2021 and 2022, we monitored the activity densities of H. armigera males using sex pheromone traps in Bt cotton and non-Bt maize fields. We assessed how much of the population variation of cotton bollworm in the fields within the Bt cotton planting area could be explained by (i) landscape composition [including the proportion of Bt cotton among total area of host crops (cotton, maize, wheat and vegetables)], (ii) landscape configuration (patch density - i.e. the number of patches within a given area) across 0.5-2.0 km scales, or (iii) the population density of the previous pest generation. Results: Cotton bollworm activity-density exhibited two distinct peaks annually (mid- to late May and mid-July each year), with the number of males caught during the second peak significantly and positively correlated with the first peak's numbers. The suppressive effect of the proportion of Bt cotton in the landscape on bollworms was more pronounced at larger scales, and patch density had a significant positive effect on bollworm activity density. Conclusions: These findings support (i) the promotion of Bt cotton in northwestern China has reduced and suppressed the occurrence of cotton bollworms at the landscape scale and (ii) the importance of controlling spring populations for effective summer outbreak management, and (iii) that cotton bollworm control should be coordinated at a large scale across multiple crop fields. © 2024 Society of Chemical Industry.

Abstract:Cotton fibers, commonly known as cotton lint, are single-celled trichomes derived from epidermal layers of cotton ovules. Despite of its importance in word trade, the molecular mechanisms of cotton fiber production is still poorly understood. Through transcriptome profiling, functional genomics, proteomics, metabolomics approaches as well as marker-assisted molecular breeding, scientists in China have made significant contributions in cotton research. Here, we briefly summarize major progresses made in Chinese laboratories, and discuss future directions and perspectives relative to the development of this unique crop plant.

Abstract:The cotton bollworm Helicoverpa armigera is a cosmopolitan pest and its diverse habitats plausibly contribute to the formation of diverse lineages. Despite the significant threat it poses to economic crops worldwide, its evolutionary history and genetic basis of local adaptation are poorly understood. In this study, we de novo assembled a high-quality chromosome-level reference genome of H. a. armigera (contig N50 = 7.34 Mb), with 99.13% of the HaSCD2 assembly assigned to 31 chromosomes (Z-chromosome + 30 autosomes). We constructed an ultradense variation map across 14 cotton bollworm populations and identified a novel lineage in northwestern China. Historical inference showed that effective population size changes coincided with global temperature fluctuation. We identified nine differentiated genes in the three H. armigera lineages (H. a. armigera, H. a. conferta and the new northwestern Chinese lineage), of which per and clk genes are involved in circadian rhythm. Selective sweep analyses identified a series of Gene Ontology categories related to climate adaptation, feeding behaviour and insecticide tolerance. Our findings reveal fundamental knowledge of the local adaptation of different cotton bollworm lineages and will guide the formulation of cotton bollworm management measures at different scales.

Effects of transgenic Bt cotton on overwintering characteristics and survival of Helicoverpa armigera

Abstract:The effects of transgenic Bt cotton on the overwintering generation of the cotton bollworm, Helicoverpa armigera (Hübner) (Lepidoptera: Noctuidae), are unknown. We hypothesized that a Bt cotton diet may adversely affect fitness of this generation and examined fresh weight, lipids, glycogens, low-molecular-weight sugars and SCPs (supercooling points) of pupae, as well as survival of larvae, diapausing pupae and adult emergence in comparison with controls. Field and laboratory experiments showed that larvae fed on Bt cotton had a decreased pupation rate, and fewer entered diapause and emerged as adults compared with larvae fed non-Bt cotton. Furthermore, larvae fed Bt cotton had reduced pupal weight, glycogen content and trehalose levels both in diapausing and in non-diapausing pupae, and only diapausing pupae had an increased SCP compared to controls. The SCPs of diapausing pupae reared on Bt cotton were significantly higher than those reared on non-Bt cotton. The trehalose levels of diapausing pupae reared on Bt cotton were significantly lower than those of larvae reared on non-Bt cotton. Thus, these results suggest that a Bt cotton diet weakens the preparedness of cotton bollworm for overwintering and reduces survival of the overwintering generation, which will in turn reduce the density of the first generation in the following year. Effects of transgenic Bt cotton on the overwintering generation of cotton bollworm appear to have significantly contributed to the suppression of cotton bollworm observed throughout northern China in the past decade.
Determining the major Bt refuge crops for cotton bollworm in North China

Abstract:Evaluation of the effectiveness of refuge strategies involved in cotton bollworm Bt resistance management would be aided by technologies that allow monitoring and quantification of key factors that affect the process under field conditions. We hypothesized that characterization of stable carbon and nitrogen isotopes in adult bollworm, Helicoverpa armigera (Hübner) (Lepidoptera: Noctuidae) moths may aid in determining the larval host that they developed upon. We found moths reared from larvae fed on peanut, soybean or cotton, respectively, could be differentiated using isotopic analyses that also corresponded to their respective larval host origins. These techniques were then used to classify feral second-generation bollworm moths caught in Bt cotton (Gossypium hirsutum) fields into different populations based on their isotopic signatures. In 2006-2007 feral moths captured in Bt cotton fields predominantly correlated with the peanut (Arachis hypogea) having served as their larval host, indicating this is the most important refuge crop for Bt-susceptible bollworm individuals (providing 58%-64% individuals) during independent moth peaks for the second generation in North China. The remaining feral moths correlated with soybean (Glycine max) (0-10%); other C3 plant (20%-22%) and non-C3 plant (12%-14%) host types also provided some Bt-sensitive moths. Field observations showed that peanut constitutes the primary refuge crop contributing to sustaining Bt-susceptible moths dispersing into cotton in North China. These results suggest that peanut may be a more effective refuge to sustain Bt-susceptible bollworm individuals and reduce the risk of development of a Bt-resistant biotype.
