Abstract:Antibacterial peptide is secreted instantaneous from blood cell and fat cell while insect infected by the bacteria,with advantage of antibacterial at wide spectra and repress the resistant strains.Defensin is a kind of small,cystin -rich and inducible antimicrobial peptides found in a large range of living organisms that contributes to host defense.Data demonstrates that it has a broad spectrum of antibacterial activity and special function mechanism.So the researches of defensin have become striking hot spot in international academic circles.The total DNA was extracted from Drosophila and defensin gene was amplified by PCR reaction for no intron in it,in which adaptors were added.Then changed these adaptors into cohesive ends,which could correctly linkage with expression vector cut by CpoⅠand NotⅠenzyme.Linked and cloned defensin gene with pHBM905B vector which including signal peptide.The positive clones,of which the chromosomes were inserted with defensin gene,were identified by the phenotype and PCR.The positive ones were transformed into Pichia pastoris yeast strain GS115 and cultured them induced by methanol.The expression product was examined by SDS-PAGE and the results displayed that its size was consistent with anticipative one.Results of agarose diffusion experiment indicated that its expression product exhibited highly efficient antimicrobial activity.Furthermore,it had extraordinary strong stability against hot,which reveal its wide foreground in medical and daily usage.Further research can be processed based upon our study.
