Abstract:Objective To investigate the genetic polymorphism of 15 STR loci in Tujia nationality in Chongqing.Methods The Genotyping technique of fluorescent dye labeling multiplex STR-PCR,capillary electrophoresis and DNA sequencer GeneScan were adopted in genotyping a sample of 115 unrelated individuals collected from Tujia nationality in Chongqing. The allele frequency of 15 STR loci was calculated, and the heterzygosity(H),the power of discrimination(DP) ,the probability of matching(Pm), the power of exclusion(PE),the polymorphism information content(PIC)and the paternity index(PI) were acquired.Results Among the Tujia population,we observed 141 alleles and 386 genotypes in 15 STR loci with their frequencies of 0.004 3-0.534 8 and 0.008 7-0.313 0.The genetype distributions meet Hardy-Weinberge equilibrium through χ~2 testing(P0.05).Statistical result showed that the heterzygosity(H) was ranged from 0.634 8 to 0.887 0.The power of discrimination(DP) was 0.796 5 -0.964 2,combined DP 0.999,999,999,9.The probability of matching(Pm) was 0.035 8-0.203 5,combined Pm was 3.12×10~ -17 . The power of exclusion(PE) was 0.334 7 - 0.768 9 ,combined PE was 0.999,997,688,3. The polymorphism information content(PIC) was 0.576 4 - 0.858 3 ,combined PIC=0.999,999,999,6. The paternity index(PI) was 1.369 0- 4.423 1 .Conclusion The Chongqing Tujia nationality 15 STR genetic data are established.It provides a basis for Tujia nationality genetic database,forensic individual identification and paternity testing.
