Abstract:Objective:To describe the application of Microsoft Excel in aided computing of pharmaceutic data analysis.Methods:The theory and methods of the pharmaceutic data analysis in Excel were analyzed and built the mathematical model of the problem calculated with the Excel's commends and functions.Results:Excel may be used in the calculating of the matrix,evaluation of the liner and non- linear equations,value inserting,differential equation,integral equation,model fitting and mathematical optimization design.Conclu- sion:By the application of Excel,the theory and methods of the data analysis in pharmaceutical calculating may be expressed clearly, understood and used casily,so the related concept,method,process and result are visualized.

Abstract:Objective To perform the program on meta-analysis of u-value, t-value, F-value, χ~2-value and Pvalue in Microsoft Excel. Method The Excel computational procedure was edited using IF statement and other relevant functions, such as SUM, NORMSINV, TDIST, FDIST and CHIDIST, etc. With real medical study data, valuate the results computed by the Excel program. Result The consolidated outcomes were generated and presented correctly with inputting quantitative-datas of different documents into the Excel calculating program, which agree with the results from the textbook. Conclusion Meta-analysis of u-value, t-value, F-value, χ~2-value and P-value can be accomplished in Excel conveniently, rapidly and accurately.

Abstract:Cross-over design is often used in clinical trials to compare the efficacy of two or more treatmentc. Two one-side test and confidence interval method can be used to test the equivalence. For 2 × 2 cross-over design,both methods can be used,the same as comparison between two treatments of m × m cross-over. In complicated occasion , just confidence interval method can be used. Abstract:In recent years, as more generic drug products become available, it is a concern not only whether generic drug products that have been approved based on the regulation of average bioequivalence will have the same quality, safety and efficacy as that of the brand-name drug product, but also whether the approved generic drug products can be used interchangeably. In its recent draft guidance, the U.S. Food and Drug Administration (FDA) recommends that individual bioequivalence (IBE) be assessed using the method proposed by Hyslop, Hsuan, and Holder to address drug switchability. The FDA suggests that a 2x4 cross-over design be considered for assessment of IBE, while a 2x3 cross-over design may be used as an alternative design to reduce the length and cost of the study. Little or no information regarding the statistical procedures under 2x3 cross-over designs is discussed in the guidance. In this paper, a detailed statistical procedure for assessment of IBE under 2x3 cross-over designs is derived. The main purpose of this paper, however, is to derive an IBE test under an alternative 2x3 design and show that the resulting IBE test is better than that under a 2x3 cross-over design and is comparable to or even better than that under a 2x4 cross-over design. Our conclusions are supported by theoretical considerations and empirical results. Furthermore, a method of determining the sample sizes required for IBE tests to reach a given level of power is proposed.

Abstract:In its 2001 guidance, the U.S. Food and Drug Administration (FDA) recommends that population bioequivalence (PBE) and individual bioequivalence (IBE) be assessed to address respectively the prescribability and switchability be- tween a brand-name drug product and its new formulation or generic copy. For IBE, the FDA recommends a 2× 4 crossover design and a statistical test procedure proposed by Hyslop, Hsuan and Holder (2000). The same method is also recom- mended in FDA (2001) for assessment of PBE under the 2 × 4 crossover design. However, we note that, asymptotically, FDA's PBE test has a size smaller than the nominal level and thus has a low power to detect PBE. In addition, the 2001 FDA guidance does not provide any statistical procedure for PBE under commonly used 2 × 2o r 2× 3 crossover designs. In this paper, an asymptotically valid statistical test is derived for PBE under the 2 × 2, 2 × 3o r 2× 4 crossover design, using the method of moments and linearization. A method of determining the sample size required to achieve a desired power of the PBE test is also proposed. Simulation results are provided to examine the performance of the proposed PBE test and FDA's test. Finally, an example is presented for illustration.

Abstract:Objective: To solve the problems of data analysis in pharmaceutical experiment by commonly used Microsoft Excel.Methods: The application of statistical function in Excel function using linear regression as example to illustrate the detailed procedure of the import of source data and the return of results.The application of descriptive statistics,t test and analysis of variance in the data analysis tool of Microsoft Excel,combination with examples to discuss its use for data analysis in pharmaceutical practice.Results: The application of statistical function within Excel in the analysis of linear regression,the method is simple and the result is reliable.The tool of data analysis in Excel is suitable for descriptive statistics,t test and analysis of variance in the normal pharmaceutical data analysis.By comparison with other data analysis software,the application of Microsoft Excel possessed the merits of fast in operation,convenient in use,accurate in calculation and easy to master.Conclusion: The merit of friendly user interface in Excel and the limpid results of statistical analysis made medicine workers feel convenient during the application of Excel data analysis soft,and it deserves to be spread in the pharmaceutical practice.

Abstract:Case-crossover design is a variation of case-control design that it employs persons' history periods as controls. Case-crossover design can be viewed as the hybrid of case-control study and crossover design. Characteristic confounding that is constant within one person can be well controlled with this method. The relative risk and odds ratio, as well as their 95% confidence intervals (CIs), can be estimated using Cochran-Mantel-Haenszel method. R codes for the calculation are provided in the main text. Readers may adapt these codes to their own task. Conditional logistic regression model is another way to estimate odds ratio of the exposure. Furthermore, it allows for incorporation of other time-varying covariates that are not constant within subjects. The model fitting per se is not technically difficult because there is well developed statistical package. However, it is challenging to convert original dataset obtained from case report form to that suitable to be passed to clogit() function. R code for this task is provided and explained in the text.

Abstract:Objective To use Excel worksheet function to achieve statistical analysis of the fourfold table qualitative data,and to improve knowledge of the.majority of researchers on the statistical capabilities of Excel.Methods The result of actual research published in Chinese Journal of Endemiology was the data source,and statistical analysis results of fourfold table data from Excel and from SAS software were contrasted.Results The worksheet function of Excel could realize the analysis fourfold table data chi-square test,obtained an accurate P values,but not just P ＞ 0.05 or P ＜ 0.05.The results of chi-square test of Excel were the same as the results from SAS software,but Excel could not obtain probability value of fisher exact probability.Conclusion As a dependent statistical analysis software,Excel is an easy to learn,easy to use,efficient and a good helper.In the absence of professional statistical software,Excel can be used to achieve data statistical analysis. On the Confidence Intervals in Bioequivalence Studies.

Abstract:A bioequivalence study is a type of clinical trial designed to compare the biological equivalence of two different formulations of a drug. Such studies are typically conducted in controlled clinical settings with human subjects, who are randomly assigned to receive two formulations. The two formulations are then compared with respect to their pharmacokinetic profiles, which encompass the absorption, distribution, metabolism, and elimination of the drug. Under the guidance from Food and Drug Administration (FDA), for a size-$\alpha$ bioequivalence test, the standard approach is to construct a $100(1-2\alpha)\%$ confidence interval and verify if the confidence interval falls with the critical region. In this work, we clarify that $100(1-2\alpha)\%$ confidence interval approach for bioequivalence testing yields a size-$\alpha$ test only when the two one-sided tests in TOST are ``equal-tailed''. Furthermore, a $100(1-\alpha)\%$ confidence interval approach is also discussed in the bioequivalence study.
Power Analysis and Sample Size Determination for Crossover Trials with Application to Bioequivalence Assessment of Topical Ophthalmic Drugs Using Serial Sampling Pharmacokinetic Data

Abstract:Objective To develop methods for determining a suitable sample size for bioequivalence assessment of generic topical ophthalmic drugs using crossover design with serial sampling schemes.Methods The power functions of the Fieller-type confidence interval and the asymptotic confidence interval in crossover designs with serial-sampling data are here derived.Simulation studies were conducted to evaluate the derived power functions.Results Simulation studies show that two power functions can provide precise power estimates when normality assumptions are satisfied and yield conservative estimates of power in cases when data are log-normally distributed.The intra-correlation showed a positive correlation with the power of the bioequivalence test.When the expected ratio of the AUCs was less than or equal to 1, the power of the Fieller-type confidence interval was larger than the asymptotic confidence interval.If the expected ratio of the AUCs was larger than 1, the asymptotic confidence interval had greater power.Sample size can be calculated through numerical iteration with the derived power functions.Conclusion The Fieller-type power function and the asymptotic power function can be used to determine sample sizes of crossover trials for bioequivalence assessment of topical ophthalmic drugs.

Abstract:Sample size calculation plays an important role in bioequivalence trials. In practice, a bioequivalence study is usually conducted under a crossover design or a parallel design with raw data or log-transformed data. In this paper, we discuss the differences in sample size calculation between a crossover design and a parallel design with raw data or log-transformed data. Formulas for sample size calculation under a crossover design and a parallel design with raw data or log-transformed data are derived. A brief discussion for the relationship among these formulas is given.

Abstract:We propose a crossover design for simultaneous significance testing of two binary endpoints, in which the AB/BA crossover design is carried out for each endpoint. An asymptotic -level test is obtained by applying the intersection-union principle to the marginal Mainland-Gart tests. Power approximations and sample size calculation are derived and implemented in R programs. An adaptive design with sample size reestimation is also presented. We demonstrate the numerical accuracy of the proposed design through an extensive simulation study. Supplementary materials for this article are available online.

Abstract:In this work, two algorithms (overlapping method and the probability-based method) for design space calculation were compared by using the data collected from extraction process of Codonopsis Radix as an example. In the probability-based method, experimental error was simulated to calculate the probability of reaching the standard. The effects of several parameters on the calculated design space were studied, including simulation number, step length, and the acceptable probability threshold. For the extraction process of Codonopsis Radix, 10 000 times of simulation and 0.02 for the calculation step length can lead to a satisfactory design space. In general, the overlapping method is easy to understand, and can be realized by several kinds of commercial software without coding programs, but the reliability of the process evaluation indexes when operating in the design space is not indicated. Probability-based method is complex in calculation, but can provide the reliability to ensure that the process indexes can reach the standard within the acceptable probability threshold. In addition, there is no probability mutation in the edge of design space by probability-based method. Therefore, probability-based method is recommended for design space calculation.

Abstract:OBJECTIVE: To develop the drug incompatibility checker based on Microsoft Excel. METHODS: According to the checking table of common drug compatibility by Chongqing Health Center for Women and Children, the drug incompatibility checker was developed using the Excel-based functions and VBA programming language, and it was compared with drug incompatibility cheched by checking table of common drug compatibility. RESULTS & CONCLUSIONS: The result calculated by the checker is consistent with checking table of common drug compatibility. The interface of the spreadsheet program is simple and clear, and it is a quick way for pharmacists in dispensing department, nurses and physicians in clinical department to inquire drug incompatibility.

Abstract:In this paper, we consider statistical tests for inter-subject and total variabilities between treatments under crossover designs. Since estimators of variance components for inter-subject variability and total variability in crossover design are not independent, the usual F-test cannot be applied. Alternatively, we propose a test based on the concept of the extension of the modified large sample method to compare inter-subject variability and total variability between treatments under a 2 x 2 m replicated crossover design. An asymptotic power of the proposed test is derived. A sensitivity analysis is performed based on the asymptotic power to determine how the power changes with respect to various parameters such as inter-subject correlation and intra-class correlation. Also the two methods for sample size calculation for testing total variability under 2 x 4 crossover design are discussed. The method based on the Fisher-Cornish inversion shows better performance than the method based on the normal approximation. Several simulation studies were conducted to investigate the finite sample performance of the proposed test. Our simulation results show that the proposed test can control type I error satisfactorily.

The kit includes integrate analysis, user-defined function of AUC, AUMC, MRT t1/2 etc.; auto-chart; vivo and vitro relationship; bioequivalence analysis; weighted line regress, plasma concentration-time data fitting, etc. The BAPP2.0 was determied by the province committee, and integrate analysis function has achieved domestic advanced level.

Abstract:Taking the goodness of fit test (Chi test) as an example, this paper attempts to calculate the Bayesian factor BF10 of n-fold Bernoulli test by the Excel software (using JASP software as the evidence). The results showed that in the range of 0.15-0.55 (the rate of samples which are all "true"), the calculated results of Excel were more accurate, and the differences between the two (Excel and JASP) were not statistically significant (P>0.3).
