Abstract:Objective To identify halothane hepatotoxicity related genes.Methods Eight female hepatic hemangioma patients undergoing resection of hemangioma were randomly divided into 2 groups(n=4 each):eontrel group(C)and halothane group(H).Epidural block was performed at T8,9 in both groups.The catheter was advanced 3.5 cm in the epidural space cephMad.Correct catheter placement was confirmed by a test dose of 5 ml of 2%lidoeaine.0.25%(in group C)or 0.125%dicaine(in group H)10 ml was given epidurally at 5 min after test dose.Epidural block was maintained with intermittent epidural 0.25%(in group C)or 0.125% dicaine(in group H)5 ml.In control group anesthesia was induced with 70%N2O in O2.Tracheal intubation was facilitated with succinyl choline 2 mg/kg after topical anesthesia of pharynx and trachea.Anesthesia was maintained with 70%N2O in O2 and infusion of succinyl choline(20-40 μg·kg-1·min-1).In group H halothune 1.5 MAC was inhaled during induction and maintenance of anesthesia in addition to N2O and infusion of succinyl chohne.The mRNA was pooled from normal liver tissue and labeled with Cyanine 5 or Cyanine 3-conjugated dUTP respectively by reverge transcription.The fluorescently labeled cDNAs were mixed and hybridized simultaneously to cDNA microarray chip.After that the cDNA chips were scanned to determine the intensity of Cyanine 5 and Cyanine 3 labels and their ratio to identity the differentially expressed genes.Results The expression of 31 genes among the 8 192 genes in group H was different from that in group C and included mainly metabolism gene,signal transduction gene and stress-related genes.Among the 31 genes,the expression of 18 was lower and 13 was higher.The expression of glyceraidehyde-3-phosphate dehydrogenase mRNA and NADH dehydregenase mRNA which are associated with mitochondrial respiration was lower while the expression of heat shock protein 70 and heine oxygenase-1 was higher.Conclusion The halothane hepatotoxicity related genes include metabolism genes,signal transduction genes and stress-related genes.
