Antioxidative Enzymes Status in Upland Cotton Callus Culture under Osmotic Stresses

Abstract:Mannitol and sorbitol are usually used as carbon sources in tissue culture engineering. However, they may act as osmolytes if they are used in higher concentration. In our present in- vitro experiment, we studied physiological and biochemical responses in cotton callus under these osmolytes. They significantly decreased the fresh biomass and enhanced soluble protein and MDA contents with the increase in their concentration levels. Oxidative metabolism in terms of antioxidant enzymes (SOD, POD, APX and CAT) quantitatively improved over related controls. Relatively lipid peroxidation in terms of MDA contents was higher than antioxidative metabolism. POD activity was higher in comparison to SOD activity showing the greater ability of cotton callus to scavenge H2O2. Our results showed that both osmotic agents caused damage to the membranous structure as well as activated antioxidant activities in the experimental cotton callus culture. Therefore, it is reasonable to speculate that ROS might have been produced.
