Abstract:A direct catalytic asymmetric aldol-type reaction of 3-substituted-2-oxindoles with glyoxal derivatives and ethyl trifluoropyruvate, catalyzed by a chiral N,N'-dioxide-Sc(OTf)(3) (Tf = trifluoromethanesulfonyl) complex, has been developed that tolerates a wide range of substrates. The reaction proceeds in good yields and excellent enantioselectivities (up to 93% yield, 99:1 diastereomeric ratio (dr), and >99% enantiomeric excess (ee)) under mild conditions, to deliver 3-(alpha-hydroxy-beta-carbonyl) oxindoles with vicinal quaternary-tertiary or quaternary-quaternary stereocenters. Even with 1 mol % catalyst loading or on scaleup (10 mmol of starting material), maintenance of ee was observed, which showed the potential value of the catalyst system. In studies probing the reaction mechanism, a positive nonlinear effect was observed and Sc(III)-based enolate intermediates were detected by using ESIMS. On the basis of the experimental results and previous reports, a possible catalytic cycle was assumed.

Abstract:Herein, we report the first intramolecular C-N bond formation of phenylpropanamide derivatives via organocatalytic oxidative reactions, affording 3,3-disubstituted oxindole derivatives with up to 99% yield. The high efficiency of this reaction is exemplified by the transition metal-free mild conditions and the ability to perform the reaction on a gram scale. Meanwhile, the DFT calculation of the catalytic oxidative transformation pathway has also been studied.

Abstract:This paper reports the reactions of o-nitrobenzamides with ketones induced by titanium reagent (TICl4-SM system). The results show that the low-valent titanium promoted a novel reductive cyclization of o-nitrobenzamides and ketones to give 1, 2-dihydroquinazohn-4 3H)-ones. The products were characterized by elementary analysis, IR, H-1 NMR and X-ray single crystal diffraction. The crystal of compound 3b belongs to monoclinic system with space group P2(1)/n, a=1.191 7(2) nm, b=0.691 10) nm, c= 1.782 1(4) nm, beta=98.81degrees, V=1.455 04(5) nm(3), Z=4, D-c=1.220 g/cm(3), mu=0.077 mm(-1), F(000) =568, and final R=0.043 3 and wR=0.107 6 for reflections [I>2sigma(I)]. In summary, a series of 1,2-dihydroquinazolin-4 3H) -ones was synthesized via reductive cyclization of o-nitrobenzamides with ketones induced by the TiCl4-Sm system. Our method possesses the advantages of easily accessible starting materials, convenient manipulation and moderate to high yield.

Abstract:C2-formyl indoles are important building blocks for the organic synthesis of alkaloids and small molecules with broad biological activities. Generally, its preparation involves both direct formylation of indoles at the activated C2-position with as unstable carbonyl sources and oxidative cleavage of tedious prepared tetrahydro- β -carbolines (TH β Cs) in the harsh conditions. In continuation of our research on the oxidative functionalization of indoles, in this letter, we report that straightforward effectively oxidation of easily available 2-methylindoles bearing C3-heteroatom functional group proceeds regioselectively C2-formyl indoles in good and excellent yields employing an oxidative combination of NCS/DMSO. This methodology presents a reasonably broad substrate scope and excellent functional group tolerance, thus enabling the preparation of highly versatile building blocks susceptible to further functionalization. An additional feature of this approach includes high efficiency, ease of operation, facile purification, and short reaction time, making it a promising strategy for organic synthesis and drug discovery.

Abstract:Asymmetric synthesis of 3-(3-indolomethyl)oxindoles through the addition of indole-substituted enolized ketoesters to 3-bromo-3-substituted oxindoles has been achieved using a N,N'-dioxide/Ho(III) complex. A number of 3-(3-indolomethyl)oxindoles, which may possess biological activity, were obtained in good yields with high diastereo- and enantioselectivities (up to 97% yield, >19 : 1 dr, 98% ee). Furthermore, time-dependent reversal of diastereoselectivity enabled access to optically active diastereomers. The product followed by facile transformations gave a new route into trigolute analogs.

Abstract:Indoles as important N-heteroaromatic skeletons are widely used in the fields of pharmaceuticals, agrochemicals and biological sciences, but the N-trifluoromethyl indoles have been rarely explored. So far, the main accesses to N-trifluoromethyl indoles include oxidative desulfurization-fluorination of dithiocarbamates and Fisher indole synthesis via the reaction of N-CF3 hydrazine and ketones. We report herein the efficient straightforward strategies for the synthesis of N-trifluoromethyl indoles through desulfurization-fluorination/cyclization of 2-alkynylaryl isothiocyanates or 2-alkynylanilines under mild conditions. These cascade reactions possess good substrate scope and functional group compatibility, providing a variety of N-trifluoromethyl indoles in moderate to high yileds.

Abstract:Herein, we describe an efficient transition-metal-free regioselective C(3)alkylation of indoles for the synthesis of bis(indolyl)methanes and 3-styryl indoles. Nitrobenzene is employed as the oxidant to oxidize the alcohols in the presence of a strong base and the reaction avoids the use of transition metals such as Ru and Mn. The protocol provides a favorable route to access biologically active compounds such as arundine, vibrindole A, and turbomycin B.

Abstract:Photoredox-catalyzed aminoarylation and thioamination of unactivated alkenes have been developed, providing novel synthetic routes to access synthetically challenging quaternary carbon-centered benzoindolizidinones and trifluoromethylthiolated piperidines using readily available starting materials. Notably, these transformations were enabled by merging amidyl radical generation from N-alkyl benzamides with oxidant incorporation. Density functional theory calculations were performed to understand the reaction mechanism and to rationalize the regioselectivities. Moreover, the newly developed catalytic aminoarylation provided a convenient synthetic route for natural product tylophorine and its gem-dimethyl analogues with greatly improved drug-like properties such as enhanced solubility and stability. Abstract:The first catalytic enantioselective dearomatization/rearomatization of 2-nitroindoles triggered by the Michael addition of 3-monosubstituted oxindoles was established. A wide range of 3-indolyl-3′-alkyloxindoles (up to 99% yield, 97% ee) and 3-indolyl-3′-aryloxindoles (up to 95% yield, 99% ee) were obtained by using an organocatalyst. This method provides an unprecedented strategy to access structurally diverse 3,3′-disubstituted oxindoles bearing a sterically congested triaryl-containing all-carbon quaternary stereocenter. Abstract:Here we report a general rhodium-catalyzed asymmetric intermolecular dearomative cyclopropanation of indoles using trifluoromethyl N-triftosylhydrazones as carbene precursors. The reaction enables the rapid construction of diverse cyclopropane fused indolines bearing a trifluoromethylated quaternary stereocenter in high enantioselectivities (up to 99% ee). This mild method exhibits a broad substrate scope, is compatible with various functional groups, and can even be utilized for the late-stage diversification of complex bioactive molecules. DFT calculations suggest that the formation of a key zwitterionic intermediate is responsible for the chiral induction. Overall, this approach opens up new possibilities for asymmetric cyclopropanation of challenging aromatic heterocyclic compounds.
