Abstract:A novel and efficient racemization catalyst, Pd/layered double-hydroxide-dodecyl sulfate anion, was prepared and used in the dynamic kinetic resolution (DKR) of arylamines. The undesired enantiomer was completely racemized at 55°C, allowing the catalyst to be compatible with biocatalysts. DKR proceeded smoothly and showed a broad substrate scope, with good conversion and high product enantiomeric excesses (eep). The system could be reused more than 30 times without loss of conversion and eep value.

Abstract:An efficient palladium-catalyzed amination of aryl halides with amines, using rac-P-Phos as the ancillary ligand and Pd(OAc)(2) as the palladium source is developed. The ligand and all of the synthetic intermediates are stable to air and moisture, allowing the easy handling. The catalyst system performed well for a large number of different substrate combinations in 80-110 degrees C to furnish aromatic amines in high yields.

Abstract:A new and efficient dynamic kinetic resolution (DKR) process of secondary aromatic alcohols was developed with acid resins as racemization catalysts. Acid resin CD8604 was shown to have excellent racemization activity and good biocompatibility. When employing CD8604 and complex acyl donors as racemization catalyst and acyl donor, respectively, enantiomerically pure aromatic acetate was obtained with excellent yield and ee values through the DKR process. It is noteworthy that the system could be reused more than 10 times with little loss of yield and ee value.

Abstract:A composite hollow mesoporous MIL-101@Pd@ZIF-8-30min (HM-MIL@Pd@ZIF-30min) nanocatalyst with hollow core and stable shell was developed in this study. Hollow mesoporous MIL-101(HM-MIL) was hydro thermally synthesized and employed as the supports of Pd nanoparticles by adopting a facile HM-MIL-template strategy to make ZIF-8 crystals uniformly overgrew on HM-MIL-101@Pd microcrystal cores. The obtained core shell catalyst possessed hollow mesoporous structure, and the shell layer of ZIF-8 had additional advantages of abundant Lewis basic sites for heterogeneous racemization reaction. Compared with unmodified HM-MIL@Pd and solid MIL-101(Cr)@Pd@ZIF-8 (S-MIL@Pd@ZIF-8), HM-MIL@Pd@ZIF-8-30min had higher racemic chemical activity (conv. 99%) and selectivity (sel. R-amide 94%) in one-pot dynamic kinetic resolution (DKR) reaction system of rac-1-phenethylamine. The catalyst showed excellent stability, and the conversion and selectivity reached 95% and 93% respectively after ten catalytic cycles.

Abstract:Herein we report a highly enantioselective kinetic resolution of tertiary benzyl alcohols via palladium/chiral norbornene cooperative catalysis. With simple aryl iodides as the resolution reagent, a wide range of readily available racemic tertiary benzyl alcohols are applicable to this method. Both chiral tertiary benzyl alcohols and benzo[c]chromene products are obtained in good to excellent enantioselectivities (selectivity factor up to 544). The appealing synthetic utility of the obtained enantioenriched tertiary alcohols is demonstrated by the facile preparation of several valuable chiral heterocycles. Preliminary mechanism studies include DFT calculations to explain the origin of enantiodiscrimination and control experiments to uncover the formation of a transient axial chirality during the kinetic resolution step.

Abstract:Knoevenagel condensation of aromatic aldehydes with active methylene compounds such as malononitrile, ethylcyanoacetate, benzimidazol-2-acetonitrile and benzothiazole-2-acetonitrile proceeded very smoothly, catalyzed by nano-Fe3O4 encapsulated-silica particles supported primary amine. Both reaction time and yield are satisfying. The advantages of this catalyst are ease of preparation, non-toxicity, low cost, ease of handling and recyclability.

Abstract:P-stereogenic tert- and sec-phosphines have wide applications in asymmetric catalysis, materials, and pharmaceutical chemistry, however, their practical synthesis still constitutes a significant challenge. Herein, a successful kinetic resolution of rac-secondary phosphine oxides via the enantioselective P-benzylation process catalyzed by the palladium/Xiao-Phos was designed. Both tert- and sec-phosphine oxides were delivered in good yield and excellent enantiopurity (selectivity factor up to 226.1). The appealing synthetic utilities are further demonstrated by the facile preparation of several valuable P-chiral compounds, precursors of bidentate ligands, as well as transition metal complexes.

Abstract:A new, efficient dynamic kinetic resolution (DKR) of secondary aromatic alcohols using self-made solid super acid TiO2/SO42- as a racemization catalyst was developed. Low-cost and easily produced TiO2/SO42- showed excellent racemization activity. When coupled with the lipase, Novozym 435, good biocompatibility was observed, and optically pure aromatic acetate (>99%) was obtained with a high yield. It is noteworthy that the system could be reused more than 10 times with little loss of yield or reduction in the enantiomeric excess (ee) value of product.

Abstract:Axially chiral amides are usually found in many biologically active compounds and are useful ligands in asymmetric catalysis. Herein, by using the dynamic kinetic resolution approach, an asymmetric allylic alkylation reaction of racemic amide naphthols is disclosed, which leads to generation of the axially chiral naphthamides in good to excellent yields (up to 97%) and enantioselectivities (up to 96:4 e.r.). Density functional theory was used to gain a theoretical understanding of the enantioselectivities in this reaction.

Abstract:An efficient method for asymmetric synthesis of acyclic α-tertiary amine derivatives has been achieved through enantioselective aminations of α-branched ynones with azodicarboxylates enabled by chiral phosphoric acid catalysis. Moreover, kinetic resolution of racemic starting material was realized under these conditions, which gave access to valuable enantioenriched α-substituted ketones. A wide array of α-aryl and alkyl-substitutions, and the substituted alkynyl groups were well compatible with this method, producing both the amination products and the recovered ketones with good to high enantioselectivities.This article is protected by copyright. Abstract:Chiral amines are key building blocks for the development of numerous bioactive compounds. In this study, we developed a concurrent chemoenzymatic cascade approach using ω-transaminase for the isomeric configuration inversion of a racemic amine mixture. One isomer was transaminated using ω-transaminase, generating coproduct ketones and an additional chiral substance. Then, the mixture underwent selective reductive amination of a ketone using a specially designed compatible nickel-based nanocatalyst, which transformed coproduct ketone to racemic amines while leaving the opposite enantiomer unchanged. The combination of the two steps in one reaction system functions as an overall isomeric configuration inversion system. Moreover, the desired chiral amines with an additional chiral substance were formed. The procedure consumed NH3 and generated H2O as the sole byproduct.

Abstract:Enantiomerically pure (R)-1-aminotetralin was prepared by dynamic kinetic resolution using a system of novel racemization catalyst combined with Novozym 435. The autocatalytic amidation reaction in the process would reduce the eeP value of the reaction. The autocatalytic amidation reaction was suppressed by changing the structure of acyl donor. The influence of acyl donor with different acid moieties and different alcohol moieties was investigated. As acyl donor became complicated,the dynamic kinetic resolution result of 1-aminotetralin could also become better. While using 4-chlorophenyl valerate as acyl donor,dynamic kinetic resolution could reach the best result with eeP value>99%and conversion>99%.

Abstract:A highly efficient kinetic resolution of racemic amino alcohols has been achieved for the first time with a chiral tin catalyst. A chiral organotin compound with 3,4,5-trifluorophenyl groups at the 3,3'-positions of the binaphthyl framework enabled this transformation with excellent yield and high enantioselectivity. Abstract:Racemization catalyst 5 c and the enzyme Candida antarctica lipase B were combined in a one-pot dynamic kinetic resolution (DKR) of primary amines in which a wide range of amines were transformed to their corresponding amides in up to 95 % isolated yield and >99 % ee. The DKR protocol was applicable with either isopropyl acetate or dibenzyl carbonate as the acyl donor. In the latter case, release of the free amine from the carbamate products was carried out under very mild conditions. The racemization of (S)-1-phenylethylamine with several different Ru catalysts was also evaluated. Catalyst 5 c, of the Shvo type, was able to selectively racemize amines and was also compatible with the reaction conditions used for DKR. A racemization study of three different amines with varying electronic properties was also performed. Competitive racemization of a 1:1 mixture of the deuterated and non-deuterated amine was carried out with 5 c and a primary kinetic isotope effect was observed for all three amines, providing support that the rate-determining step is beta-hydride elimination. The chemoenzymatic DKR protocol was applied to the synthesis of norsertraline (16) by using a novel route starting from readily available 1,2,3,4-tetrahydro-1-naphthylamine (1 o).
