Abstract:BACKGROUNDHigh content of (1,3;1,4)--d-glucan in barley grains is regarded as an undesirable factor affecting malting potential, brewing yield and feed utilization. Production of thermostable bacterial (1,3;1,4)--glucanase in transgenic barley grain or supplementation of exogenous bacterial (1,3;1,4)--glucanase has been used to improve malt and feed quality. The aim of the present study was to investigate the effect of over-expression of an endogenous (1,3;1,4)--glucanase on -glucan content and grain composition in barley.RESULTSA construct containing full-length HvGlb2 cDNA encoding barley (1,3;1,4)--glucanase isoenzyme EII under the control of a promoter of barley D-Hordein gene Hor3-1 was introduced into barley cultivar Golden Promise via Agrobacterium-mediated transformation, and transgenic plants were regenerated after hygromycin selection. The T-2 generation of proHor3:HvGlb2 transgenic lines showed increased activity of (1,3;1,4)--glucanase in grains. Total -glucan content was reduced by more than 95.73% in transgenic grains compared with the wild-type control. Meanwhile, over-expression of (1,3;1,4)--glucanase led to an increase in 1000-grain weight, which might be due to elevated amounts of starch in the grain.CONCLUSIONManipulating the expression of (1,3;1,4)--glucanase EII can control the -glucan content in grain with no apparent harmful effects on grain quality of transgenic plants. Combined Proteomic and Metabolomic Analysis of the Molecular Mechanism Underlying the Response to Salt Stress during Seed Germination in Barley

Abstract:Salt stress is a major abiotic stress factor affecting crop production, and understanding of the response mechanisms of seed germination to salt stress can help to improve crop tolerance and yield. The differences in regulatory pathways during germination in different salt-tolerant barley seeds are not clear. Therefore, this study investigated the responses of different salt-tolerant barley seeds during germination to salt stress at the proteomic and metabolic levels. To do so, the proteomics and metabolomics of two barley seeds with different salt tolerances were comprehensively examined. Through comparative proteomic analysis, 778 differentially expressed proteins were identified, of which 335 were upregulated and 443 were downregulated. These proteins, were mainly involved in signal transduction, propanoate metabolism, phenylpropanoid biosynthesis, plant hormones and cell wall stress. In addition, a total of 187 salt-regulated metabolites were identified in this research, which were mainly related to ABC transporters, amino acid metabolism, carbohydrate metabolism and lipid metabolism; 72 were increased and 112 were decreased. Compared with salt-sensitive materials, salt-tolerant materials responded more positively to salt stress at the protein and metabolic levels. Taken together, these results suggest that salt-tolerant germplasm may enhance resilience by repairing intracellular structures, promoting lipid metabolism and increasing osmotic metabolites. These data not only provide new ideas for how seeds respond to salt stress but also provide new directions for studying the molecular mechanisms and the metabolic homeostasis of seeds in the early stages of germination under abiotic stresses.
Amino acid substitutions of the limit dextrinase gene in barley are associated with enzyme thermostability

Abstract:Limit dextrinase (LD) is a key enzyme in determining the malting quality. A survey of 60 barley varieties showed a wide range of variation for the enzyme activity and thermostability. Galleon showed low enzyme activity and high thermostability while Maud showed high activity and low thermostability. Alignment of the LD amino acid sequences of Galleon and Maud identified seven amino acid substitutions Lys/Arg-102, Thr/Ala-233, Ser/Gly-235, Gly/Ala-298, Cys/Arg-415, Ala/Ser-885 and Gly/Cys-888. Genetic diversity of LD was investigated using single strand conformation polymorphism based on the amino acid substitutions. Only limited genetic variation was detected in the current malting barley varieties, although wide variation was observed in the wider barley germplasm. The Thr/Ala-233 and Ala/Ser-885 substitutions were associated with enzyme thermostability ( P < 0.0001), but no polymorphism was associated with the enzyme activity. This result was confirmed from further sequence analysis. The results will provide a tool for understanding and selection of high LD thermostability.
Comparisons of Copy Number, Genomic Structure, and Conserved Motifs for Α-Amylase Genes from Barley, Rice, and Wheat.

Abstract:Barley is an important crop for the production of malt and beer. However, crops such as rice and wheat are rarely used for malting. α-amylase is the key enzyme that degrades starch during malting. In this study, we compared the genomic properties, gene copies, and conserved promoter motifs of α-amylase genes in barley, rice, and wheat. In all three crops, α-amylase consists of four subfamilies designated amy1, amy2, amy3, and amy4. In wheat and barley, members of amy1 and amy2 genes are localized on chromosomes 6 and 7, respectively. In rice, members of amy1 genes are found on chromosomes 1 and 2, and amy2 genes on chromosome 6. The barley genome has six amy1 members and three amy2 members. The wheat B genome contains four amy1 members and three amy2 members, while the rice genome has three amy1 members and one amy2 member. The B genome has mostly amy1 and amy2 members among the three wheat genomes. Amy1 promoters from all three crop genomes contain a GA-responsive complex consisting of a GA-responsive element (CAATAAA), pyrimidine box (CCTTTT) and TATCCAT/C box. This study has shown that amy1 and amy2 from both wheat and barley have similar genomic properties, including exon/intron structures and GA-responsive elements on promoters, but these differ in rice. Like barley, wheat should have sufficient amy activity to degrade starch completely during malting. Other factors, such as high protein with haze issues and the lack of husk causing Lauting difficulty, may limit the use of wheat for brewing.

Abstract:Amylopectin is synthesized by the coordinated actions of many (iso)enzymes, including ADP-glucose pyrophosphorylase (AGPase), starch synthases (SSs), branching enzymes (BEs), and debranching enzymes (DBEs). Here, two polymorphic forms of starch synthase I (SSI) and pullulanase (PUL) in rice-developing seeds, designated as SSI-1/SSI-2 and PUL-1/PUL-2, were discovered for the first time by zymographic analysis. The SSI and PUL polymorphisms were strongly associated with the SSI microsatellite marker (p = 3.6 × 10(-37)) and PUL insertion/deletion (InDel) markers (p < 3.6 × 10(-51)). Western blotting and mass spectrometric analysis confirmed that the polymorphic bands were truly the SSI and PUL enzymes. Only one non-synonymous variation in SSI DNA sequence (the SNP A/G) causing the change of the amino acid K438 to E438 was observed, which coincided well with the polymorphic forms of SSI. Nine non-synonymous variations were found between PUL-1 and PUL-2. Two non-synonymous variations of PUL (F316L and D770E) were identified by mass spectrometric analysis, but all of the variations did not change the structure of PUL. The co-immunoprecipitation results revealed the differences in protein-protein interaction patterns, i.e., strong or weaker signals of SSI-BEI and SSI-BEIIb, between the two forms of SSI. The results will enhance our understanding of SSI and PUL properties and provide helpful information to understand their functions in starch biosynthesis in rice endosperm.
Studies on the Esterase Isoenzymes of Polygonatum Plants

Abstract:The esterase isoenzyme zymograms from leaf,rhizome and root among three species Polygonatum plants i.e.,Polygonatum filipes mear.,P.cyrtonema Hua.and P.odoratum(Mill.)Druce.,were obtained by polyacrylamide gel slab electrophoresis and distingushed by K-K-J-G electrophoresis photodensimeter scan. The results showed that there were significant differences among the esterase isozyme patterns of leaf,rhizome,and root of three species Polygonatum.At the leaf, although total band numbers of the esterase isozyme zymogram were the same(10 bands),but the differences in their peak area and distribution were found.In zone C, zymogram of P.filipes Mear.had the most bands among three species Polygon- atum.In zone B,that of P.odoratum(Mill.)Druce.had the most.In zone A,that of P.cyrtonema Hua.had the most.At the root the situation was similar,but zone D was found.It indicated that the isoenzymes were different between the organs.The paper suggested that the zymogram differences of esterase isozyme among varietal plants were decided by their genotypic matteral property,and esterase isozyme patterns thus could be used as a biochemical marker to identify plant species.
The Effect of H2O2 and Abscisic Acid (ABA) Interaction on Β-Amylase Activity under Osmotic Stress During Grain Development in Barley

Abstract:The effects of exogenous abscisic acid (ABA) and polyethylene glycol (PEG 6000) treatments on grain H(2)O(2), ABA and beta-amylase activity were studied during grain development in the spike culture experiments with variety Triumph and its ABA-insensitive mutant TL43 as the plant materials. The results showed that during grain development the two genotypes were similar in the pattern of ABA concentration change, but differed greatly in the pattern of H(2)O(2) concentration and beta-amylase activity changes. The beta-amylase activity was positively correlated with H(2)O(2) concentration, negatively correlated with ABA concentration, and it is mainly closely associated with continued high levels of ABA with respect to H(2)O(2). Water stress (PEG treatment) induced beta-amylase was associated with H(2)O(2) concentration but not with ABA concentration. Exogenous application of H(2)O(2) and Ascorbic acid (AsA) increased beta-amylase activity in Triumph but reduced that of TL43. However, the endogenous H(2)O(2) concentration in grains was always consistent with beta-amylase activity. A novel model was hypothesized from the current results to illustrate the relationship between H(2)O(2), ABA and beta-amylase synthesis for the barley exposed to abiotic stresses.
Functionality of Barley Proteins Extracted and Fractionated by Alkaline and Alcohol Methods

Abstract:ABSTRACTThis research optimized the extraction of different protein fractions from barley grains and assessed the physicochemical properties of the fractions obtained. Pearling was first used to remove the grain's outer layers (mainly bran and germ) so that the barley cytoplasmic proteins (albumin and globulin) would be enriched in the pearling flour (PF), while endosperm proteins (hordein and glutelin) would be enriched in the pearled grain flour (PGF). Salt, alcohol, and alkaline solutions were then used to extract different barley protein fractions from PF and PGF. The effects of extraction solvent type, pH, temperature, and extraction time on protein content and extraction efficiency were studied. Aqueous ethanol (55%, v/v) efficiently extracted barley hordein from PGF at 60°C, whereas pH 11.5 alkaline solution was the most efficient for extracting both cytoplasmic and endosperm proteins from barley PF and PGF at 23°C. Subunit molecular weight, amino acid composition, and the functional properties of each isolated barley protein fraction were investigated. Barley glutelin demonstrated superior oil‐binding property and emulsifying stability, whereas barley hordein exhibited good foaming capacity.

Abstract:The starch debranching enzyme limit dextrinase(LD,EC 3.2.1.41) is one of the key enzymes in malting,which catalyses the hydrolysis of α-1,6-D-glucosidic bonds in amylopectin and limit dextrin.The methods for purification and activity measurement of the enzyme have been much improved since it was discovered in 1940's.During grain development,the activity of free LD increased initially and then decreased,while the bound LD appeared later,and the increase in its activity is closely associated with the decrease in the activity of free LD.The activity of LD was quite low in mature grains.However,the total activity increased gradually during germination.Recently,the gene encoding barley LD was isolated from a genomic phage library using a barley cDNA clone as probe and the two inhibitors which could form complex with LD in grains also were identified and characterized.It is indicated that the inhibitors have dual impacts on malting process and quality.In this paper the change in LD activity during grain development and germination and its response to environmental factors,genotypic difference and methodology of assaying the activity,and isolation of the genes encoding the enzyme were reviewed.

Abstract:The degradation of starch in barley grains is a primary step of beer production. The addition of an appropriate amount of gibberellin (GA) promotes the production of fermentable sugars, beneficial to the brewing industry. However, the response of proteomics in germinating barley to GA and abscisic acid (ABA) treatments is not thoroughly understood. In this study, isobaric tags for relative and absolute quantitation (iTRAQ) proteomics analysis was performed to illustrate the change of proteins in Tibetan wild barley XZ72 and XZ95 under GA and ABA treatments during germination. XZ72 had more proteins upregulated than XZ95 under GA treatment, while under ABA treatments, XZ95 had more proteins upregulated than XZ72. Concerning the proteins involved in energy metabolism under GA treatment, XZ72 had more proteins upregulated than XZ95. Among the 174 proteins related to starch metabolism, 31 proteins related to starch hydrolysis, such as α-amylase, α-glucosidase, and β-fructofuranosidase, showed higher relative abundance in control and GA treatments in XZ72 than in XZ95. Analysis of correlation between proteins and metabolites indicated that higher hydrolase activity is beneficial for the accumulation of fermentable sugars during germination. On the other hand, 26 starch-synthesis-related proteins were upregulated in XZ95 under ABA treatment. It may be suggested that GA-induced proteins act as accelerators of starch degradation, while ABA-induced proteins inhibit starch degradation. The current results showed that XZ72 is highly capable of allocating the starch-hydrolyzing enzymes, which play important roles in starch breakdown.
The Variation of Β-Amylase Activity and Protein Fractions in Barley Grains As Affected by Genotypes and Post-anthesis Temperatures

Abstract:The variation of beta-amylase activity and protein fractions in barley grains was evaluated using 148 barley genotypes grown in the field and two cultivars under in vitro culture with two temperature treatments during grain development. The results showed that there was significant genotypic variation in beta-amylase activity and protein fraction content. Regression analysis indicated that beta-amylase activity was positively correlated with total protein and the level of each of the protein fractions, with the correlation coefficient between beta-amylase activity and hordein content being the highest. Furthermore, higher post-anthesis temperatures (32/26 degrees C, day/night) significantly enhanced beta-amylase activity and protein fraction content, presumably as a result of reduced starch content. Albumin and glutelin were the least and most affected, respectively, in comparison with the plants under lower temperature (22/16 degrees C). Temperature post-anthesis also influenced the morphology of the starch A granule and the number of B granules, suggesting the altered starch structure may also be a reason for deteriorated malting quality under high temperatures.
Electrophoretic Properties, Functionality of Barley Protein Isolates

Abstract:Three different protein fractions were isolated from barley bran and flour. Electrophoretic properties, including molecular weight and fraction patterns of barley protein isolates (BPIs), were characterized and examined by Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). The functional properties of BPIs also were investigated, and then compared with soy, and zein protein. SDS-PAGE indicated that BPI-1 was albumin and globulin fractions together; BPI-2 was mainly composed by glutelin and a little hordein; BPI-3 was totally hordein fraction. Foaming capacity of BPI-3 was 18.5% higher than soy protein, about 7 times than zein protein. Foaming stability of BPI-3 was highest among these proteins. All BPIs had the equal emulsion capacity and stability as soy, which higher than zein protein. The water holding capacity (WHC) of BPI-2 was much higher than BPI-1, zein and soy, followed by BPI-3. BPI-2 and zein had the greatest oil holding capacity among these proteins.
Aleurones from a Barley with Low [alpha]-Amylase Activity Become Highly Responsive to Gibberellin When Detached from the Starchy Endosperm

Abstract:The physiological and molecular bases for contrasting [alpha]-amylase phenotypes were examined in germinating seeds of two barley (Hordeum vulgare L.) cultivars, Morex and Steptoe. Morex is a high-quality malting barley that develops high [alpha]-amylase activity soon after germination. Steptoe is a feed barley that develops only low [alpha]-amylase activity levels during this period. The expression of all high- and low-isoelectric point (pl) [alpha]-amylase isozymes is reduced in Steptoe. The amount of [alpha]-amylase mRNA per gram of seedling tissue is correspondingly lower in Steptoe. Southern blot analysis revealed that the cultivars have the same copy number and organization for most high- and low-pl genes. Steptoe seedlings or embryoless half-seeds produce little [alpha]-amylase in response to exogenous applications of gibberellic acid (GA3) compared with Morex. However, when isolated aleurones of both cultivars are treated with GA3, they produce similar amounts of high- and low-pl [alpha]-amylase RNAs. This suggests that a factor in the starchy endosperm is responsible for lowered [alpha]-amylase response in Steptoe. The factor is probably not abscisic acid (ABA), since the two cultivars have similar concentrations of ABA during germination.
Effects of PEG stress on seed germination and seedling physiology of Malus robusta

Abstract:We investigated the influences of drought stress on the seed germination and seedling physiology of Malus robusta,which was simulated with different concentrations of PEG solution.Results show that the relative germination rate,germination potential,germination index and vigor index of the seeds processed with PEG decrease with the increase of stress intensity.SOD activity of the seedlings increases with the increase of PEG concentration,while POD activity demonstrates the opposite tendency.MDA and proline content increase with the increase of stress intensity.This indicates that higher PEG concentration cause worse damage to the seedlings of Malus robusta.

Abstract:Extracts of germinated barley (Hordeum vulgare L.) seeds of 41 different genotypes were analyzed for their activities of alpha-amylase, beta-amylase, alpha-glucosidase, and debranching enzyme and for their abilities to hydrolyze boiled soluble starch, nonboiled soluble starch, and starch granules extracted from barley seeds with water. Linear correlation analysis, used to quantitate the interactions between the seven parameters, revealed that boiled soluble starch was not a good substrate for predicting activities of enzymes functioning in in vivo starch hydrolysis as the extracts' abilities to hydrolyze boiled soluble starch was not correlated with their abilities to hydrolyze native starch granules. Activities of alpha-amylase and alpha-glucosidase were positively and significantly correlated with the seed extracts' abilities to hydrolyze all three starches. beta-Amylase was only significantly correlated with hydrolysis of boiled soluble starch. No significant correlations existed between debranching enzyme activity and hydrolysis of any of the three starches. Interactions between the four enzymes as they functioned together to hydrolyze the three types of starch were evaluated by path coefficient analysis. alpha-Amylase contributed to hydrolyses of all three starches primarily by its direct effect (noninteractive component). This direct contribution increased as the substrate progressed from the completely artificial boiled soluble starch, to the most physiologically significant substrate, native starch granules. alpha-Glucosidase contributed to the hydrolysis of boiled soluble starch primarily by its direct effect (noninteractive) yet contributed to starch granule hydrolysis primarily via its interaction with alpha-amylase (indirect effect). The contribution of beta-amylase to hydrolysis of boiled soluble starch was direct and it did not contribute significantly to hydrolysis of native starch granules.

Abstract:Water deficient or drought stress is a major factor causing deterioration or instability of malt barley quality. In the studies on the influence of drought stress during grain filling on malt quality formation or metabolic changes, it is quite difficult to obtain the uniform plant individuals and water condition in pot or field experiments. In this study, we combined barley spike in vitro culture and PEG-6000 simulated drought to determine the genotypic difference in the changes of grain metabolites and the expression level of the genes encoding β-amylase and β-glucan using two Tibetan wild barley accessions and two cultivated genotypes differing in malt quality stability under drought stress. Under simulated drought, grain weight and β-glucan content were dramatically reduced and β-amylase activity was increased, and a lot of metabolites were markedly changed for all genotypes. On the whole, the changes were relatively smaller in the wild barley. Meanwhile, the expressions of Bmy1 related to β-amylase synthesis and GSL1, GSL4 and GSL7 related to β-glucan synthesis were up-regulated and down-regulated under drought stress, respectively, being consistent with the changes of β-amylase activity and β-glucan content in the four barley genotypes. The current results showed that PEG-6000 simulated drought and spike in intro culture may provide the basically similar information on grain development or metabolites as do in the field experiments, and it is suitable for use in studies on the influence of drought stress on quality traits during grain filling stage of barley or other cereal crops.

Abstract:In this paper,15% PEG was applied to soak the seeds of Trichosanthes kirilowii for 24 h in order to promote its germination.The results show that PEG could decrease osmotic pressure of water and change reactions of physiology and biochemistry in seeds effectively.For example,PEG could bring the high activity of amylase consecutively,and increase the content of dissoluble protein and the activity of POD (Peroxidase), reduce accumulation of free radical, but decrease the content of MDA (Malondialdehyde) in seeds. Based on the experiment it might be said that PEG pretreatment could improve metabolic activity and make cell membrane system less damage in the course of absorbing water in seed,too.All these indicate that PEG can increase seed vitality and germination capacity.
Variation of Beta -Amylase Activity in Barley as Affected by Cultivar and Environment and its Relation to Protein Content and Grain Weight

Abstract:The cultivar and environmental variation of beta -amylase activity was studied using two barley cultivars with contrasting growth properties. There was a significant difference in beta -amylase activity between the two cultivars used, 92-11 being significantly higher than Xiumai 3. A significant variation in beta -amylase activity was detected between grains at different positions within a spike. The two cultivars showed the same pattern, with top grains showing the highest and bottom ones the lowest activities. The relative difference within a spike varied between the cultivars, with 92-11 being larger than Xiumai 3. Both seeding rate and timing of N application dramatically affected the beta -amylase activity. With N application at the booting stage, beta -amylase activity increased, mainly due to the significantly increased beta-amylase activity in the topmost grains. The bottom grains showed a lower response to timing of N application. The variation in protein content and grain weight between cultivars and among the various treatments was also examined. The possible influence of these factors on beta -amylase activity are discussed.
Heterologous expression and enzymatic characteristics of sulfatase from Lactobacillus plantarum dy-1

Abstract:Barley, rich in bioactive components including dietary fiber, polyphenolic compounds and functional proteins, exhibits health benefits such as regulating glucose and lipid metabolism. Previous studies have found that the content and composition of free phenolic acids in barley may be significantly changed by fermentation with the laboratory patented strain Lactobacillus plantarum dy-1 ( L. p dy-1), but the mechanism of enzymatic release of phenolic acid remains to be elucidated. Based on this, this study aimed to identify the key enzyme in L. p dy-1 responsible for releasing the bound phenolic acid and to further analyze its enzymatic properties. The Carbohydrate-Active enZYmes database revealed that L. p dy-1 encodes 7 types of auxiliary enzymes, among which we have identified a membrane sulfatase. The enzyme gene LPMS05445 was heterologous to that expressed in E. coli , and a recombinant strain was induced to produce the target protein and purified. The molecular weight of the purified enzyme was about 59.9 kDa, with 578.21 U mg −1 enzyme activity. The optimal temperature and pH for LPMS05445 expression were 40 °C and 7.0, respectively. Furthermore, enzymatic hydrolysis by LPMS05445 can obviously change the surface microstructure of dietary fiber from barley bran and enhance the release of bound phenolic acid, thereby increasing the free phenolic acid content and improving its physiological function. In conclusion, sulfatase produced by Lactobacillus plantarum dy-1 plays a key role in releasing bound phenolic acids during the fermentation of barley.

Abstract:Seed quality (i.e., emergence energy, viability, physical purity, size, weight) is a critical factor that influences the yield of crops. Poor seed quality can lead to reduced germination rates, lower plant populations, and, ultimately, lower crop yields. On the other hand, seed priming is suggested to be an effective technique for improving seeds germination and plant population. In this study, we investigated the effect of seed priming with polyethylene glycol (PEG) on the germination, growth, and yield of two varieties of canola, super canola, and sandal canola. The treatment plan includes five concentrations of PEG (i.e., 5, 10, 15, 20%), distilled water priming, and control (no priming). All of the treatments were applied in 3 replications following a completely randomized design. Our results showed that seed priming with 5%PEG (T2) significantly improved radicle length (50 and 36%), plant height (43 and 34%), chlorophyll a (44 and 43%), chlorophyll b (120 and 208%), and total chlorophyll (83 and 111%) compared to control in super canola and sandal canola, respectively. In particular, seed priming with 5%PEG resulted in the highest increase in protein contents (25 and 1.40%), oleic acid (26 and 40%), and linolenic acid (6 and 6%) compared to control in super canola and sandal canola, respectively. It is concluded that seed priming with 5%PEG is an effective treatment to improve the performance of canola crops in terms of seedling growth, yield, chlorophyll, protein, and oil content. More investigations are recommended as future perspectives using other canola varieties to declare 5% PEG as an effective treatment for canola for improvement in growth, oil, protein, and chlorophyll contents.
