Studies on the Extraction Efficiency and Stability of Antioxidant Substances of Chinese Bayberry (Myrica rubra) Fruit

Abstract:In this study, response surface methodology (RSM) was used to determine the optimum extraction conditions for ethanol extraction of antioxidant substances from bayberry fruits, where trolox equivalent were used as indexes.The effects of ultrasonic time, temperature, solid-liquid ratio, and alcohol concentration on antioxidant substances extraction were comparatively studied. Then three main factors (ultrasonic time, solid-liquid ratio, alcohol concentration) were selected to conduct a 3-level Central-Composite design on the basis of single factor analysis. The statistical analysis indicated that, for the range which was studied, ethanol concentration was the most critical factor that affected antioxidant substances extraction in bayberry. Under the optimum ultrasonic extraction conditions [ultrasonic time 54.94 min, ratio of material to liquid 1 ∶ 39.38 (g/mL), and alcohol concentration 66.72% ], the yield of antioxidant substances reached460.66 μg/g trolox equivalent. The stability evaluation of antioxidant substances indicated that it should be avoided to contact metal material and light during storage, and was more stable under acidity conditions (pH<3). Fruit color L*value, b*values were negatively correlated with the antioxidant equivalent; a*value and antioxidant equivalent have no significant correlation; anthocyanins, polyphenols and antioxidant equivalent was significantly and positively correlated.
