Abstract:A novel method to determine the degree of deacetylation of chitosan is described. In this method, the coulometric titrant OH− is generated by the electrolysis of water. The OH− reacted with the residual hydrochloric acid in chitosan solution and the degree of deacetylation was obtained by Faraday's law. The optimized experimental parameters in this study were 1.0mol/L KCl as supporting electrolyte, 15.00mA as the intensity of constant current, composite glass electrode as indicating electrode couples, double platinum generated electrode-platinum wire auxiliary electrode as working electrode pairs, pH 3.80 as the titration end-point. The degree of deacetylation in the four samples, which varied from 70 to 95% were measured. The results were similar to those from 1H NMR and the standard deviations were lower than 0.5%. With merit of simplicity, convenience, quickness, high accuracy and precision, automatic detection of titration end-point and low-cost, the proposed method will be very useful in the industrial production.

Abstract:An economical and accurate determination of degree of acetylation (DA) for highly acetylated chitin has always been a challenge for researchers dealing with chitin and chitosan. A new protocol for the first derivative UV method using concentrated phosphoric acid as a solvent for highly acetylated chitin was developed in this study. The solvent was proposed based on thorough investigation of the effects of associated reactions including chain degradation, monomer dehydration, and oxazolinium ion formation. The reproducibility and performance of the new protocol was evaluated using commercial samples and the results showed the DA values of both chitin and chitosan could be determined accurately by a single analytical technique in less than 3h.

Abstract:Chitosan is a biopolymer that can be subjected to a variety of chemical modifications to generate new materials. The properties of modified chitosan are affected by its degree of deacetylation (DDA), which corresponds to the percentage of D-glucosamine monomers in its polymeric structure. Potentiometric titration is amongst the simplest, most readily available, and most cost-effective methods of determining the DDA. However, this method often suffers from a lack of precision, especially for modified chitosan resins. This is in large part because the equation used to calculate the DDA does not consider the molecular weight of the chemically modified monomeric units. In this paper, we introduce a new equation that is especially suited for modified chitosan bearing three different types of monomers. To test this equation, we prepared naphthalene–chitosan resins and subjected them to potentiometric titration. Our results show that our new equation, which is truer to the real structure of the polymeric chains, gives higher DDA values than those of the routinely used equations. These results show that the traditional equations underestimate the DDA of modified chitosan resins.

Abstract:This paper reports on a novel method for rapid determination of the degree of deacetylation (DD) of chitosan samples by headspace gas chromatography (HS-GC). In this method, overdose of an acid is added to protonate the free amino groups in chitosan, which further reacts with bicarbonate (as an alkali titrant) in a close vial. Based on measuring the HS-GC signal for CO2 vs. the volume of HCO3- added by a stepwise titration, two transition points can be obtained. The DD value is calculated by net volume of HCO3- consumed, i.e., by the R-NH3+ groups in chitosan. The results showed that the present method has a good measurement precision (RSD < 0.82%) and accuracy (relative differences < 6.46% when compared with the results measured by a reference method). Since the HS-GC can perform an automated sample reaction equilibration and measurement, it could be much more efficient than the existing methods in the batch sample analysis.

Abstract:Two kinds of chitosans, namely N-acetylated and N-deacetylated chitosan were prepared by the modified processes. They can dissolve in both acid and alkali solution. 13C NMR was used to study the basic solution of chitosan, and XRD, FT-IR and SEM were used to study the structure of N-acetylated and N-deacetylated chitosan. The result from X-ray diffraction showed that a transformation of crystal structure occurred during the N-acetylation or N-deacetylation process with the decrease of crystallinity and expansion of crystal lattices. FT-IR spectra revealed that the intermolecular and intramolecular hydrogen bonds were destroyed by both treatments and a looser structure was observed by the SEM. The lower crystallinity, the decreased intermolecular interactions, the more disordered and looser structure were easy for the permeation of LiOH/urea aqueous solution and coordinated with the breakage of intermolecular and intramolecular hydrogen bond by LiOH at low temperature, the prepared chitosans dissolved in LiOH/urea/H2O mixture.

Abstract:A new method to determine the degree of deacetylation (DD) of alpha-chitin and chitosan in the range of 17-94% DD using X-ray powder diffraction (XRD) is proposed. The results were calibrated using (1)H NMR spectroscopy for chitosan and FTIR for chitin, in comparison with the potentiometric titration method. The results showed a good linear correlation between the CrI020 from XRD and the calibrated DD value. This method is found to be simple, rapid and nondestructive to the sample.

Abstract:A large number of hydrogen bonds is the main reason for hindering the dissolution and reaction of chitin, and a mild and green deacetylation method to prepare chitosan for a wider range of applications is urgent. As a non-toxic and degradable green solvent, the deep eutectic solvent can effectively interfere with the hydrogen bond network of chitin, making chitin more susceptible to other solvents. Therefore, a NADES system consisting of betaine and glycerol was proposed for application in the deacetylation reaction of chitin to facilitate further attack of N-acetyl groups by low concentrations of NaOH. After optimizing the reaction conditions, chitosan with 83.77 % deacetylation was prepared, requiring only a concentration of 25 wt% NaOH. The analysis of the product chitosan showed that NADES could not only effectively improve the degree of deacetylation, but also reduce the degree of damage to the molecular weight by alkali. In addition, the potential mechanisms involved in the deacetylation process by NADES were explored. The nature of the reaction was verified by FT-IR, XRD and theoretical calculations as the process of opening intra/intermolecular hydrogen bonds of chitin by NADES. More importantly, experimental and in-depth theoretical studies provide a reference for the green preparation of chitosan.

Abstract:Chitosans with various degrees of deacetylation (D.D.), which were used as standard sample for FTIR determination, were prepared from completely deacetylated chitosan by homogeneous N-acetylation reaction. By combining four probable probe bands, i.e. 1655, 1560, 1380 and 1320 cm −1 , eight probable reference bands, i.e. 3430, 2920, 2880, 1425, 1155, 1070, 1030 and 895 cm −1 and two baseline methods, the most suitable ratios A probe band / A reference band from IR spectra to determine the degree of acetylation of chitosan were evaluated from 48 combinations to be A 1560 / A 2880 , A 1560 / A 2920 and A 1655 / A 3430 ( A 1560 / A 2880 is mostly recommended). The second baseline method, i.e. linking between adjacent two valleys, was better for measuring the absorbances of 1560 and 1655 cm ?1 bands. The determination range of the D.D. (1%–100%) covered almost the whole range. The standard curves with A 1560 / A 2880 and A 1655 / A 3430 were also suitable for the determination of degree of substitution of other N-acylated chitosan, such as N-propionyl chitosan, N-butyryl chitosan and N-hexanoyl chitosan.

Abstract:Poisonous solvents were widely used in the preparation of N-succinyl-chitosan,which were easy to cause environmental pollution and difficult to remove completely from the products.These disadvantages limited their uses in biomedical materials.In the present work,a novel method was proposed to successfully synthesize N-succinyl-chitosan,which was safe and environmentally friendly.A series of N-succinyl-chitosan with different degrees of substitution were prepared in absolute ethanol by controlling the reaction time.The different degrees of substitution,molecular weight and polydispersity index of the products were measured.The products were characterized by Fourier transform infrared(FTIR) spectroscopy,X-ray diffractometry(XRD) and thermogravimetry(TG).Furthermore,the water-solubility and biocompatibility were studied.When the reaction time is 15 h,the degree of substitution and molecular weight of the product are highest.Moreover,polydispersity index of the product is smallest among all the products(10 h,12 h,15 h,18 h,20 h).In addition,the results indicate N-succinyl-chitosan has been synthesized with decreased degree of crystallization and thermostability.Water-solubility of modified chitosan is improved due to the decrease of crystallization.N-succinyl-chitosan prepared in absolute ethanol has good biocompatibility,which has great potential in biomedical fields.

Abstract:A new chitosan derivative is prepared using chitosan. Ethyl cholorocarbonate was first introduced to the hydroxyl group of phthaloylchitosan through a nucleophilic reaction. Hydrazine was then added to recover the amino groups of chitosan, and promote cross-linking. The structure of this new chitosan derivative was characterized by Fourier transform infrared (FT-IR) and proton nuclear magnetic resonance (1H NMR) spectroscopy, and its physical properties were determined by X-ray diffraction (XRD), differential scanning calorimetry (DSC), and thermogravimetric analysis (TGA). The thermal and chemical stabilities of the new derivative were improved compared with those of native chitosan. Assay of Escherichia coli adhesion on a film based on this chitosan derivative showed good adsorption and biofilm formation.

Abstract:The protonation constants of chitosans with different molecular weight (Mw) and degree of deacetylation were demonstrated by the modified Henderson-Hasselbalch model and Högfeldt's three-parameter model. The protonation constants pKa of chitosan showed a slightly decreasing from 6.51 to 6.39 when the molecular weight changing from 1370 to 60kDa. The degree of deacetylation showed a greatly effect on pKa values, which were increased from 6.17 to 6.51 with the degree of deacetylation decreasing from 94.6 to 73.3%. The degree of deacetylation influenced the balance of hydrophobic interactions and hydrogen bondings on chitsoan. The fitting of the titration data to both two methods showed that Högfeldt's three-parameter model provided a better agreement. The results obtained in this paper allowed to a better understanding of some physicochemical mechanisms and biological properties of chitosan.

Abstract:To determine the number average molecular mass of chitosan-oligosaccharide by u-sing the acetylacetone’s method based on the color-producing reaction with amino terminal . Via the tests on the repetitiveness, accuracy and the stability, supposed this method was reliable. At the same time, the contrastive results of HPLC and the traditional K3Fe(CN)6 method indicate that the acetylacetone method was more suitable for the determination of chitosan-oligosaccharide molecular mass.

Abstract:We use chitosan as raw material, use acetic acid to dissolve it and then investigate the effect of phosphotungstic acid on chitosan degradation; respectively, we studied the impact on chitosan degradation under conventional method and ultrasonic method, meanwhile we control the amount of 90% phosphotungstic acid to look into its impact on chitosan degradation. The results show that the phosphotungstic acid play a increasing role on chitosan degradation, with the dosage increased, the degradation was increased; the degradation under ultrasonic is faster than conventional.

Abstract:The chitosan samples with various degree of deacetylation were prepared by the reaction of N-acetylation between chitosan and acetic anhydride.The samples were characterized by FTIR and ultraviolet spectrum.The mechanical and fat resistance properties of hand-sheet coated by chitosan with different D.D.value were measured.The results indicated that there is a line correlation between D.D.value and the amount of the added acetic anhydride,while the molar ratio of acetic anhydride and chitosan is below 0.6;the mechanical and fat resistance properties are improved as the weight of coating arises;the resistance index is improved by 50.3% when the weight of coating reaches 4.8 g/m2;at the same amount of coating weight,the resistance index decreases gradually as the D.D.value of chitosan decreases;while the weight of coating is above 2.32 g/m2,the increasing of fat resistance index slows down;the mechanical and fat resistance properties are greatly influenced by the microstructure of the film formed by chitosan.

Abstract:In a new approach to the preparation of solid chitosan acetate, the dependence of solubility of chitsoan acetate on the mole ratio of acetic acid to GlcN residues of chitosan was evaluated from turbidity. The structure of the product chitosan acetate was characterized by titration and FT-IR. It was demonstrated that the chitosan acetate with high solubility retained the structure and antibacterial activity of chitosan. The antibacterial activities against Escherichia coli (Gram negative) and Staphylococcus aureus (Gram positive) have been exhibited by a special kind of agar and further investigated by optical density method. It was found that chitosan acetate at a concentration of 0.1% (m/V) exhibited some antibacterial activity but some of the bacteria did still grow. With a concentration of 0.15% (m/V), there were nearly no bacteria grown. Electron micrographs revealed bacterial action patterns of chitosan acetate towards E. coli and S. aureus.

Abstract:This paper introduces degradation of chitosan and maesurement of molecular weight, and get the appropriate condition.The best conditions of degradation of chitosan by nitrite sodium: 10% NaNO2 is 0.6mL, creaction temperature is 35℃, reaction time is 35min and CH3COOH concentration is 3%; The best conditions of degradation of chitosan by hydrogen peroxide: H2O2 concentration: 6%, creaction temperature is 70℃, reaction time is 6h and CH3COOH concentration is 4%; The best conditins of degradation of chitosan by common α-amylase: creaction temperature is 40℃, reaction time is 90min, dosage of enzyme 1200U/g.

Abstract:Chitosan was prepared with stressing method by blending chitin and solid alkali in a single-screw extruder at given temperature and characterized by potentiometric titration, gel permeation chromatography (GPC), infrared spectrum (IR) and carbon-13 magnetic resonance sperctroscopy (13C NMR). Chitosan with a deacetylation degree (DD) of 76.1% was obtained at a mass ratio 0.2:1:1 for H2O/chitin/NaOH at 160°C for 12 min. Compared to conventional solution method(usually 1: 10 for chitin/NaOH), the alkali assumption greatly decreased. Molecular weight of chitosan obtained by solid-phase method (S3,M w 1.54×105) was lower than that obtained by suspension method (Y2,M w 3.34 ×105). During deacetylation, molecular weight decreased with high reaction temperature and long reaction time but remained same at different initial ratios of NaOH/chitin. It might be concluded that degradation of chitosan was caused by breakout of the main chain of the oxidized chitosan catalyzed by alkali during the deactylation. IR and13C NMR showed that structures of chitosans prepared by solid-phase method were not changed.

Abstract:Schiff base of chitosan was obtained by the reaction of chitosan and aromatic aldehyde, and then Schiff base of chitosan reacted with methyl iodide to get soluble quaternary ammonium salts of chitosan which were N-metoxybenzenemethyl quaternary ammonium salt of chitosan (NM- Chitosan) and N-hydroxybenzenemethyl quaternary ammonium salt of chitosan(NH-Chitosan). The obtained chitosan derivatives were characterized by Infrared Spectra Analysis(FT-IR), Nuclear Magnetic Resonance Spectroscopy(1H NMR) and elemental analysis. Quantitative analysis showed that there was small difference between degrees of quaternization(DQ) and degrees of substitution(DS), and the solubility of chitosan derivatives was demonstrated to be better in the organic solvents.
