Role of Killer Cell Immunoglobulin-Like Receptor in Blood Transfusion

Abstract:Objective To study the role of killer cell immunoglobulin-like receptor in blood transfusion.Methods KIR genotypes of NK cells were determined by PCR-SSP after isolated from peripheral blood of healthy subjects using immuno-magnetic beads.NK cells were then mixed with the whole blood with known HLA typing,and cultured at 37 ℃ for 0,6,12,24 h.Levels of IL-2,IL-4,PGE2,TNF-α,TNF-β,IFN-γ,GM-CSF,IgA,IgM and IgG were detected by ELISA.Results Levels of IL-2,IL-4,PGE2,NF-α,TNF-β,IFN-γ and GM-CSF increased,but those of IgA,IgM and IgG decreased in a time-dependent manner(P0.05).Along with the inconsistent degree of KIR and HLA(0%,20%,40%,60% and 80%),contents of TNF-α,TNF-β,IFN-γ and GM-CSF were upregulated(P0.05),but those of IL-2,IL-4,PGE2,IgA,IgM and IgG showed unchangeable.Conclusion The immunologic function of allogeneic NK cells can be induced by the mismatch of KIR and HLA-I,leading to the reversal of transfusion related immune inhibition.
