Effect of Lead on Intracellular Free Ca~(2+) Concentration in Dissociated Hippocampal Neurons of Mouse and Action of L-type Calcium Channels on It

Abstract:Objective We explored that effect of lead(Pb) on intracellular free Ca 2+ concentration in dissociated hippocampal neurons of mouse and action of L-type calcium channeles on it.Methods Digesting with low concentration of trypsin and gently triturating mode were used to dissociate hippocampal neurons.[Ca 2+] i was measured with fura-2 double wavelength fluoremetry.Results Whether there was Ca 2+ in extracllular Tyrod's fluid or not,Pb resulted in increase of intracellular free Ca 2+ concentration[from(203.4±10.86)nmol/L to (423.3±19.26) nmol/L in resting] in dissociated hippocampal neurons of mouse;Pb inhibits the increase of intracellular free Ca 2+ concentration in dissociated hippocampal neurons of mouse induced by KCl.Nimodipine enhanced this effect of Pb,but BayK8644 reduced partly the effect.Conclusion The effect that the increase of intracellular free Ca 2+ concentration in dissociated hippocampal neurons of mouse induced by lead was related to the release of intracellular calcium stores,and the inhibition of Pb on depolarization elcited by KCl was dependent on reduce of L-type calcium channels.
